Edible insects have received considerable attention as sustainable protein sources due to their high nutritional value, economic benefits, and eco-friendly production. However, concerns regarding food fraud and allergenicity associated with edible insects have emerged with their increased consumption. In particular, allergens from edible insects, which can trigger severe reactions such as anaphylaxis, emphasize the necessity for reliable and rapid detection methods.
The objective of this study is to develop on-site detection methods for edible insects using isothermal technologies. Two innovative approaches were established: The loop-mediated isothermal amplification (LAMP) assays for the rapid detection of six edible insect species, and the LAMP-CRISPR/Cas12a assay for enhancing specificity and accuracy. The LAMP assay was designed to quickly and efficiently confirm for the presence of target edible insect DNA in diverse sample types, providing results within 30 minutes. The LAMP-CRISPR/Cas12a integrated LAMP system further enhanced the specificity by the precise cleavage mechanism of Cas12a, thereby minimizing false positives. Both methods were evaluated across a range of insect-based food products, demonstrating their capacity to accurately identify target species and distinguish them from non-target species in field-application settings.
These findings suggest the development of efficient and reliable detection methods capable of addressing allergenic risks and mitigating food fraud concerns associated with edible insects. By ensuring food safety and authenticity, these methods contribute to the development of sustainable and trustworthy food systems for a growing global population.
