      • Carboxyethylgermanium sesquioxide(Ge-132)가 항암제의 세포독성에 미치는 영향

        The cytotoxicity of carboxyethylgermanium sesquioxide(Ge-132) and anticancer drugs (cisplatin, mitomycin C, 5-fluorouracil) against tumor cell lines(L1210, SK-HepG-1) and mouse splenocytes was examined by MTT assay. Ge-132(8.5 x 10²㎕/ml, 1.7 X 10³㎕/ml, 3.4 x 10³㎕/ml) showed dose-dependent cytotoxicity against the tumor cell lines, on the contrary, increased cell proliferation against the normal mouse splenocytes. Combination of Ge- 132 with cisplatin(1.5㎕/ml, 15㎕/ml), mitomycin C(0.5㎕/ml, 1.0㎕/ml) or 5-fluorouracil (0.31㎕/ml, 1.25㎕/ml) showed additional cytotoxicity against L1210 and SK-HepG-1 cell lines, however, reduced clearly the anticancer drugs-induced cytotoxicity against the normal mouse splenocytes.

        뇌종양의 악성도 판정을 위한 분열기 세포측정에 관한 연구

        Tissue markers of cellular proliferation have been utilized as prognostic indicators in tumors of the central nervous system. Nucleolar organizer regions represent transcriptionally active sites of ribosomal deoxyribonucleic acid and can be identified by a simple argyrophilic technique. The authors describe a standardized approach to the assessment of these argyrophilic nucleolar organizer regions(AgNORs) in 25 astrocytomas(including 6 glioblastomas), 6 oligodendrogliomas and 1 xanthochromic astrocytoma, which were also estimated in association with 2-year survival post-operatively. Appropriate sections were silver stained and AgNORs were counted in 1000 cells. In cases of astrocytoma, the mean AgNORs/cell count of group survived more than 2 years after operation was significantly different from that survived less than 2 years : 1.93±1.13 vs. ≥ 4.79±3.90. And the mean AgNORs/cell count was 1.53±0.32 in low grade astrocytoma. 2.03±0.59 in grade Ⅲ astrocytoma, and ≥6.64±3.77 in grade Ⅳ astrocytoma. Among the patients with the same grade astrocytoma, the mean AgNORs/cell count was lower in the cases of long survival rather than short survival. The assessment of AgNORs can be easily performed by any surgical pathology laboratory without specialized equipment. It may be used as a valuable and independent prognosticator of biological behavior in cases of astrocytoma.

        This study is to represent basic datas of biomechanical work of human body which has measured precisely and datas on pathology and abnormal gaits paterns. 5 6th grader boys in Taejon city were attended in this experiment. Each student wore 8 landmarks on their lower leg joints and walked for 4 Km/h, 5 Km/h, and 6 Km/h on the tread mill. Mechanical energy of lower extremity segment by lower joint(thigh and shank) movement were measured in stance phase and swing phase by using 2 dimentional image analyzer, and which were evaluated energy of segment The conclusions were as fllowing; 1. Rotational kinetic energy change slightly in lower extremity segment, translational kinetic energy change in the biggest, and value of energy creat greater in swing phase than in stance phase. 2. Energy change in gait cycle increas in swing phase and decrease in stance phase. 3. Perfect energy change of lower extremity segment is not do.

        Several experiments were conducted to investigate the herbicidal character and phytotyxity of rice plants using P. C. P, O. C. P and prometryne. The experiments conducted were a laboratory test on the effect of inhibiting root growth of young radishes, a filed trial on the effect of weed control, plant growth and oield in Paddies, and a phytotoxity test on rice plants using pots. 1. Prometryne was less effective in inhibiting root growth of young radishes compared with P. C. P and O. C. P. 2. There appeared to be no difference in effectiveness of weed control in paddies among the four herbicides. And it seemed that all the herbicides tested showed no effect on the perennial weeds such as Eleocharis Kurogwai OHWI. 3. Prometryne showed severe phytotoxity and inhibited plant growth in the early stages, but this phytotoxity disappeared as plants grew in later stages and thus had no effect on the yield.

Increasing gait velocity cause toe off in early stage and swing of lower extremity happen early and quickly with angle of shank increasing in swing phase. 2. Children gait velocity grow rapidly above 5km/h, internal work of thigh mechanical energy showed value of work clearly and large. and internal work change represent greater in shank than in thigh 3. The stance period of gait is the period off set by two operated cocontracting muscles, and the later the speed of gait is, the lower the effect of the net torque.

        An Allium-odorum is difficult material to handling, because it is soft and weak as mechanical. For bundling work of allium-odorum, we must grip in the bundle shape of unit weight, however, it is difficult that grip allium-odorum by hand because the bundle bulk is very big. Especially, in packing work, the bundle shape of allium-odorum package than other vegetables is very important, because difference of a price becomes much according to the bundle shape. In this paper, we propose bundling mechanism for a quadrangle shape in bundle shape, and semi-automatic bundling mechanism that makes use of a eccentric roller and a triangularity link. We carry out mechanical modeling and analysing respectively using the CATIA V5 and SimDesigner. We manufactured the prototype of the semi-automatic bundling machine, and got satisfied result through field test. And this machine will be commercialized and announced soon.

      • 혈청 Fructosamine에 대한 임상적 유용성에 관한 연구

        Diabetes mellitus is a universal health problem and may occur at any age. The traditional biochemical measurements for initially detection patients with diabetes mellitus are random estimations of blood and urine glucose concentrations. Despite their common use both tests are fairly non-specific, being influenced by wide variety of condition. Measurement of the HbA_(1c) in a random blood specimen is a more simple screening test for diabetes, and widely used as an index of diabetic control. But unfortunately, it has difficulties with methodology. Fructosamine has been described as a new parameter for the estimation of glycosylated protein, It is based on the reduction potential of ketoamines in an alkaline medium and measures colorimethcally the concentration of all glycosylated semm proteins belonging to the class of fructosamine. It reflects the time-integrated glucose concentration over the previous 1 to 3 weeks and can be used for the follow up of diabetes mellitus. We measured serum fructosamine concentration and compared with any other items. The results are summehzed as follows : 1. Within-run and between-run imprecision were assessed by 20 analysis of 3 differential fructosamine concentration in control sera. The respective coefficients of variation were 2.9%, 6.6% in low concentration, 2.2%, 5.1% in medium concentration and 3.3%, 4.7% in high concentration. 2. The serum fructosamine concentration were 2.26±0.21 mmol/L in normal control subjects(n=29) and 3.35±0.65 mmol/L (n=62) in patients with diabetes mellitus (p<0.001). 3. In diabetic patients (n=62), the serum fructosamine concentrations correlated less significantly with glucose (Y(fructosamine) = 0.01×(glucose)+2.52, r=0.54, P<0.001) and more significantly with HbA_(1c) (Y(fructosamine) = 0.22×(HbA_(1c))+1.71 , r=0.63, P<0.001). 4. The concentrations of fructosamine in serum and EDTA plasma were not significently diferent (P<0.01). So, Plasma is as well suited to fructosamine determinations as serum(Y(serum) = 0.95×(Plasma)+0.25, r=0.96). 5. No interference was observed with lipemic sera (cholesterol) in detection of serum fructosamine(P=0.393).

      • 방선균 유전자 초고속 발현 시스템 개발

        방선균의 유전자는 그 구조의 특이성으로 외래 숙주에서는 발현이 거의 되지 않아 단백질 발현에 많은 어려움이 있다. 지금까지 다양한 벡터들이 방선균에서의 발현을 목적으로 개발되어 사용하여 왔지만, 이들은 유전자를 발현시키고자 할 때마다 발현 유전자의 특성에 맞게 벡터 및 유전자를 제조합시켜야 하는 불편함이 있었다. 따라서 본 연구에서는 방선균에서 쉽게 사용할 수 있는 두 가지의 초고속 발현 백터를 제작하였다. 제작된 pFSE와 pFSE-extra 벡터는 방선균에서 replication이 가능한 pIJ101 replication origin과 selection marker인 tsr(thiostrepton) 유전자를 가지고 있으며, 대장균에서도 replication이 가능한 shuttle vector 이다. 두 pFSE 벡터는 방선균에서 강력히 발현되는 강력 promoter ermEp를 가지고 있으며, pFSE-extra 벡터는 Streptomyces griseus에서 분비되는 단백질을 코드하는 유전자 sghC의 promoter 및 분비시그널을 포함하는 영역으로 구성되어 있어, 각각 하부에 연결된 유전자의 발현을 세포내에서 또는 세포 밖으로 분비하는 특성을 가지고 있다. 두 벡터의 발현시스템에는 AspEI으로 절단되는 제한 부위를 가지고 있으며, 효소로 절단 시 간단히 T-vector로 이요할 수 있는 초고속 발현 시스템이다. The genes of streptomycetes are hardly expressed in other host systems because of their unique structure. Many expression systems for the streptomycetes genes have been developed, yet modification of vector and target genes is the essential prerequisite, which make a limit in their use. To make expression easier in efficient and fast way, two kinds of expression systems, named pFSE and pFSE-Extra, were constructed, Both plasmids have the repication origins from pIJ101 and pBlescript and can be used as Streptomyces-E. coli shuttle vectors. The plasmid pFSE has the strong promoter derived from ermE and designed for the intracellular expression of streptomycetes genes in Streptomyces host system. pFSE-extra has the promoter region including the secretion signal originated from the sghC gene that was identified to encode massively secreted protein from Streptomyces griseus and can be used for the secretion of the expressed protein in Streptomyces also. Both plasmids can be used as T-vector cloning system after digestion with AspEI restriction enzyme, which make their application broader as the fast expression system in Streptomyces.

        유럽연합 (EU) 자료 출판 및 이용에 관한 연구

        At present, international society is in very rapidly changed situation with a new relation of cooperation and companion. As a huge community for economic profit, European Union(EU) is a significant international organization, which has an important influence on international economy and political order. EU information has been accepted as useful accademic and trade information reseources related to various areas, politics, economics, international relations, government of many countries. Also, these information has distributed to not only member nations but also non-member countries through a depository program of the EU Commission. The purpose of this study is to make an awareness of and uses of the information from the EU through the investment of EU information product system and the analysis of usage in Korea. Information activites of the major organizations of EU have been examined with a special policies to the Commission of the European Union. The Commission is the EU institution from which the majority of the EUs documentation originates. The information functions of the Commission have been studied in such categories as information production and distribution authorizing services and information service to public. Also, this research, interview method and questionnaires used for 5 depository centers and 40 non-depository institutes. The followings are the findings: 1. Most depository centers were at the university since 1990s. And they have a study of the EU or related research. 2. EU materials in depository centers were not informed well and had not a special librarian for systematic control of information. As a result, a few people have been used it. 3. The majority of non-depository institutes has purchased the EU materials since 1990s and 70% of them want to increase acquisition of the EU materials in the future. However, they have many difficulties to access and use EU materials for a lack of publication information. To solve current problems and establish a plan of the utilization of EU information, each centers perform the active promation, analysis of EU materials, support and train of EDC librarians. And, current study of EU information and their development is needed.
