Objectives: Liriope platyphylla has been traditionally used in Korean medicine for relieving cough, reducing lung heat, and nourishing the yin. However, scientific data regarding the safety of its water extract reamins insufficient. This study was conducted to evaluate the genotoxicity and acute oral toxicity of the Liriope platyphylla water extract through a bacterial reverse mutation test and a single-dose oral toxicity study in rats.
Methods: This study evaluated the genotoxic and acute oral toxicity of the Liriope platyphylla water extract. A bacterial reverse mutation assay (Ames test) was conducted using Salmonella typhimurium strains TA98, TA100, TA1535, TA1537, as well as Escherichia coli WP2 uvrA, both with and without metabolic activation (S9 mix). Additionally, a single-dose oral toxicity study was performed in Sprague-Dawley rats according to OECD Test Guideline 423.
Results: No increase in revertant colonies or dose-dependent mutagenicity was observed in any strain, regardless of the presence of S9 mix. In the acute oral toxicity test, no mortality, clinical signs, or pathological changes were found at doses up to 2,000 mg/kg.
Conclusions: These results suggest that the Liriope platyphylla water extract is non-mutagenic and does not exhibit acute oral toxicity after single-dose administration, supporting its potential safety as a herbal resource for further development
