Purpose: Fanconi anemia complementation group F (FANCF) is a key factor to maintaining the function of Fanconi anaemia/BRCA (FA/BRCA) pathway, a DNA-damage response pathway. However,the functional role of FANCF in breast cancer has not been elucidated. In the present study, we evaluated the chemosensitization effect of FANCF in breast cancer cells. Methods: We performed specific knockdown of the endogenous FANCF in breast cancer cells by transfecting the cells with an FANCF short hairpin RNA (shRNA) vector. Cell viability was measured with a Cell Counting Kit-8, and DNA damage was assessed with the alkaline comet assay. The apoptosis, cell cycle, and drug accumulation were measured by flow cytometric analysis. Protein expression levels were determined by Western blot analysis, using specific antibodies. Results: The analyses of two breast cancer cell lines (MCF-7 and MDA-MB-435S) demonstrated that the FANCF shRNA could effectively block the FA/BRCA pathway through the inhibition of Fanconi anemia complementation group D2ubiquitination. Moreover, FANCF silencing potentiated the sensitivity of cells to mitomycin C (MMC), where combined FANCF shRNA/MMC treatment inhibited cell proliferation, induced Sphase arrest, apoptosis, and DNA fragmentation, and reduced the mitochondrial membrane potential, compared with MMC treatment alone. Conclusion: Taken together, this study demonstrates that the inhibition of FANCF by its shRNA leads to a synergistic enhancement of MMC cytotoxicity in breast cancer cells. These results suggest that the inhibition of the FA/BRCA pathway is a useful adjunct to cytotoxic chemotherapy for the treatment of breast cancer.
850°C 고온에서 Alloy 800H의 저사이클 피로 거동

Alloy 800H의 고온 저사이클 피로 특성에 관한 연구의 일환으로써, 현재 연구의 목적은 850°C 고온에서 Alloy 800H의 저사이클 피로 거동을 고찰하고 피로 수명을 평가하기 위한 것이다. 본 연구에서 850°C의 고온에서 저사이클 피로 거동을 이해하기 위하여 체계적인 실험이 수행되었다. 저사이클 피로시험은 완전 양진 변형률 제어로 하였으며 0.6, 0.9, 1.2 및 1.5%의 서로 다른 네 가지 전변형률 범위에서 10-3/s의 일정한 변형률 속도 하에서 수행하였다. 저사이클 피로 수명은 전변형률 범위가 증가할수록 감소하였다. 피로 수명과 반복 응력-변형률 거동에 미치는 전변형률 범위에는 강한 영향이 있음이 관찰되었다. 또한 피로 수명을 Coffin-Manson 법칙에 의하여 평가하였다. 마지막으로 파단면을 주사 전자 현미경과 광학 현미경을 통해 분석하였다. As part of a series of studies on the high temperature low cycle fatigue properties of Alloy 800H, the objective of the present study is to investigate the low cycle fatigue behavior of Alloy 800H at high temperature of 850 °C and evaluate the fatigue life. Systematic experiments were conducted to understand the low-cycle fatigue behavior of this alloy at high temperature of 850 °C. Low-cycle fatigue tests were conducted under fully reversed axial total strain range control mode employing a triangular waveform, under constant strain rate of 10-3/s with four different total strain ranges of 0.6 %, 0.9 %, 1.2 % and 1.5 % at 850 °C. The low-cycle fatigue life decreased with increasing total strain range. Strong influence of total strain range on low-cycle fatigue life and cyclic stress-strain behavior was observed. And also, the fatigue life was evaluated by the Coffin-Manson method. Finally, the fracture surface was analyzed using scanning electron microscope (SEM) and optical microscope (OM).
A study on the risk investigation and safety of personal care robots

Personal care robots are recognized for their importance as they recognize the need for interaction between human beings and robots in non-production environments. In this study, we collected and analyzed accident cases and statistical data by type of domestic and overseas to utilize in order to establish App Guide and Test method before certified based on safety requirement standard of personal care robot. Through this research, it is aimed to summarize the types of accidents of personal assisted robots and to use them to develop the risk assessment system.
