A rapid visualization of nucleus in the oocytes from large domestic animals is described. The rapid staining method involves use of acid Tyrode`s solution, acetoethanol, basic fuchsin and acetoglycerol. The method gave high quality of chromatin preparation and was reproducible. Furthermore, the rapid staining method was fast and simple, taking only 40 min. It can be used for visualization of nuclear complex in the oocytes from mouse, pig and cow, possibly from other species where the cytoplasm of the oocyte is opaque, requiring laborious clearing procedure. With this newly developed technique, maturation and fertilization in vitro of follicular oocyte, and embryo culture in the pig and cow can be analysed within a short period of time. This would provide an opportunity to carry out subsequent experiments. For comparison, a DNA-specific dye, Hoechst 33258, was also used to study nuclear status. The results suggest that these two methods could be used complementarily depending on the conditions in the laboratory and one`s expertise.
