    The Hox genes are known to be involved in the antero-posterior (A-P) axial pattern formation by expressing at the specific time and specific position along the A-P axis during early animal development. The murine Hoxa-7 is expressed in two overlapping domains in the ectoderm derivative neural tube and spinal ganglia, and mesoderm derivative prevertebrae at day 12 of gestation. Deletion of position specific cis-acting regulatory element(CR) of Hoxa-7 causes loss of region specific expression pattern. In order to find whether this element is conserved among different species, genomic DNAs were extracted from several Koreanized species, such as Apis mellifera, Protaetia brevitarsis, Hyphantria cunea, Panax ginseng and Korean, and Southern blot analysis was performed using the CR element of murine Hoxa-7 as a probe. All three insects and human DNA generated specific band(s) homologous to the element, while ginseng, a plant, did not. In order to isolate a human homolog of murine CR element, the human genomic DNA library had been constructed and screened. Out of 3×105 plaques which are about 1.5× coverage of human haploid genome, one clone (λLHM) had been isolated. It contained about 18 kb human DNA and the highly conserved region turned out to be located in a 3.9 kb SalI fragment. The sequence conservation of non-coding region between very diverged organisms such as insets and mammals, implies that the regulatory element as well as the molecular mechanism for Hox gene expression may be conserved throughout evolution.
