Intercellular communication occurs not only through direct cell–cell contact or secreted signals such as cytokines but also via nanoscale vesicles known as extracellular vesicles (EVs). EVs carry membrane proteins derived from the plasma membrane, cytosolic proteins, and various nucleic acids, enabling them to play an important role in the transmission of physiological and pathological information. In this study, we explored strategies to enhance the delivery efficiency of EV-encapsulated cargos through EV engineering, as well as approaches that employ virus-derived fusogenic proteins to improve cytosolic delivery. [Paper Ⅰ] Efforts to utilize EV as therapeutic delivery vehicles have steadily progressed, and recent advances in genetic engineering have enabled the active loading of functional genetic cargos such as mRNA and transcription factors into EVs. However, once internalized by recipient cells, these cargos are frequently sequestered in endosomes and subsequently degraded through lysosomal pathways, leading to substantial loss and markedly reduced delivery efficiency. This challenge has highlighted the need for strategies that enable EVs to achieve endosomal–lysosomal degradation escape. Approaches to express virus-derived fusogenic proteins, such as VSV-G, on EV membranes have therefore been explored. In this work, we review diverse endosomal–lysosomal escape strategies developed to date, the analytical methods used to quantitatively assess their effectiveness, and how these approaches may be applied to the development of EV-based therapeutics. [Paper Ⅱ] Building upon these insights, this study further designed an antitumor EV (bRSVF-EV) that carries misfolded proteins—typically considered cellular debris—and expresses the respiratory syncytial virus fusion protein (RSVF), enabling these vesicles to evade endosomal–lysosomal degradation. Treatment of RSVF-expressing HEK293T cells with bafilomycin A1 inhibited lysosomal function, thereby inducing the intracellular accumulation of misfolded proteins and facilitating their incorporation into EV. The resulting bRSVF-EV selectively transplanted xenogeneic antigens onto cancer cell membranes through a nucleolin-dependent mechanism, eliciting potent innate immune activation. Moreover, by escaping the endo–lysosomal pathway, these EV directly delivered misfolded proteins into the cancer cell cytosol, triggering endoplasmic reticulum stress and inducing immunogenic cell death (ICD). This mechanism produced strong antitumor immune responses in murine tumor models, and combination therapy with PD-1 blockade further extended survival, with some mice achieving complete remission. In summary, this dissertation provides a literature-based evaluation of EV-mediated endosomal–lysosomal escape strategies and demonstrates the therapeutic potential of a tumor-targeted oncolytic EV platform that induces ICD by delivering misfolded proteins directly into the cancer cell cytosol. 세포 간 상호작용은 기존에 알려진 직접적인 세포 접촉이나 사이토카인과 같은 분비 신호뿐 아니라, 최근에는 세포 외 소포체(extracellular vesicles, EV)와 같은 나노 크기의 소포를 통해서도 이루어지는 것으로 알려져 있다. EV는 세포막에서 유래한 막단백질, 세포질 내 단백질, 그리고 다양한 핵산을 함유하고 있어 생리적·병리적 정보 전달에 중요한 역할을 한다. 본 연구에서는 세포 외 소포체 엔지니어링을 통해 이러한 내부 담지 물질(cargo)의 전달 효율을 향상시키는 전략과, 바이러스 유래 융합 단백질을 활용한 전달 효과 증진 방안을 탐구하였다. EV를 치료제 전달체로 활용하려는 시도는 꾸준히 이루어져 왔으며, 최근에는 유전공학 기술의 발달로 mRNA, 전사인자와 같은 기능성 유전물질을 EV에 능동적으로 탑재하는 다양한 전략이 개발되었다. 그러나 이러한 물질들이 수용 세포로 내재화된 이후 엔도좀에 포획되고 리소좀 분해 경로를 거치며 상당량이 소실되는 문제가 제기되었다. 이에 따라 EV가 엔도-리소좀 분해 경로를 회피(endosomal–lysosomal degradation escape)하도록 유도하는 전략의 필요성이 대두되었고, 이를 위해 VSV-G와 같은 바이러스 유래 융합 단백질을 EV 표면에 발현시키려는 접근이 시도되었다. 본 연구에서는 지금까지 개발된 다양한 엔도-리소좀 분해 회피 기술과 그 효과를 정량적으로 평가할 수 있는 분석 방법들을 검토하고, 이러한 전략들이 EV 기반 치료제 개발에 어떻게 활용될 수 있는지 논의하였다. 본 연구에서는 일반적으로 세포 찌꺼기로 간주되는 비접힘 단백질을 효과적으로 탑재하고 엔도–리소좀 분해 경로를 회피할 수 있도록, 호흡기세포융합바이러스 융합단백질(RSVF)을 발현하는 항종양성 EV(bRSVF-EV)를 설계하였다. RSVF를 발현하는 세포주에 bafilomycin A1를 처리하여 리소좀 기능을 억제함으로써 비접힘 단백질의 세포 내 축적을 유도하였고, 이를 통해 EV 내부로의 효율적인 적재가 가능해졌다. 제작된 bRSVF-EV는 nucleolin 의존적 기전을 통해 암세포막에 이종(xenogeneic) 항원을 선택적으로 표면 이식하여 강력한 선천면역 반응을 유도할 뿐 아니라, 엔도–리소좀 분해를 회피하고 비접힘 단백질을 암세포의 세포질로 직접 전달함으로써 소포체 스트레스와 면역원성 세포사멸(immunogenic cell death, ICD)을 촉발하였다. 이러한 작용기전에 의해 마우스 종양 모델에서 뚜렷한 항종양 면역 반응이 유도되었으며, 특히 PD-1 억제제와 병용 투여 시 생존 기간이 현저히 연장되고 일부 개체에서는 완전 관해에 도달하는 우수한 치료 효과가 관찰되었다. 종합하면, 본 연구는 EV 기반 전달체에서의 엔도–리소좀 회피 전략에 대한 문헌적 고찰과 더불어, 비접힘 단백질을 암세포 세포질로 직접 전달하여 ICD를 유도하는 종양 표적화 oncolytic EV 플랫폼의 치료적 가능성을 제시한다.

Here, we investigated the effect of anticancer drug-loaded functional polymeric nanoparticles on drug resistance of three-dimensional (3D) breast tumor spheroids. 3D tumor models were built using concave microwells with different diameters (300, 500 and 700 um) and nanoparticles were prepared using thermo-responsive poly(N-isopropylacrylamide) (PNIPAM)-co-acrylic acid (AA). Drug-release studies showed that 86% of doxorubicin was released from PNIPAM-co-AA nanoparticles over the course of 96 hours. Upon culturing with doxorubicin-loaded PNIPAM-co-AA nanoparticles for 96 hours, the smallest tumor spheroids (cultured in 300 um microwells) were extensively disrupted, resulting in a reduction in spheroid diameter. In contrast, the sizes of the largest tumor spheroids (cultured in 700 um microwells) were not changed. Scanning electron microscopy revealed that the surface morphology of 3D tumor spheroids treated with doxorubicin-loaded PNIPAM-co-AA nanoparticles was severely rugged and the circular shape of 3D spheroids had collapsed. Cell viability assays also demonstrated that the largest tumor spheroids (700 um microwells) cultured with doxorubicin-loaded PNIPAM-co-AA nanoparticles were highly resistant to the anticancer drug. We confirmed that tight cell-cell contacts within largest tumor spheroids significantly improved the anticancer drug resistance. Therefore, this uniform-sized 3D breast tumor model could be a potentially powerful tool for anticancer drug screening applications.

다이아몬드 내 질소-공공 결함(NV center)과 같은 고체 상태 기반 양자 광원은 장시간 유지되는 스핀 코히어런스(spin coherence)와 우수한 광학 제어 가능성 덕분에 양자 네트워크 구현을 위한 유망한 구성 요소로 주목받고 있다. 이러한 광원의 성능을 극대화하고 양자 노드 간의 확장 가능한 상호연결을 실현하기 위해서는 광자 구조와의 통합을 통해 빛-물질 간 상호작용을 강화하는 것이 필수적이다. 그러나 기존의 나노 광자 구조는 NV 센터를 식각된 표면에 매우 근접하게 배치할 수밖에 없는 구조적 특성을 가지며, 이는 표면에서 유래하는 디코히어런스 현상에 취약해져 스펙트럼 불안정성과 스핀-광자 인터페이스 성능 저하를 초래하는 한계가 있다. 이러한 문제를 해결하기 위해, 본 연구에서는 NV center와의 빛-물질 상호작용을 강화하기 위한 다이아몬드 기반 일차원 fishbone 형상 포토닉 크리스탈 공진기를 설계, 시뮬레이션, 그리고 실험적으로 구현하였다. 본 구조는 기존의 구멍을 뚫어 주기를 조절하는 방식과는 달리, 삼각형 단면의 나노빔 상에 주기적으로 굴절률 대비를 가지도록 하는 가시 구조의 핀을 수직으로 배열하여 광학 모드의 국소화를 유도한다. 이를 통해 NV center를 표면으로부터 100 nm 이상 깊숙이 매립할 수 있으며, 표면 유래 디코히어런스의 영향을 효과적으로 줄일 수 있다. 본 공진기는 페러데이 케이지 기반 플라즈마 식각 기술과 빗면 식각(angled-etching)을 결합하여 제작되었으며, 대칭적인 포토닉 거울과 이 중심에 두꺼운 결함 영역을 갖는 구조로 구현되었다. 해당 구조는 NV의 zero-phonon line(ZPL, 637 nm)과의 스펙트럼 정렬이 가능하며, 기존 나노 광자 구조에서 자주 발생하는 1 GHz 이상의 스펙트럼 확산, 전하 잡음, 기계적 변형 등의 문제를 완화할 수 있다. 실험 결과, 제작된 fishbone 공진기692.91 nm에서 공진 피크를 나타냈으며, 측정된 Q-요인은 약 107로 확인되었다. 본 연구는 fishbone 포토닉 공진기가 고체 상태 양자 네트워크에서 요구되는 스펙트럼 안정성과 스핀-광자 결합을 모두 만족시키는 유망한 통합 플랫폼이 될 수 있음을 보여준다. Solid-state quantum emitters—particularly nitrogen-vacancy (NV) centers in diamond—have emerged as promising building blocks for quantum networks due to their long-lived spin coherence and optical controllability. To harness their full potential, integration with photonic structures is essential for enhancing light-matter interaction and enabling scalable quantum interconnects. This typically involves the use of nanostructured cavities or waveguides, which require the emitter to be placed in close proximity to etched surfaces for optimal coupling. However, such shallow placement renders the NV centers highly susceptible to surface-induced decoherence, leading to spectral instability and degraded spin-photon interface performance. To address this challenge, we present the design, simulation, and experimental realization of a one-dimensional fishbone-shaped diamond photonic crystal cavity, engineered to enhance light-matter interaction with nitrogen-vacancy (NV) centers. Unlike conventional photonic crystal designs that rely on air-hole modulation, our cavity employs refractive index contrast via subwavelength dielectric fins along a triangular nanobeam, facilitating deeper NV embedding (>100 nm) and minimizing exposure to surface-induced decoherence. The cavity was fabricated using angled-etching in conjunction with Faraday cage-assisted plasma processing, yielding a suspended structure with symmetric photonic crystal mirrors and a dielectric-rich defect region. This geometry enables spectral alignment with the NV zero-phonon line (ZPL) at 637 nm while mitigating common challenges associated with shallow implantation, such as enhanced charge noise, increased strain gradients, and significant spectral diffusion (>1 GHz), often observed in conventional nanophotonic cavities. Our results demonstrate the potential of this fishbone architecture to serve as a robust and scalable platform for spectrally stable and coherent spin-photon interfaces in solid-state quantum networks. Photoluminescence measurements revealed a resonant mode at 692.91 nm with a measured Q-factor of 107. These results establish the fishbone cavity as a promising platform for scalable, coherent spin-photon interfaces essential for solid-state quantum networks.
Pressure-Induced Osteoblast Differentiation in Mesenchymal Stem Cells Using Agarose gel

기계적 자극은 중간엽 줄기세포의 운명 결정과 골분화에 있어 핵심적인 역할을 하며, 특히 압력 자극은 골조직 환경에서 발생하는 주요한 생리적 자극 중 하나이다. 이에 따라 압력 자극을 통해 중간엽 줄기세포를 골모세포로 분화시키는 연구가 활발히 진행되고 있다. 그러나 기존의 압축 시스템은 자극의 비균일성, 지지체의 물성에 의한 간섭, 낮은 재현성, 자극 후 분석의 불편함 등의 한계를 지닌다. 본 연구에서는 이러한 한계를 극복하기 위해 2D 배양 환경에서 세포에 직접적인 압력 자극을 가할 수 있는 ‘pressing gel device’를 개발하였다. 본 device는 저비용의 아가로스 하이드로겔을 압축 매체로 사용하며, 가교제나 화학적 처리 없이 단순한 온도 제어만으로 겔레이션이 가능하여 세포 생존성에 대한 생화학적 영향을 최소화한다. 또한 세포에 균일하게 직접 압축을 가할 수 있으며, 자극 종료 후 즉시 분석이 가능하여 실험적 접근성과 재현성을 크게 향상시켰다. 해당 device를 통해 중간엽 줄기세포에 압력 자극을 부여하고, ALP 활성도, 칼슘 침착, 형광 염색, qRT-PCR 등의 분석을 수행한 결과, 골분화와 관련된 다양한 유전자 및 단백질 마커들의 발현이 유의미하게 증가함을 확인하였다. 이는 물리적 압축 자극만으로도 중간엽 줄기세포의 골분화를 효과적으로 유도할 수 있음을 시사한다. 따라서 본 연구에서 개발된 pressing gel device는 기계적 자극에 의한 생물학적 반응을 평가할 수 있는 유용한 플랫폼으로, 향후 기계생물학 기반 조직 재생 및 골 재생 치료 전략에 있어 높은 응용 가능성과 임상적 전환 가능성을 지닌다. Mechanical stimulation plays a critical role in the fate determination and osteogenic differentiation of Mesenchymal Stem Cells (MSCs). Among various types of mechanical stimuli, compression is one of the primary physiological cues present in the bone tissue environment. Accordingly, extensive research has been conducted to induce osteoblast differentiation of MSCs through compression. However, conventional compression systems have faced several limitations, including non-uniformity force distribution, interference from scaffold material properties, low reproducibility, and inconvenience in post-stimulation analysis. In this study, to overcome these limitations, we developed a custom-designed ‘pressing gel device’ capable of applying direct compression to cells in a two-dimensional (2D) culture environment. This device utilizes low-cost agarose hydrogel as a compression medium, which enables gelation through simple temperature control without the need for crosslinkers or chemical treatments, thereby minimizing biochemical effects on cell viability. Furthermore, it allows for uniform and direct compression to be applied to cells, and immediate analysis is possible upon removal of the stimulus, greatly improving experimental accessibility and reproducibility. Using this device, compression was applied to MSCs, and various analyses including ALP activity assay, calcium deposition, fluorescence staining, and quantitative Real-Time Polymerase Chain Reaction (qRT-PCR) were performed. As a result, the expression of multiple gene and protein associated with osteogenic differentiation was significantly upregulated after compression conditions. This indicates that physical compression alone can effectively induce osteogenic differentiation of MSCs. Therefore, the pressing gel device developed in this study serves as a useful platform for evaluating biological responses to mechanical stimulation and holds strong potential for application in future mechanobiology-based tissue regeneration and bone repair strategies, as well as for clinical translation.
Precisely Tailored Thermoelectric Properties of Carbon Nanotubes(CNTs) via Selective Inkjet Printing

With increasing demands for self-powered wearable electronics, flexible thermoelec-tric (TE) devices harvesting electrical energy from body heat have drawn significant at-tention. Numerous promising material candidates have been explored for realizing highly reliable flexible TE devices. Among them, carbon nanotubes (CNTs) offer notable TE properties, mechanical flexibility, and solution processability which provide attractive advantages to meet these requirements. However, there have been critical bottlenecks of low electrical conductivity of CNTs, resulting in the poor Figure-of-Merit (zT). To ad-dress these challenges, extensive research has focused on enhancing the electrical con-ductivity and Seebeck coefficient of pristine CNTs through surface charge transfer dop-ing. However, most conventional doping methods, such as solution dipping or vaporized deposition, are applied across the entire CNT film surface, therefore, it difficult to pre-cisely control the thermoelectric performance. In this study, we report a doping strategy via inkjet printing to tailor the TE proper-ties of CNT films using poly(acrylic acid) (PAA) as a p-type dopant. By optimizing the concentration of the PAA inks, we achieved enhanced TE performance in CNT films. The concentration-dependent control of doping levels enabled an optimal balance be-tween electrical conductivity and the Seebeck coefficient, resulting in maximum TE per-formance at a specific concentration (30 mg mL-1). To investigate the effect of localized selective doping on the TE properties of CNT films, full-area and half-area doping were conducted at the optimized PAA concentration. Notably, the half-doped CNT film exhibited a higher Seebeck coefficient (50.479 μV/K) than full-are doped films, despite showing a lower electrical conductivity. This indicates that concentration and mobility gradients across the film contribute to the enhancement of Seebeck voltage. The observed properties in the half-doped film indicate that the spa-tial gradient played a dominant role in determining TE properties. The selective doping-induced inhomogeneity created an internal potential gradient amplifying the thermoelec-tric response. In the half-doping condition, the spatial gradients of carrier concentration and mobility were favorably tuned, resulting in an enhanced Seebeck response. Therefore, our study demonstrates that the inkjet printing-based doping method for CNTs not only enables precise control of doping levels over TE properties but also offers a scalable and versatile approach for designing high-performance TE materials.

Cancer therapies have been extensively studied, resulting in the development of various therapeutic strategies. However, traditional chemotherapy agents and small-molecule inhibitors exhibit limited specificity, causing toxicity to normal cells and subsequently yielding low therapeutic efficacy. To address these limitations, there has been an increasing demand for strategies enabling precise tumor targeting. The concept of antibody-drug conjugates (ADCs), first proposed by Paul Ehrlich in 1900, represents one such approach. ADCs have continuously evolved, leading to the FDA approval of 13 ADC therapeutics by 2024, firmly establishing ADCs as one of the most actively researched targeted cancer therapy platforms. Nevertheless, existing ADCs have inherent drawbacks, including poor tumor penetration, complex manufacturing processes, immunogenicity, and dose-limiting toxicity. To overcome these issues, this study aims to exploit the superior tumor penetration, rapid intracellular uptake, reduced immunogenicity, and simplified manufacturing process associated with protein-drug conjugates (PDCs) to facilitate more precise and effective targeted therapy. In this study, Cluster of Differentiation 47 (CD47) was selected as the target protein. CD47 is overexpressed in tumor cells, inducing immune evasion through its interaction with signal regulatory protein alpha (SIRPα) on macrophages. Targeting CD47 can disrupt immune evasion by tumor cells, thereby activating immune responses and significantly enhancing anticancer efficacy. To achieve this, we utilized a variant SIRPα (vSIRPα), engineered to exhibit approximately 50,000-fold higher binding affinity to CD47 compared to the wild-type SIRPα, thus ensuring high specificity and low off-target effects. Monomethyl auristatin E (MMAE), a potent payload currently utilized in four FDAapproved ADCs, was chosen asthe anticancer drug. MMAE exhibits 100?1,000 times higher cytotoxic potency than doxorubicin and functions as an antimitotic agent by inhibiting tubulin polymerization, thereby arresting cell division. Furthermore, MMAE possesses high cell membrane permeability and an effective bystander effect, thus promising robust anticancer efficacy against solid tumors. Nevertheless, to reduce nonspecific toxicity to normal cells, a tumor-specific release system is crucial. Consequently, MMAE was conjugated to vSIRPα via a cathepsin B-cleavable valine-citrulline (VC) linker. Cathepsin B is a lysosomal enzyme overexpressed in tumor cells, enabling selective intracellular release of MMAE and minimizing systemic toxicity while maximizing therapeutic efficacy. Overall, this study focuses on designing a CD47-targeted PDC utilizing vSIRPα and employing a linker system tailored to precisely control the potent cytotoxicity of MMAE. The ultimate objective is to maximize tumor specificity, thus overcoming the limitations of existing ADC technologies and laying a foundation for the next generation of targeted cancer therapeutics. 암 치료제는 지속적으로 연구되어 왔으며, 다양한 치료 전략이 개발되었으나, 전통적인 화학항암제 및 소분자 억제제는 낮은 특이성으로 인해 정상 세포에도 독성을 유발하며 치료 성공률이 낮다. 이를 극복하기 위해 종양세포를 정밀하게 표적화하는 치료 전략이 요구되었으며, 1900 년 Paul Ehrlich 에 의해 최초로 항체-약물 접합체 (antibody-drug conjugate, ADC)의 개념이 제안되었다. 이후, ADC 는 발전을 거듭하며 2024 년까지 총 13 종이 FDA 승인을 받았고, 현재 가장 활발히 연구되고 있는 표적 항암제 플랫폼 중 하나로 자리 잡았다. 그러나 기존 ADC 는 낮은 종양 침투력, 복잡한 제조공정, 면역원성, 투여량 제한 독성 does limiting toxicity)등의 한계를 가지고 있다. 따라서 본 연구에서는 단백질약물 접합체 (protein-drug conjugate, PDC)의 더 높은 종양 침투력, 더 빠른 세포내유입, 낮은 면역원성과 더 간단한 제조공정 특성을 활용하여 보다 정밀하고 효과적인 표적 치료연구를 진행하려 한다. 본 연구에서는 Cluster of Differentiation 47 (CD47)을 암 표적 단백질로 선정하였다. CD47 은 종양세포에서 과발현되어 대식세포와의 SIRPα/CD47 상호작용을 통해 면역 회피를 유도하는 단백질이다. CD47 을 표적화하면 종양세포의 면역 회피를 차단하고, 면역세포를 활성화하여 항암 효과를 극대화할 수 있다. 이를 위해 기존 wild-type SIRPα 대비 약 50,000 배 높은 CD47 결합력을 가지는 변형 단백질 (varient SIRPα, vSIRPα)을 활용하여, 높은 특이성과 낮은 비표적 효과를 기대하였다. 도입될 항암 약물 (payload)로는 FDA 승인된 ADC 4 종에서 사용되는 monomethyl auristatin E (MMAE)를 적용하였다. MMAE 는 doxorubicin 보다 100~1000 배 강력한 세포독성을 가지며, 튜불린 (tubulin) 중합 억제를 통해 세포분열을 저해하는 항유사분열제이다 [10]. 또한, 높은 세포막 투과성과 bystander 효과를 통해 고형암 내 강력한 항암 효과를 기대할 수 있다. 그러나 일반세포에 대한 비특이적 독성을 줄이기 위해 종양 특이적 방출 시스템이 필수적이다. 이를 위해 MMAE 를 vSIRPα에 cathepsin B cleavable Valine-Citrulline (VC) linker 를 통해 도입하였다. Cathepsin B 는 종양세포의 lysosome 에 과발현되는 효소로, 이를 이용하면 종양세포 내부에서 선택적으로 MMAE 를 방출할 수 있어 비특이적 독성을 최소화하면서도 강력한 치료 효과를 기대할 수 있다. 본 연구에서는 vSIRPα를 이용한 CD47 표적 PDC 를 설계하고, MMAE 기반의 강력한 세포독성을 조절하는 linker 시스템을 적용하여 종양 특이성을 극대화한 항암제 개발을 목표로 한다. 이를 통해 기존 ADC 의 한계를 극복한 새로운 표적 치료제를 제시하며, 차세대 항암제 연구의 기초를 마련하는 데 기여하고자 한다.
