        15d-PGJ2 inhibits NF-kB and AP-1-mediated MMP-9 expression and invasion of breast cancer cell by means of a heme oxygenase-1-dependent mechanism

        Activation of peroxisome proliferator-activated receptor  (PPAR) serves as a key factor in the proliferation and invasion of breast cancer cells and is a potential therapeutic target for breast cancer. However, the mechanisms underlying this effect remain largely unknown. Heme oxygenase-1 (HO-1) is induced and overexpressed in various cancers and is associated with features of tumor aggressiveness. Recent studies have shown that HO-1 is a major downstream target of PPAR. In this study, we investigated the effects of induction of HO-1 by PPAR on TPAinduced MMP-9 expression and cell invasion using MCF-7 breast cancer cells. TPA treatment increased NF-B /AP-1 DNA binding as well as MMP-9 expression. These effects were significantly blocked by 15d-PGJ2, a natural PPAR ligand. 15d-PGJ2 induced HO-1 expression in a dose-dependent manner. Interestingly, HO-1 siRNA significantly attenuated the inhibition of TPA-induced MMP-9 protein expression and cell invasion by 15d-PGJ2. These results suggest that 15d-PGJ2 inhibits TPA-induced MMP- 9 expression and invasion of MCF-7 cells by means of a heme oxygenase-1-dependent mechanism. Therefore, PPAR/HO-1 signaling- pathway inhibition may be beneficial for prevention and treatment of breast cancer.

        정신분열병에 대한 리스페리돈의 효과 및 안정성

        연구목적 : 본 시험의 목적은 임상시험 시작전에 연구자들을 대상으로 PANSS Workshop을 통하여 PANSS, ESRS에 대한 국내에서의 표준화 작업을 구축하고 새로운 정신병 치료제인 리스페리돈의 효과와 안정성을 재확인하여 리스페리돈 사용에 대한 적정화를 이루는데 있다. 연구방법 : 1996년 4월부터 1996년 9월까지 국내 39개 대학병원 정신과에 입원중인 혹은 증상이 악화되어 입원하는 정신분열병 환자 377명을 대상으로 다시설 개방 연구를 시행하였다. 1주일간의 약물 배설기간을 가진후, 리스페리돈을 8주간 투여하였고, 기준점, 1주, 2주, 4주, 그리고 8주후에 평가되었다. 용량은 제1일에는 리스페리돈 1mg씩 1일 2회, 제2일에는 2mg씩 1일 2회, 제3∼7일에는 3mg씩 1일 2회 투여하였다. 이후 환자의 임상상태에 따라 임의로 증량할 수 있으며, 최대 일일 16mg을 초과하지 않도록 하였다. 추체외로 증상을 조절하기 위한 투약을 허용하였다. 임상증상 및 부작용의 평가는 PANSS(Positive and Negative Syndrome Scale), CGI(Clinical Global Impression) 그리고 ESRS(Extrapyramidal Symptom Rating Scale)을 사용하였다. 연구결과 : 377명중 343명(91%)이 8주간의 연구를 완결하였다. 치료 종결시점인 8주후 PANSS 총점수가 20% 이상 호전된 경우를 약물 반응군으로 정의할때, 약물반응군은 81.3%였다. 리스페리돈에 반응하는 예측인자로는 발병연령, 이전의 입원 횟수, 유병기간이 관련 있었다. 리스페리돈은 1주후부터 PANSS양성, 음성, 및 일반정신병리 점수상에 유의한 호전을 보여 효과가 빨랐다. CGI의 경우도 기준점에 비해 1주후부터 유의한 감소를 나타내었다. ESRS의 경우, 파킨슨 평가점수는 기준점과 비교해 투여 1주, 2주, 4주후 유의하게 증가되었다가 8주후 기준점과 차이가 없었다. Dystonia 평가점수는 1주후만 유의한 증가를 보였으며, dyskinesia 평가점수는 유의한 차이가 없었다. 혈압, 맥박수의 생명징후 및 일반 혈액학 검사, 생화학적 검사, 심전도 검사에서 유의한 변화는 없었다. 결 론 : 이상의 다시설 개방 임상 연구를 통해 리스페리돈은 정신분열병 환자에서 양성증상뿐만 아니라 음성증상 및 전반적인 증상에도 효과적인 것으로 사료된다. 보다 명확한 평가를 위해서는 다른 항정신병약물과의 이중맹검 연구가 필요할 것으로 생각되며, 또한 장기적 치료에 대한 평가도 함께 이루어져야 하겠다. Objective : The purpose of this study was to investigate the efficacy and safety of risperidone in the treatment of Korean schizophrenic patients. Method : This multicenter open study included 377 schizophrenic patients drawn from 39 university hospitals. After a wash-out period of 1 week, the schizophrenic patients were treated with risperidone for 8 weeks and evaluated at 5 points ; at baseline, and 1, 2, 4 and 8 weeks of treatment. The dose was increased from 2mg/day(1mg twice daily) to 6mg/day(3mg twice daily) during the first week and adjusted to a maximum of 16mg/day over the next 7 weeks according to the patient's clinical response. Medication to control extrapyramidal symptoms was permitted. The psychiatric and neurological status of the patients was assessed by PANSS, CGI, and ESRS scales. Results : 343(91%) of 377 patients completed the 8-week trial period. Clinical improvement, as defined by a 20% or more reduction in total PANSS score at end point, was shown by 81.3% of patients. The predictors of response to risperidone were associated older age, shorter duration of illness, fewer previous hospitalization. Risperidone had rapid onset of action ; a significant decrease of the total PANSS and three PANSS factor(positive, negative, general), and CGI was already noticed at the end of first week. For the ESRS, parkinsonism rating scores were significantly increased until week 4 comparing with baseline. Dystonia rating scores were significantly increased until week 1, and dyskinesia rating scores were not significantly changed during the study. Laboratory parameters including vital sign, EKG, hematological, and biochemical values showed no significant changes during the trial. Conclusions : This study suggests that risperidone is generally safe and effective against both the positive and negative symptoms in our group of patients.

      • 열처리 및 트립신 첨가로 야기된 유청 단백질의 전기영동적 변화

        In order to study the reaction behavior of the mixtures of bovine (β-Lg) with holo-α-lactalbumin (α-La) and apo-α-locatalbumin during heat treatment, samples were heated at 85℃ for 10 min, added trypsin and trypsin inhibitor, and then they were analyzed using sodium dodecylsulfate-polyacrylamide gel electrophoresis. In the other trial, the β-Lg was pre-heated at 85℃ for 10 min, mixed with α-La, heated at 85℃ for 10 min, and treated the same as above described. The electrophoretic pattern showed that the intensity of loss of β-Lg was higher than the a α-La with increasing reaction time. Loss rate of apo-α-La-β-Lg mixture was higher than that of holo-α-La-β-Lg mixture. When the mixture was heated and added trypsin, appeared some bands of about 26.3 kDa, and 14.0 kDa which is smaller than the α-La(14.2 kDa). When the β-Lg was pre-heated 85℃ for 10min, mixed with α-La, and treated the same as other samples, appeared some bands of about 26.3 kDa, 21.5 kDa, and 14.0 kDa which is smaller than that of the α-La. The loss rate of the apo-α-La-β-Lg mixture was higher than that of holo-α-La-β-La mixture. It is assumed that the β-La was already denatured by pre-heating and influenced little on the apo-α-La, and there for the apo-α-La-β-La mixture was more easily attacked by the enzyme.

      • 전기화학적 방법을 이용한 저온형 연료전지용 고분산 Pt 촉매의 제조

        Pt was deposited on carbon support by chronoamperometry. The catalytic activity for methanol oxidation was checked by CV (cyclic voltammetry) method. The amount of Pt loading was calculated by integration of I-t curve obtaining the total current used for the Pt deposition. The ECA (electrochemical active surface area) for loaded Pt was calculated from the hydrogen adsorption/desorption peaks in 0.5 M H₂SO₄solution. Pt was deposited on clean carbon electrode surface with various concentration of Pt solution (0.1 mM) was used. As the concentration of Pt increased, the specific catalystic activity of Pt decreased suggesting that, in the electrochemical reaction of Pt ion with electrode surface, Pt particles were smaller and more dispersed in low concentration of Pt solution. With this condition (0.1 mM Pt), Pt was electrodeposited on a commercial carbon support, Vulcan XC-72. Electrochemically loaded Pt catalyst had the higher ECA, and showed higher specific catalytic activity for mathanol oxidation than a commercial Pt/C catalyst.

        This review covers selected physicochemical and functional properties of α-lactalbumin and β-lactoglobulin. An understanding of chemical structure, heat stability, conformational changes of the two major whey proteins Is important to utilize for better dairy products and food ingredients. β-Lactoglobulin is more reactive and sensitive during heat treatment than α-lactalbumin due to a free sulfhydryl residue, which catalyzes formation of intermolecular disulfide bonds. The molecular mechanism of thermal interaction of individual and mixed proteins during processing, cooking and storage should be more systematically elucidated. Interactions between α-lactalbumin and β-lactoglobulin upon heating milk affect the extent of α-lactalbumin denaturation and aggregation. The functional properties of the two proteins are strongly influenced by pH, ionic composition, chemical structure, surface hydrophobicity and amino acid residue.

      • 만성 B형 간염환자에서 인터페론 치료 후 간 조직검사 소견의 변화와 Tumor Necrosis Factor의 임상적 의의

        Background: Interferon has been widely used in the treatment of chronic hepatitis B, but its effectiveness is debatable. The clinical indices as the loss of HBeAg, HBV DNA, and improvement of liver function are applicated to identify the effectiveness of interferon therapy, but the mechanism of hepatohistological change is not well known. We investigated the changes of histologic finding and plasma tumor necrosis factor(TNF) in the patients with chronic hepatitis B after treatment with interferon-alpha. Methods: 11 patients with chronic hepatitis B who had treated by interferon-alpha were enrolled for this study. Liver biopsy was done before and after treatment of interferon and plasma TNF was evaluated at the same time. A semiquantitative study using a histologic scoring system was applied on pre and post liver biopsy specimens and the changes of HBeAg and HBV DNA were also evaluated. Results: Among subjects, the response group, recurrence group, and non-response group based on serologic response with clearance of HbeAg were6, 3, and 2 patients, respectively. The changes of serum ALT level and HBV DNA titer on pre and post therapy decreased significantly in the response group. The change of mean plasma TNF level in the response group decreased insignificantly to 19.4 from 19.9. The histological improvements of grading and staging were shown 4 patients in the response group. Conclusions: Serologic response with clearance of HBeAg was associated with histolofical improvement. The loss of HBsAg in the serum was significant indicator of reduction in the amount of HBV core antigen and HBV surface antigen in the hepatocytes. Plasma TNF level was not associated with response of interferon-alpha therapy and histologic activity.

        Glucose Oxidase가 척수지각신경절세포에 미치는 영향과 천마의 효과에 관한 연구

        Objectives : To clarify the cytotoxic effect of glucose oxidase(GO) and protective effect of gastrodiae Rhizoma (GR) on spinal sensory ganglion(DRG) neurons, neurotoxicity mediated by GO was measured by MTT assay and neurofilament enzymeimmuno assay(EIA). Methods : DRG neurons were cultured in the media containing various concentrations of GO for 3 hours. In addition, neuroprotective effects of GR, on GO-induced neurotoxicity in DRG neurons were examined after DRG neurons were preincubated with various concentrations of GR for 2 hours before 15mU/ml GO for 3 hours. Results : GO decreased remarkably cell viability in dependently in these cultures, and also RG increased cell viability and amount of neurofilament in DRG neurons damaged by GO. Conclusion : It is suggested that GO has toxic effect in cultured mouse DRG neurons, and also RG was effective in the protection of GO-induced neurotoxicity in these cultures.

      • 말기환자 관리에 대한 의사들의 태도

        Background : Nowadays, it is possible to prolong the life. And to improve the quality of life of the dying patients, they need to receive not only the physical, pscychological, social and spiritual care but also comprehensive and continuous care. Also, doctors have to consider many ethical issues especially in the dying patients. The purpose of this study is to analyze the doctors' attitude of those ethical issues and emphasize the importance of hospice. Methods : During September 1996, doctors who worked for a university hospital participated in this study by responding to the pre-made questionnaire. It dealt with the most suffering pain, the preferred death place, the use of life maintaining equipments, the application of cardiopulmonary resuscitation(CPR) to terminal patients and the doctors' experience and attitude euthanasia. Results : ① The most suffering symptom in dying patients was pain(60.0%). ② The death place was recommended by doctors was home(58.8%) and hospital (15.4%) in order. ③ 56.9% of doctors answered 'the life maintaining equipments isn't needed', 29.2% of doctors answered' according to the case', and 'needed' was 12.3%. ④ The application of CPR to the terminal patients, without the DNR chart, the only 15.4% of doctors answered "Do not CPR." But with the DNR chart, the percents of 'Do not CPR' was increased to 69.2%. The resident group did more CPR than specialist group(p<0.01). ⑤ The 50.8% of doctors agreed with the passive euthanasia. Conclusions : Many doctors follow the family member's opinion in the ethical issues of the dying patients. Euthanasia is forbidden by the law in Korea, but many doctors agree with the passive euthanasia. Hospice can solve the debating ethical issues including the euthanasia.
