전세계적으로 가장 유명한 피부병인 아토피피부염은 면역부작용과 피부장벽 장애의 특징을 가지고 있다. 최근 논문에 의하면 12 리폭시게나아제가 피부장벽기능장애를 유발하는 중요한 요인으로 밝혀졌다. 아라키돈산으로부터 유래된 12 hydroxyeicosatetraenoic acid (HETE)는 피부염증의 중요한 지질 매개인자로 알려져 있으며 아토피피부염에서 T 림프구의 활동을 조절하고 있다. 여러 편의 논문이 아토피피부염과 면역기능장애의 중요한 관계에 대해서 발표하였음에도 불구하고, 아직 아토피피부염에서의 HETE의 역할이 밝혀지지 않았다. 이 실험의 목적은1-chloro-2,4-dinitrochlorobenzene (DNCB)로 아토피피부염을 유발시킨 BALB/c 마우스 모델의 생체시료에서 12 HETE 거울상이성질체를 키랄 액체 크로마토그래프 탠덤 질량분석기를 이용하여 분리하고 정량화 시키는 것이다. BALB/c 마우스에 AOO (Acetone/Olive oil)이나 DNCB를 일정한 간격으로 도포한 후 혈장, 피부, 비장, 림프노드를 분리하여 키랄 액체 크로마토그래프 탠덤 질량분석기를 이용하여 12 HETE를 정량화하였다. 반복적인 DNCB 투여로 DCNB 투여군은 인간의 아토피 피부염과 비슷한 임상적 증상 (피부건조, 홍반, 부종, 미란)을 보였으며 조직학적으로는 상피층이 두꺼워지고 과각화증과 염증세포침윤 등을 관찰할수 있었다. 혈청 IgE 또한 Control 군에 비해 DNCB 투여군 에서 확실한 증가가 관찰되었다. 아토피피부염이 유발된 DNCB 투여군의 생체시료에서 모든 12(R), (S) HETE의 현저한 증가가 관찰되었으며, 이것으로 인하여 12(R), (S) HETE가 아토피피부염의 발생기전과 밀접한 관련이 있는 것을 확인할 수 있었다. 본 연구에서 얻어진 결과는 HETE와 아라키돈산이 아토피피부염 발병기전과 관련이 있으며 12 HETE 정량화를 통하여 아토피피부염의 치료 연구에도 도움이 될 것이라 사료된다 Atopic dermatitis, one of the most important skin diseases, is characterized by both skin barrier impairment and immunological abnormalities. Recent studies indicate that 12 lipoxygenases (Lox) play a crucial role in skin barrier dysfunction.12-Hydroxyeicosatetraenoic acid(HETE), derived from arachidonic acid, is a potent lipid mediator of skin inflammation and a modulator of T-lymphocyte activity in atopic dermatitis. Although several studies have shown a significant relationship between atopic dermatitis and immunological abnormalities, the role of HETE in atopic dermatitis remains unknown. The objective of this study was to develop chiral methods to characterize 12-HETE enantiomers in 1-chloro-2,4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis mouse model and to evaluate the effects of 12-HETE on atopic dermatitis. In this study, BALB/c mice were applied with either DNCB or acetone/olive oil (AOO) to induce atopic dermatitis. 12(R) and (S)-HETEs in plasma, skin, spleen, lymph nodes were quantified by chiral liquid chromatography-tandem mass spectrometry. Repeated application of the DNCB model showed clinical (skin dryness, erythema, edema, and erosion) and histological symptoms (epidermal thickening, hyperkeratosis, and inflammatory cell infiltration) that were very similar to those seen in human atopic dermatitis. Serum IgE levels were elevated in this model compared to those in the control group. 12(R) and (S)-HETEs in biological samples of DNCB-induced atopic dermatitis mice also increased significantly compared to those in the AOO group, reflecting the involvement of 12(R) and (S)-HETEs in atopic dermatitis. These results are useful to understand the pathogenesis of atopic dermatitis as it relates to HETE and arachidonic acid.
Effect of gut microbiota on the development of atopic dermatitis in NC/Nga mice
Recent researches on gut microbiome provided better understanding of relationship between gut microbiota and host pathology, including atopic dermatitis, of which occurrence and development are known to be associated with gut microbiota but not characterized so far. To identify intestinal microorganisms related with atopic dermatitis, specific antibiotic cocktails were developed to change the two major phylotypes of gut microbiota, of which composition was examined by pyrosequencing using 16S rRNA gene tags. NC/Nga mice, model animal for atopic dermatitis, were administered two different antibiotic cocktails targeting either Gram negative (GNA) or Gram positive (GPA) bacteria by either ad libitum drinking water or oral gavage for 6 weeks. The GPA group showed an increased serum IgE level accompanied with more severe skin damage compared to the control and GNA groups. Bacterial community analysis data also revealed dramatic changes in the gut microbiota composition of the GPA group with a significant increase in abundance of the species Enterobacter cowanii and Enterobacter hormaechei belonging to the Proteobacteria. The involvement of the two bacteria in the pathology of atopic dermatitis was tested by oral administration to NC/Nga mice. Only the mouse group colonized with Enterobacter hormaechei showed an aggravation of skin damage and an increased serum IgE level. Our data suggest that enterobacteria, Enterobacter hormaechei, may play an important role in development and severity of atopic dermatitis in NC/Nga mice

Analysis of Colonization and Genotyping of Exotoxin of Staphylococcus aureus in Patients with Atopic Dermatitis Background: The skin of atopic dermatitis patients has a high susceptibility to Staphylococcus aureus and toxins produced by S. aureus may aggravate the atopic dermatitis by acting as superantigens. Objectives: Purpose of this study was to evaluate the relationship of skin barrier function, colonization of S. aureus and clinical severity in atopic dermatitis. We also examined the predominant toxin genes produced in patients with atopic dermatitis. Methods: Thirty-nine patients with atopic dermatitis were evaluated for the clinical severity and skin barrier function by using SCORAD(Severity Scoring in Atopic Dermatitis) index and TEWL(transepidermal water loss). S. aureus was isolated from forearm, popliteal fossa and anterior nares of atopic dermatitis patients, and toxin genes were analyzed by using multiplex PCR(polymerase chain reaction). Results: TEWL showed a statistically significant correlation with clinical severity in patients with atopic dermatitis(p <0.05). TEWL was correlated with number of S. aureus colonization sites and nasal colonization but these results were not statistically significant. Of the 39 patients, S. aureus strains was isolated in 48.7%. Among them, the predominant toxin gene was sea only, which was produced in 52.6% and followings were coexistence of sea and tsst-1(42.1%) and tsst-1 only(5.3%). Conclusions: Skin barrier function, measured by TEWL revealed a statistically significant correlation with clinical severity in pateints with atopic dermatitis. The most common toxin gene was sea in Korean atopic dermatitis patients and this might be an important role in the pathogenesis of atopic dermatitis. 아토피피부염 환자에서 황색 포도알균의 배양과 독소 유전자 분석 배경 : 아토피피부염 환자의 피부는 피부 장벽기능의 이상으로 인해 황색 포도알균에 대해 높은 감수성을 가지며 황색 포도알균에 의해 분비되는 독소가 초항원으로 작용하여 아토피피부염을 악화시키는 것으로 알려져 있다. 목적 : 본 연구는 아토피피부염에서 피부 장벽기능과 황색 포도알균의 집락 형성과 임상적 중증도와의 관련성을 확인하는 것이다. 또한 아토피피부염에서 가장 흔히 연관되는 독소 유전자를 분석하고자 하였다. 방법 : 39명의 아토피피부염 환자를 모집하여 경표피 수분 손실량과 SCORAD (Severity Scoring in Atopic Dermatitis) index를 이용하여 피부 장벽기능과 임상적 중증도를 평가하였다. 아토피피부염 환자의 아래팔과 다리의 굽힘쪽, 그리고 코점막으로부터 황색 포도알균을 배양하였고 multiplex PCR을 통해 독소 유전자를 분석하였다. 결과 : 아토피피부염 환자의 중증도에 따른 경표피 수분 손실량은 통계학적으로 유의한 관계을 보였다.(p <0.05) 각 중증도별 그룹에서 경표피 수분 손실량과 황색 포도알균의 집락 형성 부위의 개수 및 코점막에서 집락 형성과의 관계를 조사한 결과 경표피 수분 손실량은 집락 부위의 개수가 많을수록, 코점막에서 집락이 형성될수록 높아졌으나 통계학적으로 유의하지는 않았다. 전체 환자의 48.7%에서 황색 포도알균이 배양되었으며 그 중에서 가장 많이 검출된 독소 유전자는 sea 단독생산이 52.6%에서 확인되었고 sea와 tsst-1이 동시생산된 경우가 42.1%였으며 tsst-1 단독생산이 5.3%로 확인되었다. 결론 : 아토피피부염 환자에서 임상적 중증도와 경표피 수분 손실량을 통해 측정한 피부 장벽기능은 통계학적으로 유의한 상관관계를 보였다. 한국인 아토피피부염에서 가장 흔히 검출된 독소 유전자는 sea였으며 아토피피부염의 병인에 있어 중요한 역할을 할 것으로 생각한다.

Atopic dermatitis (AD) is a chronic inflammatory disease by barrier defects combined with modified immune responses of the innate and the adaptive immune system to exogenous or endogenous factors. However, the network of mechanisms leading to the manifestation of AD is far from being completely understood. Cordyceps bassiana (C. bassiana) is a kind of insect-borne mushrooms that have been reported to exhibit a variety of ethnopharmacological activities such as strengthening the immune system, improving athletic performance, reducing the aging and promoting longevity. Although numerous pharmacological potentials of C. bassiana have been known, anti-atopic effect of C. bassiana has not been sufficiently demonstrated yet. In this study, I obtained positive results that Cm-WE, water extract of C. bassiana without polysaccharide, affects anti-atopic efficacy in DNFB-induced NC/Nga mice and antiinflammatory activity in RAW264.7 macrophage cell. To confirm anti-atopic efficacy of Cm-WE in another animal model, I used oxazolone-induced hairless mouse model and found that Cm-WE improves dermatitis symptoms and signs - skin thickness, over-expressed immunoglobins (IgE, IgG1 and IgG2a) and mast cell infiltration. To evaluate signaling regulatory mechanism of its inhibitory activity, I employed various cells involved in AD such as T-Lymphocyte, keratinocyte and mast cell and found out decrease the mRNA levels of IL-4, IL-6, IL-13 and IFN-γ. The inhibitory effect of Cm-WE seemed to be due to phosphorylation suppression of p38, JNK through syk kinase. Moreover, I isolated a new compound, 1,9-dimethylguanine (1,9-DMG), reported for the first time from C. bassiana, and demonstrated anti-atopic efficacy, such as improvement of dermatitis symptoms, skin thickness, over-expressed IgE and mast cell infiltration in DNFB-induced NC/Nga murine AD model. Based on anti-inflammatory and antiatopic effects of Cm-WE and 1,9-DMG, I investigated pharmacokinetic properties through development of a rapid, sensitive and reliable analytical method for 1,9-DMG in rat plasma. The present data will expand more successful strategies for prevention and treatment of skin inflammatory disease and can be expected as a potential herbal medicine and a new drug for atopic dermatitis.

<국문초록> 아토피성 피부염은 일반적으로 알러지 질환의 소인 또는 가족력을 가진 환자에게서 주로 발생하는 만성 염증성 피부질환이다. 특히 소년기에 아토피성 피부염은 재발되는 만성 염증의 특징을 가지며, IgE의 과생산 및 환경인자와도 관련되어 있다고 보고되어 있다. 그러나 현재까지 아토피성 피부염의 정확한 발병기전에 대해선 밝혀지지 않았으며, 최근 아토피성 피부염 연구에 있어서 NC/Nga 마우스의 동물 모델의 사용이 적합한 것으로 알려져 있다. 백련(Nelumbo nucifera)은 주로 인도, 한국, 중국 및 일본 등에 분포되어 있으며, 다양한 효능과 약리작용 등이 보고되어져 있다. 본 연구에서는 NC/Nga 마우스의 등 피부에 DNCB를 반복적으로 접촉시켜 atopic dermatitis-like 피부손상을 유발시킨 후, 연의 잎(NL), 꽃(NF)과 뿌리(NR) 추출물을 이용하여 피부개선효과를 검정하였다. NL, NF와 NR 추출물의 효능에 대한 평가는 조직병리학적 검사와 혈중 IgE 농도, TEWL의 측정, scratching behavior 및 Clinical symptoms severity 측정 등의 방법을 사용하였다. 아토피 피부염이 유발된 NC/Nga 마우스에게 NL, NF와 NR 추출물(50 mg/mice/day)을 4주 동안 경구 투여한 결과, clinical severity의 억제, TEWL의 감소, scratching behavior의 호전 및 혈중 IgE 농도의 감소되었다. 또한, 조직병리학적 분석 결과, 진피층의 증식과 과각질화를 유발하는 탈과립 된 비만세포와 호산구 등의 염증세포 침윤이 NL군에서 유의성 있게 감소되었다. HaCaT 세포내 MMPs의 발현을 평가하기위해 gelatin zymography를 실행 하였으며, HaCaT 세포의 증식과 이동은 MTT와 wound healing assay를 이용하여 측정하였고, HaCat 세포내 TARC의 분비는 INF-γ과 TNF-α로 유도되었고, TARC kit을 이용하여 측정하였다. 그 결과, NL와 NF 추출물을 처리한 군에서 농도 의존적으로 HaCaT 세포의 증식과 이동을 억제하였으며, MMP-2와 MMP-9의 발현 또한 NL와 NF 추출물의 농도가 증가할수록 감소시키는 것으로 나타났고, NL 추출물을 처리한 HaCaT 세포 내 TARC의 분비를 농도의존적으로 억제하였다. 본 연구 결과, 아토피 피부염에 대한 NL 추출물의 예방효과는 농도 의존적인 경향을 나타냈으며, NL 추출물의 지속적인 투여는 아토피 피부염의 개시와 진행을 현저하게 감소시키는 것으로 나타났다. <Abstract> Atopic dermatitis (AD) usually develops in patients with an individual or family history of allergic diseases, and is characterized by chronic relapsing inflammation seen especially in childhood, association with Ig E hyperproduction and precipitation by environmental factors. However, the exact etiology of AD has been unclear. A suitable animal model is required to explore the pathogenesis and treatment of AD. Nelumbo nucifera (Nympheaceae), native of India, China, Korea and Japan has been reported to exhibit different health promoting effects. The objective of this study was to examine the effects of leaf (NL), flower (NF) and root (NR) of Nelumbo nucifera on the atopic dermatitis-like skin lesion induced by repeated epicutaneous application of 2,4-dinitrochlorobenzene (DNCB) on the dorsal skin of NC/Nga mice. The efficacy of NL, NF and NR was judged by histopathological examination, the blood Ig E level, measurement of transepidermal water loss (TEWL), scratching behavior and clinical severity score. The oral administration of NL, NF and NR (50 mg/day) for 4 weeks into NC/Nga mice with overt dermatitis resulted in the suppression of clinical severity score, TEWL, scratching behavior and blood Ig E level. Histopathologic analyses revealed that infiltration of inflammatory cells, especially degranulated mast cells and eosinophils, thickening of the epidermis, and prominent hyperkeratosis, were significantly reduced in NL group. Moreover, the preventive effect of NL extract on the dermatitis was dose-dependent and continuous intake of NL extract significantly decreased its onset and development. To evaluate the expression of MMPs in HaCaT cell line, gelatin zymography was performed. Similarly, proliferation and migration of HaCaT was carried out by following MTT and wound healing assays. The release of Thymus and activation regulated chemokine (TARC) in HaCaT cells induced by Interferon-γ (INF-γ) and Tumour necrosis factor-α (TNF-α) was determined by using TARC kit. NL and NF dose dependantly inhibited the proliferation and migration of HaCaT cells. Moreover, NL and NF inhibited the expression of MMP-2 and MMP-9 in HaCaT in a dose dependant fashion. The pretreatment of Nelumbo nucifera leaf extract (NL) at concentrations of 250 and 500 μg/ml suppressed the secretion of TARC in HaCaT cells stimulated with 10 ng/ml of INF-γ and 10 ng/ml of TNF-α. These results suggest that lotus leaf extract may be a useful resource in the management of atopic dermatitis.
Study on the Efficacy of Exosome-Based Therapeutics in Atopic Dermatitis

Atopic dermatitis (AD) is a chronic, relapsing inflammatory skin disease characterized by overactivation of Th2 immune responses, impaired skin barrier function, and elevated serum IgE levels. Although conventional treatments such as topical corticosteroids and monoclonal antibody-based biologics (e.g., dupilumab) offer therapeutic benefits, their long-term use is limited by adverse effects and variability in patient response, highlighting the need for alternative strategies. Exosomes, nanoscale vesicles mediating intercellular communication, have recently emerged as promising next-generation immunomodulatory therapeutics due to their anti-inflammatory and tissue-regenerative properties. In the part 1, This dissertation integrates findings from two independent studies evaluating the therapeutic potential of exosome-based treatments in AD. In the first study, a DNCB and SDS-induced mouse model of AD was used to compare the efficacy of umbilical cord-derived mesenchymal stem cell exosomes (MSC-EVs) with dupilumab. Clinical symptoms, serum IgE levels, histopathological features, and Th2 cytokine expression were analyzed. MSC-EVs exhibited anti-inflammatory effects comparable to or exceeding those of dupilumab. Notably, exosome treatment was associated with modulation of inflammation-related microRNA (miR-146a), suggesting a therapeutic mechanism involving miRNA regulation. In the part 2, an antigen-specific AD model was established by repeated application of Dermatophagoides farinae (house dust mite) extract to the skin. Exosome treatment led to significant improvement in skin lesion size and thickness, dermal inflammatory infiltration, and serum IgE levels. Histopathological analysis revealed reduced epidermal thickness and restoration of skin barrier structure. The in vivo model confirmed the direct anti-inflammatory and skin-protective effects of exosomes, and therapeutic efficacy was also observed in spleen-associated immune modulation, indicating high clinical applicability. In the part 3, exosomes loaded into an F127 hydrogel were topically applied in a D. farinae–induced AD model. Compared with systemic administration, hydrogel-based local delivery led to significant improvement in clinical skin severity scores and serum IgE levels, as well as restoration of skin barrier proteins. This confirmed that exosomes can effectively alleviate AD symptoms through topical application, supporting their clinical applicability. These findings collectively demonstrate that exosome-based therapeutics exert dual effects—immune regulation and barrier restoration—in atopic dermatitis. This suggests their potential as a promising next-generation treatment strategy capable of complementing or replacing current biologic therapies. 아토피 피부염은 만성 재발성 염증성 피부 질환으로, Th2 면역반응의 과활성과 피부 장벽 손상, 혈중 IgE 상승 등이 병태생리의 핵심으로 작용한다. 기존 치료제인 국소 스테로이드제나 단일클론 항체 기반의 생물학적 제제(예: 듀필루맙)는 일정 수준의 효과를 보이지만, 장기적 부작용과 치료 반응의 개인차로 인해 새로운 치료 전략에 대한 필요성이 대두되고 있다. 엑소좀은 세포 간 신호전달을 매개하는 나노 입자로, 항염증 및 조직재생 기능을 바탕으로 차세대 면역조절 치료제로 주목받고 있다. 본 논문에서는 아토피 피부염에서 엑소좀 치료제의 가능성을 다각도로 평가하기 위해 수행된 두 편의 연구를 통합하였다. 제1연구에서는 DNCB 및 SDS를 반복 도포하여 아토피 피부염을 유도한 마우스 모델을 대상으로, 제대 젤리 유래 중간엽 줄기세포 엑소좀(MSC-EVs)과 듀필루맙의 치료 효과를 비교하였다. 임상적 병변, 혈청 IgE, 조직학적 염증소견, Th2 사이토카인 발현을 분석한 결과, 엑소좀은 듀필루맙과 유사하거나 더 우수한 염증 억제 효과를 나타냈다. 특히 엑소좀 투여군에서는 염증 관련 마이크로RNA(miR-146a-3p)와 염증 경로 억제가 밀접하게 연관되어 있음을 보여주었다. 이러한 결과는 엑소좀이 miRNA 조절을 통해 아토피 피부염을 완화시킬 수 있는 치료 기전을 가짐을 시사한다. 제2연구에서는 집먼지진드기(Dermatophagoides farinae) 추출물을 반복적으로 피부에 도포하여 유도한 아토피 모델에서 엑소좀의 치료 효과를 평가하였다. 치료 후 피부 병변 면적과 두께, 진피 침윤, 혈청 IgE 수치 등이 유의하게 개선되었으며, 표피의 두께 감소 및 장벽 구조 복원이 조직병리학적으로 확인되었다. in vivo 모델에서 엑소좀의 직접적인 항염 및 피부보호 효과를 입증하였으며, 엑소좀이 비장을 통해 유의미한 치료 효능을 보였다는 점에서 임상 적용 가능성이 높음을 시사한다. 제3연구에서는 집먼지진드기(Dermatophagoides farinae) 추출물을 반복적으로 피부에 도포하여 유도한 아토피 모델에서 F127 하이드로젤에 로딩된 엑소좀의 치료효과를 평가하였다. 치료 후 아토피 임상적 병변, 혈청 IgE 등의 수치에서 주사한 그룹과 비교하여 유의하게 개선되었으며 피부 단백질 복원이 회복되는 것을 확인하였다. 이를 통해 엑소좀이 피부 국소 방식을 통해 아토피 증상 개선 효과가 있다는 것을 입증하여 임상 적용 가능성이 높음을 시사한다. 이러한 결과는 엑소좀 기반 치료제가 아토피 피부염에서 면역 염증 조절과 피부 장벽 회복을 동시에 유도할 수 있는 이중 작용 기전을 통해, 기존 생물학적 제제를 보완하거나 대체할 수 있는 유력한 차세대 치료 전략임을 보여준다.
Studies on the role of HDAC6 molecular network in allergic inflammation and atopic dermatitis

HDAC6 is a class IIb histone deacetylase and deacetylates lysine residues in non-histone proteins. HDAC6 is primarily expressed in the cytoplasm and acts as a regulator of microtubule-mediated processes such as cell migration, autophagy, stress granule formation, and misfolded protein degradation. HDAC6 has been proposed as a therapeutic target for various cancers, neurodegenerative diseases, and innate immune responses. However, the molecular mechanism of HDAC6 in hypersensitivity immune reactions has not yet been identified. To determine the role of HDAC6 in allergic inflammation, two models of allergic inflammation were used: anaphylaxis and atopic dermatitis. In anaphylaxis models, early growth response 3 gene (EGR3) is most expressed in antigen-stimulated RBL2H3 cells and regulates the expression of HDAC6 by binding to the HDAC6 promoter. HDAC6 mediates in vitro allergic inflammation reactions and in vivo anaphylaxis models. In anaphylaxis models, IL-27 is an inflammatory cytokine directly regulated by HDAC6 and mediates allergic inflammation in vitro and in vivo. In the inflammatory tumor microenvironment, IL-27 is present in exosomes secreted from activated mast cells and mediates intercellular interactions to promote tumorigenic and invasive ability of cancer cells and TAM polarization of macrophages. Atopic dermatitis is a chronic allergic inflammatory disease mediated by mast cells, immune cells, and skin component cells. In atopic dermatitis, inhibition of HDAC6 attenuates the symptoms of atopic dermatitis and reduces the recruitment of mast cells to the inflammatory site and the expression of inflammatory mediators in tissues. HDAC6 regulates the expression of sirtuin 1 (SIRT1) by regulating the expression of miR-9. SIRT1 and miR-9 regulate autophagy and reactive oxygen species (ROS) generation accompanied by AD in keratinocytes. CXCL13 is an inflammatory cytokine and regulated by HDAC6 in atopic dermatitis. CXCL13 mediates skin inflammation and is present in exosomes secreted by activated skin mast cells. CXCL13 regulates the intercellular interactions among skin mast cells, keratinocytes, and dermal fibroblasts, and acts as a major mediator of atopic dermatitis. Taken together, these findings indicate that the EGR3-HDAC6-IL27 molecular network modulates allergic inflammatory responses and the inflammatory tumor microenvironment, and HDAC6 mediates atopic dermatitis by regulating miR-9 and CXCL13. This provides evidence that HDAC6 can be a target for anti-allergy or anti-atopic drugs by understanding the molecular networks of allergic inflammation and atopic dermatitis. HDAC6는 히스톤 탈 아세틸화 효소 중 하나로서, 비히스톤 단백질의 탈 아세틸화를 통해서 다양한 생물학적 과정을 조절함이 알려져 있다. HDAC6는 주로 미세소관이 매개하는 생물학적 과정의 주 조절자로서 작용하며, 다양한 암 질환, 신경 퇴행성 질환 및 선천적 면역 반응의 치료 표적으로서 제시된다. 본 연구에서는 과민성 면역반응인 알레르기성 염증 및 아토피 피부염에서 HDAC6의 역할 및 분자적 조절 메커니즘을 밝히기 위해 연구를 수행하였다. RBL2H3 세포에 항원을 자극하여 알레르기성 염증 반응을 유도한 뒤, 가장 많이 발현이 변화하는 유전자를 RNA-seq 및 차등 발현 유전자(DEG) 분석을 통하여 발굴하였다. EGR3는 유사 분열 자극에 의해 유도되는 초기 성장 반응 유전자 패밀리 중 하나이며, 전사 조절자로서 널리 알려져 있다. 본 연구에서는 EGR3가 HDAC6유전자의 프로모터에 결합하여 HDAC6의 발현을 전사 수준에서 조절함을 밝혔다. EGR3는 HDAC6의 발현을 조절함으로써 시험관 및 생체 내 아나필락시스 모델을 매개하는 주 조절자로서 작용하였다. 아나필락시스 모델에서 HDAC6의 조절은 알레르기성 염증반응에 의해 증가되는 사이토 카인인 IL-27의 발현을 조절하였다. IL-27은 염증성 또는 항 염증성 성향을 가지고 있는 사이토 카인으로, 면역계에서T세포의 분화를 유도하며 IL-10의 발현을 조절함이 알려져 있다. 본 연구에서 IL-27은 항원에 의해 발현이 중가 하며, 시험관 및 생체 내 알레르기 염증반응을 매개하였다. 또한, 암세포의 종양 형성 및 침습 능력과 대식세포의 분극화를 조절함으로써 염증성 종양 미세환경의 주요 조절 매개체로 작용하였다. 만성 알레르기성 염증질환 모델인 아토피 피부염은 각질세포, 진피 섬유아세포 및 비반 세포를 포함하는 다양한 면역세포들이 관여하는 염증반응으로 심한 가려움증, 피부 건조증 및 습진 등의 증상이 유발되는 대표적인 알레르기 질환이다. 본 연구에서는 아토피 피부염 모델에서 아토피 피부염 유발물질인 DNCB에 의해 HDAC6의 발현이 유도되며, HDAC6의 작용 및 발현 저해는 아토피 피부염의 임상 증상을 약화시키고 피부 염증 부위로 비반 세포 및 면역 세포의 모집을 감소시킴을 밝혔다. MicroRNA array 및 Cytokine array를 통한 HDAC6의 분자적 기전 연구는 HDAC6가 miR-9의 발현을 조절하여 SIRT1의 발현을 직접적으로 조절하며, 염증성 사이토 카인인 CXCL13의 발현 및 분비를 조절함을 밝혔다. MiR-9는 SIRT1을 표적으로 조절하여 아토피 피부염 증상을 약화시켰고, CXCL13은 아토피 피부염을 매개하는 주 조절자로서 작용하였다. 또한, SIRT1과 miR-9는 각질세포에서 손상으로 인하여 발생하는 자가포식 반응을 조절하고, CXCL13은 비반 세포에서 분비하는 엑소좀에 존재하여 비반세포, 대식세포, 각질세포 및 진피 섬유아세포 사이의 세포간 상호작용을 매개함으로써 아토피 피부염의 주 매개자로서 작용하였다. 본 연구에서는 알레르기성 염증 및 염증성 종양 미세환경에서 EGR3-HDAC6-IL27 분자 네트워크와 아토피 피부염에서 miR-9 및 CXCL13을 조절하는 HDAC6의 역할을 조사하였다. 이러한 발견은 HDAC6이 항 알레르기 또는 항 아토피 약물의 표적이 될 수 있음을 시사한다.
케르세틴이 집먼지진드기 추출물로 유도된 아토피피부염 NC/Nga 마우스의 피부 병변에 미치는 영향

Background : Atopic dermatitis is a chronic inflammatory skin disease accompanied by severe itching, and its prevalence is continuously increasing worldwide. The cause of atopic dermatitis is not clear. However, it is known that inflammation caused by the penetration of allergens into the skin due to a decrease in skin barrier function is associated with the development of atopic dermatitis. The stratum corneum is important for the skin barrier function, and this structure is formed through the keratinocyte differentiation process. Since there is no drug that can cure atopic dermatitis, the treatment of atopic dermatitis is symptomatic treatment using drugs such as immunosuppressants or antihistamines. However, these agents have limited effects, and long-term use may cause side effects. Therefore, it is necessary to develop new agents for treating atopic dermatitis. Flavonoids, which are secondary metabolites of plants, are known to have various physiological activities such as anti-cancer, antihypertensive, antioxidant and antibacterial activities, and thus have a high possibility of being used as a disease treatment. Quercetin, a type of flavonoid, is known to have physiological activities such as anticancer, antioxidant, and anti-allergic effects, and many studies using quercetin are being conducted. Objective : The purpose of this study was to investigate the effect of quercetin on skin lesions in atopic dermatitis. Methods : NC/Nga mice were used as experimental animals for atopic dermatitis. Atopic dermatitis was induced by applying house dust mite cream(AD cream) to the skin of NC/Nga mice. The experimental group was divided into three groups: the control group, the atopic dermatitis group, and the atopic dermatitis+quercetin group. Quercetin dihydrate(30 mg/kg BW/time) was injected intraperitoneally in mice 3 times per week for 4 weeks. During the experimental period, skin symptoms were observed with the naked eye, and the number of skin scratches was measured by CCTV at 2 and 4 weeks after quercetin injection. After 4 weeks of quercetin injection, the experimental animals were sacrificed, the skin tissue findings were observed, and the expression levels of genes related to skin keratinocyte differentiation and the amount of lipid peroxides in skin tissues were measured. Results :The number of scratches was 264.3 ± 21.9 times/hr in the atopic dermatitis group and 62.3 ± 11.1 times/hr in the atopic dermatitis + quercetin group, a decrease of 77% in the atopic dermatitis + quercetin group. In the atopic dermatitis + quercetin group, skin rashes, papules, and eczema symptoms were mostly improved, and the size and number of crusts were also reduced. In the skin histological findings, most of the stratum corneum was not observed in the atopic dermatitis group, but the stratum corneum was well observed in the atopic dermatitis + quercetin group. The content of TBARS in skin tissue was 257.1 ± 27.1 μM/g tissue in the atopic dermatitis group, 375% higher than that of the control group, and 55% lower in the atopic dermatitis + quercetin group than the atopic dermatitis group. The mRNA levels of Sprr1b, Sprr2d, Sprr2f, Sprr2h, Sprr2k, and Tgm5 genes related to keratinocyte differentiation in skin tissues were decreased in the atopic dermatitis group than in the control group, and increased in the atopic dermatitis + quercetin group than in the atopic dermatitis group. The mRNA levels of LCE1F, LCE1G, LCE1J, and LCE1K genes related to late cornified envelope in skin tissues were decreased in the atopic dermatitis group than in the control group, and increased in the atopic dermatitis + quercetin group than in the atopic dermatitis group. The mRNA levels of Il1b, Il1f6, and S100a8 genes related to inflammatory response in skin tissues were increased in the atopic dermatitis group than in the control group, and decreased in the atopic dermatitis + quercetin group than in the atopic dermatitis group. As a result of the above experiments, it was found that quercetin improved skin symptoms of atopic dermatitis and improved skin itching. In addition, quercetin was found to be effective in restoring the skin barrier function by improving the expression level of genes related to keratin differentiation and restoring the stratum corneum. CONCLUSIONS: Quercetin improves atopic dermatitis-associated skin lesion, and this effect shows the possibility of use as a treatment for atopic dermatitis. It is thought that additional preclinical and clinical studies are needed to use quercetin clinically.
