풍진 바이러스에 감염된 Vero 세포주의 유전자 발현 양상의 변화의 검색 = (The) research on the gene expression profiles of the rubella virus infected Vero cells

국문 초록 (Abstract)

풍진 바이러스 감염을 확인하는데 이용 할 수 있는 cDNA 라이브러리를 만들기 위해 Vero 세포를 풍진 바이러스로 감염시킨 후 이들 세포로부터 발현에 변화를 보이는 유전자들을 검색하였다.
Vero 세포를 2주 이상 풍진 바이러스와 함께 배양하고, 감염된 세포를 RT-PCR 및 직접면역형광법으로 확인하였다. 풍진 바이러스에 감염된 Vero 세포에서 total RNA를 추출하였고 differential display reverse transcription을 수행하였다. 그 결과 상대적으로 높거나 낮은 발현을 나타내는 cDNA를 선택하여 클로닝하였다.
풍진 바이러스에 감염된 Vero 세포에서 발현이 증가된 28개의 유전자와 발현이 감소된 22개의 유전자를 클로닝하였다. 발현이 증가된 28 유전자 중에서 4 유전자가 풍진 바이러스에 감염된 Vero 세포에서 새롭게 발현됨을 나타냈다.

To make a cDNA ribray that can be used for detect rubella infection, Vero cells were infected with rubella virus and mRNAs of those cells were extracted and screened by DDRT-PCR.
Vero cells were cultured with rubella virus for more than 2 weeks and rubella infection was confirmed by RT-PCR and direct immunofluorescent technique.
From rubella infected Vero cells, total RNA was extracted and differential display reverse transcription was performed, and alternatively expressed cDNAs were selected and cloned
In the rubella infected Vero cells, 28 cDNAs that show increased gene expression and 22 cDNAs that show decreased gene expression were cloned. In the 28 up-regulated cDNAs, 4 newly expressed cDNA was cloned.
