    • Neutrophil pyroptosis mediates pathology of P.aeruginosa lung infection in the absence of the NADPH oxidase NOX2

      대식세포에서 세포 내 병원균을 효과적으로 처리하기 위한 작용기작으로 NLRC4 inflammasome의 활성화에 의한 caspase-1 의존적인 pyroptosis가 중요한 역할을 한다는 사실은 알려져 있다. 하지만 세균의 급성 감염에서 중요한 역할을 하는 호중구에서의 NLRC4 inflammasome에 의한 pyroptosis에 대한 연구는 아직까지 잘 되어 있지 않다. 따라서 본 연구를 통해 녹농균으로 인한 폐 감염 시 호중구 특이적인 조절 작용과 병적인 역할에 대해 알아보려고 하였다. Nox2 결손 유전자 마우스 모델을 이용해서 확인한 결과 nox2가 없을 시 녹농균에 대한 호중구의 항균 작용이 현저하게 감소되어 있었으며 pyroptosis 또한 증가되어 있었다. 그리고 이런 호중구의 pyroptosis 증가는 녹농균의 병원성 물질 중 하나인 flagellin에 의한 것이었음을 flagellin이 결손된 균주를 이용해서 확인하였다. 녹농균으로 인한 호중구의 pyroptosis가 NLRC4와 TLR5 의존적이라는 사실 또한 각각의 유전자 결손 마우스 모델을 통해 확인할 수 있었다. 본 연구를 통해 녹농균으로 인한 폐 감염 모델에서 호중구 pyroptosis의 작용기전을 밝혀내었다. 이 결과는 호중구의 기능에 문제가 있는 환자들에게서 급성 감염이 발생할 경우 호중구의 pyroptosis가 중요한 역할을 할 수 있을 것이라는 가능성을 제시한다. Nod-Like Receptor family, CARD domain-containing 4 (NLRC4)-inflammasome activation is required for efficient clearance of intracellular pathogens through caspsase-1-dependent pyroptosis in macrophages. Although neutrophils play a critical role in protection from P. aeruginosa infection, the mechanisms regulating NLRC4-inflammasome-mediated pyroptosis in neutrophils and its physiological role are largely unknown. I sought to determine the specific mechanisms regulating neutrophil pyroptosis in P. aeruginosa strain PAO1 (PAO1) lung infection and to identify the pathological role of this process. Nox2-/- models with reduced neutrophil antibacterial activity exhibited increased neutrophil pyroptosis, which was mediated by flagellin, a pathogenic PAO1 component. I also demonstrated that PAO1-induced pyroptosis in neutrophils was dependent upon NLRC4 and Toll-like receptor 5 (TLR5), using animals deficient of Nlrc4-/-, or Tlr5-/-. This study reveals previously unknown mechanisms and physiological role of neutrophil pyroptosis during P. aeruginosa lung infection. Furthermore, those findings regarding neutrophil pyroptosis in the context of neutrophil dysfunction may explain the causes of acute infectious diseases in patients with incompetent neutrophils.

    • Molecular Mechanisms of Streptococcus pneumoniae-induced Pyroptosis in Microglia : Involvement of NLRP3 Inflammasome and Autophagy

      Streptococcus pneumoniae is a Gram-positive, extracellular bacterium that is responsible for significant mortality and morbidity worldwide, causing pneumonia, otitis media and meningitis. During pneumococcal meningitis, resident immunocompetent cells recognize S. pneumoniae by pattern recognition receptors including Toll-like receptors (TLRs) and nucleotide-binding oligomerization domain-like receptors (NLRs). Among various NLR proteins, NLRP3 has been known to be activated by pneumococcus and organize inflammasome that triggers the activation of caspase-1 and subsequent maturation of caspase-1-dependent cytokines, IL-1β and IL-18 and eventually induces pyroptosis. Recent studies have suggested that several intracellular bacteria which trigger pyroptosis also stimulate autophagy to protect host cells by eliminating intracellular pathogens. However, the involvement of autophagy in S. pneumoniae-induced pyroptosis is remained unknown. In the present study, I investigated the molecular mechanisms of pyroptosis in S. pneumoiae-infected BV-2 microglial cells and the involvement of NLRP3 inflammasome and autophagy in this process. The present work revealed that S. pneumoniae induced pyroptosis through caspase-1 activation and subsequent maturation and secretion of IL-1β and IL-18. Potassium efflux was also required for pneumococcus-induced pyroptosis. In addition, it was found that the activation of caspase-1 and the maturation of IL-1β and IL-18 in S. pneumoniae-triggered pyroptotic cell death process was mediated by NLRP3 inflammasome. Autophagy was also induced by S. pneumoniae. Pneumococcal infection increased the expression of autophagy-related genes and induced autophagosome formation resulting in more cells with GFP-LC3+ dots. To determine the role of autophagy in S. pneuomoniae-induced pyroptosis, autophagy was inhibited by the treatment with autophagy inhibitor, 3-MA or Atg5 specific siRNA. It is also found that pyroptosis was further induced by the inhibition of autophagy, indicating that the protective role of autophagy in S. pneumoniae-induced pyroptosis of BV-2 microglia. The current study also investigated the involvement pf ROS in S. pneumoniae-induced pyroptosis and autophagy. At 4 h of post-infection, the generation of ROS by pneumococcal infection inhibited caspase-1 activation and stimulated autophagy. However, after 8 h of infection, the release of IL-1β was increased and caspase-1-denpendent cell death was finally induced by ROS production. Next, I examined whether pneumolysin, a pore-forming cytolysin and one of the most important virulence factor of pneumococcus, is involved in S. pneumoniae-triggered pyroptosis. The present study elucidated that the activation of caspase-1 and the production of IL-1β was induced by pneumolysin and the toxin eventually resulted in the pyroptotic cell death of BV-2 microglial cells. Consistent with in vitro experiments, S. pneumoniae induced caspase-1 activation and caspase-1-dependent cytokine maturation in mouse meningitis model. In addition, it was found that depletion of pneumolysin in pneumococcus caused the reduced expression of activated caspase-1 fragment and subsequent cytokine levels. In conclusion, these results demonstrated that S. pneumoniae induces pyroptosis in murine microglia and NLRP3 inflammasome are critical for caspase-1 activation and caspase-1-dependent cytokine maturation in pneumococcus-triggered pyroptotic cell death process. Furthermore, autophagy is stimulated and protects pneumococcus-infected microglia from pyroptosis. 폐렴구균(Streptococcus pneumoniae)의 감염은 전 세계적으로 높은 질병률과 사망 률을 초래하며 폐렴, 중이염, 그리고 뇌수막염과 같은 질병들을 유발한다고 알려져 있다. 폐렴구균성 뇌수막염이 발병하게 되면 뇌수막에 존재하는 면역세포들은 Toll-like receptors(TLRs) 또는 Nucleotide-binding oligomerization domain-like receptors(NLRs)과 같은 양식 인지 수용체(Pattern recognition receptors)들을 통하여 폐렴균을 인지하게 된다. 다양한 NLR 단백질들 중에서 NLRP3는 폐렴균에 의해서 활성화 되며, NLRP3 inflammasome을 조직하여 caspase-1의 활성화 및 caspase-1 의존성 cytokine인 IL-1β와 IL-18의 성숙을 유도하고, 최종적으로는 pyroptosis를 일으킨다고 알려져 있다. 최근에 발표된 연구들에 따르면, 몇몇의 세포 내 세균들은 숙주세포의 pyroptosis를 일으키는 동시에 자가포식(Autophagy)을 유도한다고 보고되었다. 하지만 폐렴균에 의해 유도된 pyroptosis 과정 중에 autophagy가 연관되어 있는지에 관한 연구는 아직 미흡한 상태이다. 따라서, 본 연구에서는 폐렴균 감염에 의한 소교세포의 pyroptosis 작용 기전과 그 과정에서의 NLRP3 inflammasome 및 autophagy의 관련성을 밝히고자 하였다. 연구 결과, 폐렴균 감염 시 caspase-1이 활성화되고 IL-1β와 IL-18이 성숙 및 분비되며 pyroptosis가 유도되었다. 또한 pyroptosis가 일어나는 과정에서 칼륨의 유출이 요구되며, caspase-1의 활성화와 IL-1β및 IL-18의 성숙에 NLRP3 inflammasome이 관여함을 확인하였다. Autophagy 관련 인자들의 발현과 autophagosome 형성이 증가함을 통하여 폐렴균 감염에 의해 Autophagy가 일어남을 확인하였고, 3-MA 또는 siAtg5를 처리하여 autophagy를 억제시킨 결과, pyroptosis가 더 강하게 유도되는 것을 확인함으로써 autophagy가 pyroptosis로부터 소교세포를 보호함을 확인하였다. 또한 pyroptosis 과정에서 ROS의 역할을 알아본 결과, 감염 초기에는 생성된 ROS가 autophagy를 촉진하여 caspase-1의 활성화를 억제하나 감염 후기에는 IL-1β의 분비와 pyroptosis를 유도함을 확인하였다. 다음으로, 폐렴균의 강력한 독성 인자 중 하나인 pneumolysin이 폐렴균 감염에 의한 pyroptosis에 관련되어있는지 알아보았다. 그 결과, pneumolysin이 caspase-1을 활성화시키고 IL-1β의 분비를 촉 진하며 최종적으로 소교세포의 pyroptosis를 유도함을 확인하였다. 이러한 in vitro 실험 결과와 마찬가지로, 쥐 폐렴구균성 뇌수막염 모델에서도 caspase-1이 활성화되고 caspase-1 의존성 cytokine들이 성숙되는 것을 확인하였고, pneumolysin이 결핍된 돌연변이 폐렴균 감염 시 caspase-1의 활성화와 그에 따른 cytokine들의 분비가 감소되는 것을 확인하였다. 이상의 연구결과들을 통하여, 본 연구는 폐렴균 감염 시 소교세포에서 pyroptosis가 일어나며 그 과정에서 NLRP3 inflammasome이 caspase-1의 활성화 및 caspase-1 의존적인 cytokine의 성숙에 관여함을 밝혔다. 또한 소교세포를 pyroptosis로부터 보호하기 위한 기작으로 autophagy가 일어난다고 사료된다.

    • Inhibition of ASMase Induces Pyroptotic-lysis of Reactive Astrocytes

      Reactive astrocytes are known to persist in neuroinflammatory environments, contributing to disease progression. In this study explored the impact of LPS on pyroptosis-related markers and gene expression in primary astrocytes and the C8-D1A cell line. LPS stimulation resulted in the upregulation of pyroptosis-associated proteins and genes related to reactive astrocytes, leading to significant morphological alterations in both models. Furthermore, peripheral administration of LPS induced astrocyte activation and pyroptosis in the hippocampus, suggesting a link between peripheral immune stimuli and central nervous system (CNS) inflammation. Additionally, we studied the role of an acid sphingomyelinase (ASMase), in this context. Inhibition of ASMase enhanced pyroptosis by disrupting the repair of GSDMD pores, effectively inducing the death of reactive astrocytes. This outcome correlated with a decrease in glial fibrillary acidic protein (GFAP) expression in the mouse brain, suggesting a potential mechanism to mitigate reactive astrocyte-mediated neuroinflammation. These findings provide a deeper understanding of the mechanisms underlying LPS-induced pyroptosis and the regulation of reactive astrocytes in the CNS. Keywords: astrocytes, reactive astrocytes, pyroptosis, acid sphingomyelinase 반응성 성상세포는 신경염증 환경에서 사멸하지 않고, 질병을 악화시킨다고 알려져 있다. 본 연구에서는 쥐 뇌 조직 유래 성상세포와 C8-D1A 쥐 성상세포 세포주에서 파이롭토시스 관련 유전자 발현 및 세포사멸 유도에 미치는 영향을 조사하였다. LPS 자극은 파이롭토시스와 관련된 마커 및 반응성 성상세포와 관련된 유전자의 발현을 증가시키며, 유의미한 형태학적 변화를 유도하였다. 말초에 LPS를 투여한 결과, 해마에서 성상세포의 활성화와 파이롭토시스가 유도되어 말초 면역 자극과 중추 신경계 염증 간의 연관성을 시사하였다. 더불어 산성 스핑고미엘린 분해효소 저해를 통해 게스다민 구멍 복원을 저해하여 반응성 성상세포의 사멸을 유도함을 확인하였다. 이러한 효과는 생쥐 뇌에서 GFAP 형광 발현을 감소시켰으며, 이는 반응성 성상세포 매개 신경염증 진행을 저해할 수 있는 가능성을 보였다. 이러한 연구를 통해 파이롭토시스 및 반응성 성상세포 사멸 조절 메커니즘에 대한 새로운 통찰을 제공하며, 추가 연구의 필요성을 제안한다. 대표어 : 별아교세포, 활성별아교세포, 파이롭토시스, 산성 스핑고미엘린 분해효소 

    • 폐 편평상피세포암에서 pyroptosis 에 의한 lurbinectedin 의 항암 작용 기전 이해

      Lurbinectedin is a chemotherapeutic DNA alkylating agent used to treat metastatic small-cell lung cancer. However, there is insufficient research on the mechanism of action of anticancer agents and their extended application to other lung cancer types. In this study, we investigated the potential of lurbinectedin as a treatment for squamous cell carcinoma based on its anticancer mechanisms. Lurbinectedin treatment exhibited anticancer effects similar to those observed in small cell lung cancer, even in non-small cell lung cancer containing adenocarcinoma and squamous cell carcinoma. Interestingly, lurbinectedin treatment showed cell swelling, large membrane blebs, and membrane rupture, such as pyroptosis. In consistent with these results, lurbinectedin treatment increased GSDME N-Terminal (GSDME-NT) fragmentation in a time or dose-dependent manner. Furthermore, we found that GSDME-NT was attenuated by caspase inhibitor upon treatment with Lurbinectedin, suggesting that GSDME activation is induced by the caspase pathway. GSDME knockdown via shRNA blocked pyroptotic morphology and reduced cell death in lurbinectedin-treated cells. These anticancer effects were confirmed in organoids derived from squamous cell carcinoma. Collectively, our findings suggest that lurbinectedin may be a novel treatment for squamous cell carcinoma through a previously unidentified mechanism called pyroptosis. Lurbinectedin은 전이성 소세포폐암 치료에 사용되는 DNA 알킬화제이다. 하지만 현재까지 이 약물에 대한 추가적인 작용 기전이나 이를 다른 유형의 폐암에 확장 적용하는 연구는 아직 부족한 실정이다. 이를 위해, 본 연구에서는 lurbinectedin의 항암 기전에 기반하여 폐편평상피세포암(squamous cell carcinoma)에 대한 치료 가능성을 탐색하였다. 결과적으로, lurbinectedin은 소세포폐암에서와 유사한 항암 효과를 비소세포폐암(non-small cell lung cancer) 유형인 선암과 편평상피세포암에서도 나타냈다. 흥미롭게도, lurbinectedin 처리 시 세포 팽창, 세포막 돌기 형성 및 파열 등 pyroptosis(염증성 세포사멸)와 유사한 형태학적 변화가 관찰되었다. 이러한 형태학적 변화 결과와 일치하게, lurbinectedin은 농도와 시간에 의존적으로 GSDME N-말단 단편(GSDME-NT)의 생성을 증가시켰다. 나아가, caspase 억제제를 함께 처리했을 때 GSDME-NT 단편화가 감소하는 것을 확인하였으며, 이는 GSDME의 활성화가 caspase 경로에 의해 유도됨을 시사하였다. 또한, shRNA를 이용한 GSDME 유전자 억제는 pyroptosis 형태를 억제하고 lurbinectedin에 의한 세포사멸을 감소시켰다. 이러한 항암 효과는 폐편평상피세포암 유래 오가노이드 모델에서도 확인되었다. 따라서 이를 종합적으로 보았을 때, 본 연구는 lurbinectedin이 기존에 알려지지 않은 pyroptosis라는 새로운 세포사멸 기전을 통해 폐편평상피세포암에 대한 치료제로서의 가능성을 가질 수 있음을 제시한다.

    • 비알콜성 지방간염에서 NLRP3 인플라마좀과 pyroptosis 에 대한 S IRT 1 의 역할

      Saturated fatty acid (SFA) exposure causes nonalcoholic steatohepatitis (NASH). Inflammation is the leading cause of pathologies of nonalcoholic fatty liver disease, containing NASH. However, it is uncertain how SFA induces inflammation. Increasing evidence has demonstrated that caspase-1 activation and pyroptosis contribute to the development of NASH. In addition, we found that SIRT1 activity was associated with palmitic acid-induced elevation of NLRP3 inflammasome, attenuating caspase-1 expression, and reducing LDH release, and apoptosis. Interestingly, ROSA26-Cre; SIRT1fl/fl (exon 4) conditional knockout contributed to liver damage due to NLRP3 inflammasome, caspase-1 expression, LDH release, and pyroptotic cell death. The human liver cancer cell line HepG2 cells were exposed to palmitic acid (PA). Cells also induced NLRP3 inflammasome or GSDMD/pyroptosis during PA treatment for 24 hours. Thus, ROSA26-Cre; SIRT1fl/fl (exon 4) conditional knockout mouse displayed hyperactivated NLRP3 inflammasome, which resulted in hyperactivation of inflammasomes and increased mortality in NASH models. These findings define a unique regulatory mechanism of NLRP3 inflammasome activation from GSDMD/pyroptosis.

    • S100A8/A9에 의한 1차 배양 미세아교세포 및 성상교세포에서 증가되는 S100A8의 TLR4/NF-κB/NLRP3 경로를 통한 pyroptosis 조절

      염증 매개체 중 하나인 S100A8/A9는 알츠하이머병(AD) 또는 외상성 뇌손상(TBI)과 같은 여러 신경퇴행성 질환에서 증가하며 염증의 신뢰할 수 있는 마커로 간주된다. S100A8/A9는 위험 관련 분자 패턴(DAMPs)에 속하며 면역 반응을 유도할 수 있다. 미세아교세포와 성상교세포는 중추신경계(CNS)의 비신경 세포에 해당된다. 미세아교세포는 염증과 손상이 일어나는 동안 대식세포로 기능하며 중요한 식세포 역할을 한다. 또한 성상교세포는 여러 사이토카인의 분비 및 신경독성 물질의 방출을 담당한다. 염증과 관련된 세포 사멸 중 파이롭토시스(pyroptosis)는 유전적으로 암호화된 괴사의 경로 중 하나이며 비염증성 형태의 세포 사멸과는 다른 유형의 세포 사멸이다. 지금까지 다른 연구들에선 S100A8/A9를 사용한 암의 세포 사멸을 확인했지만 뇌 질환에서는 현재까지 밝혀진 바가 많지 않다. 본 연구에서는 S100A8/A9 처리에 의한 S100 A8 발현이 1차 미세아교세포 및 성상교세포의 파이롭토시스에 영향을 미치는지 조사하였고 그 결과, 1차 미세아교세포와 성상교세포에서 S100A8/A9에 의해 유발된 caspase-1의 활성화와 특정 사이토카인의 성숙은 TLR4/NFκB/ NLRP3 경로를 통해 파이롭토시스 세포 사멸을 유도함을 확인했다. 또한 S100 A8 short-hairpin RNA (shS100A8)는 ninjurin-1 (NINJ1) 및 GSDMD와 같은 파이롭토시스 관련 단백질의 발현을 억제하였다. 요약하면, 본 연구 결과는 1차 소교 세포 및 성상교세포에서 S100A8/A9에 의한 증가된 S100A8 발현이 파이롭토시스를 유도한다는 것을 밝히는 새로운 관점을 제안한다. One of the inflammatory mediators, S100A8/A9 is increased in several neurodegenerative diseases such as Alzheimer’s disease (AD) or traumatic brain injury (TBI) and is regarded as a reliable marker of inflammation. S100A8/A9 belongs to the danger -associated molecular patterns (DAMPs) and types of molecules that can elicit an immune response. Microglia and astrocytes are non-neuronal cells of the central nervous system (CNS). During inflammation and injury, microglia function as macrophages and play an important phagocytic role. Astrocytes take charge of secretion of multiple cytokines or release of neurotoxic amounts. Among the cell death associated with inflammation, pyroptosis is one of the genetically encoded pathways of necrosis and it is a different type of cell death from the non-inflammatory form of apoptosis. So far, other studies have confirmed cell death through S100A8/A9 in cancer, but not in brain disease. As a heretofore not revealed, I investigated whether S100A8 expression by each treatment of S100A8 and S100A9 affect the pyroptosis of primary microglia and astrocytes. The activation of caspase-1 and maturation of specific cytokines in the S100A8/A9-triggered primary microglia and astrocytes induced pyroptotic cell death through TLR4/NF-κB/ NLRP3 pathway. Furthermore, S100A8 short-hairpin RNA (sh S100A8) inhibited the expression of pyroptosis-related proteins like ninjurin-1 (NINJ1) and GSDMD. In summary, our findings suggest a novel perspective, revealing that the increased S100A8 expression by S100A8/A9 in primary microglia and astrocytes induce pyroptosis.

    • Role of NLRP3 inflammasome activation in the pyroptotic cell death of glial cells

      Inflammasome is a caspase-1-activating multi-protein complex composed of sensor molecules such as NOD-like receptor family, pyrin domain-containing 3 (NLRP3), apoptosis-associated speck-like protein containing a caspase recruitment domain (ASC) and procaspase-1. The inflammasome complex plays a crucial role in the induction of inflammatory responses mainly in myeloid cells such as macrophages. Assembly of the inflammasome complex induces the activation of caspase-1, which leads to the maturation and release of pro-inflammatory cytokines, such as interleukin (IL)-1β and IL-18. Also, active caspase-1 cleaves gasdermin D (GSDMD) which initiates generation of pores in plasma membrane and leads to ‘pyroptosis’, a form of programmed lytic cell death. Recent studies have proposed that NLRP3 inflammasome possibly contribute to neuroinflammation and neurodegenerative diseases. Also, NLRP3 inflammasome complexes are activated in glial cells and glial inflammasome activation is critical to various neurologic disorders. However, the effects of glial pyroptosis in neuroinflammation are still poorly understood. Here, I examined the effect of mixed glial pyroptosis upon NLRP3 inflammasome activation. Active caspase-1 and IL-1β was secreted by lipopolysaccharide (LPS) and ATP stimulation in mixed glia. However, pyroptosis of mixed glia with LPS plus ATP treatment were reduced, not like bone marrow-derived macrophages (BMDMs). Furthermore, attenuated pyroptosis of mixed glia was not related to GSDMD, the effector of pyroptosis. Next, microglia were isolated from mixed glia and examined NLRP3 inflammasome-mediated pyroptosis. Interestingly, pyroptosis of microglia present in mixed glia showed a weaker response than pyroptosis of isolated microglia. Additionally, mixed glia showed less toxicity to SH-SY5Y, dopaminergic neuroblastoma cell line, than BMDMs upon LPS/ATP stimulation with coculture system. Collectively, mixed glia showed NLRP3 inflammasome-mediated attenuated pyroptosis than BMDMs and interaction between glial cells potentially reduced microglial pyroptosis.

    • Inhibitory effect of microRNA-762 on pyroptotic cell death of mesenchymal stem cell after transplantation in ischemic heart

      The high death rate of cells transplanted into damaged hearts due to exposure to harsh conditions, such as hypoxia and high reactive oxygen species (ROS) concentrations, hampers the efficacy of stem cell-based therapy. Preventing cell death after transplantation is an essential requirement for using cell therapy to treat damaged hearts. Thus, the long-term survival of transplanted cells is a major goal in optimizing cell therapeutic applications. In addition to necrosis, apoptosis, and anoikis, pyroptosis has recently been recognized as a form of programmed cell death in the myocardium. Pyroptosis is mediated by cell lysis and the release of inflammatory cytokines, such as interleukin (IL)-1β and IL-18. Furthermore, cysteine-dependent aspartate-specific proteases 1 (caspases 1) are sequentially cleaved by pyroptosis-mediated cell death. Suppressing pyroptosis to improve transplanted stem cell survival in the infarcted myocardium has not been well studied. Thus, we inhibited pyroptosis to increase the survival rate of transplanted cells in the damaged myocardium. In the present study, IL-1β staining demonstrated that pyroptosis occurred in hearts with ischemia-reperfusion (I/R) injury. In addition, the expression of F4/80, a specific macrophage marker, was increased in the damaged myocardium, thus indicating macrophage recruitment. Next, increased pyroptosis-specific gene expression levels were found in RAW264.7 cells treated with lipopolysaccharides (LPS) and interferon (IFN)-γ. These data show that M1 macrophages are specifically involved in pyroptosis in hearts with I/R injury. Furthermore, specific microRNAs (miRs) have been identified to inhibit pyroptosis as well as improve the survival of stem cells transplanted into damaged hearts. miRs were screened by differentially expressed gene (DEG) analysis, and miRNA-762, which inhibits IL-1β expression by binding to its 3’ untranslated region (UTR), was identified. Significantly decreased pyroptosis-related gene and protein expression levels were detected in mesenchymal stem cells (MSCs) transfected with miR-762. Finally, cardiac fibrosis was ameliorated by transplanting miR-762-transfected MSCs into hearts with I/R injury. In conclusion, pyroptosis is an essential mechanism that induces inflammation and cell death. In addition, inhibiting the pyroptosis factor IL-1β by transfecting miR-762 into MSCs could be used in cell therapy for treating damaged hearts.

      Liver cancer is the leading cause of cancer mortality worldwide. Despite the improved treatment of HCC, the 5-year survival rate is relatively lower than other cancers, therefore further research to identify a new target is needed. Caspase-10 is a human-only protein that is highly homologous to caspase-8. Both caspase-10 and caspase-8 are known to activate apoptosis. Given that caspase-10 is highly homologous to caspase-8, it has been reported that there is overlap between the substrate of caspases. However, recent studies reported that caspase-10 might have a distinct role in cancer cell death that differs from that of caspase-8. To elucidate the mechanism of caspase-10 in cancer, we generated a caspase-10 knock-out cell line and performed RNA-seq analysis. In this study, we identified that caspase-10 induces pyroptosis. We show that caspase-10 mediates pyroptosis by cleaving GSDME under the treatment of sorafenib. Recently, switching from apoptosis to other types of programmed cell death such as pyroptosis has emerged as a new strategy for cancer treatment. Our findings suggest that targeting caspase-10 can be a potential therapy for patients with liver cancer. 간암은 전 세계적으로 높은 사망률을 보이는 암 종이다. 간암에 대한 많 은 치료법이 개발되었지만 암 5년 상대 생존율은 타 암 종에 비해서 낮은 수준을 보이고 있다. 따라서 간암 치료법에 대한 더 많은 연구가 이루어 져야 한다. Caspase-10은 인간에게만 있는 프로테아제로서, 유전자 서열에 있어 caspase-8과 높은 상동성을 가지고 있다. Caspase-10과 caspase-8은 apoptosis를 유도하는 것으로 이미 알려져 있다. 때문에 이 효소에 대한 기질 또한 비슷할 것으로 생각되고 따라서 비슷한 역할을 할 것으로 생각 되었다. 하지만 최근 연구들에서 caspase-8과 구별되는 caspase-10만 의 역할이 있다고 보고되고 있으며 우리는 이 연구를 통하여 암세포에서 caspase-10의 역할을 규명하고자 하였다. 이를 위해 먼저 간암 세포 주인 HepG2세포를 이용하여 caspase knock-out 세포 주를 제작하였다. 그 후 제작한 caspase knock-out 세 포 주에 대한 RNA-seq을 수행하였다. 이 연구를 통해 caspase-10이 GSDME를 활성화 시킴으로써 파이롭토 시스를 일으키는 것을 확인 할 수 있었다. 또한 이러한 세포 사멸은 sorafenib 자극을 주었을 때 유도될 수 있음을 확인할 수 있었다. 이전까지 암 치료 요법으로서 암세포의 apoptosis를 유도하는 방법이 일반적으로 많이 이용되어왔으나, 암세포의 apoptosis 저항성과 같은 한 계점이 존재한다. 근 보고되는 연구에서 암 치료 요법으로서 pyroptosis 와 같은 다른 유형의 세포사멸을 유도하는 것이 새로운 치료방법으로 떠 오르고있다. 이 연구를 통하여 Caspase-10를 조절하는 것이 새로운 간암 치료 방법 이 될 수 있음을 시사하였다.

    • Novel microwave plasma treated solutions (MPTS) trigger GSDME induced pyroptosis through intracellular ROS signaling in anaplastic thyroid cancer

      Anaplastic thyroid cancer (ATC) is one of the most aggressive cancers that is accompanied by a poor prognosis and resistance to multimodal treatments. Various drugs have a short response time and are likely ineffective against anaplastic thyroid cancer. Therefore, new molecular therapies and novel targets are needed to treat anaplastic cancers. Recently, plasma medicine has been used not only in basic fields, but also in biomedical applications on skin, wound healing, and cancer treatment of the liver, colon, and head and neck. However, direct plasma has difficulty treating, sustaining, and permeating the various affected areas. In this study, we prepared microwave plasma treated solutions (MPTS) and investigated their effect on ATC cells. Treatment with MPTS reduced the cell viability of ATC cells, which exhibited cell swelling and bubble formation morphologically. Gasdermin E(GSDME) was cleaved by MPTS and induced pyroptosis in ATC cells. Inhibiting caspase activity and knock down of GSDME showed reduced caspase-3 dependent cleavage of GSDME, consequently increasing cell viability and decreasing lactate dehydrogenase release. Furthermore, MPTS enhanced the production of intracellular reactive oxygen species (ROS) and induced phosphorylation of IRE1α that is associated with endoplasmic reticulum (ER) stress. Therefore, GSDME-mediated MPTS induced pyroptosis downstream of the ROS/ER stress pathway and activated caspase-3. We then confirmed tumor shrinkage through in vivo and histological analysis, with a focus on the increasing GSDME-N terminal. In summary, these results demonstrate that MPTS induces pyroptosis via GSDME activation and ROS generation in vitro and in vivo in anaplastic thyroid cancer models, suggesting a novel mechanism and indicating a potential chemotherapeutic approach for the treatment of ATC. 최근 발생률이 급증하고 있는 갑상선 암은 분화 갑상선암 (differentiated thyroid cancer), 미 분화 갑상선암 (undifferentiated or anaplastic thyroid cancer), prooly differentiated carcinoma, 갑상선 수질암(medullary thyroid cancer로 분류되며, 그중 anaplastic thyroid cancer는 예후가 좋지 않고 다중치료에 대한 내성을 동반하는 가장 위험한 암 중 하나입니다. 이러한 anaplastic thyroid cancer의 우선적인 치료는 TSH 억제요법, 전이나 재발 병소에 대한 수술적 치료, 방사선 치료 등과 같은 다양한 치료법을 사용해도 반응 시간이 짧고 ATC에 효과가 없을 가능성이 높습니다. 따라서 ATC를 치료하기 위해서는 새로운 molecular therapy와 새로운 타겟으로 하는 약물의 개발이 필요합니다. 최근 의료관련 분야에 플라즈마 의학이 적용되고 있다, 특히, 저온 상압 플라즈마는 인체조직에 열 손상을 입히지 않을 뿐만 아니라, 암 치료, 살균, 치아미백, 지혈, 상처치유 등에서 높은 효율성을 보이고 있다. 그러나 이러한 저온 상압 플라즈마는 다양한 영향을 받는 부위에 치료, 유지 및 침투하는 데 어려움이 있습니다. 본 연구에서는 기존 저온 상압 플라즈마를 개선한, Microwave plasma treated solutions (MPTS)을 만들고 ATC에 미치는 영향을 연구했습니다. MPTS를 이용한 치료는 형태학적으로 세포 팽창과 거품 형성을 나타내며 ATC 세포의 생존력을 감소시켰습니다.또한, MPTS를 처리시, 새로운 암세포의 사멸중 하나인, pyropotosis를 유도하며, Gasdermin E(GSDME)가 MPTS에 의해 절단되어, ATC에서 pyroptosis를 유도하였습니다. Caspase 활성 억제 및 GSDME의 knock down은 caspase-3 의존적 GSDME cleavage 감소를 보여 결과적으로 세포 생존력을 높이고 LDH방출을 감소시켰습니다. MPTS는 세포 내 활성 산소 종(ROS)의 생성을 강화하고 소포체(ER) 스트레스와 관련된 IRE1α의 인산화를 유도했습니다. 따라서 GSDME 매개 MPTS는 ROS/ER 스트레스 경로의 하류로 pyroptosis를 유도하고 활성 caspase-3를 유도했습니다. 요약하자면, 이러한 결과는 MPTS가 GSDME 활성화와 ROS 생성을 통해 pyroptosis를 유도하여 새로운 메커니즘을 제안하고 ATC 치료를 위한 잠재적 치료법을 나타낸다는 것을 보여줍니다.
