        황금(黃芩)의 4-VO로 유발한 흰쥐뇌허혈에 대한 신경방어효과

        Scutellariae Radix(SR), the root of Scutellaria baicalensis G_EORGI, whose property, flavour and channel tropism is bitter in flavour, cold in property(寒), acting on the lung, gallbladder, stomach and large intestine channels(歸?,?,?,大??), has the effects of clearing away heat and dampness(淸熱??), purging fire, detoxicating(?火解?), stopping bleeding(止血) and preventing miscarriage(安?). This drug is one of the popular drugs in traditional Korean medicine, which has been used to antipyretic, antibacterial, antitoxic and antihypersensitive effects. So this study was planned to check the neuroprotective effect of SR on the global ischemia induced by 4-vessel occlusion in Wister rats. and SR extract was lyophilized after extraction with 70% methanol. We induced 4-vessel occlusion for 10 minutes and reperfused again. The number of CA1 pyramidal neurons were counted after 7 days of reperfusion under the cresyl violet staining. The result obtained that in 4-VO ischemia, SR showed significantly neuroprotective effects(1,000 and 500 ㎎/㎏ of SR extracts, p<0.05) compared with control group. Each neuroprotective ratio was about 27.4 %, 23.2 % respectively. Consequently, Scutellariae Radix has neuroprotective effects on the global ischemia induced by 4-vessel occlusion in Wistar rats.

      • 출아효모에서 TAR 클론닝법을 이용한 고등동물의 게놈으로부터 특정 염색체 부위의 분리

        복잡한 게놈 분석에 용이하도록 효모의 인공 염색체(YAC) 클론닝 시스템은 발달되어왔다. YAC은 박테리아의 인공 염색체(BAC)보다 더 큰 단편을 클론닝할 수 있고, 또한 클론된 단편을 쉽게 변형시킬 수 있다. 형질전환과 연계된 재조합법(Transformation-Associated Recombination; TAR)은 게놈 라이브러리를 만들지 않고 직접 게놈 DNA로부터 분리하고자 하는 유전자나 특정 염색체 부분을 클론닝 할 수 있는 방법이다. 이 방법은 spheroplast transformation을 수행하는 동안, 목적으로 하는 유전자의 5' 그리고 3' 염기 배열(hooks)을 지닌 TAR 벡터와 게놈 DNA 사이에서 일어나는 상동성 재조합에 의해 이루어진다. 효모 내의 in vivo 재조합을 이용한 TAR 클론닝법은 복잡한 게놈으로부터 목적의 염색체 부분을 원형 YAC의 형태로서 선택적으로 분리할 수 있다. 그러므로 TAR 클론닝 법은 특정 염색체로부터 YACs을 만드는데 매우 유용하여, 전체 게놈으로부터 특이적 유전자나 유전자의 family를 분리하는데 효과적인 방법으로 사료된다. Yeast artificial chromosome (YAC) cloning systems have advanced the analysis of complex genomes considerably. They permit the cloning of larger fragments than do bacterial artificial chromosome systems, and the cloned material is more easily modified. Transformation-associated recombination (TAR) is a cloning technique that allows specific chromosomal regions or genes to be isolated directly from genomic DNA without prior construction of a genomic library. This technique involves homologous recombination during spheroplast transformation between genomic DNA and a TAR vector that has 5' and 3' gene targeting sequences (hooks). Using in vivo recombination in yeast, TAR cloning selectively isolates, as circular YACs, desired chromosome segments or entire genes from complex genomes. We propose that TAR cloning can provide an efficient means for generating YACs from specific chromosomes and that TAR cloning may be useful for isolating families of genes and specific genes from total genome DNA.

      • 질편모충 항원분석을 위한 단백빌 용출방법의 비교

        The purification methods such as bead-beater destruction, trichloroacetic acid(TCA) precipitation and 3-[(3-cholamidopropyl) dimethylammonio]-1-propane sulfonate(CHAPS) solubilization of Trichomonas vaginalis antigens on sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) and enzyme-linked immunoelectrotransfer blotting (EITB) were evaluated. Total TCA-precipitated proteins and CHAPS-solubilized extract of T. vaginalis possessed common protein bands whose molecular weights were at 28 kDa, 35 kDa, 40 kDa, 50 kDa, 60 kDa, 100 kDa, 120 kDa, 130 kDa, and 160 kDa, and bead-beater treated trichomonal proteins represented some different spectrum of protein bands from TCA- or CHAPS-treated ones in that they didn't show high M.W. protein bands(〉100 kDa) on SDS-PAGE. But similar antigenic bands with the molecular weights of 60 kDa, 110 kDa, 120 kDa and 140 kDa by EITB were identified in TCA-, CHAPS- and bead-beater treated proteins. In conclusion, TCA-precipitation and CHAPS-solubilization were more stable methods to analyse trichomonal proteins than extraction with bead-beater, and the result will be useful for antigen analysis of Trichomonas vaginalis to elucidate a mechanism of pathogenesis in host-parasite relationship.

        Recent reports have proposed that Ephedrae Herba may modulate the immune response on allergy or asthma. Human nasal mucosal fibroblasts are a rich source cytokines, inflammatory mediators, and chemokines. Chemokines are important for the recruitment of leukocytes to sites of infection, which is essential in host defense. Objectives : The objective of this study was to investigate the effect of Ephedrae Herba on the release of the chemokines such as monocyte chemotactic protein(MCP)-1, MCP-2, and MCP-3 in human nasal mucosal fibroblasts after stimulation with cytokines like tumor necrosis factor -α(TNF-α), interferon-γ(IFN-γ), and interleukin -1β(IL-1β).

        Performance of Hybrid Laser Diodes Consisting of Silicon Slab and InP/InGaAsP Deep-Ridge Waveguides

        The fundamental transverse mode lasing of a hybrid laser diode is a prerequisite for efficient coupling to a single-mode silicon waveguide, which is necessary for a wavelength-division multiplexing silicon interconnection. We investigate the lasing mode profile for a hybrid laser diode consisting of silicon slab and InP/InGaAsP deep ridge waveguides. When the thickness of the top silicon is 220 nm, the fundamental transverse mode is lasing in spite of the wide waveguide width of 3.7m. The threshold current is 40 mA, and the maximum output power is 5 mW under CW current operation. In the case of a thick top silicon layer (1 m), the higher modes are lasing. There is no significant difference in the thermal resistance of the two devices.

        오적산 병용투여가 혈당강하 치료를 받고 있는 제2형 당뇨병 환자의 혈당에 미치는 영향

        Objective: This study was conducted to investigate the hypoglycemic effect and safety of Ojeok-san in patients with type 2 Diabetes Mellitus. Methods: We investigated type 2 diabetes mellitus patients at Kyung-Hee University Korean Medical Hospital who were administered Ojeok-san for at least one day between January 2012 and September 2020, basal characteristics and laboratory tests were reviewed retrospectively. The hypoglycemic effect of Ojeok-san was assessed by comparing fasting blood sugar (FBS) and two hours post-prandial plasma glucose (PP2) levels from before and after taking Ojeok-san. Subgroup analyses were conducted according to baseline hypoglycemic treatments and glycated hemoglobin levels (< or ≥6.5%). The safety of Ojeok-san was assessed by comparing levels of aspartate aminotransferase, alanine aminotransferase, γ-glutamyl transferase, blood urea nitrogen, and creatinine. Results: After Ojeok-san administration, FBS and PP2 were significantly reduced to an average of 14.33 mg/dL and 27.67 mg/dL respectively. In the subgroup analysis, PP2 in patients receiving metformin mono therapy was significantly reduced to 28.67 mg/dL, and those receiving a dual therapy of metformin and DPP-4 inhibitor, it was significantly reduced to 32.33 mg/dL. In patients with glycated hemoglobin of lower than 6.5%, FBS was significantly reduced to 12.20 mg/dL, and both FBS and PP2 were significantly reduced, to 15.50 mg/dL and 40.00 mg/dL, respectively, in those with glycated hemoglobin levels of more than 6.5%. The safety profile showed no significant difference after Ojeok-san administration. Conclusions: Ojeok-san has significant hypoglycemic effects in patients with type 2 diabetes mellitus who are also taking hypoglycemic agents.

        Prmt7 regulates the JAK/STAT/Socs3 signaling pathway in postmenopausal cardiomyopathy

        Protein arginine methyltransferases (PRMTs) modulate diverse cellular processes, including stress responses. The present study explored the role of Prmt7 in protecting against menopause-associated cardiomyopathy. Mice with cardiac-specific Prmt7 ablation (cKO) exhibited sex-specific cardiomyopathy. Male cKO mice exhibited impaired cardiac function, myocardial hypertrophy, and interstitial fibrosis associated with increased oxidative stress. Interestingly, female cKO mice predominantly exhibited comparable phenotypes only after menopause or ovariectomy (OVX). Prmt7 inhibition in cardiomyocytes exacerbated doxorubicin (DOX)-induced oxidative stress and DNA double-strand breaks, along with apoptosis-related protein expression. Treatment with 17β-estradiol (E2) attenuated the DOX-induced decrease in Prmt7 expression in cardiomyocytes, and Prmt7 depletion abrogated the protective effect of E2 against DOX-induced cardiotoxicity. Transcriptome analysis of ovariectomized wild-type (WT) or cKO hearts and mechanical analysis of Prmt7-deficient cardiomyocytes demonstrated that Prmt7 is required for the control of the JAK/STAT signaling pathway by regulating the expression of suppressor of cytokine signaling 3 (Socs3), which is a negative feedback inhibitor of the JAK/STAT signaling pathway. These data indicate that Prmt7 has a sex-specific cardioprotective effect by regulating the JAK/STAT signaling pathway and, ultimately, may be a potential therapeutic tool for heart failure treatment depending on sex.

      • 겔여과크로마토그라피에 의한 질트리코모나스의 항원분획 분석

        This study was aimed to analyze the antigenicity of T. vaginalis using a high performance gel filtration chormatography(HPLC), enzyme-linked immunosorbent assay (ELISA) and enzyme-linked immunoelectotransfer blot (EITB). Soluble antigen was prepared using a bead beater. Antiserum was collected from rabbit immunized previously. The results obtained are as follows: the crude antigen of T.vaginalis was separated into 7 peak by HPLC and the fraction 3 revealed the highest reactivity in DLISA. The immunogenic bands of fraction 3 were visualized at 38, 45, 47, 55, 64, 78, 94, 115, 126 and 180 KDa and the bands at 38KDa and 180KDa showed stronger reactivity than that of equivalents of the crude antigen in EITB. In conclusion the fraction 3 of the crude antigen of T. vaginalis obtained by chromatographic separation may have a high antigenicity and proteins ay 38KDa and 180KDa of the fraction 3 may play an important role in antigen-antibody reaction in T. vaginalis infection.

        Cdon suppresses vascular smooth muscle calcification via repression of the Wnt/Runx2 Axis

        Osteogenic transdifferentiation of vascular smooth muscle cells (VSMCs) is a risk factor associated with vascular diseases. Wnt signaling is one of the major mechanisms implicated in the osteogenic conversion of VSMCs. Since Cdon has a negative effect on Wnt signaling in distinct cellular processes, we sought to investigate the role of Cdon in vascular calcification. The expression of Cdon was significantly downregulated in VSMCs of the aortas of patients with atherosclerosis and aortic stenosis. Consistently, calcification models, including vitamin D3 (VD3)-injected mice and VSMCs cultured with calcifying media, exhibited reduced Cdon expression. Cdon ablation mice (cKO) exhibited exacerbated aortic stiffness and calcification in response to VD3 compared to the controls. Cdon depletion induced the osteogenic conversion of VSMCs accompanied by cellular senescence. The Cdon-deficient aortas showed a significant alteration in gene expression related to cell proliferation and differentiation together with Wnt signaling regulators. Consistently, Cdon depletion or overexpression in VSMCs elevated or attenuated Wnt-reporter activities, respectively. The deletion mutant of the second immunoglobulin domain (Ig2) in the Cdon ectodomain failed to suppress Wnt signaling and osteogenic conversion of VSMCs. Furthermore, treatment with purified recombinant proteins of the entire ectodomain or Ig2 domain of Cdon displayed suppressive effects on Wnt signaling and VSMC calcification. Our results demonstrate a protective role of Cdon in VSMC calcification by suppressing Wnt signaling. The Ig2 domain of Cdon has the potential as a therapeutic tool to prevent vascular calcification.

      • 절편모충(Trichomonas vaginalis)에 대한 단일클론항체의 생산과 그 특성

        Trichomonas vaginalis, mucosal protozoan parasite of the urogenital tract, is responsible for the most common clinically recognized sexually transmitted disease. Surface antigen of parasitic protozoa play the important roles in disease pathogenesis by serving as attachment factors, direct virulence factors, or mediating evasion of the host immune response through direct suppressoin or antigenic variation. In this study seven monoclonal antibodies against plasma membrane of T. vaginalis HY-1 were prepared and were investigated the effector function of them. Seven monoclonal antibodies were produced and their isotypes were Ig G1 (Tv1, Tv7), Ig G2a (Tv4), Ig M(Tv2, Tv3, Tv5, Tv6). All of the seven monoclonal antibodies recognized the determinants located on the surface of live organism by IFAT. Although these surface reactive seven monoclonal antibodies agglutinated T. vaginalis trophozoites, especially MAb Tv1 and Tv4 could agglutinate eight isolates of T. vaginalis. In immunoblot assay, MAb Tv1 & Tv4 detected single peptide with molecular weight of 60 kDa, and Tv7 detected 80 kDa. On the other hand Tv2 and Tv6 detected two (40 kDa & 120 kDa) or more peptides (62 kDa, 64 kDa & 120 kDa), and Tv3 and Tv5 did not bind any pepetide. From these results, MAb Tv1 reacted with 60 kDa, major surface membrane protein of T. vaginalis and agglutinated trichomonads.
