감열지용 Oil Red O 시약의 제조방법에 관한 연구

본 실험에서는 감열지 발색층에 인쇄된 문자정보의 지워짐 없이 지문을 현출할 수 있는 ORO 시약을 제조하기 위하여 Beaudoin의 ORO 시약 제조방법을 기준으로 methanol과 NaOH의 함량을 조절하였다. 그 결과 Beaudoin의 ORO 시약 제조방법에서 methanol의 함량을 50%로 줄였을 때 발색층의 문자정보를 보존하면서 지문을 현출할 수 있었다. Methanol의 함량이 50%보다 적거나 methanol을 물에 희석할 경우 지문현출에 소요되는 시간이 길어지고 이는 감열지의 훼손을 일으켰다. 또한 NaOH의 함량이 줄어들면 현출된 지문의 융선이 번지거나 지문을 현출할 수 없었다. aging된 지문에서 현출력을 시험하기 위해 감열지의 발색층과 지지체에 지문을 유류하고 1, 3, 7, 14일 보관한 후 Beaudoin의 ORO 시약 또는 본 연구에서 개발한 감열지용 ORO 시약을 적용한 결과, 3일까지는 두 시약간의 차이가 없었으나 7일 aging된 지문부터 본 연구에서 개발한 감열지용 시약이 Beaudoin의 ORO 시약에 비해 지문현출력이 뛰어났으며 지지체에 유류된 지문의 경우에는 두 시약간의 차이가 없었다.
감열지에 유류된 잠재지문의 외부노출 여부에 따른 현출기법에 관한 연구

감열지는 사용의 편리성이 좋기 때문에 다양한 방면에서 사용되고 있으며 그로 인하여 일상에 사용되는 빈도가 높아짐에 따라 범죄 현장이나 용의자를 추적하는 과정에서 쉽게 접할 수 있는 증거물 중 하나이다. 감열지에는 잠재지문이 유류 되어 있을 확률이 높기 때문에 과학수사 분야에서 매우 중요한 물적증거로 인식되고 있다. 감열지는 대부분 영수증으로 사용되고 있으며 영수증에 기록되어 있는 내용 때문에 영수증을 보관하는 경우가 많다. 따라서 저자는 영수증 보관 시 보관 과정에서 외부환경에 노출 여부에 따라 감열지에 유류 된 지문 성분의 양의 차이에 따른 지문 현출 시약의 현출력의 차이가 있을 것이라고 예상하여, 외부환경 노출 여부에 따른 감열지에 유류 된 지문 현출 결과의 차이를 알아보고자 하였다. 감열지는 일반 종이와는 다르게 열과 극성용매에 반응하여 흑화 현상을 보이는 종이이기 때문에 흑화 현상이 진행되면 현출 된 지문이 보이지 않아 지문을 관찰하기가 어렵다. 따라서 저자의 연구에서는 일반종이에 적용하는 시약이 아닌 감열지에 적용할 수 있는 1,2-IND/PVP, 아세톤으로 pre-washing 후 Ninhydrin, DMAC, Iodine fuming method로 총 4가지의 시약을 사용하였다. 실험 검체는 감열지의 감열면에 세로로 split하여 기름지문을 유류한 뒤 왼쪽은 공기와 빛을 차단하기 위해 두꺼운 책 사이에 끼워 보관하고 오른쪽은 일반적인 실온환경에서 보관하였다. 감열지에 지문을 유류 한 뒤 1, 3, 5, 7, 10, 14, 21일 동안 보관하였다. 실험 결과, 대체적으로 aging 기간이 오래될수록 외부환경에 노출되지 않은 지문보다 외부에 노출된 지문이 상대적으로 지문의 융선을 관찰하기 어렵다는 것을 알 수 있었고, 실험에 사용한 4가지 시약 중 1,2-IND/PVP와 아세톤으로pre-washing 후 Ninhydrin으로 처리한 지문은 노출여부와 관계없이21일차까지 번짐 없이 선명하게 지문을 현출하여 가장 우수한 현출력을 보이는 것을 확인하였다. 이는 아세톤이 pre-washing과정에서 지문 유류물을 훼손하는 정도가 크지 않음을 추측할 수 있고, Ninhydrin이 아미노산의 α, β 모두와 반응하여 더욱 높은 현출력을 보인 것으로 예상한다 따라서 범죄 현장에서 감열지가 발견된 경우, 영수증의 내용을 알 수 없어 지문이 유류 되고 얼마만큼의 시간이 경과됐는지 알 수 없고 감열지의 보관환경을 짐작할 수 없을 때 1,2-IND/PVP와 아세톤으로 pre-washing 후 Ninhydrin을 담금법으로 처리하는 것을 권장한다. Thermal paper is used in various aspects because it is easy to use and so as the frequency of daily use increases, it is one of the most easily seen evidence in the process of tracing crime scenes or suspects. Since thermal paper is likely to have latent fingerprints, it is recognized as a very important physical evidence in the field of forensic science. Thermal paper is mostly used as a receipt, and receipts are often kept because of the contents recorded on them. The authors expected that there will be differences in fingerprinting depending on the differences in environment in which receipts are kept and the differences in the ingredients of fingerprints on the thermal paper. Therefore the authors wanted to investigate the differences in fingerprints on the thermal paper due to exposure to the external environment. Since thermal paper, unlike ordinary paper, appears darkening reacting to heat and polar solvent, it is difficult to observe fingerprints when darkening occurs. Therefore, we have applied various reagents that can be applied to thermal paper rather than reagents applied to ordinary paper, especially, 1,2-IND / PVP, Ninhydrin (after pre-washing with acetone), DMAC and Iodine, four reagents were used. The test specimens were constructed by following: 1. Thermal paper were split vertically and oily fingerprints are set on the thermal surface. 2. The left side was kept between the thick books to block the air and light, and the right side was kept in a normal room temperature environment. 3. The fingerprints were stored in the thermal paper for 1, 3, 5, 7, 10, 14, and 21 days. As a result, it was found that fingerprints exposed to the outside of the fingerprints were harder to observe than the fingerprints that were not exposed to the external environment as the aging period became longer. In addition, among the four reagents used in the experiment, the fingerprints treated with 1,2-IND/PVP and Ninhydrin (after prewashing with acetone) were found to have the best output with clear fingerprints without blotting until 21 days regardless of exposure.
다양한 시약을 이용한 감열지 상의 혈흔지문증강에 관한 연구

지류증거물은 사건현장에서 감정물로 많이 수집되어진다. 그 중 감열지는 감열면은 반다공성의 감열지 뒷면은 다공성의 서로 다른 특성을 가지며, 감열면은 극성용매와의 접촉, 산화-환원반응의 발생, 열처리시 흑화현상을 일으킨다. 현재 사용되는 대다수의 시약들은 극성용매를 포함하고 있어, 감열지는 법과학적으로 감정하기 어려운 증거물중 하나이다. 법과학 분야에서 감열지를 주제로한 선행연구들은 감열면에서의 땀지문 현출을 목적으로 한 것들이 대부분이며, 혈흔 지문 현출을 연구한 논문은 단 2건에 불과하다. 이 2건의 선행논문은 흑화를 방지하기위해 LCV, Amido black, Hungarian red의 물 기반 시약들만 사용하였다는 한계점을 갖는다. 그러나 감열면의 흑화현상을 막는 PVP시약이 개발됨에 따라, 물 기반 시약 외에도 극성용매를 기반으로 한 시약과 열처리를 필요로 하는 기법이 적용 가능해졌다. 본 연구에서는 Amido black (water based), Acid violet 17, Acid yellow 7, Crowle's stain, Hungarian red, Amido black (methanol based), Coomassie blue, DFO/PVP, 1,2-IND/PVP, Ninhydrin/PVP, DMAC등 혈흔 시약을 포함한 감열지에서의 지문 현출 기법들을 사용하여 혈흔지문 증강 결과를 관찰하고자 하였으며, 감열지의 상태에 따라 깨끗한 상태의 감열지와 흑화가 발생한 감열지의 두 가지 경우를 나눠 실험을 진행하였다. 깨끗한 상태의 감열지에서는 Amido black (methanol based)과 Ninhydrin/PVP이 감열면과 비감열면에서 전혈, 희석혈액 상관없이 현출이 잘 되었으며 그 다음으로는 Amido black (water based)와 1,2-IND/PVP, Coomassie blue, Hung arian red순으로 현출되었다. 흑화상태의 감열면에서 시약 처리 결과, 흑화 현상의 발생과 혈흔 유류 시점이 전혀 영향을 미치지 않는 것으로 나타났다. Amido black (methanol based), Ninhydrin/PVP 처리시, 추가적인 PVP처리 없이도 좋은 대조비의 혈흔지문을 얻을 수 있었다. 물기반 시약의 경우 10% PVP를 후처리해야 되는 것으로 나타났다. 예상과 달리 메탄올 기반의 혈흔시약 처리시 감열면에서 흑화현상이 발생하지 않았는데 이는 시약처리 과정에서 루이코 염료가 메탄올에 씻겨나갔기 때문으로 추정된다. 메탄올 기반 시약은 유해하나 PVP시약 처리 없이도 흑화현상 제거가 가능하며, 메탄올기반 아미도블랙의 경우 다른 시약과 비교하였을 때 혈흔지문 증강시 감도가 좋은 것으로 나타났다. 그러나 감열면의 프린트가 사라지므로, 시약 처리 전 사진 촬영이 반드시 선행되어야 할 것이다.
Propylene glycol monomethyl ether를 이용한 새로운 1,2-indanedione/zinc 조성에 관한 연구

잠재지문(latent fingerprint)은 범죄현장에서 발견되어 개인식별과 범죄해결을 위한 증거로 사용된다. 그러나 눈에 보이지 않아 여러 방법을 이용하여 잠재지문을 현출(development)하는 과정이 필요하다. 1,2-indanedione/zinc(1,2-IND/Zn)은 종이, 감열지 그리고 테이프가 부착된 종이에서 지문을 현출할 수 있는 시약이다. 하지만 가장 기본적인 1,2-IND/Zn만을 사용하여 감열지나 테이프가 부착된 종이에 적용하기 위해서는 추가 시약이 필요하며 여러 단계를 거쳐야 지문 현출이 가능하고, 각 시약의 조성이 다르기 때문에 실무적으로 사용이 복잡하고 시약의 낭비로 이어질 수 있다는 단점이 존재한다. 본 연구에서는 leuco 염료에 대한 용해성을 향상시키거나 감열지의 글씨를 지우는 데 사용되는 제품의 주요 물질로 알려진 propylene glycol monomethyl ether(PGME)를 1,2-IND/Zn에 추가하여 여러 목적에 one-step으로 사용할 수 있는 새로운 1,2-IND/Zn 조성을 알아보고자 하였다. 1,2-IND/Zn 조성에 PGME를 추가한 PGME-IND/Zn의 최적 조성(optimized formulation)을 탐색하였고, 이 최적 조성이 테이프가 부착된 종이에서의 지문 현출에 어떠한 영향을 미치는지 알아보았다. 마지막으로 PGME-IND/Zn 작업용액의 보관기간(storage period)이 지문 현출 효과에 미치는 영향을 알아보았다. 실험 결과, PGME-IND/Zn의 최적 조성을 확인하였고, 테이프가 부착된 종이에서 효과적으로 지문을 현출할 수 있었다. 또한, 작업용액 제조 후 30일까지는 그 효과가 크게 감소하지 않았다. Latent fingerprints are found at crime scenes and used as evidence for personal identification and crime-solving. However, they need to be developed using various methods to visualize because they are invisible. 1,2-indanedione/zinc(1,2-IND/Zn) is a reagent that can reveal fingerprints from paper, thermal paper, and paper with tape. However, using the most basic 1,2-IND/Zn to thermal paper or paper with tape requires additional reagents, and it has to go through the multiple steps. Because the formulations of 1,2-IND/Zn are different with purposes, it is complicated to use practically and can lead to waste of the reagent. In this study, the composition of new IND reagents that can be used as a one-step for several purposes by adding propylene glycol monomethyl ether(PGME), known to improve the solubility in leuco dyes or as the main substance in products used for wiping out all the details on thermal paper, to 1,2-IND/Zn was investigated. PGME was added to the 1,2-IND/Zn formulation(PGME-IND/Zn) to search for an optimized formulation of PGME-IND/Zn. Also, how it affects the development of fingerprints on paper with tape and with the storage period of PGME-IND/Zn working solution was investigated. Finally, the optimized formulation of PGME-IND/Zn effectively developed fingerprints on paper, thermal paper and paper with tape. Also, the effect of developing fingerprints did not decrease significantly until 30 days after preparation of the working solution.
택배 운송장이 부착된 박스 표면에서 잠재지문 현출에 관한 연구

잠재지문 현출 기법을 선정함에 있어 표면의 특성은 매우 중요한 고려 사항이다. 특히 서로 다른 두 가지 이상의 표면이 동시에 존재하는 검체의 경우, 하나의 기법을 선정하여 처리하는 데는 어려움이 있다. 택배 운송장은 송·수하인의 정보, 운임 및 지불조건, 품목, 운송일자 등을 포함한 물류정보시스템의 기반을 이루는 핵심 문서이다. 운송장은 감열지로 만들어진 표면지와 글라신지로 만들어진 이형지로 이루어져 있으며, 두 재질 모두 반다공성 표면의 특성을 갖는다. 운송장은 일반적으로 다공성 표면인 택배 박스에 부착된 상태로 운송된다. 따라서 본 연구에서는 운송장의 감열지와 글라신지, 그리고 박스 표면에 남겨진 잠재지문을 동시에 현출할 수 있는 최적의 방법을 모색하고자 하였다. 네 개 택배사의 운송장에서 감열지와 글라신지를 일정한 크기로 잘라 박스에 부착한 후 지문 공여자 3명으로부터 자연지문, 땀지문, 기름지문을 유류하게 하였고, 이를 1일, 3일, 7일간 보관하였다. 이후 형광 자성분말(Dazzle orange), 1,2-IND/Zn의 acetone 전처리(pre-washing) 및 후처리(post-washing) 방식, 그리고 DMAC 등 총 네 가지의 현출 기법으로 지문을 현출한 후 현출된 지문을 비교·평가하였다. 실험 결과, 네 가지 현출 기법 중 운송장이 부착된 박스 표면에 남겨진 잠재지문을 동시에 현출할 수 있는 단일 시약 및 기법은 확인되지 않았다. 그러나 표면별로 효과적인 현출 기법이 각각 확인되었다. 감열지에 남겨진 지문은 DMAC으로, 글라신지와 박스 표면에 남겨진 지문은 1,2-IND/Zn로 처리하는 것이 가장 우수한 품질의 지문이 현출되었다. 이러한 결과에 따라 시약을 단계적으로 처리하는 것이 본 실험 조건에서 최적의 현출 방법으로 판단되었다. 구체적으로는 먼저 DMAC으로 검체를 처리하여 감열지에 남겨진 지문을 현출한 후, 1,2-IND/Zn를 처리하여 글라신지 및 박스 표면에 남겨진 지문을 추가로 현출하는 것이 가장 효과적인 처리 순서로 판단된다. The surface characteristics of an object are an important consideration in determining appropriate latent fingerprint development techniques. In particular, when a sample contains two or more different types of surfaces simultaneously, it is difficult to select a single technique that can effectively develop fingerprints on all surfaces. A courier waybill is a key documents in logistics information systems, containing details such as sender and recipient information, delivery fees and terms, item descriptions, and shipping dates. It consists of a top layer made of thermal paper and a release liner made of glassine paper, both of which have semi-porous surface properties. Courier waybills are generally affixed to porous surfaces such as cardboard boxes during transit. Therefore, this study aimed to identify the optimal method for simultaneously developing latent fingerprints left on the thermal paper, glassine paper, and box surface. Thermal papers and glassine papers were cut from waybills issued by four different courier companies and affixed to cardboard boxes. Three donors deposited natural, sweat, and sebaceous fingerprints onto the samples, which were then stored for 1, 3, and 7 days. Afterwards, the fingerprints were developed using four different techniques, including Dazzle orange, 1,2-IND/Zn with acetone pre-washing and post-washing, and DMAC, and the developed fingerprints were compared and evaluated. The results showed that no single reagent or technique could simultaneously develop latent fingerprints across all three surfaces. However, effective development methods were identified for each individual surface. DMAC yielded the best results for thermal paper, while 1,2-IND/Zn was most effective for both the glassine paper and the cardboard surface. Based on these findings, a sequential application of reagents was determined to be the optimal development method under the experimental conditions. Specifically, the most effective sequence involved first applying DMAC to develop fingerprints on thermal paper, followed by 1,2-IND/Zn to additionally develop those on the glassine paper and box surface.
새로운 지질 조성을 활용한 인공 잠재지문 수용액 개발 및 프린팅 적용

과학 기술의 발전은 지문 현출 시약들의 개발로 이어졌으며, 현재 다양한 지문 현출 시약들이 과학수사에 활용되고 있다. 이러한 지문 현출 시약들은 현출력을 평가하기 위해 일정한 잠재지문 용액에 대한 시약의 적용 및 검증이 필요하다. 선행연구에서 제시한 인공 잠재지문 용액은 아미노산 성분으로 국한되어 있거나, 극소수의 현출 기술에만 반응하는 단순 혼합물이라는 한계가 있었다. 따라서 본 연구에서는 인체에서 유래한 지질 성분을 활용하여 한국인의 피지샘 땀 성분과 유사한 지질 잠재지문 수용액을 개발하였고, 이를 기존 아미노산 잠재지문 수용액과 혼합하여 새로운 인공 잠재지문 수용액을 개발하였다. 또한, 새로운 인공 잠재지문 수용액을 프린팅에 적용하여, 다양한 지문 현출 시약에 대하여 정량적으로 평가할 수 있는 통제된 실험 조건을 구성하기 위한 기틀을 마련하였다. 사람의 땀 성분은 대부분 물로 이루어져 있으나, 소수성이 높은 비극성 지질 분자들을 용질로 사용하여 용액의 용해도를 높이기 위해 극성 및 비극성을 모두 띠는 n-hexane과 isopropyl alcohol(1:1, v/v)을 용매로 사용하였다. 프린터의 빈 카트리지에 지질 용액과 아미노산 용액을 채운 후, A4 복사 용지, OHP 필름, 감열지에 실제 지문 크기의 인공 잠재지문을 프린팅하였다. 다공성 검체에는 ninhydrin 및 1,2-indandione(1,2-IND)과 oil red o(ORO) 및 nile red(NR) 시약을 적용하였고, 감열지에는 ThermaNin®과 1,2-IND+polyvinylpyrrolidone(1,2-IND+PVP) 및 감열지용 ORO 시약과 NR 시약을 적용하였으며, 비다공성 검체에는 cyanoacrylate(CA) fuming 후 basic yellow 40(BY 40) 염색 시약과 흑색 및 형광 분말을 적용하였다. 그 결과, 다공성 표면에서는 ninhydrin, 1,2-IND, ORO 시약을 적용한 결과가 잘 현출되었으며, 감열지에서 ThermaNin®, 1,2-IND+PVP 및 감열지용 ORO 시약을 처리한 결과 또한 잘 현출되었다. 비다공성 표면의 결과는 CA fuming 후 BY 40 염색의 결과와 흑색 및 형광 분말을 결과가 모두 잘 현출되었다. 본 연구의 다양한 표면에서 프린팅한 지문 이미지는 각 표면에 적합한 시약을 처리하여 현출한 결과, 모든 시료에서 지문의 융선과 특징점이 뚜렷하게 관찰되었으며, 현출한 지문에 대한 품질 평가 결과, 일부 특수 감열지를 제외하고 모두 높은 점수를 획득하였다. Previous artificial latent fingerprint solutions have been limited to amino acid-based components or simple mixtures responsive to only a few development techniques. This study developed a new artificial latent fingerprint solution by utilizing lipid components derived from the human body, resembling the sebaceous and sweat composition of Koreans, and combining it with existing amino acid-based fingerprint solutions. The new solution was applied in printing to evaluate its quantitative effectiveness across various fingerprint development reagents. While human sweat is primarily water-based, to increase the solubility of non-polar lipid molecules with high hydrophobicity, a solvent that combines polar and non polar properties, n-hexane and isopropyl alcohol in a 1:1 (v/v) ratio, was used. The lipid and amino acid solutions were loaded into empty printer cartridges and printed as actual fingerprint-sized images on A4 copy paper, OHP film, and thermal paper. For porous substrates, fingerprints were developed using ninhydrin, 1,2-indandione(1,2-IND), oil red o(ORO), and nile red(NR) reagents, while non-porous substrates were treated with cyanoacrylate(CA) fuming followed by basic yellow 40(BY 40) staining and application of black and fluorescent powders. On thermal paper, fingerprints were developed using ThermaNin®, 1,2-IND+polyvinylpyrrolidone(1,2-IND+PVP), as well as thermal paper-specific ORO and NR reagents. Results showed successful fingerprint development on porous surfaces using ninhydrin, 1,2-IND, and ORO, while NR did not exhibit a strong fluorescent response. On non-porous surfaces, clear fingerprint images were observed with CA fuming followed by BY 40 staining, black powder, and fluorescent powder applications. For thermal paper, clear development was achieved with ThermaNin®, 1,2-IND+PVP, and thermal paper-specific ORO, while NR again did not produce a strong fluorescent response. Across various surfaces, the fingerprints produced in this study showed distinct ridge details and minutiae, and identity assessments confirmed all developed fingerprints to be identical.
