      • 蜂毒藥鍼이 Writhing reflex 및 脊髓內 痛症關聯 神經細胞의 活性에 미치는 影響

        Introduction : In spite of the use of Bee Venom aqua-acupuncture in the clinics, the scientific evaluation on effects is not enough. Bee Venom aqua-acupuncture is used according to the stimulation of acupuncture point and the chemical effects of Bee Venom. The aims of this study is to investigate the analgegic effects of the Bee Venom aqua-acupuncture, through the change of writhing reflex and the change of c-fos in secondary neurons in the spinal cord. Materials and Methods : Pain animal model was used acetic acid method. The changes of writhing reflex of the mice which were derived pain by injecting acetic acid into the abdomen, after stimulating Bee Venom aqua-acupuncture on Chungwan(CV12) and non acupuncture point on the backside were measured. We used Fos immunohistochemical technique to study the neuronal activity in the spinal cord. Results : 1. It showed that the writhing reflex were appeared on the groups which injected acetic acid only, and saline-acetic acid group(sample I), but not on the group bee venom-saline group(sample II). 2. The change of writhing reflex by Chungwan(CV12) Bee Venom aqua-acupuncture showed significant decrease in the order of Chungwan(CV12) Bee Venom aqua-acupuncture group III(2.5×10-3g/kg), II(2.5×10-4g/kg), and I(2.5×10-5g/kg), compared with control group. There were significant decrease of number of writhing reflex in 5∼10, 10∼15 and 15∼20 minutes intervals of Chung wan(CV12) Bee Venom aqua-acupuncture group I, and in 0∼5, 5∼10, 10∼15 and 15∼20 minutes intervals of II and III, compared with control group. 3. The change of writhing reflex by non acupuncture point Bee Venom aqua-acupuncture showed significant decrease in the 0∼5 and 5∼10 minutes intervals and the total number of writhing reflex in 2.5×10-4g/kg group, compared with control group 4. The effects of writhing reflex of Chungwan(CV12) Bee Venom aqua-acupuncture group showed significant decrease, compared with non acupuncture point Bee Venom aqua-acupuncture group. 5. Expression of c-fos in superficial dorsal horn(SDH), nucleus proprius(NP) and neck of dorsal horn(N) on 6∼9th thoracic spine decreased significantly at 2.5×10-4g/kg Bee Venom aqua-acupuncture, compared with saline-acetic acid group. 6. The numeral change of Fos-LI neurons on the NP, N, and ventral gray(V) on 6∼9th thoracic spine, SDH on 9∼11th thoracic spine, and SDH and V on 11∼13th thoracic spine decreased significantly at Chungwan(CV12) Bee Venom aqua-acupuncture, compared with saline-acetic acid group. 7. The correlation between the numbers of writhing reflex and Fos-LI neurons in T6-13 segment was statistically statistically significant at Chungwan(CV12) Bee Venom aqua-acupuncture. Conclusion : This study shows that the Bee Venom aqua-acupuncture on Chungwan(CV12) decreases the numbers of writhing reflex and Fos-LI neurons. As the analgegic effects of Bee Venom aqua-acupuncture is recognized. Bee Venom aqua-acupuncture treatment is expected for pain modulation. In order to use it in many ways, more researches are needed for the dose and stability of Bee Venom aqua-acupuncture.

      • 봉독약침이 제2형 콜라겐유도 관절염에 미치는 영향

        Bee venom (BV) has traditionally been used in Oriental medicine to relieve pain and to treat inflammatory disease such as rheumatoid arthritis (RA). Autoimmunity to type II collagen (CII) may involve in the pathogenesis of RA. This study was performed to evaluate the effect of BV on type II collagen induced arthritis (CIA) with the naked eye, a immunohistochemical method and the examination of histology. Male mice were immunized by subcutaneously injection of an 200㎍ emulsion mixed with bovine CII and complete Freund's adjuvant (CFA) twice for two weeks. In the control group, normal saline was injected, and in the experimental group, BV was applied. The incidence of arthritis, the mean arthritis index and the number of the arthritic limbs of the BV group were all significantly lower than those of the control group. Among the pro-inflammatory cytokines, the production of TNF-in the BV group was also suppressed compared with the control group, but IL-1was not. The examination on the histopathology of joints of CIA mice showed the effect of Bee Venom Herbal Acupuncture on the arthritis. Treatment with BV resulted in inhibition of development of arthritis and immune responses to CII.

      • 蜂毒藥鍼과 淸熱瀉濕湯이 제 Ⅱ 형 collagen 유도 關節炎에 미치는 영향

        Bee-venom(BV) and Cheongyeolsaseup-Tang (Qingrexieshi-tang) are famous method that treated arthritis. So that the purpose of this study was to investigate effects of Bee-venom and Cheongyeolsaseup-Tang extract (CST) on type Ⅱ collagen-induced arthritis rats. Experimental groups were divided into 5 groups ; Sham group were daily administered DW 1.0 ㎖ to normal rats for 28 days, Control group were daily administered DW 1.0 ㎖ to arthritic rats for 28 days, experimental group Ⅰ were daily injected BV 0.1 ㎎/㎏ to arthritic rats for 28 days. experimental group Ⅱ were daily administered CST 500 ㎎/㎏ 1.0 ㎖ to arthritic rats for 28 days, experimental group Ⅲ were daily administered CST 500 ㎎/㎏ 1.0 ㎖ and daily injected BV 0.1 ㎎/㎏ to arthritic rats for 28 days. The present author observed inhibitory effect of edema, analgetic effects by hot plate, analysis of blood chemistry (WBC, ALT, AST), western blot (IL-6, cPLA2), histopathological and radiological changes. The results were as follows : 1. Group Ⅰ, Ⅱ, Ⅲ were significantly inhibited the rate of paw edema compared with control group. Group Ⅲ were most significantly increased the edema inhibition rate. 2. Group Ⅰ, Ⅱ, Ⅲ were significantly decreased WBC and the time of heat threshold compared with control group, and group Ⅲ were most significantly decreased. 3. Group Ⅰ, Ⅱ, Ⅲ were significantly decreased AST (Asparatate transaminase) and ALT (Alanine transaminase) compared with control group, and only group Ⅲ were significantly decreased ALT. 4. Group Ⅰ, Ⅱ, Ⅲ were significantly decreased the mass of IL-6, cPLA_(2) in synovia compared with control group, and group Ⅲ were most significantly decreased in immunological test. 5. Group Ⅰ, Ⅱ, Ⅲ were relieved and reproduced the erosion of arthritic site compared with control group, and group Ⅲ were most effective compared with group Ⅰ, Ⅱ with naked eye on the histopathological view. 6. Group Ⅰ, Ⅱ, Ⅲ were improved on the radiological view, and group Ⅲ were most effective compared with group Ⅰ, Ⅱ. The present author thinks that Bee-venom and Cheongyeolsaseup-Tang can be used for curing rheumatoid arthritis, anti-inflammatory effects will be somewhat better in combination of BV and CST.

      • 봉독약침이 Monosodium Iodoacetate로 유발된 골관절염 통증 모델에서 중추신경내 NOS, C-fos, Serotonin, Substance P 발현에 미치는 영향

        This study was carried out to investigate the effects of Bee-venom treatment on the expression of NOS, c-fos, serotonin and substance P in central nerve system of monosodium iodoacetate(MIA)-induced osteoarthritic pain model. Arthritis was induced by injection of MIA(0.5 ㎎) into knee joint cavities of rats. Arthritic rats were divided into control(n=8) and treated(n=8) group. Control group was injected with normal saline once a day for 20 days, while treated group was injected with Bee-venom extract once a day for same duration. Injected point of Bee-venom and saline was subcutaneous tissue at Tokpi(ST35) acupoint. Histochemical study of NADPH-d and immunohistochemical study of c-fos, serotonin and substance P in brain and spinal cord were performed at 20 days after MIA injection. The results were as follows ; - The numbers of NADPH-d positive cells in superficial dorsal horn of spinal cord of treated group(24±4) was significantly (p<0.01) decreased compared with control(33±5). The numbers of NADPH-d positive cell in dorsolateral periaqueductal gray matter of treated group(114±18) was significantly(p<0.01) decreased compared with control(143±14). - The numbers of c-fos positive cells in dorsal periaqueductal gray matter of treated group(56±8) was significantly(p<0.01) decreased compared with control(78±13). The numbers of c-fos positive cells in paraventricular thalamic nucleus of treated group(64±14) was significantly decreased compared with control(88±27). - The numbers of serotonin positive cells in median raphe nucleus of treated group(104±15) was significantly(p<0.05) decreased compared with control(139±34). On the basis of these results, we concluded that Bee-venom treatment has inhibiting effects on the pain transmission in monosodium iodoacetate-induced osteoarthritic pain model in rat.
