        Serial values for hematologic and biochemical analysis after myocardial infarction in rats

        To diagnose acute myocardial infarction(MI), many cardiac markers have been used in hematologic and biochemical analysis, and many studies have been published for hema-tologic and biochemical analysis associated with human acute MI. However, after occurrence of acute MI, the serial investigation for values in hematologic and bioche-mical analysis including chronic MI has rarely been performed. To observe the change of the serial values in hematologic and biochemical analysis, we induced artificial MI. The left main descending artery(LMDA) of the left coronary artery was ligated during the progression(day 1, 3, 5, 7, 14 and 30) of MI. Total 66 Sprague-Dawley rats were divided into the sham group(n=24, thoracotomy without LMDA ligation) and the experimental(MI) group(n=42, with LMDA ligation). And all individual in each group was sacrified at day 1, 3, 5, 7, 14 and 30 for the hematologic and biochemical analysis. In comparison of hematologic analysis between the sham and MI groups, the mean values of red blood cell(RBCs), hemoglobin and hematocrit(HCT) showed a steady increase. In biochemical analysis, the mean values of glucose, choles-terol, total creatine kinase(CK) and isoenzyme MB, and lactate dehydrogenase(LDH) were increased in all MI groups compared with the sham groups. The results of this study suggest that early hematologic and biochemical mean values occurred after acute MI are similar to those of human acute MI. In conclusion, we could observe the alterations and serial values in hematologic and biochemical analysis to the extent of chronic status after acute MI.

        Alterations of Proteins in Artificially Induced Chronic Myocardial Infarction in Rats

        We investigated the changes of protein in chronic MI which was occurred with long-term ischemia, without reperfusion. Sprague Dawley (SD) rats were divided into the sham group and the experimental groups (MI groups). The sham group was treated only thoracotomy without ligation for left main descending artery (LMDA) of left coronary artery (LCA), and the experimental groups (MI7d, ligation of LMDA for 7 days and MI30d, ligation of LMDA for 30 days) were conducted an artificial chronic MI. The change of proteins according to passage of times was compared and analyzed on first and second dimension (1 and 2D) sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis. Among total 46 spots expressed differentially in the sham group versus MI7d and MI30d groups on 2D gel, we selected proteins that the volume of spot was increased in the MI7d and MI30d groups compared with the sham group. After that, the proteins were identified through liquid chromatography/tandem mass spectrometry (LC-MS/MS) analysis. In result, we could obtain many proteins as follows; albumin, glucose regulated protein 58 KDa, similar to tripartite motif protein 50, ubiquinol-cytochrome c reductase core protein Ⅱ, sarcomeric mitochondrial creatine kinase, ATP synthetase alpha chain (mitochondrial precursor) and creatine kinase. In conclusion, we suggest many changed proteins shown at chronic ischemia after artificial MI and consider that these proteins play an important role in the function of heart after MI.

        만주어 사전 구축을 위한 『동문유해(同文類解)』의 활용 가능성 모색

        본고는 한(漢)-조(朝)-만(滿) 어휘집인 『同文類解』의 체제와 그 특징을 살핌으로써 이를 새로운 만주어 사전 구축에 활용할 수 있는 방안을 살피는 것을 목적으로 한다. 『同文類解』는 상당한 수의 만주어 핵심 어휘를 중심으로 그와 형태적·의미적으로 연관된 관련어들에 대한 많은 정보를 담고 있다. 특히 자료에 직접적으로 드러나 있는 어휘 의미나 문법에 대한 정보를 비롯하여 이차적으로 파악되는 단어들 간의 파생 관계나 연어 정보들은 만주어를 보다 효과적으로 이해하는 데 도움을 준다. 그럼에도 불구하고 그동안 『同文類解』는 만주어 사전을 만드는 데 적극적으로 활용되지 못했다. 본고에서는 『同文類解』에 담겨 있는 만주어 어휘에 대한 정보들이 앞으로 새롭게 만들어질 만주어 사전에 풍부하고 가치 있는 요소를 제공해 줄 것이라 기대하며 그 사전적 활용 가능성을 모색하였다. This study aims to search for possibilities of using Dong-Mun-Yu-Hae(同文類解), a wordbook of Chinese-Korean-Manchu, in order to make new Manchu dictionary by considering the system and features of this book. Dong-Mun-Yu-Hae(同文類解) has many information about connected words, which are related to headwords either morphologically or semantically. Especially the additional information about grammar or meaning shown directly in the book, the derivational relationship among words, and the patterns of collocation in certain words are very useful for understanding Manchu vocabulary more systematically. Considering the situation, Dong-Mun-Yu-Hae(同文類解) has not been actively used in making Manchu dictionary so far, we are expecting that the various contents in this book enable newly made future Manchu dictionary to have plentiful and valuable elements in showing Manchu vocabulary.

        『동문유해(同文類解)』의 만주어(滿洲語)한글 표기 체계에 대하여

        본고는 조선시대 간행된 한조만(漢朝滿) 어휘집인 『同文類解』(1748)에 나타난 한글 표기 및 구별 기호에 대해 문헌 전체의 용례를 검토하여 그 특징을 개관하고 구별 기호의 성격을 파악해 보고자 하였다. 만주어 한글 전사의 구체적인 모습은 청학서(淸學書) 가운데 하나인 『同文類解』를 통해 살펴볼 수 있는데, 여기에 사용된 한글의 조합 방식이 매우 독특하다. 또한 『同文類解』의 전사에는 문자적 요소인 한글뿐만 아니라 이와 구분되는 몇 가지 구별 기호들이 사용되었는데, 각기호들은 그 모양과 위치에 따라 서로 다른 기능을 가진 것으로 보인다. 본고에서는 먼저 『同文類解』의 만주어 한글 표기의 체계를 각각 초성, 중성, 종성으로 나누어 살펴보고, 만주어의 한글 표기를 전산화하기 위해 구별 기호(°, ┚)가 포함된 한글 표기에 대한 적절한 입력 방안을 마련해 보았다. 이어서 구별 기호를중심으로 그 종류 및 기능에 대해 논의하였다. 본고는 연구 방법의 측면에서 만주어의 한글 표기에 사용된 구별 기호에 대해 『同文類解』의 전체 용례를 정리하고 검토하였다는 점에서 연구사적 의의를 지닌다. This study aims to look into the transcription system of Manchurian using Hangeul in Dong-Mun-Yu-Hae(同文類解), especially the diacritics presenting the various phonetic properties of Manchurian. It is presumed that the diacritics have diverse functions according to their forms and positions. Nevertheless, so far there has been no clear explanation about it. It is important to focus on the fact that Hangeul was used to transcribe Manchurian in the Joseon Dynasty since 18th century. This Hangeul transcription of Manchurian could offer the different and specific information about Manchurian and help us to understand the phonological aspect of Manchurian. In this study, we look into the whole text of Dong-Mun-Yu-Hae(同文類解) and classify the functions of diacritics used in this text. In addition, we propose the way of computerizing the diacritics with simple signs consisting of circle and line. Although we could not make clear the functions of diacritics completely, this kind of study will lay the foundation for further study of Manchurian.

        Apoptotic effect of $IP_6$ was not enhanced by co-treatment with myo-inositol in prostate carcinoma PC3 cells

        Inositol hexaphosphate ($IP_6$) is a major constituent of most cereals, legumes, nuts, oil seeds and soybean. Previous studies reported the anticancer effect of $IP_6$ and suggested that co-treatment of $IP_6$ with inositol may enhance anticancer effect of $IP_6$. Although the anticancer effect of $IP_6$ has been intensively studied, the combinational effect of $IP_6$ and inositol and involved mechanisms are not well understood so far. In the present study, we investigated the effect of $IP_6$ and myo-inositol (MI) on cell cycle regulation and apoptosis using PC3 prostate cancer cell lines. When cell, were co-treated with $IP_6$ and MI, the extent of cell growth inhibition was significantly increased than that by $IP_6$ alone. To identify the effect of $IP_6$ and MI on apoptosis, the activity of caspase-3 was measured. The caspase-3 activity was significantly increased when cells were treated with either $IP_6$ alone or both $IP_6$ and MI, with no significant enhancement by co-treatment. To investigate the effect of $IP_6$ and MI of cell cycle arrest, we measured p21 mRNA expression in PC3 cells and observed significant increase in p21 mRNA by $IP_6$. But synergistic regulation by co-treatment with $IP_6$ and MI was not observed. In addition, there was no significant effect by co-treatment compared to $IP_6$ treatment on the regulation of cell cycle progression although $IP_6$ significantly changed cell cycle distribution in the presence of MI or not. Therefore, these findings support that $IP_6$ has anticancer function by induction of apoptosis and regulation of cell cycle. However, synergistic effect by MI on cell cycle regulation and apoptosis was not observed in PC3 prostate cancer cells.

        Disease Occurrence in Transgenic Rice Plant Transformed with Silbene Synthase Gene and Evaluation of Possible Horizontal Gene Transfer to Plant Pathogens

        Genetic engineering is being used to enhance disease resistance and nutritional value of crops including rice plant. Considering the fast-growing agricultural biotechnology and rapidly increasing global area of transgenic crops, the risk evaluation on environment is necessary. In this study, we surveyed the difference of disease occurrence between transgenic rice variety, Iksan526 transformed with peanut stilbene synthase gene and non-transgenic rice varieties, Dongjin and Nampyeong in the field. Moreover, the possibility of gene transfer from transgenic rice to bacterial and fungal pathogens was investigated. The results of this study indicated that there was no significant difference in the occurrence and severity of the diseases between Iksan526 and Dongjin or Nampyeong. In addition, the results suggested that rice pathogen, such as Xanthomonas oryzae pv. oryzae, Rhizoctonia solani and Magnaporthe grisea did not take up stilbene synthase and bar genes under natural conditions. Moreover the transformed DNA was not transferred to the pathogens even in repetitive contacts.

        로즈마리 추출물 및 분획물의 항균ㆍ항산화 활성

        허브류의 하나인 로즈마리를 추출, 분획하여 각 용매별 항산화와 항균효과를 관찰하였다. 로즈마리의 항산화 효과를 측정하기 위해 DPPH radical 소거능과, ABTS radical 소거능 및 FRAP 활성을 측정하였다. DPPH radical 소거능을 측정한 결과 ethyl acetate분획물의 RC??값이 3.22 ㎍/㎖로 강력한 항산화능을 나타냈으며, water 분획물을 제외한 나머지 분획물(methanol: 5.05 ㎍/㎖, hexane: 6.28 ㎍/㎖, chlorofrom: 5.32 ㎍/㎖, buthanol: 5.74 ㎍/㎖)에서도 강력한 항산화능을 보였다. FRAP활성 또한 ethyl acetate 분획물이 5.9±0.3 μM/μg의 높은 Fe²?함량을 나타났으며 water 분획물을 제외한 나머지 분획물에서 높은 Fe²?함량을 나타내어 DPPH radical 소거능과 유사한 경향을 보였다. ABTS radical 소거능은 ethyl acetate분획물의 RC??값이 5.05 ㎍/㎖, methanol 추출물의 RC??값이 8.72 ㎍/㎖으로 강력한 항산화능을 나타냈으며 그 외 나머지 분획물들은 DPPH radical 소거능보다 상대적으로 낮은 항산화능을 나타냈다. 로즈마리 단일물질로는 rosmarinic acid, carnosic acid가 DPPH radical 소거능과 FRAP활성에 유사한 경향을 보인 반면 ABTS radical 소거능은 rosmarinic acid만이 43.16 μM에서 RC??값을 나타내었다. 로즈마리의 항균 활성은 Paper disc method를 이용한 Inhibition zone과 MIC (최소 저해 농도)를 측정하여 항균력을 알아본 후 hand plate와 resazurin 환원 실험을 통해 항균제와 식품 보존제로서의 이용 가능성을 알아보았다. 항균 활성 검색 결과 methanol 추출물과 chloroform 분획물이 S. aureus에 대해서 2.5 ㎎/disc의 농도에서 inhibition zone이 각각 15.3 ㎜, 15.5 ㎜로 높게 나타났으며 E. coli에 대해서 14.3 ㎜, 13.7 ㎜로 나타나 강력한 항균 효과를 나타내었다. MIC는 S. aureaus 균주와 E. coli 균주 모두 methanol 추출물에서 5 ㎎/㎖의 MIC값을 나타내었고 chloroform 분획물은 2 ㎎/㎖에서 MIC값을 나타내었다. 항균활성의 경우 methanol 추출물과 소수성의 hexane, chloroform 분획물에서 높은 활성을 보였다. 위의 결과를 통해 항균제와식품 보존제 등의 상품화는 경제적이고 그 활성이 우수한 methanol 추출물의 이용이 가장 이상적인 것으로 판단되어 methanol 추출물을 이용하여 항균제와 식품 보존제로의 이용 가능성을 측정한 결과 항균제로는 hand plate를 통해 5 ㎎/㎖에서 식품 보존제로는 resazurin 환원 실험을 통해 1 ㎎/㎖에서 이용이 가능하리라 생각된다. This study was performed to evaluate the antioxidant and antimicrobial activity of methanol extract from Rosmarinus officinalisL. and its fractions. The ethyl acetate fraction of rosemary had a higher antioxidant activity in both DPPH (3.22 ㎍/㎖) and ABTS (5.05 ㎍/㎖) compared to other extracts and fractions. Based on the results of the FRAP assay, the ethyl acetate fraction of rosemary showed a value of 5.9±0.3 μM/㎍, and buthanol fraction and rosmarinic acid exhibited values of 4.8±0.2 μM/㎍ and 5.1±0.1 μM/μM, respectively. Measurements of the antimicrobial activities of the extracts, fraction against gram positive, negative bacteria revealed that the methanol extract, hexane, ethyl acetate, and chloroform fraction of rosemary caused Staphylococcus aureusand Escherichia colito form clear zones greater than 12 mm. Furthermore, the methanol extract and chloroform fraction showed high antibacterial activity, with inhibition zone exceeding 13 ㎜. The methanol extract and chloroform fraction of rosemary had broad antimicrobial spectrums and low MIC values. Therefore, methanol extracts of rosemary could serve as potential antibacterial agents to inhibit pathogen growth in food and hand sanitizers.
