YRp7 (5.8 kb) carrying amr, Tcs, yeast gene TRO1 and pC194 (2.9kb) carrying Cmr gene were digested with Hind III. The ligated DNA was transformed into E. coli HB101. Plasmid DNAs from several transformants with Apr Cmr Tcs were isolated. The size of the plasmid corresponded to the sum of Hind III fragments of YRp7 and pC194. This recombinant plasmid was named pYE311. pYE311 transformed E. coli CSH30 (trpC, thi) into Trp- but did not transform B. subtilis ATTCC33608 (trpC, hisB) into Trp- . pYE311 also transformed E. coli HB101 into Amr Cmr and B. subtilis 168 into Cmr. pYE311 was stably maintained by Rec- recipient cells in an extra-chromosomal state. These properties make pYE311 potentially useful as a shuttle vector for E. coli and B. subtilis.
