Inflammatory Responses Induced by S100A8 and S100A9 in Human Keratinocytes

Atopic dermatitis (AD) is an inflammatory skin disorder caused by genetic factors and immunological deficiency. Dryness, infectious agents such as bacteria, germs, and fungi, as well as environmental and psychological factors can contribute to AD, but the most direct causative factor has yet to be identified. In this study, I examined whether IL-6, IL-8, and MCP-1 protein expressions are increased by S100A8 and S100A9 treatment in HaCaT cells in time- and dose-dependent manners. Alterations of cytokine expression were examined by performing ELISA with various signal protein-specific inhibitors. Activation of the MAPK pathway and NF-κB was evaluated by using western blotting and NF- κB activity test. The expressions of IL-6, IL-8, and MCP-1 increased after treatment with S100A8 and S100A9, and the increase was significantly blocked by specific signal inhibitors such as TLR4i, rottlerin, PD98059, SB203580, and BAY-11-7085. Because these inhibitory effects indicate the association of ERK and p38 MAPK with S100A8 and S100A9 mechanisms, the activation of ERK and p38 MAPK was evaluated; both were activated in a time-dependent manner after treatment with S100A8 and S100A9. Phosphorylations of ERK and p38 MAPK were blocked by TLR4i and rottlerin. S100A8 and S100A9 induced translocation of NF- κB in a time-dependent manner, and the activation of NF- κB was inhibited by TLR4i, rottlerin, PD98059, SB203580, and BAY-11-7085. In addition, S100A8 and S100A9 decreased the expression of the skin barrier proteins filaggrin and loricrin. These results contribute to elucidating the pathogenic mechanism for AD and to developing strategies for controlling clinical features of AD. 아토피 피부염은 가장 흔한 만성 염증성 피부질환으로 유전적인 요소와 면역체계 이상으로 인한 염증성 피부질환이다. 그 외에 피부가 건조하거나 정상인에 비해 쉽게 피부 가려움증을 느끼는 증상이 있으며, 환경적 요인으로 세균·바이러스·곰팡이 등에 의한 감염과 정서적 요인이 복합적으로 작용하여 발병하는 것으로 알려져 있지만, 명확한 원인은 아직 밝혀지지 않았다. 본 연구에서는 사람 정상 각질세포주인HaCaT 세포에서 S100A8 및 S100A9 처리에 의해 IL-6, IL-8 및 MCP-1 이 시간 및 용량 의존적으로 증가하는지 여부를 조사하였다. HaCaT 세포주에서 S100A8 및 S100A9 처리한 후에 다양한 신호전달 억제제를 사용하여 ELISA를 이용한 사이토카인의 변화를 조사 하였다. Western blotting과 NF-κB 활성도 실험을 통해서 MAPK 경로와 NF-κB 활성화를 보았다. HaCaT 세포주에서 S100A8 및 S100A9 처리 후 IL-6, IL-8 및 MCP-1은 증가하였고, 증가한 IL-6, IL-8 및 MCP-1은 TLR4i, rottlerin, PD98059, SB203580 및 BAY-11-7085와 같은 특이적 신호전달 억제제에 의해 현저히 줄어들었다. 이러한 억제 효과가 ERK 및 p38 MAPK와 S100A8 및 S100A9의 연관성을 나타내기 때문에 ERK 및 p38 MAPK의 활성화를 알 수 있다. HaCaT 세포주에서 S100A8 및 S100A9두 가지 모두 처리한 후에 시간의존적으로 활성화되었다. 인산화된ERK와 p38 MAPK는 TLR4i와 rottlerin에 의해 억제되었다. HaCaT 세포주에서 S100A8 및 S100A9는 시간에 따라 NF-κB 의 활성화를 유도하였고 NF-κB 의 활성화는 TLR4i, PP2, rottlerin, PD98059 및 SB203580에 의해 억제되었다. 또한, HaCaT 세포에서 S100A8과 S100A9는 피부 장벽 단백질인 filaggrin과 loricrin의 발현을 감소시켰다. 이러한 결과는 AD의 병인기전을 밝히고 AD에 임상적으로 제어할 수 있게 기여한다.
급성 피부장벽 손상 후 회복에 대한 Protein kinase C의 영향
The skin is a physical, chemical, mechanical and microbial barrier; preventing water and electrolytes loss and protecting human body from various outside assaults. The barrier function of the skin is mainly carried out by the outermost layer of the epidermis, stratum corneum (SC); The intercellular lipid located between the corneocytes in SC plays a significant role in maintaining the homeostasis of permeability barrier function. Intercellular lipid contents are provided by the lamellar bodies that are originated form the granulocytes in epidermis through the exocytosis to the stratum granulosum and SC junction.There is a calcium gradient in the epidermis and these calcium ions act as an important signaling factor for epidermal permeability barrier function homeostasis. Protein kinase C(PKC) which comprises a family of serinethreonine kinases plays a central role in the proliferation and differentiation of keratinocytes. In the process of differentiation of the keratinocytes, calcium-induced PKC activation plays a central role in the expression of the keratinocyte differentiation-specific marker proteins, such as involucrin, loricrin, and filaggrin. Recently, it has been reported that, in cultured human keratinocytes, the PKC activation is associated with the enzyme activation of ceramide which is one of the intercellular lipid contents. In this study, we investigated the effect of PKC in the recovery of epidermal permeability barrier function using hairless mouse animal experiment model. We examined the expression and localization of various PKC isotypes in hairless mouse skin after acute skin barrier disruption using tape-stripping treatment. We also investigated the effects of PKC inhibitors on the recovery of barrier function by measuring trans-epidermal water loss (TEWL).As a result, we observed the expression of PKC-α, -β, -δ, -η proteins in a hairless mouse skin. The rate of recovery in acutely disrupted skin barrier treated with PKC inhibitor is shown to be decreased. The secretion of lamellar bodies were inhibited and there was a significant change in intercellular lipid structure. Also, PKC inhibitors suppressed the loss of normal epidermal calcium gradient after acute skin disruption and increased the intracellular calcium ion in cultured keratinocytes. It was inhibited by calcium channel blocker. In consequence, PKC inhibitors increase the calcium ion density in keratinocyte thus suppress the barrier recovery process after acute skin barrier disruption. Therefore PKC is thought to be related with the recovery process of acute skin disruption. 피부는 생물체와 외부 환경 사이에 존재하는 물리적, 화학적, 기계적, 미생물 방어 장벽으로서 외부로의 과도한 수분과 전해질의 손실을 억제하고, 다양한 외부의 공격으로부터 인체를 보호하는 장벽 기능을 수행한다. 이러한 장벽 기능은 주로 피부 표피층 최외각의 각질층에 의하여 수행되며, 특히 각질세포 사이에 존재하는 각질세포간 지질은 표피 내 투과장벽 기능의 항상성 유지에 중요한 역할을 수행하는 것으로 알려져 있다. 각질세포간 지질의 공급은 주로 표피 과립층에 있는 층판소체가 과립층과 각질층의 경계 부위에서 세포 외 유출작용을 통하여 분비되면서 이루어진다. 피부에는 표피 내 칼슘 기울기가 존재하며, 칼슘 이온은 피부장벽 기능의 항상성 유지에 매우 중요한 신호전달 인자로 작용한다. 각질형성세포의 증식과 분화에 있어 핵심적인 역할을 수행하는 효소인 protein kinase C (PKC)는 serinethreonine kinase 에 속하는 효소로서 각질형성세포를 포함한 많은 종류의 세포에서 다양한 세포 내 반응을 조절한다. 각질형성세포의 분화 과정에서 칼슘에 의한 PKC의 활성화는 involucrin, loricrin, filaggrin 등과 같은 분화 단백질의 발현에 핵심적인 역할을 하는 것으로 알려져 있다. 배양된 각질형성세포에서 PKC의 활성화가 각질세포간 지질 성분중 하나인 세라마이드(ceramide)의 합성에 관여하는 효소의 활성화와 연관되어 있다는 보고가 있으나 아직까지 PKC와 피부장벽 기능간의 연관성에 대한 연구는 이루어진 바 없다. 본 연구에서는 무모 생쥐의 동물실험 모델을 이용하여 PKC가 피부장벽의 회복에 미치는 영향을 관찰하였다. 이를 위하여 반복적인 테이프 스트리핑 방법을 이용하여 8-12 주의 무모 생쥐의 피부장벽을 급성 손상시킨 후 Western blot과 면역조직화학염색을 시행하여 피부에서의 PKC 단백질 발현 양상을 관찰하였다. 또한 피부장벽을 급성 손상시킨 후 PKC 억제제를 도포하여 피부장벽의 회복율을 측정하고, 각질층내 지질의 변화를 관찰하였으며, 이들 억제제가 표피 내 칼슘 기울기에 미치는 영향을 관찰하였다. 이와 함께 배양된 각질형성세포에 PKC 억제제를 처리하여 세포 내 칼슘 이온 농도의 변화를 측정하였다. 실험 결과, 표피에서 PKC-α, -β, -δ, -η의 발현을 관찰할 수 있었다. PKC 억제제를 도포한 피부에서 급성 손상 후 피부장벽의 회복율이 저하되었으며, 층판소체의 분비가 억제되고 각질층의 지질구조의 변형이 나타나는 것을 관찰하였다. 또한, PKC 억제제가 피부장벽 손상 후 나타나는 정상적인 표피 내 칼슘 기울기의 소실을 억제하였고, 배양된 각질형성세포에서 세포 내 칼슘 이온의 농도를 증가시켰다. 이러한 세포 내 칼슘 이온의 증가는 칼슘 통로 차단제에 의하여 억제되었다. 급성 피부장벽 손상 후 PKC 억제제만을 처리한 경우 피부장벽의 회복이 저해되었으나, 칼슘 통로 차단제를 함께 도포한 후에는 피부장벽 회복의 저해가 억제되었다. 결론적으로 PKC 억제제는 과립세포 내의 칼슘의 농도를 증가시켜, 급성 피부장벽 손상 후 나타나는 피부장벽의 회복 과정을 억제한다. 그러므로 PKC의 각 아형별 역할은 알 수 없으나 일부 아형이 세포 내로의 칼슘 이동에 영향을 미쳐 피부장벽 기능의 급성 손상 후 회복 과정에 관여할 수 있을 것으로 생각된다.

Milk fat globule membrane (MFGM) is a lipid carrier in mammals including humans that consists mainly of polar lipids, like phospholipids, glycolipids, and sphingolipids. In the first study, a process to enrich polar lipids in commercial butter and whey powder, including polar lipids of MFGM, was developed. Whey protein concentrate (WPC) 60 was selected as the most suitable raw material based on the yield, phospholipid, protein, and lactose content of the polar lipid fraction obtained by ethanol extraction of two WPC (WPC60 and WPC70) and two buttermilk (A and B). After fractionation under optimum conditions, the polar-lipid enriched fraction from WPC60 contained 38.56% phospholipids. The content of glycolipids, cerebroside, lactosylceramide, ganglioside GM3, ganglioside GD3, was 0.97%, 0.55%, 0.09%, and 0.14%, respectively. Rancimat results showed that the oxidation stability of fish oil increased with an increase in the polar-lipid fraction by more than 30 times. In addition, the secretion of interleukin (IL)-6 and tumor necrosis factor (TNF)-α decreased in a concentration-dependent manner after treatment of RAW 264.7 cells with 0.1 to 100 ppm of the polar lipid fraction. In this study, polar lipid concentrates with antioxidant and anti-inflammatory activity were prepared from milk processing by-products. In the second study, we isolated the milk sphingolipid-enriched fraction (MSEF) of sweet buttermilk powder and conducted a clinical trial for evaluating its efficacy in skin barrier recovery. MSEF was isolated via solvent extraction of buttermilk powder, and further concentrated by removing the phospholipids and neutral lipids. A cream containing 1% MSEF was used during clinical trials to assess for water holding and skin barrier recovery capacities. The main components of the MSEF were sphingomyelin, glucosylceramide, and lactosylceramide, confirmed by thin layer chromatography (TLC), high-performance liquid chromatography (HPLC), mass spectrometry (MS), and nuclear magnetic resonance (NMR). The MSEF-cream-treated group had a significantly higher (p < 0.05) water holding capacity, compared to the base-cream (vehicle) group. Compared to that in the base-cream group, trans-epidermal water loss (TEWL) recovery increased in the presence of the sphingolipid-containing MSEF-cream (MSEF group), with a significant difference (p < 0.05) recorded on day 14. Taken together, these results showed the MSEF-cream contributed to improving the water holding capacity and skin barrier recovery of damaged skin. Therefore, sphingolipid-containing MSEF can be useful for strengthening or repairing skin barrier function.

피지의 참치 산업은 국가 경제에서 중요한 역할을 하고 있으나, 가공 과 정에서 두부, 내장, 껍질, 뼈, 프레임(frames), 절단 부산물(trimmings), 혈 수(bloodwater)와 같은 대량의 유기성 부산물이 발생한다. 이러한 물질들은 적절히 관리될 경우 가치 있는 바이오자원으로 활용될 수 있음에도 불구하고, 현재까지 상당 부분이 폐기물로 취급되고 있다. 이에 본 연구는 피지 산업형 참치 어업 부문에서 발생하는 유기성 참치 부산물을 관리하기 위한 순환경제 (Circular Economy) 프레임워크를 개발하는 것을 목적으로 한다. 2025년 산업 데이터를 활용한 분석 결과, 피지의 참치 가공시설은 단순 가공공장이 아니라 유기성 부산물이 집중적으로 발생하는 핵심 거점으로 나타 났으며, 특히 수출량이 국내 어획량을 초과하는 시기에 이러한 현상이 더욱 두드러졌다. 특히 PAFCO와 같은 대형 가공시설은 현지 어획량뿐 아니라 외부 반입 원료까지 처리하고 있어 대규모 부산물 발생 지점으로 기능하고 있었다. 또한 연구는 피지가 직면한 주요 장애요인은 부산물의 분리수거 체계 미비, 취약한 냉동 및 냉장 저장 시스템, 품질검사 부족, 그리고 수산 부산물에 대 한 불명확한 정책적·제도적 처리인 것으로 나타났다. 본 연구는 아이슬란드의 “100% Fish” 모델, 유럽연합(EU)의 동물성 부 산물 규제 체계, 일본의 식품재활용법 기반 순환체계를 참고하여, 피지의 현 실적 여건을 고려한 단계적 순환경제 프레임워크를 제안하였다. 첫 단계에서 는 부산물의 분리수거, 냉동 및 저장 시스템 구축과 같은 기초 인프라를 강화 하고, 이후 fishmeal, fish oil, fertilizer와 같은 저부가가치 활용을 확대 하며, 장기적으로는 collagen, gelatin, protein hydrolysates 및 nutraceuticals와 같은 고부가가치 산업으로 발전시키는 전략을 제안하였다. 결론적으로, 피지 참치 산업의 순환경제 전환은 첨단 바이오 기술 이전에 저장 및 냉동 기반의 초기 관리체계 구축이 선행되어야 하며, 이를 위해 정부 의 정책적 지원과 제도적 정비가 필수적임을 제시하였다. 주요어: 순환경제(Circular Economy), 피지(Fiji), 참치 산업(Tuna Fisheries), 수산부산물(Fisheries By-products), 냉동저장체계(Cold Chain), 블루이코노미(Blue Economy) Fiji’s tuna fisheries are important to the national economy, but processing activities produce large amounts of organic by-products such as bones, heads, skin, viscera, frames, bloodwater, and trimmings. Much of this material is still treated as waste, although it can be used as a valuable bioresource if it is properly handled. This thesis develops a Circular Economy Framework to manage organic tuna by- products in Fiji's industrial tuna fisheries sector. Using 2025 industry data, the study shows that processing facilities are the main sites where by-products are generated, especially when export volumes exceed local landings. This indicates that processing hubs such as PAFCO handle large volumes of tuna from both local and external sources, creating concentrated waste streams. The study finds that Fiji’s main barriers are limited source separation, weak freezing and cold storage systems, lack of quality testing, and unclear policy treatment of fish by-products. Drawing on examples from the European Union, Japan, and Iceland, this research proposes a phased approach that first improves segregation, freezing, and storage before moving toward value pathways such as fishmeal, fertilizer, fish oil, collagen, gelatin, and protein hydrolysates.
