        The Step-by-Step Model Technology Considering Nonlinear Effect Used for Construction Simulation Analysis

        Long-span spatial structures have advantages over other structural systems in terms of novelty and aesthetics in architectural form. However, the complexity of stress distribution causes these kinds of structures to change significantly between the construction and utilization stages. One-time loading method was used in the traditional design method to calculate and evaluate structural stress and structural deflection, either of which could cause problems in component size and residual stress during construction process. Therefore, construction simulation analysis of these structures must be performed. Element birth and death technology and step-by-step model technology (SSMT) are the main approaches in the finite element method used in construction analysis. This paper proposes a theory basis for the SSMT that was used in the constructional analysis of the Yujiapu Railway Station Building. Results obtained by the proposed method were compared with those by element birth and death technology. In addition, the preset construction deformation value was calculated to validate the correctness and efficiency of the proposed method.

      • Genetic variant in TP63 on locus 3q28 is associated with risk of lung adenocarcinoma among never-smoking females in Asia

        <P>A recent genome-wide association study (GWAS) of subjects from Japan and South Korea reported a novel association between the TP63 locus on chromosome 3q28 and risk of lung adenocarcinoma (p = 7.3 10(-12)); however, this association did not achieve genome-wide significance (p 10(-7)) among never-smoking males or females. To determine if this association with lung cancer risk is independent of tobacco use, we genotyped the TP63 SNPs reported by the previous GWAS (rs10937405 and rs4488809) in 3,467 never-smoking female lung cancer cases and 3,787 never-smoking female controls from 10 studies conducted in Taiwan, Mainland China, South Korea, and Singapore. Genetic variation in rs10937405 was associated with risk of lung adenocarcinoma [n = 2,529 cases; p = 7.1 10(-8); allelic risk = 0.80, 95% confidence interval (CI) = 0.74-0.87]. There was also evidence of association with squamous cell carcinoma of the lung (n = 302 cases; p = 0.037; allelic risk = 0.82, 95% CI = 0.67-0.99). Our findings provide strong evidence that genetic variation in TP63 is associated with the risk of lung adenocarcinoma among Asian females in the absence of tobacco smoking.</P>

      • A Multi-keyword Ranked Search over Encrypted Cloud Data Supporting Semantic Extension

        With the emergence of cloud computing, many data owners outsource their local data to cloud server so as to enjoy high-quality data storage services. For the protection of data privacy, sensitive data has to be encrypted before outsourcing, which makes effective data utilization a challenging task. Although existing searchable encryption technologies enable data users to conduct secure search over encrypted data, the functionality of these schemes need to be further improved. In this paper, we construct a secure and efficient multi-keyword ranked search scheme which supports both the semantic extension search and the multi-keyword ranked search. The semantic extension is achieved through the mutual information statistical analysis of keywords. And the multi-keyword ranked search is achieved through a balanced binary tree whose nodes are the vectors of term frequency (TF) values. The splitting operation and secure transformation are utilized to encrypt the vectors of index and query. Note that, the encrypted vectors can be well used to calculate accurate relevance scores. Phantom terms are added to the index vector to blind the search results to resist statistical attacks. Due to the use of tree-based index structure, the proposed scheme can achieve the sub-linear search time. Finally, the experiments are conducted to demonstrate the efficiency of the proposed scheme.

        Inhibition of Fatty Acid β-Oxidation by Fatty Acid Binding Protein 4 Induces Ferroptosis in HK2 Cells Under High Glucose Conditions

        Background: Ferroptosis, which is caused by an iron-dependent accumulation of lipid hydroperoxides, is a type of cell death linked todiabetic kidney disease (DKD). Previous research has shown that fatty acid binding protein 4 (FABP4) is involved in the regulation of ferroptosis in diabetic retinopathy. The present study was constructed to explore the role of FABP4 in the regulation of ferroptosis in DKD. Methods: We first detected the expression of FABP4 and proteins related to ferroptosis in renal biopsies of patients with DKD. Then, we used a FABP4 inhibitor and small interfering RNA to investigate the role of FABP4 in ferroptosis induced by high glucosein human renal proximal tubular epithelial (HG-HK2) cells. Results: In kidney biopsies of DKD patients, the expression of FABP4 was elevated, whereas carnitine palmitoyltransferase-1A (CPT1A), glutathione peroxidase 4, ferritin heavy chain, and ferritin light chain showed reduced expression. In HG-HK2 cells, the induction of ferroptosis was accompanied by an increase in FABP4. Inhibition of FABP4 in HG-HK2 cells changed the redox state, suppressing the production of reactive oxygen species, ferrous iron (Fe2+), and malondialdehyde, increasing superoxide dismutase, and reversing ferroptosis-associated mitochondrial damage. The inhibition of FABP4 also increased the expression of CPT1A, reversed lipiddeposition, and restored impaired fatty acid β-oxidation. In addition, the inhibition of CPT1A could induce ferroptosis in HK2 cells. Conclusion: Our results suggest that FABP4 mediates ferroptosis in HG-HK2 cells by inhibiting fatty acid β-oxidation.

        Bayesian methods for dealing with missing data problems

        Missing data, a common but challenging issue in most studies, may lead to biased and inefficient inferences if handled inappropriately. As a natural and powerful way for dealing with missing data, Bayesian approach has received much attention in the literature. This paper reviews the recent developments and applications of Bayesian methods for dealing with ignorable and non-ignorable missing data. We firstly introduce missing data mechanisms and Bayesian framework for dealing with missing data, and then introduce missing data models under ignorable and non-ignorable missing data circumstances based on the literature. After that, important issues of Bayesian inference, including prior construction, posterior computation, model comparison and sensitivity analysis, are discussed. Finally, several future issues that deserve further research are summarized and concluded.

        Defense responses of sulfur dioxygenase to sulfide stress in the razor clam Sinonovacula constricta

        Background Sulfde is a well-known toxicant widely distributed in the culture environment. As a representative burrowing benthic bivalve, the razor clam Sinonovacula constricta is highly sulfde tolerant. Mitochondrial sulfde oxidation is an important way for sulfde detoxifcation, where sulfur dioxygenase (SDO) is the second key enzyme. Objective To investigate the mechanism of sulfde tolerance in S. constricta, the molecular characterization of its SDO (designated as ScSDO) was studied. Methods The cDNA sequence of ScSDO was cloned by RACE technique. The response of ScSDO in gills and livers of S. constricta was investigated during sulfde exposure (50, 150, and 300 μM sulfde) for 0, 3, 6, 12, 24, 48, 72, and 96 h by qRTPCR. Moreover, the temporal expression of ScSDO protein in S. constricta gills after exposure to 150 μM sulfde was detected by Western blot. The subcellular location of ScSDO was identifed by TargetP 1.1 prediction and Western Blot analysis. Results The full-length cDNA of ScSDO was 2914 bp, encoding a protein of 304 amino acids. The deduced ScSDO protein was highly conserved, containing the signature HXHXDH motif of the metallo-β-lactamase superfamily and two metalbinding sites, of which metal-binding site I is known to be the catalytically active center. Subcellular localization confrmed that ScSDO was located only in the mitochondria. Responding to the sulfde exposure, distinct time-dependent increases in ScSDO expression were detected at both mRNA and protein levels. Moreover, the gills exhibited a higher ScSDO expression level than the livers. Conclusions All of our results suggest that ScSDO plays an important role in mitochondrial sulfde oxidation during sulfde stress, making S. constricta highly sulfde tolerant. In addition, as a respiratory tissue, the gills play a more critical role in sulfde detoxifcation.

        Analysis on the influence of type and proportion of aggregates on the performance of PU based polymer concrete

        In this work, the fabrication and improvement of a lightweight and corrosion-resistant polyurethane (PU) based polymerconcrete are studied. The aggregate in traditional polymer concrete is generally crushed stone. Due to the low bonding strengthbetween polymer matrix and crushed stone, it is easy to fall off between polymer matrix and crushed stone after long-termuse, resulting in the degradation of the performance of polymer concrete. In order to overcome the low bond strength betweencrushed stone and polymer matrix in polymer concrete, modified polyetheramine particles were used to enhance the finalmechanical parameters of PU based polymer concrete. A series of mechanical and environmental tests were carried out, andthe effects of aggregates addition ratio and type on the mechanical properties of polymer concrete were analyzed. Theexperimental results determined the influence mechanism of aggregates proportion and type on the mechanical properties ofpolymer concrete, and obtained the appropriate aggregates type and addition proportion. The current work reveals that usingmodified polyetheramine particles as aggregates of polymer concrete can significantly enhance the bond strength anddurability between polymer matrix and aggregates. The density of this polymer concrete proposed in this paper is about 1.2g/mm3; The density of traditional polymer concrete is about 1.5-1.8 g/mm3, the weight can be reduced by 25%, and there isno problem of falling off between polymer matrix and aggregate.

        Vitellogenin’s putative role in Tegillarca granosa’s cadmium detoxification

        The bloody clam, Tegillarca granosa, is a commercial benthic bivalve, having a strong accumulation ability and torrelence to cadmium. To investigate whether vitellogenin (Vg) is involved in cadmium (Cd) detoxification, the full-length cDNA of T. granosa Vg was cloned, and its expression pattern in response to cadmium exposure was studied compared with the reference metallothionein (MT) gene. The full T. granosa Vg sequence consisted of 8988 bp, including a 6930-bp open reading frame that encoded a 2309 amino acid polypeptide. The deduced Vg protein contained a Vg N-terminal domain, domain of unknown function (DUF1943), SbcC domain, and von Willebrand factor typeD domain (VWD). Multiple metal-binding sites were predicted in the deduced T. granosaVg protein, suggesting its potential in functioning as a metal-binding protein. In addition, Vg expression increased in the T. granosa digestive gland and hemolymph in time-dependent manner after exposure to 1, 3, 6 and 9 lg/L Cd for 28 days.MTexpression was measured in parallel with Vg expression, and the latter was more sensitive to Cd induction than the former. Together, results of the present research suggested that Vg may play an important role in T. granosa metal detoxification.
