천아오린 Gachon University 2022 국내박사
Research on higher education is related to national development and national progress, and is an important issue that China needs to solve urgently under the new normal. Moocs, as a new education model recognized by China's higher education circle, has gradually risen to the height of guaranteeing the future development of the country, and become the general trend of the development of the world's education industry. Art has now developed into an independent comprehensive discipline in China, among which the MOOC education of dance has become an increasingly important direction in the research on the cultivation of dance art talents in universities. The construction of MOOCs for dance majors in Chinese universities should not only highlight the two-way combination of theory and practice, but also be treated as a long-term dynamic research. Especially in the face of the complex situation of the current global novel coronavirus pneumonia epidemic, the MOOC teaching of dance majors in Chinese universities has shown the characteristics of relative lag and imperfection, clarify and integrate the element resources of its internal environment and external environment. For this reason, this research will explore the construction path of MOOCs for dance majors in Chinese universities from the perspective of constructivism, take representative universities in China that offer dance majors as the research objects, analyze their current situation and existing problems, and realize research with clear goals. The main research contents are as follows: First of all, combined with research needs, the definition and characteristics of dance MOOC are established by sorting out the concept and connotation of MOOC, and the related theories of MOOC construction are expounded and interpreted, so that practitioners can track both theoretical reference and practical operation. It can be followed, which will lay the foundation for the continuous improvement of the "quality" and "quantity" of the MOOCs for dance majors in universities in the future. Secondly, using the dance MOOCs on the Chinese MOOC platforms of the nine major Chinese MOOCs as research samples, the information and data of the platform, proportion, content distribution, teaching units, number of delivery and teaching mode are collected. The purpose is to It clearly depicts the overall development of the current Chinese dance professional MOOC. Thirdly, comprehensive universities under the overall management of the Ministry of Education of China - Minzu University of China, normal colleges - Capital Normal University, art colleges - Beijing Dance Academy, higher vocational colleges - Guangdong Foreign Languages Art Vocational College as an example, analyzes in detail from the five dimensions of MOOC teaching environment, teacher structure ratio, professional characteristics division, classroom content display and after-class evaluation mechanism. The reason is that in the five major fields of Chinese folk dance, dance teaching principles, Chinese classical dance, dance history and theory, and children's dance divided by the Chinese dance academic circle, the teaching results obtained by these four types of colleges have been recognized by the industry. It is generally recognized as the most plentiful and authoritative and leading, especially the distinctive features that each shows in the current construction of Chinese dance professional MOOCs is quite representative. For example, the Chinese ethnic and folk dance courses offered by the Minzu University of China have always been in a demonstration and leading position in China, and the demonstration and construction of them in the form of MOOCs are still effective. It is unique; the content explanation and analysis of the dance education ontology teaching method and principle MOOC offered by Capital Normal University is the clearest and most detailed. In addition, the person in charge's own construction experience and related academic resources are rich, making the dance professional MOOC in its colleges and universities. The construction results are currently the most comprehensive in Chinese universities; the construction of Chinese classical dance teaching and dance history and theory MOOCs of Beijing Dance Academy has continued the advantages of its strong disciplines, so it is very important for the construction of corresponding MOOCs in other Chinese universities. Important reference value; the dance MOOC construction of Guangdong Foreign Language Arts Vocational College fully considers the teaching audience of its own universities, and creates a relevant MOOC course centered on children's dance teaching and creation, which reflects the targeted The nature and practicability provide construction directions and ideas for dance MOOCs in other higher vocational colleges in China. At the same time, these four types of colleges also have common phenomena in the construction of MOOCs for dance majors in Chinese universities, which are confirmed by the results of in-depth interviews with the leaders and participants of MOOC construction in various universities in this study. For example, the MOOC teaching environment needs to be improved in terms of top-to-bottom policy support, updating of teaching equipment, and professional talent training; and other issues are also more prominent. The division of professional characteristics is not clear and reasonable, resulting in the inability to highlight the professional characteristics of dance MOOC construction in colleges and universities, and lack of innovation. In terms of classroom content display, most of them continue to use the offline course model, failing to incorporate learners' psychological acceptance and satisfaction into the consideration of the process of constructing classroom content, and failing to make good use of the relevant advantages of MOOCs. As a result, the presentation of classroom content is the same and boring. The after-class evaluation mechanism also relies too much on the evaluation template set by the MOOC platform, the interaction with learners after class is not active enough, the after-class discussion is more perfunctory, and the evaluation results are not paid enough attention. Compared with the construction of MOOCs for dance majors in other colleges and universities, these phenomena are typical to a certain extent. They can comprehensively outline the current situation of MOOC construction of dance majors in Chinese universities, and then find out the problems. Finally, based on the constructivist learning theory, this study attempts to discuss the construction strategies of MOOCs for dance majors in Chinese universities, in order to provide reference and quantitative basis for the construction strategy direction selection, policy formulation and follow-up research practice of dance majors in Chinese universities. Carry out rigorous and objective analysis from different dimensions, put forward corresponding improvement plans, and provide ideas for managers and future research.

With the advent of the information age, the rapid development of the Internet of things and the rise of big data make everything possible in the 21st century. All this is inseparable from the development and innovation of science and technology. However, the progress of science and technology has not brought the synchronous development of social morality. A series of social problems caused by the lack of morality, especially the dishonesty, eager for quick success and instant benefit in academic research in Colleges and universities Ignoring people's safety behavior has become the focus of attention. Therefore, the study of ethical leadership in Colleges and universities is not only a profound theoretical problem, but also an urgent practical problem. The rise and development of ethical leadership is conducive to the healthy and sustainable development of organizations and plays a positive role in promoting social stability and development. Then, what effect the urgently needed ethical leadership in the organization will have on the innovative behavior of middle school students, and what is its mechanism, which are worthy of our in-depth study. Under the theoretical framework of organizational behavior, this paper makes an empirical study on the mediating rolebetween ethical leadership and graduate innovation behavior. After a questionnaire survey of 300 teachers and students, this paper makes an empirical analysis ofthe data and draws a conclusion. The specific ideas are as follows. Firstly, find the direct impact of ethical leadership on graduate students' innovative behavior. Secondly, it test the direct effect of ethical leadership on graduate students' trust. Then it discusses the impact of trust on graduate innovation behavior. Finally, we anlaysis the mediating effect of graduate trust. The conclusion shows that trust plays a complete mediating role in ethical leadership on graduate students' innovative behavior. It shows that under the background of China's unique moral culture, inheriting and carrying forward ethical leadership has important theoretical and practical significance. The research results ofthis paper are as follows: First, Ethical leadership has a positive impact on students' innovative behavior, trust and students' innovative behavior. Therefore, in daily academic research practice, in order to make students innovate continuously, we should pay attention to persuading people with morality in the way of leadership, have full respect and tolerance for students, pay attention to openness in the way of communication, and make the communication smooth whether from top to bottom or from bottom to top. Second, Ethical leadership has a positive impact on students' trust. This shows that ethical leadership has a great impact on students' individual psychological motivation and organizational interpersonal communication, and moral leaders can promote the formation of higher leadership ability and improve the operation efficiency of the organization. Therefore, in the daily academic research process of colleges and universities, the moral factors of leaders should be required as an important assessment index to strengthen the moral quality of leaders. Thirdly, through data verification, this paper shows that student’s trust plays an mediating role between ethical leadership and students' innovative behavior. Therefore, in the future academic research practice, we should pay attention to these two factors and strengthen the construction of students' trust through a series of academic research measures. For example, we can select students with high trust and strengthen the cultivation of students' trust through the trust oriented assessment system. In addition, in the daily communication process, we should strengthen the openness ofcommunication, so that communication can be unimpeded from top to bottom or from bottom to top. This paper studies the impact of ethical leadership on students' innovative behavior under the unique cultural background of China, which is highly targeted, ethical leadership should be better developed in its own soil. Chinese colleges and universities should also cultivate a group of Chinese ethical leaders according to their own cultural characteristics and carry forward the traditional academic research culture, So as to create the unique cultural characteristics of Chinese colleges and universities. These are the uniqueness of this paper. However, a series of problems, such as how ethical leadership will affect other factors in the process of academic research in Colleges and universities, how to effectively train moral academic researchers, and how to develop moral measurement questionnaires with Chinese characteristics, still need our continuous research.

Abstract Research on the Impact of Environmental Regulation on Enterprises Green Innovation Zhang Xinwen Supervised by Prof. Zhi Dalin Dept.of Global Business & Entrepreneurship Graduate School of Gachon University Environmental protection has always been a severe challenge to China's social and economic development. However, in recent years, the CPC Central Committee, the State Council, and relevant departments have issued many important decisions and arrangements, strengthening the important position of environmental protection in national development, and China's environmental protection cause has achieved significant progress. However, at the enterprise level, the further introduction of environmental regulation policies also has an important impact on enterprise development, and the impact of environmental regulation on enterprise green innovation has become a hot issue in the academic circle. Based on existing literature and relevant theories, this paper systematically studies the main factors influencing enterprises' green innovation ability from the perspective of micro-enterprises; and discusses the influence mechanism of environmental regulation, social media attention, and rent-seeking factors on enterprises' green innovation ability. At the same time, this paper deeply analyzes the threshold effect of government environmental regulation on enterprise green innovation. Therefore, this paper constructed a theoretical model including environmental regulation, media attention, corporate rent-seeking, and corporate green innovation, and combined with the collected empirical data to test the relevant theories, in order to verify the hypotheses proposed in the paper. In this paper, environmental regulation, enterprise green technology innovation, and the impact of environmental regulation on enterprise green innovation are briefly analyzed, and the relevant theoretical results are summarized. Then, the paper defines the concepts and categories of environmental regulation and enterprise green innovation and puts forward the research hypothesis based on the variables selected from system theory, reputation theory, and rent-seeking theory. Finally, using the statistical data of China's heavy pollution industries from 2010 to 2019, the panel regression method is used to test the research hypothesis proposed in this paper to explore the impact of environmental regulation on enterprise green innovation and its threshold effect. Meanwhile, the moderating effect of media attention and rent-seeking on green innovation is studied from two aspects: external focus and internal choice. The results show that the government's environmental regulation has an important impact on the green innovation of heavily polluting enterprises in China. The specific research results are as follows: (1) Different types of environmental regulations have different effects on enterprise green innovation. Command-based and market-based environmental regulations have a significant promoting effect on green innovation, while public-participatory environmental regulations have no significant impact on green innovation. (2) The intensity of different types of environmental regulations has different impacts on enterprises' green innovation. The impact of command-based environmental regulations on enterprises' green innovation presents an inverted U-shaped relationship, while that of market-based environmental regulations does not exist this structural relationship, which is mainly still a linear relationship. (3) Under command-based and market-based environmental regulation, media attention plays a positive moderating role between government environmental regulation and enterprise green innovation. (4) Internal rent-seeking not only has a negative moderating effect between command-type environmental regulation and enterprise green innovation, but also has a negative moderating effect between market-type environmental regulation and enterprise green innovation, but its moderating effect on public participatory environmental regulation is not significant.

ABSTRACT Synergistic anti-proliferation effects of all-trans retinoic acid with paclitaxel on autosomal polycystic kidney disease cell line. Que Thanh Thanh Nguyen Supervisor: Prof. Jae Young Kim Department of Life Science Graduate School of Gachon University Autosomal dominant polycystic kidney disease (ADPKD) has been well-known as a monogenic inherited disorder and characterized by cyst development in the kidney. In recent years, the mechanism in autosomal dominant polycystic kidney disease (ADPKD) associated to genetic factors and non-genetic factors have been widely studied to get deep knowledge about ADPKD. In renal tubules, renal epithelial cells over-growth due to lower intracellular calcium level and the mitogenic effect of cAMP (cyclic adenosine monophosphate), is one of the hallmark features that originated cystogenesis. In this study, for the first time, we investigated the synergistic effects of combinatory treatment between all-trans retinoic acid (ATRA), an active form of vitamin A, and paclitaxel, a high effective anticancer drug, on the autosomal dominant polycystic kidney cell line. Cells were treated with a combination of ATRA with paclitaxel for 48 h and then analyzed by cell viability assay. In combinatory treatment groups, our data indicated that the percentage of cell viability was significantly decreased in the comparison with DMSO (control) and ATRA/paclitaxel (alone) treatment. Cell cycle arrest showed that the combinatory treatment of ATRA with paclitaxel induce ADPKD cell line arrest at the G2/M phase. Furthermore, apoptotic cells were detected in our study by Annexin V/PI staining. The up-regulation of p53 protein expression and caspase activation could be involved in apoptosis signal and cell cycle arrest. We revealed that the intra-calcium level was sharply increased by combined treatment of ATRA and paclitaxel after 72 h of treatment and this enhance was blocked by RARα/RXRα antagonist or calcium channel blockers. Besides, ADPKD cell proliferation was controlled through the ERK signaling pathway by in phosphorylated ERK. Our result suggested the function of combinatorial treatment of ATRA with paclitaxel which expresses synergistic anti-proliferation effect on ADPKD cell proliferation by inducing cell cycle arresting, apoptosis and suppressing the ERK signaling pathway. Keywords: Autosomal polycystic kidney disease (ADPKD), ATRA, paclitaxel, cell cycle arrest, apoptosis, intracellular calcium, phosphorylated ERK1/2.

(The) impact of work family conflict on the career growth of female employees in Chinese context
Zhang, Xiao Gachon University 2022 국내박사
Abstract The impact of work family conflict on the career growth of female employees in Chinese context Zhang Xiao Supervised by Prof. Yi Yang Dept. of Global Business & Entrepreneurship Graduate School of Gachon University In recent years, the career growth of employees is more and more important. It is worth noting that gender differences tend to fade in the competition for talents compared with the last century. With the rapid development of society, more and more women are becoming independent, which is especially reflected in the continuous improvement of women’s participation rate in the social labor force. The career growth of female employees also cannot be ignored. In the modern society, the expectations and requirements for women's roles are increasing. They are not only expected to play the role of good wife and mother, but also they are expected to make contributions at work. Therefore, the women face an increasingly serious dilemma between family and work. In China, there has been a cultural concept of male superiority over female superiority for a long time because of the Confucian culture. Based on the still dominant traditional division of gender roles, the burden of family education and domestic affairs still falls mainly on women. With the aging of the population, the comprehensive implementation of the two-child policy, the introduction of the three-child policy and the rising proportion of dual-worker families and other social phenomena tend to be normal, the female employees’ work and family roles demand pressures continue to increase. It is hard for them to balance work and family roles with the limited time and energy. Thus, the work family conflict problem is becoming more and more seriously. The serious work family conflict not only affects the women’s physical and mental health, but also it hinders the career growth of female employees. For the current research, the female employees’ career growth from a role conflict perspective research is still inadequate. It is necessary to expand the research. In view of this, this study uses the existing theories to answer and explain the work and family problems faced by female employees in the current society. The study included 2 sub-studies. In the first study, we explored the cross-effects of work family segmentation preference on work family two-way conflict and work family facilitation among female employees based on boundary theory. With the development of the mobile internet technology and the wide popularity of all kinds of convenient mobile devices, the boundary between work and non-work fields is increasingly blurred, and the interaction between the two fields is more close than before. Family is a very important non-work area in people’s life, and the interaction between work and family is complicated. There are both negative and positive, the negative relations, namely “work family conflict”, and positive relations, namely “work family positive spillover”. In this study, we study the effects of preferred segmentation of the work domain from the family domain on women’s work family two-way conflict and work family facilitation. In the second study, the data of 331 female professional employees were collected from the perspective of work family two-way conflict. The factors such as organization support, family support, job involvement and female career growth are taken as the moderating, mediating and outcome variables of the study. A model is established to analyze the direct influence mechanism of work family conflict of two-way influence on women’s career growth. Firstly, it integrates and sorts out various variables, and fully combines the research status, puts forward the research hypothesis, and establishes the theoretical model on this basis. Secondly, a small sample survey questionnaire is prepared, and we make the final questionnaire on the basis of the initial small sample survey questionnaire. Then, we distribute the questionnaire on line in order to collect the data . Finally, using the SPSS22.0 data processing tool to test the reliability, validity and other aspects of the effective questionnaire, analysis the data and vertify the hypothesis step by step. The results showed that: (1) Work family segmentation preference is negatively related with work family conflict. (2) Work family segmentation preference is negatively related with family work conflict. (3) Work family segmentation preference is positively related with work family facilitation. (4) Work family two-way conflict has a significant negative impact on the career growth of female employees. (5) Job involvement plays a mediating role in the influence mechanism of work family two-way conflict and career growth of female employees. (6) The relationship between work family two-way conflict and job involvement can be directly influenced by the moderating effect of family support and organization support. This study suggests that female employees can adjust themselves to balance work and family, reduce work family conflict and improve work family facilitation. Family support from spouse, parents and relatives can well alleviate work family conflict, so that female employees can devote themselves to work and contribute to their career growth. Enterprises should actively care about and attach importance to the career growth of female employees, provide fair career growth conditions and environment for female employees, take targeted actions, increase social humanistic care for female employees, and further build policies in line with the career growth of female employees.

There is a certain value tension or conflict between economic globalization and cultural pluralism. One of the critical problems facing China's urbanization is the scarcity of cultural resources. The biggest problem we face is how to transform intangible cultural heritage into cultural resources and cultural capital that can support the globalization and modernization of cities and transform national and regional intangible cultural heritage into global cultural resources with universality. Kite culture is not only widely used for flying, competition, and entertainment but also has taken a big step into the hall of art and become an artwork, collection, and tourist souvenir to beautify people's life. As the "Kite Capital of the World," Weifang City, driven by the influence of the Weifang International Kite Festival, has expanded the types and styles of kite sports, made a qualitative leap in the dissemination of kite culture, and greatly improved the development of kite industry. As a traditional Chinese folk art, kite flying is an intangible cultural heritage of China, full of Chinese traditional spirit and the memories of many people's childhood. However, compared with the vigorous development of kite flying practice, the original living environment of kite flying art no longer exists today. The road of inheritance, protection, and exploitation faces many difficulties. This requires the academic community to re-examine the heritage protection and exploitation of contemporary kite flying. Then, how should intangible cultural heritage resources be better preserved, developed, and utilized under the global cultural preservation perspective? What kind of heritage protection and exploitation effect should be achieved in this process? This point needs to be explored and investigated more deeply. For this reason, this paper selects Weifang kites as the object for empirical research and mainly uses research methods such as visiting and research, literature research, comparative research, and mathematical and statistical analysis, and combines theories of global cultural conservation, cultural change theory and digital survival of intangible heritage to conduct an in-depth exploration on the heritage protection and exploitation of Weifang kites cultural resources. Cultural heritage resources of Weifang; the general and unique characteristics of Weifang kites as intangible cultural heritage resources are discussed; the current situation of inheritance protection and exploitation of intangible cultural heritage resources at home and abroad is sorted out; taking Japanese and Korean kites and Weifang kites as examples, and taking Weifang, the international capital of kite culture, as the representative The kites of Weifang are compared with those of Beijing, Tianjin, and Nantong, and the unique features and problems of Weifang kites are analyzed and summarized. The research results show that although the Weifang kite intangible cultural heritage resources under the Global Cultural Conservation vision have achieved some positive results in the inheritance, protection, and exploitation, under the current new development situation, the inheritance, protection, and exploitation of the Weifang kite intangible cultural heritage resources still have some hidden dangers in many places, which may lead to the protection crisis of cultural, ecological inheritance in the long run. After cognitive analysis, it is found that this situation is mainly because there are still different levels of problems in the inheritance protection and exploitation of Weifang kite intangible cultural heritage resources in terms of inheritance complex, cultural genes, propaganda efforts, and digital dissemination. Under the circumstance that the independent development of the local kite genre is restricted, the concept of global cultural conservation is established, and the spatial and temporal tension of kite art dissemination is expanded to seek a new development way out. Combining the preliminary interviews and research, under the guidance of global cultural conservation theory, cultural change theory, cultural capital theory, industrial integration theory, digital survival, and sustainable development theory of intangible heritage, the author proposes that the spiritual connotation of intangible heritage resources should be deeply explored, policy support should be increased to promote the development of intangible heritage, the diversified system should cultivate inheritance talents, precise positioning of kite cultural heritage brand marketing, implementation of The author proposes countermeasures such as diversified marketing and integrated communication to create awareness and digital experience center to deeply explore the inheritance, protection, and exploitation of Weifang kites and let the kite culture shine, and also provide new ideas for the inheritance, protection, and exploitation of related intangible cultural heritage categories. With the increasing globalization of the business environment, the acceleration of technological progress and the evolution of consumer behavior, the key factors determining the enterprise survival and development are no longer technology and product innovations, but business model innovation (Johnson et al., 2008; Schneider and Spieth, 2013). The Chinese government also noted in the Outline of the National Innovation-Driven Development Strategy released in 2016, it is essential to “encourage enterprises to carry out business model innovation" to achieve national innovation-driven development. In the complex and rapidly changing environment, how to break through the new disadvantages of small and medium-sized enterprises to carry out business model innovation has become an urgent problem to be solved. Clayton Christensen, the author of disruptive innovation theory, believes that resource allocation will have an impact on disruptive business model innovation, and it is better to set up independent small organizations to implement destructive strategies (Christensen et al., 2013). Therefore, it is suitable for small and medium-sized enterprises to seek business model innovation through entrepreneurship learning. In recent years, scholars have began to study the influence of entrepreneurial learning on enterprise business model innovation from different perspectives, scholars from all over the world came to realize that entrepreneurial learning for enterprise business model innovation and promoting the enterprise business model innovation is of great significance. Plenty of research in this field has been done and entrepreneurial learning has gradually become an important means to promote innovation, improve the competitive advantage of enterprises and promote enterprise business model innovation. In view of the above problems, business model innovation is "an activity in which an enterprise develops entrepreneurial opportunities to achieve value creation by reforming or reconstructing elements or themes of the activity system (Guo Hai and Shen Rui, 2014)". Entrepreneurial orientation and entrepreneurial learning play an important role in promoting enterprise business model innovation and enhancing enterprise business model innovation ability. It is a common concern of the enterprise community to explore the mechanism of entrepreneurial orientation and business model innovation of SME, and strive to find ways and methods to effectively improve entrepreneurial learning. It can be seen that the business model innovation of SME can be realized through entrepreneurial learning, while the knowledge and creative thinking required by entrepreneurship learning need to be obtained through entrepreneurial orientation. Based on above statements, this research in entrepreneurship theory, entrepreneurship learning theory, enterprise business and destructive innovation theory on the basis of the innovation theory, through the review and sorting of relevant literature, combining with the practice of small and medium-sized enterprises, entrepreneurship orientation, entrepreneurship learning and business model innovation into a theoretical framework, deeply explore the mutual influence between variables and boundary conditions. On the issue of the business model innovation of small and medium-sized enterprises, on the one hand, China is learning from the mature experience model of developed countries and striving to provide a suitable environment for small and medium-sized enterprises, and on the other hand, entrepreneurs are also actively exploring the sustainable development of small and medium-sized enterprises with Chinese characteristics. For the above purposes, this study mainly includes the following sections: First, we review and sort out the literature and research related to basic theories such as entrepreneurial orientation, entrepreneurship learning and business model innovation. In the process of sorting out the entrepreneurship-oriented theory, the relevant theoretical development process of entrepreneurship and entrepreneurship orientation are elaborated in detail, and the connotation and definition of entrepreneurship orientation are defined. The development of entrepreneurship learning theory is expounded in the part of entrepreneurship learning theory. In the theoretical research related to destructive innovation, the entrepreneurial study theory and destructive innovation theory are combined together, discussing the entrepreneurial study behavior suitable for small and medium-sized enterprises in China, which enriches the entrepreneurial learning theory and destructive innovation theory. Second, the correlation of entrepreneurial orientation, entrepreneurship learning and enterprise business model innovation are analyzed, and the dimensions of each variables are determined. It analyzed the relationship between entrepreneurial orientation and its sub-dimensions and enterprise business model innovation, entrepreneurship orientation and sub-dimensions and entrepreneurship learning, and the relationship between entrepreneurial learning and enterprise business model innovation. In addition, the intermediary role of entrepreneurship learning in the relationship between entrepreneurship orientation and enterprise business model innovation is also discussed, and formed the theoretical framework. Third, practical research conducted. The questionnaires of this study were distributed and collected on site, and a sample of senior leaders in small and medium-sized enterprises and students studying for MBA and EMBA at Jilin University. A total of 271 questionnaires were issued, with 228 recovered at 84%. Among them, 201 were valid with an effective rate of 88%. Various studies have shown that nonalcoholic fatty liver diseases (NAFLD), the aggregation of lipid droplets in the liver (hepatosteatosis) followed by inflammatory complication, appears to be chronic consequences of overnutrition. In the present research, we examined the anti-hepatosteatosis effects of fermented Cordyceps militaris (CM) extracts in AML-12 hepatocytes as well as in mice with short-term high-fat diet (HFD) via activation of fatty acid oxidation (FAO). The chemical components, adenosine and cordycepin, of CM extract (CME), CM grown on germinated soybean (GSCE) and fermented GSC by Pediococcus pentosaceus ON188 (ON188E) were assessed. In hepatocytes, the expression of CPT1α and PPARα, FAO genes were upregulated only by ON188E treatment. In contrast, SCD1, a lipogenic gene was downregulated by ON188E within no cytotoxic range. Regardless of oleic acid treatment to mimic fatty liver condition, mRNA and proteins of FAO genes were upregulated by ON188E in a dose-dependent manner. Formation of intracellular lipid droplets by addition of oleic acid was reduced by ON188E to the levels in WY14643-treated cells. When cells were treated with ON188E, SPHK2 and its phosphorylated product sphingosine 1-phosphate (S1P) elevation activated FAO in hepatocytes. In consistent with this, in animal results, ON188E extract did not alter the food intake while body weight and inflammatory factors in plasma of HFD mice were controlled comparable to those of NCD mice. Moreover, ON188E treatment significantly upregulated hepatic SPHK2 level in mice fed HFD. The elevated SPHK2 activated hepatic FAO genes such as CPT1α and PPARα but downregulated DGAT2, a triglyceride biosynthetic gene. We found that ON188E treatment reduced lipid droplets in liver and the hypertrophy of adipocytes in epididymal white adipose tissue (WAT) in a dose-dependent manner. Activation of SIRT1and PPARγ in WAT suggests that ON188E could modulate browning process of adipocytes which induces body thermogenesis. Collectively, the fermented CM extracts not only reduce risk of tissue damage but activate hepatic FAO via elevation of S1P synthesis in hepatocytes and induce WAT browning as well. This suggests ON188E can be applied as a potential functional food preventing obesity and hepatosteatosis.
