The etiological agent of Hemorrinagic fever with renal syndrome (HFRS) had not been succeeded to isolate for more than 6 decades since its first description in Russia in 1913. For the first time by Lee et al., the causative antigen was isolated from lung tissue of Apodemus agrarius in Korea in 1976 by immunofluorescent antibody technique and named it as Hantaan virus in 1980.
Lung section from Apodemus mice infected with Hantaan virus was utilized as antigen in serological diagnostic test of HFRS but lung tissues from the mice might have the problems of contamination with reovirus and other respiratory viruses.
A successful gorwth of the virus in an in vitro system, a human cell line designated A549 was first reported by French et al. in 1981.
We have propagated Hantaan virus in Vero E6 cells, and studied antigenicity, multiplication and antibody response in various animals after intramuscular inoculation of the virus cultured in Vero E6 cells.
The summary of this study were as follows:
1. Hantaan virus and Seoul virus were isolated in Vero E6 cells and well multiplied.
2. Hantaan virus isolated from lung tissues of infected Apodemus mice and patient's blood were not contaminated with reovirus, which was studied by immunofluorescent antibody technique and antibody response of S.D. rats.
3. After intramuscular inoculation of Hantaan virus cultured in A549 cells and Vero E6 cells into Apodemus mice and rats, multiplication of the virus was demonstrated in the lung tissues in partial numbers of Apodemus mice but not in rats and antiboy titers were revealed higher in rats than in Apodemus mice.
