    Polyetheretherketone (PEEK)은 골 조직 공학에 있어 우수한 생체적합성과 물리적 특성을 가지고 있다. Polydopamine (D)은 화학적 고정화 방법에서 지지 물질 역할을 하며 고분자 주쇄에 하이드록시 그룹과 아미노 그룹을 가지고, 이로 인해 여러 가지 변형 반응이 가능한 물질이다. PEEK-D디스크는 dopamine의 자가 중합을 하였고, 그 후 collagen 또는 insulin을 PEEK-D디스크의 표면에 고정시켰다.
    본 연구에서는 PEEK-D-Col 디스크와 PEEK-D-In 디스크의 제조와 표면 특성에 대해 설명하였다. 이 디스크의 사용 가능성은 조골 세포 (MC3T3-E1)를 사용하여 세포의 접착, 독성, 증식 및 분화를 알아보았다.
    PEEK-D 디스크는 dopamine 자체 중합된 것과 collagen 또는 insulin 이 디스크에 고정화 된 것을 CBB염색과 접촉각 측정 분석, FT-ATR-IR, XPS분석에 의해 확인하였다. PEEK 디스크에서 조골 세포의 세포 독성은 live & dead 염색을 통해 평가하였다. 세포의 증식은 MTT와 BrdU 분석을 통해 측정하여 평가하였다. Alizarin Red S 염색법과 Von Kossa 분석법, ALP 염색법을 통해 얻은 결과에 따르면 조골 세포의 분화는 PEEK-D-In 에서 배양한 세포가 PEEK 와 PEEK-D, PEEK-D-Col 디스크에서의 분화보다 훨씬 우수하였다.

    Polyetheretherketone (PEEK) has good biocompatibility and physical properties for bone tissue engineering. Polydopamine (D) plays a supporting material role in chemical immobilization method. It has free hydroxyl groups and amino groups onto the polymer backbone and this makes it an excellent candidate to several modification reactions. PEEK-D discs were prepared using the dopamine self-polymerization method. After complete self-polymerization of the dopamine from the disc, collagen or insulin was then immobilized on the surface of PEEK-D discs.
    In this study, the preparation and surface characteristics of PEEK-D-Col and PEEK-D-In discs were described. The potential for use of this discs was assessed by using osteoblast cells (MC3T3-E1) in terms of adhesion, cytotoxicity, proliferation and differentiation. The results obtained in this study could be summarized as follows. PEEK-D discs were successfully prepared by the dopamine self- polymerization. Next, immobilizing of collagen or insulin on the discs was confirmed by CBB staining, contact angle analysis, FTIR and XPS analysis. The cytotoxicity of osteoblast cells on the PEEK discs evaluate by live & dead staining. The proliferation of osteoblast cells on the PEEK discs evaluate by measuring the MTT and BrdU assay. And the differentiation of osteoblast cells cultured on the PEEK-D-In was much better than that on the PEEK, PEEK-D and PEEK-D-col discs by using the Alizarin Red S staining, Von Kossa assay and ALP staining.
