      • 전단지의 특성이 중국 소비자의 구매의도에 미치는 영향에 관한 연구

        본 연구에서는 전단지를 활용한 촉진이 중국 소비자들의 구매의도에 어떤 영향을 미치는지에 대해서 두 차례의, 실험연구를 통해서 분석하였다.본 연구에서는 전단지의 제품 관련 속성과 촉진 관련속성을 선정하여, 두 차례의 실험 설계를 통해서 전단지를 활용한 촉진이 중국 소비자들의 지각된 가치와 지각된 위험, 그리고 구매의도에 미치는 영향을 조사하였다.첫 번째 실험연구에서는 전단지의 제품 관련 속성으로서 할인 폭과 특성광고의 효과를 살펴보았으며, 두 번째 실험연구에서는 전단지의 촉진 관련 속성으로 브랜드 프로모션 유무와 연관상품 촉진 유무에 대해서 2x2의 실험 설계를 통해 조사하였다. 연구 결과, 전단지의 특성을 활용한 촉진은 중국 소비자들의 구매의도에 유의한 영향을 미치는 것으로 나타났다. 반면 지각된 위험은 구매의도에 유의한 영향을 미치지 못하는 것으로 나타났는데, 이는 중국 소비자들이 가격을 더 중시하기 때문으로 추정할 수 있다. 마지막으로 이에 따른 시사점과 한계점들을 서술하였다. This study presents two experiments to investigate how store flyer promotion affects Chinese customers`` purchase intention. The particular interest is whether store flyers have different influences in different countries. This paper analyzes China, one of the world’s fastest-growing economies as a representative to explore the issue. Specifically, this study proposes a promotion and product characteristics for empirically analyzing the effect of store flyer promotion on Chinese consumers`` purchase intention. Tested through two experiments, the results demonstrate that store flyer promotion has a significant positive effect on Chinese consumers’ perceived value and purchase intention. These findings indicate that Chinese consumers are price-oriented rather than quality-oriented, and store flyer promotion are effective marketing tools for Chinese retailers.

        Effect of cadmium exposure on antioxidant enzyme catalase in different tissues of Acrossocheilus fasciatus

        Catalase (CAT) is an essential antioxidant in organisms, its can eliminate hydrogen peroxides produced in the cellular environment. Previous studies have focused mainly on the functions of CAT by pathogen challenge, but the functions of CAT by metals stress remain unclear. In order to investigate the function of CAT in Acrossocheilus fasciatus (AfCat), we cloned CAT complete cDNA sequences and analyzed its functions by cadmium (Cd) stress. qPCR analysis illustrated that the expression of CAT changed rapidly and dynamically in response to Cd from 0.05 mg L-1 to 1.25 mg L-1. Enzyme activity assay result showed that CAT activity was up-regulated significantly in response to Cd from 0.05 mg L-1 to 1.25 mg L-1. Histochemistry analysis showed that the structure of liver, testis and kidney were severely damaged by exposed to Cd of 1.25 mg L-1. Collectively, the results of our study indicate that AfCat may play an indispensable role in maintaining redox balance under Cd exposure, but AfCat expression will be inhibited when Cd accumulation reaches to the critical concentration. This study will illuminate the potential functions of AfCat in the defense mechanisms of reproductive and metabolic systems under Cd exposure in A. fasciatus.

        Assembly tightness detection of bolt connections using gray-level images with high-order cumulants

        Bolt connections are widely used in aero-engine rotors and gas turbines because of their load-transferring and detachable characteristics. However, bolts are prone to looseness due to the influence of fatigue, shock, and thermal loads, which decreases the reliability of the bolted connection structure. Therefore, detecting the assembly tightness of bolted connections is critical to ensure structural integrity during the assembly phase. A high-order cumulant-gray-level image feature (HOC-GLI) method is proposed to detect the assembly tightness of bolt connections. The core of this new method is to obtain high-order cumulant images of vibration signals to eliminate noise and reserve rich nonlinear information. The amplitude distribution of the third-order cumulant reflects the energy distribution of the vibration signal in 3D images. Then, the third-order cumulant 3D images are converted to 2D gray-level images to extract the texture feature. Finally, the root entropy index of the normalized gray-level cooccurrence matrix (GLCM) based on gray-level images is used to indicate the complexity of image information. Experimental studies on six bolt connection states of the aero-engine rotor are conducted. The relationship between the root entropy index and bolted connection status is obtained to verify the effectiveness of the proposed method for assembly tightness detection in different bolt connection statuses. The result shows that the root entropy index has a decreasing trend with the loosening of the bolts, which can quantitatively detect the assembly tightness of the bolt connection structure.

        Optimized Transformation of Streptomyces sp. ATCC 39366 Producing Leptomycin by Electroporation

        Streptomyces sp. ATCC 39366 produces leptomycin derivatives. Leptomycin B, a potent and specific inhibitor against the export of nuclear proteins, is the main product; however, the introduction of DNA into this strain is almost impossible, which has impeded its further use. We developed a Streptomyces sp. ATCC 39366 transformation protocol to introduce foreign DNA via electroporation. Various conditions were examined, including treatments of the cell wall with weakening agents, electroporation parameters, and DNA content. We found that only plasmid DNA isolated from a dam- ET12567 strain resulted in successful transformation. The mycelium growing in a yeast-peptone-dextrose medium supplemented with 1% glycine at 28°C on a rotary shaker (220 rpm) was more dispersed than those without supplementation and prone to electroporation. The maximum transformation efficiency of 8×102 CFU/μg plasmid DNA was obtained at a field strength of 13 kV/cm with a time constant of 13 ms (25-μF capacitor; parallel resistance, 600 Ω) using 1-mm electrocuvettes. The results of the transformations of two other Streptomyces species indicated that the optimized conditions established in this study might only be applicable to Streptomyces sp. ATCC 39366. However, this is the first report of successful transformation of Streptomyces sp. ATCC 39366, and will facilitate the construction of a gene knockout mutant in Streptomyces sp. ATCC 39366 to produce series of new leptomycin derivatives.

        Molecular characterization and expression analysis of putative odorant carrier proteins in Adelphocoris lineolatus

        The alfalfa plant bug Adelphocoris lineolatus (Goeze) (Hemiptera: Miridae), a serious pest of several agricultural crops, is extremely attracted by flowering plants to perform season host switching. Generally, chemoreception of A. lineolatus plays a crucial role in detecting food sources, locating mates and oviposition sites. In addition, odorant carrier proteins are believed to participate in the initial chemical communication and perception in insects. In this work, we identified 31 putative odorant carrier protein genes from the antennal transcriptomes of A. lineolatus, including 17 new odorant binding proteins (OBPs), 10 new chemosensory proteins (CSPs) and four Niemann-Pick type C2 proteins (NPC2s). Phylogenetic analysis demonstrated that the OBP and CSP genes from four mirid bug species (A. lineolatus, Adelphocoris suturalis, Apolygus lucorum and Lygus lineolaris) had a relatively close evolutionary relationship. Regular patterns and key conserved motifs of OBPs and CSPs in Hemiptera insects are also identified by using Multiple EM for Motif Elicitation (MEME) tool. Tissue expression profiles analysis showed that nine of the 17 OBPs, 10 CSPs and two of the four NPC2s were primarily or uniquely expressed in the adult antennae suggesting their putative roles in chemoreception. Our study provides the first insight into the roles of odorant carrier proteins in chemoreception of A. lineolatus, which will help to facilitate biological functions of odorant carrier proteins and develop novel insect behavioral regulation strategy

      • Discovery of a novel fragrant allele and development of functional markers for fragrance in rice

        Aroma in rice (Oryza sativa L.) results from the loss of function of the betaine aldehyde dehydrogenase (Badh2) gene on chromosome 8. An 8-bp deletion in exon 7 of Badh2 was reported to be the main allele functionally associated with fragrance. The discovery of new functional alleles will provide additional genetic resources to improve fragrant rice. In this study, we sequenced the Badh2 gene in 30 rice accessions and filtered the Badh2 polymorphisms from whole genome re-sequence data of 295 rice accessions. Seven alleles were detected from the sequence data. Six of the seven were known alleles and one was a novel allele (badh2-E12). The novel allele was a 3-bp deletion in exon 12. Five functional markers, targeting six of the seven alleles, were identified. Fourteen accessions were selected to test the utility of these markers. The five molecular markers reliably distinguishing this fragrant rice from other fragrant or non-fragrant rice accessions. Analysis of two F2 rice population validated the genetic markers FME12-3 and FME14I as functional markers. These two markers co-segregated with the fragrance phenotype. These markers will be used in a Badh2 diversity study and to breed improved fragrant rice accessions via marker-assisted selection.

        Perceptions of a night float system for intern doctors in an internal medicine program: an Asian perspective

        Long duty hours have been associated with significant medical errors, adverse events, and physician “burn-out”. An innovative night float (NF) system has been implemented in our internal medicine program to reduce the negative effects of long duty hours associated with conventional full-call systems. However, concerns remain if this would result in inadequate training for interns. We developed a structured questionnaire to assess junior doctors’ perceptions of the NF system compared to full calls, in areas of patient safety, medical training, and well-being. Ninety-seven (71%) of the 137 doctors polled responded. Ninety-one (94%) felt the NF system was superior to the full call system. A strong majority felt NF was beneficial for patient safety compared to full call (94% vs. 2%, p<0.001). The NF system was also perceived to reduce medical errors (94% vs. 2%, p<0.001) and reduce physician “burn-out” (95% vs. 5%, p<0.001). Beyond being a practical solution to duty-hour limitations, there was a significant perceived benefit of the NF system compared to the full call in terms of overall satisfaction, patient safety, reducing medical errors and physician “burn-out”.

      • Antiproliferative Effects of Celecoxib in Hep-2 Cells through Telomerase Inhibition and Induction of Apoptosis

        Background: To investigate the effect of celecoxib on telomerase activity and apoptosis in a human laryngeal squamous carcinoma cell line (Hep-2 cells). Materials and Methods: The growth inhibition rate of Hep-2 cells in vitro was measured by MTT assay, and apoptosis by TUNEL assay and flow cytometry (FCM). The TRAP-ELISA method was used to determine telomerase activity in Hep-2 cells. The mRNA expression of human telomerase RNA component(hTR), human telomerase reverse transcriptase (hTERT) and human telomerase-associated protein(hTEP1) was determined by RT-PCR assay. Expression of Bax and Bcl-2 proteins was assessed by Western blotting. Results: Celecoxib can inhibit proliferation and induce apoptosis in a dose- and time-dependent manner, repress telomerase activity, decrease hTERT mRNA and Bcl-2 protein expression and increase Bax protein expression, PGE2 had no effect on telomerase. Conclusions: Celecoxib had the antiproliferative and pro-apoptotic effect in Hep-2 cells. Apoptosis was accompanied by a decrease in telomerase activity which was directly correlated with hTERT mRNA and up-regulation of Bax/Bcl-2. Bcl-2 may thus play an important role in telomerase activity as well as apoptosis.

      • Association of MDR1 Gene Polymorphisms with Susceptibility to Hepatocellular Carcinoma in the Chinese Population

        Objective: The objective of this study was to evaluate the association of MDR1 gene polymorphisms with susceptibility to hepatocellular carcinoma (HCC). Methods: A total of 689 HCC patients and 680 cancer-free subjects were enrolled. Human MDR1 gene polymorphisms were investigated by created restriction site-polymerase chain reaction (CRS-PCR) and DNA sequencing methods. Multiple logistic regression models were applied to estimate the association between MDR1 gene polymorphisms and susceptibility to HCC. Results: We detected a novel c.4125A>C polymorphism and our findings suggested that this variant was significantly associated with susceptibility to HCC. A significantly increased susceptibility to HCC was noted in the homozygote comparison (CC versus AA: OR=1.621, 95% CI 1.143-2.300, ${\chi}^2$=7.4095, P=0.0065), recessive model (CC versus AC+AA: OR=1.625, 95% CI 1.167-2.264, ${\chi}^2$=8.3544, P=0.0039) and allele contrast (C versus A: OR=1.185, 95% CI 1.011-1.389, ${\chi}^2$=4.4046, P=0.0358). However, no significant increase was observed in the heterozygote comparison (AC versus AA: OR=0.995, 95% CI 0.794-1.248, ${\chi}^2$=0.0017, P=0.9672) and dominant model (CC+AC versus AA: OR=1.106, 95% CI 0.894-1.369, ${\chi}^2$=0.8560, P=0.3549). Conclusions: These findings suggest that the c.4125A>C polymorphism of the MDR1 gene might contribute to susceptibility to HCC in the Chinese population. Further work will be necessary to clarify the relationship between the c.4125A>C polymorphism and susceptibility to HCC on larger populations of diverse ethnicity.
