PD-L1 Aptamer-Conjugated Liposomal Cocktail with SAHA and siRNA for Anti-Lung Cancer Immunotherapy

Co-delivering a chemotherapeutic agent and small interfering RNA (siRNA) as a new therapeutic modality provides a promising strategy for cancer treatment. In this study, we developed programmed death-ligand 1 (PD-L1) targeted aptamer-conjugated liposomes that containing suberoylanilide hydroxamic acid (SAHA) and complexing with PD-L1 siRNA for synergistic anticancer immunotherapeutic effect. In Lewis lung carcinoma 2 (LL/2) cancer cells, PD-L1 aptamer-conjugated liposomes (aptamosomes, aptaplex) demonstrated a remarkable cellular association ability. The combined delivery of PD-L1 aptamosomes[SAHA] and aptaplex[siPD-L1] were also more cytotoxic than only one drug treatment. In vivo inhibition of combination with aptamosome[SAHA] and aptaplex[siPD-L1] significantly reduced LL/2 tumor growth rate. Moreover, this combination therapy notably induced secretion of chemokines that stimulate migration of activated T-cells into tumor tissue in vivo. These PD-L1 aptamer-conjugated liposomes for dual delivery (SAHA and PD-L1 siRNA) have promising potential for clinical applications and possibly provides a well-controlled design for delivery system. 현재 항암 면역 치료 연구에서 화학요법제와 짧은 간섭 RNA인 siRNA를 동시에 전달하는 것은 암 치료에 유망한 전략으로 여겨져 활발하게 연구되고 있다. 본 연구에서는 항암 면역 치료효과를 위해 HDAC 저해제인 SAHA와 PD-L1 siRNA(siPD-L1)를 동시에 전달하였다. 이는 약물이나 핵산을 전달하는 데에 최적의 방법인 리포좀을 이용하여 전달하였으며, 암세포에 특이적으로 타겟팅 할 수 있도록 anti-PD-L1 앱타머를 리포좀에 접합시켜 보다 높은 항암 면역 치료 효과를 유도하였다. Anti-PD-L1 앱타머 접합 리포좀인 aptamosome과 aptaplex는 폐암 세포인 LL/2에 대해 세포 유입과 전달 능력이 상당히 우수하였으며, 이 2가지의 전달체를 이용하여 SAHA와 PD-L1 siRNA를 동시에 처리한 방법은 단일 약물 치료 방법보다 더 높은 세포독성 및 항암효과를 이끌어냈다. 또한, aptamosome[SAHA]와 aptaplex[siPD-L1]의 조합은 생체 내에서도 LL/2 종양의 성장속도를 유의하게 감소시켰으며, 병합된 치료방법에 의해 약물 SAHA로 인한 T 세포-유도 chemokine 분비도 현저하게 증가시키면서 향상된 능동면역반응에 따른 항암효과를 나타냈다. 결론적으로 SAHA와 siRNA를 이중으로 전달하기 위한 PD-L1 앱타머-접합 리포좀은 임상 응용 분야에 대해 유망한 잠재력을 가지고 있으며, 훌륭한 전달 시스템 디자인을 제공할 수 있을 것으로 기대된다.
S100A8/A9에 의한 1차 배양 미세아교세포 및 성상교세포에서 증가되는 S100A8의 TLR4/NF-κB/NLRP3 경로를 통한 pyroptosis 조절
염증 매개체 중 하나인 S100A8/A9는 알츠하이머병(AD) 또는 외상성 뇌손상(TBI)과 같은 여러 신경퇴행성 질환에서 증가하며 염증의 신뢰할 수 있는 마커로 간주된다. S100A8/A9는 위험 관련 분자 패턴(DAMPs)에 속하며 면역 반응을 유도할 수 있다. 미세아교세포와 성상교세포는 중추신경계(CNS)의 비신경 세포에 해당된다. 미세아교세포는 염증과 손상이 일어나는 동안 대식세포로 기능하며 중요한 식세포 역할을 한다. 또한 성상교세포는 여러 사이토카인의 분비 및 신경독성 물질의 방출을 담당한다. 염증과 관련된 세포 사멸 중 파이롭토시스(pyroptosis)는 유전적으로 암호화된 괴사의 경로 중 하나이며 비염증성 형태의 세포 사멸과는 다른 유형의 세포 사멸이다. 지금까지 다른 연구들에선 S100A8/A9를 사용한 암의 세포 사멸을 확인했지만 뇌 질환에서는 현재까지 밝혀진 바가 많지 않다. 본 연구에서는 S100A8/A9 처리에 의한 S100 A8 발현이 1차 미세아교세포 및 성상교세포의 파이롭토시스에 영향을 미치는지 조사하였고 그 결과, 1차 미세아교세포와 성상교세포에서 S100A8/A9에 의해 유발된 caspase-1의 활성화와 특정 사이토카인의 성숙은 TLR4/NFκB/ NLRP3 경로를 통해 파이롭토시스 세포 사멸을 유도함을 확인했다. 또한 S100 A8 short-hairpin RNA (shS100A8)는 ninjurin-1 (NINJ1) 및 GSDMD와 같은 파이롭토시스 관련 단백질의 발현을 억제하였다. 요약하면, 본 연구 결과는 1차 소교 세포 및 성상교세포에서 S100A8/A9에 의한 증가된 S100A8 발현이 파이롭토시스를 유도한다는 것을 밝히는 새로운 관점을 제안한다. One of the inflammatory mediators, S100A8/A9 is increased in several neurodegenerative diseases such as Alzheimer’s disease (AD) or traumatic brain injury (TBI) and is regarded as a reliable marker of inflammation. S100A8/A9 belongs to the danger -associated molecular patterns (DAMPs) and types of molecules that can elicit an immune response. Microglia and astrocytes are non-neuronal cells of the central nervous system (CNS). During inflammation and injury, microglia function as macrophages and play an important phagocytic role. Astrocytes take charge of secretion of multiple cytokines or release of neurotoxic amounts. Among the cell death associated with inflammation, pyroptosis is one of the genetically encoded pathways of necrosis and it is a different type of cell death from the non-inflammatory form of apoptosis. So far, other studies have confirmed cell death through S100A8/A9 in cancer, but not in brain disease. As a heretofore not revealed, I investigated whether S100A8 expression by each treatment of S100A8 and S100A9 affect the pyroptosis of primary microglia and astrocytes. The activation of caspase-1 and maturation of specific cytokines in the S100A8/A9-triggered primary microglia and astrocytes induced pyroptotic cell death through TLR4/NF-κB/ NLRP3 pathway. Furthermore, S100A8 short-hairpin RNA (sh S100A8) inhibited the expression of pyroptosis-related proteins like ninjurin-1 (NINJ1) and GSDMD. In summary, our findings suggest a novel perspective, revealing that the increased S100A8 expression by S100A8/A9 in primary microglia and astrocytes induce pyroptosis.
택사(Alismatis rhizoma) 추출물의 MCF-7 유방암세포 증식 억제 효과

본 연구 목적은 택사(Alismatis rhizoma)추출물의 유방암 세포 성장 억제 효과와 그 기전을 규명하는 것이다. 택사(Alismatis rhizoma)추출물에 대해 유방암에도 의 성장 억제 효과는 WST-1분석법으로 측정하고, 관련 단백질 및 유전자에 미치는 영향은 Real-time PCR을 이용하여 분석하였다. 그 결과 택사(Alismatis rhizoma)은 유방암 세포의 성장을 유의하게 억제했으며, 억제 효과는 용량 및 처리 시간과 양의 상관관계가 있는 것으로 나타났다. 또한 택사(Alismatis rhizoma)은 세포 주기 단백질 D1(Cyclin D1)의 발현 수준을 유의하게 감소시켰다. 또한 택사(Alismatis rhizoma)은 세포의 이동 및 침입 능력을 억제다. 종합하면 택사(Alismatis rhizoma)은 유방암 세포의 성장을 억제할 수 있는 잠재력을 가지고 있으며 세포 주기를 조절하고 세포 이동 및 침입을 억제하는 방식으로 작용할 수 있다. This study aimed to investigate the inhibitory effect of Alismatis rhizoma on MCF-7 breast cancer cells and its underlying mechanisms. MTT and WST-1 assays were conducted to evaluate the growth inhibitory effect of Alismatis rhizoma on MCF-7 cells,and Real-time PCR were used to analyze its impact on relevant proteins and genes. The results showed that Alismatis rhizoma significantly suppressed the growth of MCF-7 cells, and the inhibition effect was positively correlated with dosage and treatment time. Furthermore, Alismatis rhizoma markedly decreased the expression levels of Cyclin D1. Additionally, Alismatis rhizoma inhibited the migration and invasion ability of the cells. In conclusion, Alismatis rhizoma possesses the potential to inhibit the growth of MCF-7 cells, possibly through regulating the cell cycle and suppressing cell migration and invasion.

고초균 유래 hydroxymethylbutenyl diphosphate synthase (IspG)의 이인산 부위와의 결합 특성 임상병리학과 방 유 진 지도교수 이 기 석 이소프레노이드는 모든 생명체에 존재하는 필수 대사산물로, 세균에 서는 주로 methylerythritol phosphate (MEP) 경로를 통해 합성된다. Hydroxymethylbutenyl diphosphate synthase (IspG)는 [4Fe–4S] 클러 스터를 포함하는 효소로서, MEP 경로의 마지막 전 단계 반응을 촉매한 다. 지금까지 IspG에 대한 연구는 주로 그람 음성균을 중심으로 이루어 졌으며, 그람 양성균 유래 IspG는 아미노산 서열 상동성이 낮아 잠재적 인 기능적 차이가 존재할 가능성이 제기된다. 본 연구에서는 그람 양성 균인 Bacillus subtilis 유래 IspG (BsIspG)를 대상으로 주요 기질 부분 인 이인산과의 결합 특성을 규명하고자 하였다. 열 안정성 분석(thermal shift assay, TSA)을 통한 BsIspG와 이인산 간의 해리상수(KD)는 12.68 mM로 산출되었으며, 이인산 농도가 증가함에 따라 BsIspG의 열 안정성 이 농도 의존적으로 증가함을 확인하였다. 상동 모델링 결과, BsIspG의 전체 구조는 상동 단백질들과 유사한 형태를 보였으며, 아미노산 서열 상동성이 상대적으로 낮음에도 불구하고 이인산 결합에 관여하는 잔기들 은 모두 보존되어 있었다. BsIspG와 이인산 간의 도킹 시뮬레이션에서 의 결합 자유 에너지는 –5.5 kcal/mol로 계산되었으며, R60, R106, K181, E209, S239 잔기들이 기질 결합 부위에서 이인산 결합에 관여할 것으로 예측되었다. 돌연변이 연구에서, N137A, E209A 및 S239A 돌연 변이체는 야생형과 유사한 KD 값을 보였지만, 이인산과의 결합효율은 약 1.5배 감소하여 이들 돌연변이가 이인산과의 결합에 약간 영향을 미 치는 것으로 나타났다. 특히, R60A, R106A, K181A 돌연변이체에서는 이인산 결합에 큰 영향을 미치는 것으로 확인되어, 해당 잔기들이 BsIspG의 기질 인식에 필수적인 역할을 수행함을 보여주었다. 종합적으 로, 본 연구 결과는 기질 모이어티로서 이인산 인식에 대한 이해를 확장 하는 BsIspG와 이인산 간의 상호작용에 대한 생화학적 통찰을 제공한 다. 주제어 : 이소프레노이드, MEP 경로, 고초균, IspG, 이인산, 기질 모이어 티
