      • 배추 시스테인 단백질 분해 효소 억제 유전자 BCPI-1의 발현과 종자 발아 및 유묘 생장과의 연관성

        Phytocystatins are protein inhibitors of cysteine proteinases of the papain family that have been identified in both monocot and dicot plants. A cDNA encoding a phytocystatin, BCPI-1 (Brassica Cysteine Proteinase Inhibitor-1) has been isolated from Chinese cabbage (Brassica campestris L. ssp. pekinensis) flower buds. Here, we tested whether BCPI-1 transcription is regulated by hormones, and could be involved in regulating cysteine proteinases during and after germination. BCPI-1 was sensitive to exogenous GA_(3) and ABA, which are important factors controlling seed germination, suggesting that the expression of BCPI-1 is hormonally regulated. We introduced a recombinant plasmid containing the full-length BCPI-1 cDNA under the control of cauliflower mosaic virus 35S promoter into rice embryogenic calli using the particle delivery method, and regenerated a number of transgenic rice plants. Constitutively over-expressed BCPI-1 caused changes in overall plant growth and development, including reduced germination and seedling growth. These data support the role of the BCPI-1 in the regulation of endogenous proteinases during both seed germination and subsequent seedling development. Phylocystatins은 papain계열의 cysteine 단백질 분해 효소 활성을 특이적으로 억제하는 억제자로 다양한 식물 종으로부터 분리되었다. 본고에서는, 배추 화아 cDNA library로부터 분리된 phytocystatin인 BCPI-1(Brassica Cysteine Proteinase Inhibitor-1)의 생체 내 기능에 대해 연구하였다. 먼저, BCPI-1 전사체는 발아 조절에 중요한 영향을 미치는 GA₃와 ABA에 의해 예민하게 증가, 혹은 감소되는 반응을 보임으로써, BCPI-1이 식물 호르몬의 영향을 받으며, 특히 발아나 유묘의 생장 조절에 관련이 있음을 알 수 있었다. Particle bombardment 방법을 통하여 BCPI-1을 벼의 배 형성 세포 내에 도입, 재분화 시켜 형질 전환 벼를 생성하였다. CaMV 35S promoter의 조절에 의해 지속적인 BCPI-1 발현을 보이는 형질 전환 벼의 경우, 발아와 유묘 생장이 현저히 지연되었다. 위의 결과들을 바탕으로, BCPI-1이 식물체 내에서 생성되는 cysteine 계열의 단밸질 분해 효소 활성을 조절하여 종자의 발아와 유묘의 생장에 영향을 미치는 것으로 추측한다.

      • Over-expression of Chinese cabbage calreticulin 1, BrCRT1, enhances shoot and root regeneration, but retards plant growth in transgenic tobacco

        Calreticulin (CRT) is a ubiquitously expressed, high capacity Ca^(2+)-binding protein that is involved in intracellular Ca^(2+) homeostasis and molecular chaperoning in the endoplasmic reticulum (ER). A cDNA encoding a calreticulin, BrCRT1 (Brassica rapa Calreticulin 1), has been isolated from Chinese cabbage (B. rapa subsp. pekinensis) flower bud. Constitutive over-expression of the BrCRT1 gene promotes robust shoot production and root formation at sub-optimal concentrations of BA/NAA, which are important factors controlling plant regeneration in tissue culture. In contrast, the suppressed BrCRT1 line exhibited a slight reduction of shoot and root regeneration. In spite of enhanced regeneration in tissue culture, the seedling and plant growth rate was inhibited in soil. The steady state level of BrCRT1 transcripts was sensitive to exogenous auxins and cytokinins, and rapidly accumulated within 30 min, and this induction required de novo protein synthesis. Together with the results of transgenic tobacco plants and mRNA analysis in Chinese cabbage, our data suggest that BrCRT1 genes may up-regulate the competency of vegetative tissue to respond to hormonal signals involved in shoot and root regeneration processes.

      • Expression of the Tobacco Mosaic Virus Movement Protein Alters Starch Accumulation in Nicotiana benthamiana

        Nicotiana benthamiana plants were transformed with the movement protein (MP) gene of tobacco mosaic virus (TMV), using Agrobacterium-mediated transformation. Plants regenerated from the transformed cells accumulated 30-kDa MP and complemented the activity of TMV MP when infected with chimeric TMVs containing defective MP. These transgenic plants displayed stunting, pale-green leaves, and starch accumulations, indicating that TMV MP altered the carbon partitioning for leaves involved in TMV cell-to-cell movement.

        Over-expression of Chinese cabbage calreticulin 1b, BrCRT1b, induces initiation of organogenesis in Arabidopsis

        Calreticulin (CRT) is a major Ca2+-binding/buffering protein in the lumen of the endoplasmic reticulum. A cDNA encoding CRT, BrCRT1b (Brassica rapa CRT1b), has been isolated from Chinese cabbage flower bud. Recombinant BrCRT1b21-418 protein showed 45Ca2+-binding and chaperone activity. BrCRT1b transcripts accumulated to high levels in the root, young leaf and flower. Constitutive over-expression of BrCRT1b promoted robust shoot production and root formation at sub-optimal concentrations of auxin/cytokinin. In spite of enhanced regeneration in tissue culture, the seedling and plant growth rates were slightly inhibited in soil. Thus, our findings suggest that BrCRT1b is related to organogenesis during in vitro culture.

        <P>A broken symmetry is generally believed to be a prerequisite for plasmonic electromagnetically-induced transparency (EIT), since the asymmetry allows the excitation of the otherwise forbidden dark mode. Nevertheless, according to the picture of magnetic plasmon resonance (MPR)-mediated plasmonic EIT, we show that plasmonic EIT can be achieved even in symmetric structures based on the second-order MPR. This not only sharpens our understanding of the existing concept, but also provides a profound insight into the plasmonic coherent interference in the near-field zone.</P>

      • Tunable dual-band perfect absorbers based on extraordinary optical transmission and Fabry-Perot cavity resonance.

        <P>Magnetic resonance is considered to be a necessary condition for metamaterial perfect absorbers, and dual-band absorbers can be composed of a pair of metallic layers with anti-parallel surface currents. We designed and fabricated a tunable dual-band perfect absorber based on extraordinary-optical-transmission (EOT) effect and Fabry-Perot cavity resonance. The idea and the mechanism are completely different from the absorber based on the near-field interaction. The important advantage of our structure is that we can switch a single-band absorber to a dual-band absorber by changing the distance between two metallic layers and/or incident angle. The peak originating from the EOT effect becomes significantly narrower, resulting in an increase of the Q-factor from 16.88 to 49. The dual-band absorber can be optimized to be insensitive to the polarization of the incident electromagnetic wave by slightly modifying the absorber structure.         Heavy concrete shielding properties for carbon therapy

        As medical facilities are usually built at urban areas, special concrete aggregates and evaluation methods are needed to optimize the design of concrete walls by balancing density, thickness, material composition, cost, and other factors. Carbon treatment rooms require a high radiation shielding requirement, as the neutron yield from carbon therapy is much higher than the neutron yield of protons. In this case study, the maximum carbon energy is 430 MeV/u and the maximum current is 0.27 nA from a hybrid particle therapy system. Hospital or facility construction should consider this requirement to design a special heavy concrete. In this work, magnetite is adopted as the major aggregate. Density is determined mainly by the major aggregate content of magnetite, and a heavy concrete test block was constructed for structural tests. The compressive strength is 35.7 MPa. The density ranges from 3.65 g/cm<sup>3</sup> to 4.14 g/cm<sup>3</sup>, and the iron mass content ranges from 53.78% to 60.38% from the 12 cored sample measurements. It was found that there is a linear relationship between density and iron content, and mixing impurities should be the major reason leading to the nonuniform element and density distribution. The effect of this nonuniformity on radiation shielding properties for a carbon treatment room is investigated by three groups of Monte Carlo simulations. Higher density dominates to reduce shielding thickness. However, a higher content of high-Z elements will weaken the shielding strength, especially at a lower dose rate threshold and vice versa. The weakened side effect of a high iron content on the shielding property is obvious at 2.5 µSv=h. Therefore, we should not blindly pursue high Z content in engineering. If the thickness is constrained to 2 m, then the density can be reduced to 3.3 g/cm<sup>3</sup>, which will save cost by reducing the magnetite composition with 50.44% iron content. If a higher density of 3.9 g/cm<sup>3</sup> with 57.65% iron content is selected for construction, then the thickness of the wall can be reduced to 174.2 cm, which will save space for equipment installation.

        Plasma metabolites associated with physiological and biochemical indexes indicate the effect of caging stress on mallard ducks (Anas platyrhynchos)

        Objective: Cage rearing has critical implications for the laying duck industry because it is convenient for feeding and management. However, caging stress is a type of chronic stress that induces maladaptation. Environmental stress responses have been extensively studied, but no detailed information is available about the comprehensive changes in plasma metabolites at different stages of caging stress in ducks. We designed this experiment to analyze the effects of caging stress on performance parameters and oxidative stress indexes in ducks. Methods: Liquid chromatography tandem mass spectrometry (LC/MS-MS) was used to determine the changes in metabolites in duck plasma at 5 (CR5), 10 (CR10), and 15 (CR15) days after cage rearing and traditional breeding (TB). The associated pathways of differentially altered metabolites were analyzed using Kyoto encyclopedia of genes and genomes (KEGG) database. Results: The results of this study indicate that caging stress decreased performance parameters, and the plasma total superoxide dismutase levels were increased in the CR10 group compared with the other groups. In addition, 1,431 metabolites were detected. Compared with the TB group, 134, 381, and 190 differentially produced metabolites were identified in the CR5, CR10, and CR15 groups, respectively. The results of principal component analysis (PCA) show that the selected components sufficiently distinguish the TB group and CR10 group. KEGG analysis results revealed that the differentially altered metabolites in duck plasma from the CR5 and TB groups were mainly associated with ovarian steroidogenesis, biosynthesis of unsaturated fatty acids, and phenylalanine metabolism. Conclusion: In this study, the production performance, blood indexes, number of metabolites and PCA were compared to determine effect of the caging stress stage on ducks. We inferred from the experimental results that caging-stressed ducks were in the sensitive phase in the first 5 days after caging, caging for approximately 10 days was an important transition phase, and then the duck continually adapted.         Optimal Design of Multi-DOF Deflection Type PM Motor by Response Surface Methodology

        This paper uses response surface methodology as the optimization method of torque of multi-DOF deflection type PM motor. Firstly, the application of Taguchi algorithm selects structural parameters affecting the motor torque largely which simplifies the optimization process greatly. Then, based on the central composite design (CCD), response surface equation numerical model is constructed by the finite element method. With the aid of experiment design and analysis software, the effects of the interaction among factors on the index are analyzed. The results show that the analytical method is efficient and reliable and the experimental results can be predicted by response surface functions.
