Polyphyllin D (PD), an active component from a traditional medicinal herb Paris polyphylla, which has long been used for the treatment of cancer in Asian countries, has been found to hold significant antitumor activity in vivo or in vitro. However, there were few reports on the effects and underlying mechanism of PD on apoptosis in U87 human glioma cells. The present study was conducted to evaluate apoptotic induction of PD in U87 human glioma cells, and explore its underlying pathway. U87 glioma cells were cultured and treated with varied concentrations of PD (from 10- 8 to 10- 4 M). The inhibition of U87 glioma cell proliferation by PD was assessed by MTT assay. The apoptosis of U87 glioma cells was detected by flow cytometry, and western blot analysis was used to examine human B-cell leukemia/lymphoma 2 (Bcl-2), human Bcl-2 associated X protein (Bax), caspase-3, total-c-jun NH2-terminal kinase (t-JNK), and phosphorylation-JNK (p-JNK) protein expression in U87 human glioma cells. The treatment with PD for 24 h significantly inhibited the proliferation of U87 human glioma cells in a concentration-dependent manner. PD increased apoptosis and significantly upregulated the expression of Bax, caspase-3, and p-JNK associated with apoptosis, but downregulated antiapoptotic Bcl-2 expression in U87 human glioma cells. Our data provided evidences that PD induces apoptosis in U87 human glioma cells. This effect might be associated with the JNK pathway.

Anticancer effects of kimchi fermented for different times and containing added ingredients in HT-29 human colon carcinoma cells were studied. Optimally ripened kimchi showed higher anti-proliferation activities than fresh and over-ripened kimchi. Anticancer-ingredientsadded kimchi exhibited higher activities than standardizedrecipe kimchi in HT-29 cells. Optimally-ripened anticancer kimchi suppressed expression of Bcl-2 and promoted expression of Bax, caspase-9, and caspase-3 to promote apoptosis, and reduced mRNA expressions of proinflammatory factors, exhibiting the strongest anticancer effect. Fermentation and ingredients are important factors to increase in vitro anticancer functionalities.
Antiobesity Effects of Bamboo Salt in C57BL/6 Mice

Antiobesity effects of bamboo salt (BS) were evaluated compared with those of purified salt and solar salt by oral administration in a diet-induced obesity model using C57BL/6 mice. Compared with other salts, BS, especially nine times baked BS (BS-9Ⅹ), significantly reduced body weight, food efficiency ratio, and weights of epididymal adipose tissue and liver in high-fat diet–fed mice. Furthermore, BS suppressed the expression of adipogenic factors, such as CCAAT/enhancer binding protein alpha (C/EBPα), peroxisome proliferator-activated receptor gamma (PPARγ), and sterol regulatory elementbinding protein 1c (SREBP-1c). Therefore, BS may suppress obesity by downregulating adipogenesis.

Objective: To evaluate the feasibility of diffusion-derived vessel density (DDVD) for the preoperative prediction of lymphovascular space invasion (LVSI) in endometrial cancer (EC) and to explore whether the integration of DDVD with intravoxel incoherent motion (IVIM)-derived parameters and clinicopathologic (CP) variables could improve the prediction. Materials and Methods: This exploratory retrospective study included 194 patients with histopathologically confirmed EC (126 LVSI-negative and 68 LVSI-positive) who underwent preoperative multi-b-value diffusion-weighted magnetic resonance imaging. The apparent diffusion coefficient, true diffusion coefficient (D), pseudodiffusion coefficient (D*), perfusion fraction (f), and DDVDb0b10 (derived from b = 0 and 10 s/mm2 images) were calculated. Univariable and multivariable logistic regression analyses were performed to identify independent predictors of LVSI and to develop predictive models. The predictive performance was assessed using receiver operating characteristic (ROC) curve analysis. Differences in areas under the ROC curves (AUCs) among models were evaluated using DeLong’s test, with the Bonferroni correction applied to adjust for multiple comparisons. Results: The f, DDVDb0b10, and three CP factors, including the depth of myometrial invasion, histologic grade, and tumor stage, emerged as independent predictors of LVSI. DDVDb0b10 yielded a higher AUC than f (0.772 vs. 0.692), although the difference was not statistically significant after Bonferroni correction (adjusted P = 1.000). Predictive performance improved significantly when DDVDb0b10 was combined with the retained CP factors (AUC, 0.895 vs. 0.772; P < 0.001). The model combining the f, DDVDb0b10, and CP predictors achieved the highest AUC of 0.904, although it was not significantly different from that of the DDVDb0b10 + CP model after Bonferroni correction (adjusted P = 1.000). Conclusion: DDVDb0b10 may be a feasible imaging biomarker for the preoperative prediction of LVSI in EC. Multivariable approaches integrating DDVDb0b10 with CP features and IVIM-derived f may enhance preoperative risk stratification in EC. Further prospective multicenter validation is warranted.

The comparing of shuidouchi, natto, and cheonggukjang were evaluated using physicochemical experiment, Ames test, and in vitro anticancer experiment. Cheonggukjang contained the highest amount of aroma components of amino and ammonia type nitrogens and the lowest amount of γ-glutamyltranspeptidase. Cheonggukjang showed the better sensory qualities than shuidouchi and natto. Three short-term fermented soybean foods were associated with high degree of antimutagenic activities. At a concentration of 2 mg/mL, the growth inhibitory rate of TCA8113 cells treated with cheonggukjang determined by a MTT assay was reduced by 78%; this rate of inhibition was higher than that achieved with shuidouchi (67%) and natto (55%). Cheonggukjang significantly induced apoptosis in cancer cells by upregulating Bax, caspase-3, and caspase-9, and downregulating Bcl-2 by RT-PCR and Western blotting. Our results demonstrated that cheonggukjang had the most potent in vitro anticancer effect, induced apoptosis as well as in terms of physico-chemical properties.
