    • Optimizing CRISPR-mediated gene activation system in Chinese hamster ovary cells

      배경: Chinese hamster ovary (CHO) 세포는 바이오 의약품 산업에 서 선호되는 세포주이다. 그에 따라 수많은 연구자들이 CHO 세포의 성 능을 향상시키기 위한 노력을 기울이고 있으며, 이를 위한 주요 전략 중 하나가 생장 및 생산성과 관련된 유전자의 발현이다. CRISPR 활성화 (CRISPRa) 시스템은 유전자 스크리닝 시스템 중 하나이다. 이 시스템 을 통해 가이드 RNA (gRNA)를 변경함으로써 간단하고 빠르게 여러 유전자의 발현이 가능하다. 선행 연구에서는 이 시스템이 다양한 유형의 인간과 마우스 세포주에서 효과적임을 입증하였지만, CHO 세포에서는 체계적인 연구가 부족하였다. 따라서, 이 연구에서는 CHO 세포에 유용 한 유전자들을 탐색하는데 도움을 주기 위하여 CRISPRa 시스템이 CHO세포에서도 잘 활용될 수 있도록 최적화하였다. 결과: CRISPRa의 효과는 여러가지 요인에 따라 달라진다. 따라서, 이 요인들을 조절하기 위하여 형광 발현 유전자를 CRISPRa 시스템을 활용해 활성화했고, CHO세포에서 형광 발현 정도가 최대가 되는 조건을 최적 조건으로 판단하였다. 그리고 찾아낸 최적의 조건을 적용하여 CHO 세포의 내인성 유전자를 활성화하였다. 결과적으로, 해당 유전자의 전사량이 크게 증가한 것을 확인할 수 있었다. 또한, CRISPRa에 의한 유전자 활성화 효과는 강력한 유전자 발현 효과로 알려져 있는 형질 전 환 유전자 발현 방법과 비교했을 때 충분히 효과적이었다. 또한, CRISPRa에 의한 유전자 활성화 효과는 강력한 유전자 발현 효과로 알 려져 있는 형질 전환 유전자 발현 방법과 비교했을 때 충분히 효과적이 었다. 결론: 본 연구에서는 CRISPRa 시스템의 효과에 영향을 미치는 여러 가지 요인들을 조절하여 CHO cell에서 최적의 효과를 낼 수 있는 조건 을 찾았다. 실제로 이 조건을 사용하였을 때, 내인성 유전자가 잘 활성 화되었음을 확인할 수 있었다. 따라서, CRISPRa가 유전자 스크리닝 도 구로서 잘 활용될 수 있음을 보여줄 수 있었다. 나아가, 본 연구에서 찾 아낸 최적화된 CRISPRa의 조건은 앞으로 CHO세포에서 이 방법을 사 용하여 cell engineering에 유용한 유전자들을 쉽고 빠르게 찾아내는데 도움이 될 것이라고 생각한다. Background: Chinese hamster ovary (CHO) cells are the preferred cell lines in the biopharmaceutical industry. Thus, many researchers have made efforts to improve the performance of CHO cells. One of the primary strategies for this is the expression of genes related to their growth and productivity, and through gene screening, these useful genes can be identified. The CRISPR activation (CRISPRa) system is one of gene screening tool. This system allows for the easy and rapid expression of multiple genes, by simply altering the guide RNA (gRNA). While previous studies have demonstrated its effectiveness in various types of human and mouse cell lines, it has not been widely studied in CHO cells. Therefore, the purpose of this study is optimizing CRISPRa system for CHO cell to facilitate the screening of genes useful for improving CHO cell performance. Results: The effectiveness of CRISPRa depends on several factors. Therefore, by controlling these factors, this study activated the fluorescent gene using the CRISPRa system and determined the optimal conditions that maximally activate the expression of fluorescence in CHO cells. The optimal conditions found were applied to the activation of the endogenous gene in CHO cells. As a result, the transcription level of the target gene was successfully increased. Also, the gene activation effect of CRISPRa was sufficiently effective when compared to the effect of transgene expression, which is known for its strong gene expression effect. Conclusion: In this study, the optimal conditions for CRISPRa system in CHO cells were identified by controlling various factors that influence its effectiveness. When the identified conditions were applied, successful activation of endogenous genes was confirmed. Therefore, this result demonstrated the possibility of CRISPRa as an effective gene screening tool. Furthermore, the optimized CRISPRa conditions identified in this research will be helpful in quickly and easily searching useful genes for CHO cell engineering in the future.

    • Production of glycoengineered CTLA4-Ig with enhanced sialylation in Chinese hamster ovary cells

      Chinese hamster ovary (CHO) 세포주는 다른 세포주에 비하여 높은 단백질 생산성을 보이며 인간과 유사한 glycosylation을 수행하는 등의 장점이 있다. CHO 세포는 이러한 장점을 갖고 있어 바이오의약품을 생산하는 세포주의 70%를 차지하고 있다. 블록버스터 바이오의약품은 당단백질이 다수를 차지하고 있다. 특히 당단백질을 구성하고 있는 글리칸은 비교적 작은 분자량을 차지함에도 불구하고 바이오의약품의 효능에 중요한 영향을 미치게 된다. 본 실험에서 CHO 세포를 이용하여 생산하는 cytotoxic T-lymphocyte associated protein 4-Ig (CTLA4-Ig)는 CTLA4와 항체의 Fc 부분이 융합된 Fc-fusion protein이다. CLTA4-Ig는 CTLA4부분에 2개의 N-glycan과 Fc 부분에 1개의 N-glycan을 갖고 있는 당단백질이다. CLTA4-Ig는 Fc 부분과의 융합을 통하여 체내 반감기가 증대되지만 CTLA4 부분에 부착된 2개의 N-glycan에 시알산이 부가되지 않을 경우 체내 반감기가 감소하게 된다. 따라서 본 연구에서는 CHO 세포에서 생산하는 CTLA4-Ig의 시알산을 증대시키기 위하여 유전자 조작, 배양중 첨가물의 사용, 세포사멸 억제의 방법을 적용하였다. CTLA4에 위치한 N-글리칸은 최대 3개의 안테나 구조가 형성된다. 따라서 안테나를 형성하는 유전자인 GnT-IV와 GnT-V중 3개의 안테나 구조에 주요한 효소를 선별하고 이와 함께 인간형 시알산을 부가하는 ɑ2,6-sialyltransferase (ɑ2,6-ST)를 과발현하여 최종적으로 시알산이 증가된 형태의 CTLA4-Ig를 생산하는 것을 목표하였다. GnT-IV와 GnT-V가 과발현된 세포주를 비교하였을 때, GnT-IV가 과발현된 세포주에서 가장 tri-antennary 구조가 높게 형성되는 것을 알 수 있었다. 또한 GnT-IV와 ɑ2,6-ST를 동시에 과발현 하였을 때 가장 높은 시알산 함량을 얻을 수 있었다. 이를 통하여 CTLA4-Ig의 시알산 및 tri-antennary 구조를 증가시키기 위하여 GnT-IV와 ɑ2,6-ST를 과발현하는 것이 효율적인 전략임을 확인하였다. Glycoengineering 기술이 적용된 세포주를 배양할 때 시알산을 부가하는 donor substrate를 공급하기 위하여 N-acetylmannosamine (ManNAc)을 첨가하였다. 20 mM ManNAc이 첨가됨에 따라 wild type의 세포주에 비하여 1.5배의 시알산이 증대된 것을 확인하였다. 또한 glycoengineering의 적용과 ManNAc의 첨가가 당단백질의 시알산 함량 증대에 시너지 효과를 보이는 것을 알 수 있었다. 마지막으로 세포사멸로 인하여 방출되는 sialidase에 의하여 배양중 시알산이 부가된 당단백질이 분해되는 것을 억제하기 위하여 suramin을 첨가하였다. Suramin을 첨가하였을 때 세포의 사멸이 억제되어 생산성이 1.32배 증가하였다. 또한 suramin의 첨가는 CTLA4-Ig에 부착된 시알산을 최대 1.15배 증가시키는 것으로 확인되었다. 그러므로 세포사멸이 억제됨에 따라 방출되는 sialidase가 감소하게 되어 이러한 긍정적인 효과를 얻은 것으로 확인되었다. 결론적으로, 본 실험을 통하여 glycoengineering 기술을 적용하여 CTLA4-Ig의 시알산을 증대시키기 위한 효율적인 유전자를 선별하였다. 또한 rate-limiting step으로 작용하는 CMP-sialic acid를 증대시키기 위하여 ManNAc을 배양중 첨가하였고 glycoengineering이 적용된 세포주에 시너지적인 효과를 주어 시알산 함량의 증대를 확인하였다. 마지막으로 배양중 세포사멸에 의하여 방출된 시알산의 분해를 억제하기위해 suramin을 첨가하여 효율적으로 시알산 분해를 억제하였다. 이러한 다양한 방법을 CHO 세포배양에 적용하였을 때 높은 수준의 시알산이 부가된 CTLA4-Ig를 생산할 수 있는 전략을 수립할 수 있을 것으로 기대된다. Chinese hamster ovary (CHO) cell line has higher protein productivity than other cell lines and has the advantage of human-like glycosylation. For these reasons, CHO cells have accounted for 70% of the cell lines producing biopharmaceuticals. The majority of biopharmaceuticals is composed of glycoproteins. In particular, glycans constituting glycoproteins have an important effect on the efficacy of biopharmaceuticals despite their relatively small molecular weight. In this study, cytotoxic T-lymphocyte associated protein 4-Ig (CTLA4-Ig) produced using CHO cell cultures is an Fc-fusion protein in which the Fc region of antibody and CTLA4 are fused. CLTA4-Ig is a glycoprotein with two N-glycans in the CTLA4 region and one N-glycan in the Fc region. Although CLTA4-Ig has an increased in vivo half-life through fusion with the Fc region, sialic acid is not added to the two N-glycans attached to CTLA4 which results in short in vivo half-life. In this study, genetic engineering, media additives, and inhibition of apoptosis were applied to increase the sialic acid of CTLA4-Ig produced in CHO cells. N-glycans located in CTLA4 was mainly formed with three antennary structures. To determine which GnT is the major enzyme in forming tri-antennary structure, GnT-IV and GnT-V were transfected into CHO cells. And, glycoengineered cell lines were developed with co-overexpression of ɑ2,6-sialyltransferase (ɑ2,6-ST), thereby producing CTLA4-with increased antennary structure and sialylated N-glycan. Comparing GnT-IV and GnT-V overexpressed cell lines, GnT-IV overexpressed cell lines showed the highest tri-antennary structure. In addition, the highest sialic acid content was obtained by co-overexpression of GnT- IV and ɑ2,6-ST simultaneously. These results showed that co-overexpression of GnT-IV and ɑ2,6-ST is an effective strategy to increase the sialic acid and tri-antennary structure of CTLA4-Ig. N-acetylmannosamine (ManNAc) was added to provide a donor substrate to enhance sialyation during glycoengineered cell line culture. As 20 mM ManNAc was added in glycoengineered cell line, it was confirmed that sialic acid was increased by 1.5-fold compared to the wild type cell line without ManNAc. In addition, combining addition of ManNAc with glycoengineering showed synergistic effect on the increase in sialic acid content of glycoprotein. Finally, suramin, which had positive effects on CHO cell cultures, was investigated. Addition of suramin caused an increase in viable cell density, cell viability, and titer of the CTLA4-Ig. Moreover, suramin had no impact on protein aggregation and enhanced the sialic acid contents of Fc-fusion protein by 1.15-fold. The enhanced sialylation was not caused by the increased nucleotide sugar level but by the inhibition of sialidase activity. These results showed that suramin inhibited apoptosis and had positive impacts on the productivity and quality of Fc-fusion protein. In conclusion, glycoengineering technology was applied to select efficient genes to increase sialic acid and antennarity in CHO cells producing CTLA4-Ig. In addition, ManNAc was added during the culture to increase CMP-sialic acid, which acts as a rate-limiting step. Finally, suramin was added to reduce sialidase activity released by apoptosis in culture, thereby effectively inhibiting sialic acid degradation. These strategies will contribute to produce CTLA4-Ig with highly sialylated N-glycan.

    • 형질전환된 Chinese hamster ovary 세포에서 silkworm gland hydrolysate를 이용한 albumin-erythropoietin의 생산성과 세포사멸 확인

      형질전환된 Chinese hamster ovary(CHO) 세포는 포유동물 세포를 숙주로 하는 재조합 단백질의 상업적인 생산에 있어서 가장 널리 사용되는 세포주이다. CHO 세포는 증폭 시스템으로 dihydrofolate reductase(DHFR) 유전자와 methotrexate(MTX)를 통해 재조합 단백질을 높은 수준으로 발현할 수 있다. 또한 복잡한 단백질의 경우 요구되는 효율적인 post-translational modification(PTM) 과정을 갖추고 있고, 인간에서 질병을 일으키는 polio, HIV, hepatitis B 등은 CHO 세포에서 증식을 하지 않는다. 하지만 이러한 CHO 세포는 미생물에 비하여 낮은 단백질의 생산성이 있다. 최근의 CHO 세포 배양에서 다양한 전략으로 목적 단백질의 specific productivity를 증가시키는 연구가 진행되고 있다. Silkworm Gland Hydrolysate(SGH)은 silkworm hemolymph(SH)에서 유래된 섬유 단백질이다. 항산화제인 SGH는 포유동물 세포 배양에서 세포 증식과 재조합 단백질의 생산량을 증가시키는 영양소로 사용되고 있다. 더욱이 줄기세포와 관련된 많은 참고 문헌에는 세포 성장 측면에서 fetal bovine serum(FBS)과 유사한 효능을 보이고 있는 것을 확인할 수 있다. Erythropoietin(EPO)는 세계적으로 연간 10 달라(10억불) 이상의 시장규모를 형성하는 블록버스터 제품이다. Human EPO는 3개의 N-linked와 1개의 O-linked oligosaccharide chain으로 형성된 당단백질로서 asialo-EPO가 간에 있는 hepatic asialoglycoprotein receptor(ASGPR)에 반응하면 체내에서 빠르게 제거되기 때문에 sialic acid가 체내의 생물학적인 활성에 중요한 작용을 한다. 최근에 짧은 반감기를 가지는 peptide의 albumin 융합을 통해 체내 순환을 오래 유지할 수 있는 기술이 사용되고 있다. 이 기술을 이용한 albumin-EPO(Alb-EPO)는 human EPO에 albumin을 유전적으로 형질전환 시킨 100 kDa의 단백질로 체내 반감기의 증가와 향상된 약물 전달력 그리고 1회 투여시 요구되는 약물 감소와 같은 장점을 가지고 있다. 본 연구에서는 Alb-EPO를 생산하는 형질전환된 CHO 세포 배양에서 SGH 첨가에 따른 생산성의 효과를 확인하고자 하였다. 그 결과 CHO 세포의 viable cell density와 생존도가 증가하였으며 Alb-EPO의 생산량도 현저하게 증가하였다. 또한 CHO 세포배양에서 사용되는 세포사 저해 물질인 dextran sulfate와 N-acetylcystein과 비교하여도 SGH를 첨가하였을 때 가장 높은 세포 생존도를 보였다. 결론적으로 SGH를 CHO 세포에 첨가해서 세포의 생존도가 증가하였으며 그로 인해, 목적 단백질인 Alb-EPO의 생산량이 증가됨을 확인할 수 있었다.

    • 재조합 CHO 세포 무혈청 현탁 배양에 의한 헌터증후군 효소치료제 Iduronate 2-sulphatase의 생산성 향상에 관한 연구

      Iduronate 2-sulphatase(IDS)는 인체의 결체조직을 만드는 구성 물질 중의 하나인 mucopolysaccharides heparan sulfate (HS)와 dermatan sulfate (DS)의 분해에 관련된 lysosomal enzyme 중의 하나이다. IDS 효소 활성의 유전적 결핍은 Hunter syndrome으로 알려진 mucopolysaccharidosis type II(MPSⅡ, Hunter syndrome)를 초래하고 성염색체 열성 유전양식을 갖는다. 결핍된 IDS에 의해 불완전 분해 된 mucopolysaccharide가 세포내의 lysozome에 축적되고 소변으로 과도하게 배설되면서 각 장기의 기능 장애를 초래하게 되고, 이에 따라 점차적으로 신경학적, 육체적인 퇴행을 보이게 된다. 재조합 단백질의 산업적 수준의 생산을 위해 고생산성 세포의 확립과 배양공정의 최적화가 요구된다. 최근 대량생산을 위해 현탁 배양 공정을 많이 사용하고 있다. 따라서 본 연구에서는 Hunter 증후군 치료 방법 중의 하나인 효소 보충법을 위해 IDS의 대량생산을 목적으로 IDS 생산성 향상을 위해 상용되고 있는 무혈청 배지에 재조합 CHO 세포를 적응 시켰고 그중 세포 성장과 IDS 생산성이 가장 뛰어난 배지를 선택하였다. 재조합 단백질의 생산성을 높이기 위한 또 다른 접근 방법으로 저온 배양을 수행하였다. 온도는 세포의 성장과 재조합 단백질 생산에 영향을 주는 중요한 인자로 CHO세포를 포함한 대부분의 동물세포는 37℃에서 주로 배양 하지만 본 연구에선 재조합 단백질 생산성 향상을 위해 32℃의 낮은 온도에서 재조합 CHO 세포를 현탁 배양하였다. 또, anti apoptotic agents로 STR-G를 배양하는 동안 첨가하여 세포사멸을 억제함으로써 CHO 세포 성장과 IDS 생산성을 증진시키고자 하였다. 대량 배양을 위해 shaker flask에서 시간별 배지교환으로 semi-perfusion을 수행하였고 재조합 CHO 세포의 IDS 생산성을 향상시켰다. Iduronate 2-sulphatase (IDS) is a lysosomal enzyme involved in the sequential degradation of the glycosaminoglycans heparan sulphate and dermatan sulphate. Mucopolysaccharidosis type II (MPSII, Hunter syndrome) is an X-linked, recessively inherited lysosomal storage disorder that caused by a deficiency of iduronate 2-sulphatase. This X-linked inheritance pattern results in a skewed prevalence in males (predominant) compared to females (very rare). Thus industrial production of IDS is required for the treatment of Hunter syndrome. The establishment of high productivity cell line and the optimization of culture condition is prerequisite to develop an efficient animal cell culture process producing IDS by recombinant CHO cells. Suspension culture is preferred for its simplicity and scalability. Thus it is current trend to employ suspension culture in serum free media in large scale animal cell culture process. The recombinant CHO cells were adapted to grow in suspension in three commercial serum-free medium, and an optimum serum-free medium was selected in terms of cell density and productivity. The optimum operating temperature profile and the effect of adding apoptosis inhibitor were also investigated to enhance IDS productivity. Optimum medium change strategy were investigated to operate long-term culture of recombinant CHO cells. For the large scale industrial processes, a high cell density serum-free suspension culture system is required for enhancing the productivity and yield of culture. Therefore, it was shown in this study that a suspension adaptation of anchorage dependent cell and semi-perfusion culture system for the bioreactor process could be applicable to large scale cell culture system.

    • Re-adaptation of r-CHO-S Cells to Adhesive State Enhances Long Term Cell Viability and Protein Productivity

      Recombinant Chinese hamster ovary (rCHO) cells are widely used in the industry for the production of recombinant therapeutic proteins because of their characteristics: rapid growth rate, high productivity of recombinant protein, and high stability of foreign gene expression. CHO cells are originally anchorage-dependent cells, which mean that cell adhesion is necessary for maintaining cell survival, proliferation, and functions. Three dimensional suspension culture is, however, preferred for the mass production of recombinant proteins because it can increase the surface area for cell survival, so CHO cells have been adapted to grow in suspension to overcome the problem of two dimensional culture. In this study, r-CHO-S cells were forced to be re-adapted to adhesive state and the changes while cells were being re-adapted were investigated. r-CHO-S cell transfected with human EPO gene was used as a model cell line and fetal bovine serum (FBS) was used to supply the adhesion molecules for the attachment of the cells to the layer. The result showed that cells in re-adaptation process proliferated better and survived longer than the normal cells during whole culture period. The interesting fact is that almost every cell which survived longer was anchorage-dependant cells. For these adhesive cells, PLGA microspheres were fabricated to provide enlarged surface area. The result showed the enhanced effect of microspheres on prolonged cell viability mostly maintained by adhesive cells. EPO protein productivities of the groups in re-adaptation process also showed higher figures than the control group. FACS data confirmed that cells in re-adaptation process showed continued appearance of higher G1 phase distribution, which means that cells produced proteins vigorously. All of these results indicate that re-adapting cells to adhesive states can enhance the long term cell viability and recombinant protein productivity.

    • Characterization of Therapeutic Protein Capabilities in GS/DHFR dKO CHO-K1 Cells

      The Chinese hamster ovary (CHO) cell line is one of the most widely used mammalian hosts for the production of therapeutic recombinant proteins. With the continuous expansion of the global biopharmaceutical market, there is a growing demand for enhanced productivity in CHO cell-based expression systems. Knockout (KO) CHO-K1 cell lines targeting glutamine synthetase (GS) and dihydrofolate reductase (DHFR), along with gene amplification strategies, have contributed to improving the stability and efficiency of therapeutic protein production. However, the efficient isolation of high-producing clones from large cell populations remains a major challenge. In this study, we transfected bevacizumab and omalizumab expression vectors into suspension-adapted GS KO, DHFR KO, and GS/DHFR double knockout (dKO) CHO-K1 cell lines established using CRISPR/Cas9 technology. In both antibody systems, the GS/DHFR dKO cell line consistently showed higher monoclonal antibody (mAb) productivity than the single KO cell lines. In particular, when expressing bevacizumab, the antibody titers were quantitatively compared under methionine sulfoximine (MSX) and methotrexate (MTX) selection pressures. The GS/DHFR dKO cells exhibited superior titers compared to GS KO cells under 10 μM MSX treatment and maintained higher productivity than DHFR KO cells following MTX adaptation. Furthermore, when treated with 30 μM MSX and 20 nM MTX simultaneously, and subsequently adapted to 200 nM MTX in a stepwise manner, the dKO cells continuously maintained high productivity. These findings suggest that dual amplification using the GS/MSX and DHFR/MTX systems can effectively enhance transgene expression. To evaluate their potential as production cell lines, we performed single cell cloning (SCC) on the GS/DHFR dKO cells and selected high-producing clones, which achieved up to 660 mg/L of antibody titer in fed-batch culture. These results demonstrate that the GS/DHFR dKO CHO-K1 cell line is a promising alternative platform capable of replacing conventional commercial cell lines for the efficient production of therapeutic antibodies. 중국 햄스터 난소(Chinese hamster ovary; CHO) 세포주는 치료용 재조합 단백질 생산에 가장 널리 사용되는 포유류 세포주이다. 전 세계 항체의약품 시장이 지속적으로 확대됨에 따라, 이에 대응하기 위한 CHO 세포주의 생산성 향상에 대한 수요도 증가하고 있다. 글루타민 합성효소(GS) 및 디하이드로폴레이트 환원효소(DHFR) 유전자를 표적으로 한 CHO-K1 결손(KO) 세포주의 개발과 유전자 증폭 전략은 치료용 단백질 생산의 안정성과 효율성 향상에 기여해왔다. 그러나 대규모 세포 집단에서 고생산 클론을 효율적으로 선별하는 일은 여전히 중요한 과제로 남아 있다. 본 연구에서는 CRISPR/Cas9 기술을 활용해 확립한 GS 결손, DHFR 결손, GS/DHFR 이중 결손의 부유배양(suspension) CHO-K1 세포주에 베바시주맙(bevacizumab) 및 오말리주맙(omalizumab) 발현 벡터를 형질전환하였다. 두 항체 시스템 모두에서 GS/DHFR 이중 결손 세포주는 단일 결손 세포주에 비해 일관되게 높은 단일클론 항체(monoclonal antibody, mAb) 생산성을 보였다. 특히 베바시주맙을 발현한 경우, 메티오닌 설포시민(MSX) 및 메토트렉세이트(MTX) 선택압 조건에서 항체 생산성을 정량적으로 비교하였다. 그 결과, GS/DHFR 이중 결손 세포주는 GS 결손 세포주 대비 MSX 10 μM 처리 시 더 높은 역가를 나타냈으며, DHFR 결손 세포주보다 MTX 적응 후에도 우수한 생산성을 유지하였다. MSX 30 μM 및 MTX 20 nM을 병용 처리한 후 MTX 농도를 200 nM까지 단계적으로 증가시킨 조건에서도 높은 생산성을 지속적으로 유지하였다. 이는 GS/MSX 및 DHFR/MTX 기반의 이중 증폭 시스템이 유전자 발현을 효과적으로 증진시킬 수 있음을 시사한다. 또한 생산세포주로서의 가능성을 평가하기 위해, GS/DHFR 이중 결손 세포주에 단일 세포 복제(single cell cloning; SCC)를 수행하여 고생산 클론을 확보하였으며, fed-batch 배양을 통해 최대 660 mg/L의 항체 역가를 달성하였다. 이러한 결과는 GS/DHFR 이중 결손 CHO-K1 세포주가 기존 상업용 세포주를 대체할 수 있는 유망한 생산 플랫폼임을 보여준다.

    • Research for the Production of Therapeutic Protein using GS/DHFR Double Knockout CHO Cell Line

      중국 햄스터 난소(Chinses hamster ovary; CHO) 세포는 단일클론 항체(mAb)를 포함한 치료용 단백질 생산에 가장 널리 사용되는 산업용 세포주이다. 글루타민 합성 효소(Glutamine synthetase; GS) 와 다이하이드로폴레이트 환원 효소(Dihydrofolate reductase; DHFR)의 녹아웃을 이용한 세포주 개발 및 유전자 증폭 기술은 치료용 단백질 생산의 안정성, 생산성 및 시간 절약에 많은 기여를 하였다. 하지만, 수 많은 단일클론 중에서 고생산성 세포주를 선별하는 것은 많은 시간이 소요될 뿐만 아니라, 매우 어려운 일이다. 현재 보편적으로 사용되는 GS 녹아웃, DHFR 녹아웃 CHO-K1 세포주를 Clustered Regularly Interspaced Short Palindromic Repeats/Cas9(CRISPR/Cas9) 유전자 편집기술을 이용하여 자체 개발하였으며, 나아가 CRISPR/Cas9 유전자 편집기술을 이용하여 GS/DHFR 이중 녹아웃 CHO-K1 세포주를 최초로 확립하였다. 본 연구에서는 각각의 세포주 특징을 분석하고, 치료용 항체 발현 벡터를 도입하여 세포주 간에 항체 발현율을 비교 분석하였다. 결과적으로 GS/DHFR 이중 녹아웃 CHO-K1 세포주는 치료용 항체 발현을 위한 벡터 2종을 동시에 도입할 수 있었으며, 글루타민(Gln) 과 히포크산틴, 티민(HT)를 이용한 이중 선별과정을 통해 각각의 벡터가 안정적으로 삽입된 생산 세포주 후보군을 빠르게 구축할 수 있었다. 항체 발현 역가(力價)를 분석한 결과, GS/DHFR 이중 녹아웃 세포주가 CHO-K1 일반형, GS 녹아웃, DHFR 녹아웃 CHO-K1 세포주 보다 뚜렷하게 높은 항체 생산성을 가지는 것을 확인하였다. GS 녹아웃 및 DHFR 녹아웃 CHO-K1 세포주는 GS 코딩영역(엑손 4)와 DHFR 코딩 영역(엑손1)을 각각 표적하는 CRISPR/Cas9 유전자 편집 기술을 이용하였고, 다종(6종)의 후보군의 형태학적 특징 및 성장률을 분석하여, CHO-K1 일반형과 유사한 단일클론 녹아웃 세포주를 확립하였다. GS/DHFR 이중 녹아웃 CHO-K1 세포주는 기확립된 DHFR 녹아웃 CHO-K1 세포주에 GS 코딩영역(엑손4)를 표적하는 CRISPR/Cas9 유전자 편집 기술을 추가로 사용하여 확립하였다. 이를 통해, GS/DHFR 이중 녹아웃 CHO-K1 단일클론 후보군(6종)를 선별하였으며, 형태학적 특징 및 성장률이 CHO-K1 일반형과 유사한 단일클론 녹아웃 세포주를 확립하였다. 치료 단백질의 대량 생산을 위해선 부착 세포를 무혈청 배지에서 부유 세포로 적응시키는 것이 필수 불가결하다. 따라서, GS 녹아웃, DHFR 녹아웃, GS/DHFR 이중 녹아웃 CHO-K1 부착 세포를 무혈청배지에서 부유 배양 적응을 통해 부유 세포로 적응 시켰다. GS 녹아웃, DHFR 녹아웃, GS/DHFR 이중 녹아웃 CHO-K1 세포주는 형태학적 차이 없이 CHO-K1 일반형과 유사한 성장률을 가진 것을 확인하였다. 또한, 글루타민(Gln) 및/또는 히포크산틴 과 티민(HT)의 유무에 따른 세포주의 성장이 명확하게 다른 것을 정치 배양 및 부유 배양에서 확인하였다. 해당 연구를 통해, 세포주의 염색체 내부에 안정적으로 삽입된 치료용 항체 발현 벡터의 카피 수가 항체 생산성과 상관관계가 있음을 확인하였다. GS/DHFR 이중 녹아웃 CHO-K1 세포주는 GS 녹아웃 세포주가 사용하는 유전자 증폭시스템인 메티오닌 설폭시민(MSX)과 DHFR 녹아웃 세포주가 사용하는 유전자 증폭시스템인 메토트렉세이트 (MTX)를 모두 사용할 수 있기 때문에 치료 단백질의 생산량 차이가 단일 녹아웃 CHO-K1 세포주와 더 커질 것으로 보인다. 이를 통해, GS/DHFR 이중 녹아웃 CHO-K1 세포주가 기존에 상용화된 단일 녹아웃 CHO-K1 세포주를 대체할 새로운 고생산성 세포주가 될 것으로 기대한다 Chinese hamster ovary (CHO) cells are the most widely used industrial host for the manufacture of therapeutic proteins, including monoclonal antibodies (mAb). The development of CHO-K1 knockout (KO) cell lines for glutamine synthetase (GS) and dihydrofolate reductase (DHFR) and gene amplification technology improved the stability, productivity, and time-saving of the production of therapeutic proteins. However, it is a great challenge to select only the high-producing clones among numerous clones. We developed CHO-K1-based GS KO, and DHFR KO cell lines using Clustered Regularly interspaced Short Palindromic Repeats (CRISPR)/Cas9 gene-editing system, and furthermore, we first developed the GS/DHFR double knockout (dKO) cell line with CHO-K1 using CRISPR/Cas9 gene editing system. The auxotroph characteristics of each cell line were analyzed, and a comparison of antibody titer with each cell line was conducted after the transfection of the therapeutic protein expression vector. As a result, we were able to construct a stable GS/DHFR dKO transfected pool by simultaneous transfection with two vectors (GS and DHFR co-expressing therapeutic protein expression vector) with a double selection process using glutamine (Gln), and hypoxanthine and thymine (HT). Through these experiments, we confirmed that the GS/DHFR dKO transfected stable pool had significantly higher antibody productivity than CHO-K1 WT, GS KO, and DHFR KO transfected stable pool. GS KO and DHFR KO cell lines were developed using the CRISPR/Cas9 system targeting the DHFR coding region (exon 1), and GS coding region (exon 4). GS KO and DHFR KO top clones were selected by analyzing morphological differences and the growth rate of 6 different single clones. GS/DHFR dKO cell line was developed using CRISPR/Cas9 gene-editing techniques targeting GS coding region (exon 4) in a pre-established DHFR KO cell line. GS/DHFR dKO top clone was selected by analyzing morphological differences and growth rate of 6 different single clones. In order to mass-produce therapeutic proteins, it is essential to adapt attached cells to suspension cells with serum-free media. Therefore, adherent GS KO, DHFR KO, and GS/DHFR dKO cells were adapted to suspension cells through serum-free media suspension adaptation. All suspension-adapted cell lines were showing similar growth rates compared to CHO-K1 WT with no morphological differences. And, most importantly, glutamine (Gln) and/or hypoxanthine and thymidine (HT) auxotroph phenotype in GS/DHFR dKO cell line was clearly identified in static and suspension cultures compared to CHO-K1 WT. In this study, we found that copy number of therapeutic protein genes correlates with therapeutic protein production. GS/DHFR dKO cell line is able to use both Methionine sulfoximine (MSX) and Methotrexate (MTX) gene amplification system; GS/MSX-mediated gene amplification system, and DHFR/MTX-mediated gene amplification system. By using the GS/DHFR dKO cell line, it is expected that the difference in the productivity of therapeutic proteins will be greater than that of GS KO, and DHFR KO cell lines when using a gene amplification system. Through this, we anticipate our GS/DHFR dKO cell line will be a new high-productivity cell line to replace the previously commercialized therapeutic protein production cell lines.

    • Development of Chemically Defined Media for CHO Cell Culture

      For decades, Chinese hamster ovary (CHO) cells have been the most widely used mammalian host for large-scale commercial production of therapeutic proteins. Development and optimization of cell culture media is a crucial step for higher growth and productivity in CHO cell-based manufacturing process. In the industrial market, the demand for chemically defined media(CDM) is increasing due to good reproducibility and understanding of components. In this study, we showed the initial process to develop CDM through optimization of the composition, and screening key components on Plackett-Burman design(PBD). First, we optimized the concentration of amino acids and energy sources in SJ-PFM 2.0, an in-house medium, by comparing to commercial media. Based on this medium, a chemically defined medium was developed. Then, Plackett-Burman design was adopted to screen significant component and to attain higher growth and productivity. Finally, we found that vitamin B and polyamine groups were the most critical factor for growth and production for CHO cell culture. With the optimization of concentration and culture process of found key factors, the results showed that IVCD was 3.3-fold and IFN-β productivity was 6.3-fold improvement over the control culture. In conclusion, these results suggested that optimization of glucose and amino acids was crucial for developing chemically defined media. Moreover, PBD was an efficient way for screening effective composition to develop CDM in CHO cells. 수십년 동안 CHO세포는 치료용 단백질의 대규모 상업적 생산에 가장 널리 사용되는 포유류 숙주였다. 세포 배양 배지의 개발 및 최적화는 CHO세포 기반 제조공정에서 더 높은 성장과 생산성을 위한 중요한 단계이다. 산업 시장에서는 우수한 재현성과 구성 요소에 대한 이해로 인해 화학적 정의 배지(CDM)에 대한 수요가 증가하고 있다. 본 연구에서는 성분 최적화 및 플라켓-버만 설계 (PBD)의 핵심 구성 요소 스크리닝을 통해 CDM을 개발하는 초기 프로세스를 보여주었다. 먼저 자체 제작 배지인 SJ-PFM 2.0의 아미노산과 에너지원 농도를 상용 배지와 비교하여 최적화하였다. 이 배지를 기반으로 화학적으로 정의된 배지를 개발했고, 더 높은 성장과 생산성을 위한 성분을 선별하기 위해 플라켓-버만 설계를 사용했다. 비타민 B와 폴리아민은 CHO세포 배양을 위한 CDM의 성장과 생산에 가장 중요한 요소였다. 비타민 B 군과 폴리아민 군이 CHO 세포 배양의 성장과 생산에 가장 중요한 요소임을 발견했다. 발견된 주요 인자의 농도 및 배양 과정을 최적화하여 IVCD는 3.3배, IFN-β 생산성은 대조군 배양에 비해 6.3배 향상되었다. 결론적으로, 이러한 결과는 글루코즈와 아미노산의 최적화가 화학적으로 정의된 배지를 개발하는 데 중요하다는 것을 시사한다. 더욱이, PBD를 수행하는 것은 CHO 세포를 위한 화학적으로 정의된 배지에서 조성물을 스크리닝하는 효율적인 방법이다.

    • The optimization of serum-free medium for CHO-S cells

      Stimulating cell growth and proliferation is vital for design of serum-free animal cell culture medium. In most of the serum-free animal cell culture media, insulin-like growth factor-1 (IGF-1) is being used as a growth factor to stimulate the cell growth and proliferation in place of serum. LongR3IGF-1 has been specifically engineered for the enhancement of cell culture performance replacing the use of IGF-1. First, in this study, longR3 IGF-1 and the lipid signaling molecule lysophosphatidic acid (LPA) were evaluated as growth factors in CHO cell cultures. Second, pre formulated serum-free in-house medium (SJ-PFM2.0) optimized to CHO-DG44 cells. The CHO-S cell line is now widely used in cell engineering. Optimization of each components by examining CHO-S cells growth. Based on these results, newly fortified SJ-PFM2.0 and addition of longR3 IGF-1 and LPA enhance cell growth of CHO-S cells. The findings made from this study can be applied to the development of serum-free media including antibodies, protein drugs, therapeutic cells, vaccines and gene therapy products particularly based on the animal cell technology.

    • A study of fed-batch culture and glycosylation analysis for CHO-DG44 cells producing IgG

      The aim of the cell culture process is to achieve an increase in volumetric productivity by increasing the viable cell density, cell-specific productivity, cell culture duration, and bioreactor utilization in manufacturing. The fed-batch process is commonly used to produce recombinant proteins, such as monoclonal antibody from Chinese hamster ovary (CHO) cells, owing to its simplicity of operation and higher yield, as compared with the batch and perfusion culture methods. In this study, the fed-batch culture conditions of CHO-DG44 producing therapeutic chimeric IgG antibody for leukemia were optimized to enhance the volumetric productivity during the cell culture process. First, a commercial chemically defined media screening experiment suitable for CHO-DG44 was performed in a shake flask, and PowerCHO™ 2 was selected. Then, the fed-batch conditions in the shake flask were optimized through a major energy source feeding experiment, feed medium concentration optimization, and hypothermic temperature optimization. Compared with the batch culture, the peak viable cell density (PVCD) increased by a factor of 2.3, the cell culture duration was extended by 8 days, and the final antibody concentration increased by a factor of 6.1. Based on the optimized process conditions obtained from the previous flask experiment, a scale-up experiment was performed in a stirred tank bioreactor (STR) and an orbitally shaken bioreactor (OSR) equipped with a 3D square-shaped container. Consequently, OSR yielded a 1.5-fold increase in PVCD over that for STR, and a cell culture duration of 4 days longer. Furthermore, the final antibody concentration for OSR was 2.1-fold higher than that for STR. Therefore, OSR was confirmed to be more suitable for CHO-DG44 than STR. Finally, the glycosylation of IgG in the fed-batch process was compared between using the flask and using the bioreactors. The glycosylation of IgG for OSR was comparable to that for the flask. Taken together, this fed-batch culture study reveals that it is possible to increase volumetric productivity using OSR while maintaining IgG quality. 세포 배양 공정은 일반적으로 생존 세포 농도 (Viable cell density), 세포 비 생산성 (Cell specific productivity), 세포 배양 기간을 향상 시키고 세포배양 시 바이오리액터 활용도를 증가시켜 부피 생산성 (Volumetric productivity)을 높이는 것을 목표로 한다. 유가식 배양 공정은 회분식 배양과 관류식 배양 방법에 비해 배양방법이 단순하고 수율이 높기 때문에 Chinese hamster ovary (CHO) 세포에서 재조합 항체와 같은 바이오의약품을 생산하는 데 일반적으로 사용된다. 본 연구에서는 급성 백혈병 치료용 항체를 생산하는 CHO-DG44 세포주의 생산성을 향상시키기 위해 유가식 배양 조건을 최적화했다. 우선, 본 세포주에 적합한 화학적으로 정의된 상용화 배지 (Chemical-defined media) 스크리닝 실험을 수행하였고 PowerCHO™ 2를 최적배지를 선택했다. 이 배지를 바탕으로 주요 에너지원 공급 실험, 고농축 배지 공급 농도 최적화 그리고 저온배양 온도 최적화를 수행하였다. 그 결과 회분식 배양에 비해 최고 세포 농도 (peak viable cell density)는 2.3배 증가했고 세포 배양 기간은 8일 연장되었으며 최종 항체 농도는 6.1배 증가하였다. 이를 통하여 얻은 최적화된 공정 조건을 기반으로 교반 탱크 바이오리액터(Stirred tank bioreactor, STR) 와 3D 사각형 일회용 백이 장착된 궤도형 쉐이킹 바이오리액터(Orbitally shaken bioreactor, OSR)에서 스케일 업 실험을 수행하였다. OSR에서 세포배양을 진행하였을 때, STR에서 세포배양을 진행한 것 보다 최고 세포 농도가 1.5배 더 높았고, 세포 배양 기간은 4일 더 길었으며 최종 항체 농도는 2.1배 더 높다는 것을 확인하였다. 따라서 STR보다 OSR이 본 세포주에 더 적합한 바이오리액터임을 확인할 수 있었다. 최종적으로, 플라스크에서 진행한 유가식 배양과 바이오리액터에서 진행한 유가식 배양에서 얻은 재조합 항체의 당사슬을 분석하였고 STR보다 OSR에서 플라스크와 더 유사한 당 사슬을 갖는다는 것을 확인하였다. 결론적으로 유가식 배양 연구를 통해 높은 부피 생산성을 얻음과 동시에 당사슬을 유지하여 항체의 품질을 확인할 수 있었다.
