Regulatory Role of SFN Gene in Hepatocellular Carcinoma and Its Mechanism

Purpose: This study aims to explore the differential expression of SFN gene and its regulatory role in different hepatocarcinoma cells, and the impact on hepatocarcinoma. Materials and Methods: High and low SFN expression cells were screened by qRT-PCR and western blotting methods. SFN over expression and interference vectors were constructed. Cell viability was detected by CCK8 kit, cell cycle and apoptosis were detected by flow cytometry. Cell invasion and migration were detected. CCNB1 and CDK1 expression levels were detected by qRT-PCR and Western blotting methods. Results: The high SFN expression BEL7402 cells and the low SFN expression Hep3B cells were screened from Hep3B, HepG2, and BEL7402 cells. The activity of Hep3B cells overexpression vector SFNpcDNA3.1(+) decreased and apoptosis increased, the ratio of G0/G1 decreased and the ratio of S phase increased. The activity of BEL7402 cells transfected with SFN siRNA decreased and apoptosis increased, the ratio of G0/G1 decreased and the ratio of G2/M increased. Interference and overexpression vectors have little effect on the invasion and migration of the two cells. The expression of CDK1 in Hep3B cells decreased significantly, the expression of CDK1 and CCNB1 in BEL7402 cells increased significantly. Conclusions: The differentially expressed SFN gene can regulate the growth of the two hepatocarcinoma cells, high expression of SFN gene can inhibit their growth. The mechanism may be achieved by regulating CCNB1 and CDK1 expression.

To determine the potential for productive efficiency and genetic improvement in the blood clam Tegillarca granosa, four offspring populations (ZZ, ZK, KZ and KK) were produced from a diallel mating of two different geographical stocks (Z and K). The levels of genetic diversity and population structures of four populations were analyzed using 14 polymorphic microsatellites. The results showed that the mean observed heterozygosities (Ho) of reciprocal cross populations (ZK and KZ) was higher than those of pure populations(ZZ and KK). The largest values of genetic differentiation coefficient ( Fst = 0.067) and Nei’s unbiased genetic distance (Dc = 0.263) were between ZK and KZ, and the smallest ( Fst = 0.020, Dc = 0.116) were between ZZ and KK, which revealed that the largest genetic divergence was between the two reciprocal cross populations and the smallest was between two pure populations. This study demonstrated that the reciprocal cross populations of T. granosa had an extensive genetic difference and improvement, which may be advantageous for future breeding studies.
Multi-objective optimization of cycloidal gear based on segmental modification of pressure angle

In order to get the optimal profile of cycloid gear after sectional modification, a multi-objective optimization design method is proposed that considers both the modification parameters and macro-parameters of the cycloid gear. An algorithm of meshing force, meshing efficiency and anti-gluing ability between the cycloid gear and the pin gear was derived, and the related independent parameters were extracted as optimization variables. Taking high efficiency, high strength and light weight as the objective, the mathematical models of doubleobjective and three-objective optimization were established, and the influence of key design variable on the objective function was analyzed, and NSGA-II multi-objective genetic algorithm was used to solve the Pareto optimal solution of the optimization mathematical model. Results show that the optimized parameters can significantly improve the meshing efficiency, reduce the volume and meet the design requirements of high strength, high efficiency and lightweight on the premise of ensuring the strength of cycloid gear surface.

In this paper, a full-length cDNA-encoding cytosolic nicotinamide adenine dinucleotide phosphatedependent isocitrate dehydrogenase was isolated from maize and named ZmICDH. Analysis of the deduced protein sequence revealed that this gene had considerably high homologies with other plant ICDH genes such as those from rice, Arabidopsis and poplar. The gene was transcribed in all tissues tested, with the highest amount of transcript in root. Subcellular localization results indicated that ZmICDH was localized in cytosol. In an attempt to further understand the role of ZmICDH in maize, the functional characterization of the gene was conducted. The results indicated that the expression of ZmICDH was induced by drought and salt stresses. ZmICDH enhanced salt tolerance in transgenic Arabidopsis.

Traceable Dynamic Public Auditing with Identity Privacy Preserving for Cloud Storage

In cloud computing era, an increasing number of resource-constrained users outsource their data to cloud servers. Due to the untrustworthiness of cloud servers, it is important to ensure the integrity of outsourced data. However, most of existing solutions still have challenging issues needing to be addressed, such as the identity privacy protection of users, the traceability of users, the supporting of dynamic user operations, and the publicity of auditing. In order to tackle these issues simultaneously, in this paper, we propose a traceable dynamic public auditing scheme with identity privacy preserving for cloud storage. In the proposed scheme, a single user, including a group manager, is unable to know the signer's identity. Furthermore, our scheme realizes traceability based on a secret sharing mechanism and supports dynamic user operations. Based on the security and efficiency analysis, it is shown that our scheme is secure and efficient.
Incorporating uplift in the analysis of shallowly embedded pipelines

Under large storm loads sections of a long pipeline on the seabed can be uplifted. Numerically this loss of contact is extremely difficult to simulate, but accounting for uplift and any subsequent recontact behaviour is a critical component in pipeline on-bottom stability analysis. A simple method numerically accounting for this uplift and reattachment, while utilising efficient force-resultant models, is provided in this paper. While force-resultant models use a plasticity framework to directly relate the resultant forces on a segment of pipe to the corresponding displacement, their historical development has concentrated on precisely modelling increasing capacity with penetration. In this paper, the emphasis is placed on the description of loss of penetration during uplifting, modelled by 'strainsoftening' of the force-resultant yield surface. The proposed method employs uplift and reattachment criteria to determine the pipe uplift and recontact. The pipe node is allowed to become free, and therefore, the resistance to the applied hydrodynamic loads to be redistributed along the pipeline. Without these criteria, a localised failure will be produced and the numerical program will terminate due to singular stiffness matrix. The proposed approach is verified with geotechnical centrifuge results. To further demonstrate the practicability of the proposed method, a computational example of a 1245 m long pipeline subjected to a large storm in conditions typical of offshore North-West Australia is discussed.

4-Amino-1,2,4-triazole copper complex (4-ATzCu) was synthesized, and its thermal behaviors, nonisothermal decomposition reaction kinetics were studied by DSC and TG-DTG techniques. The thermal decomposition reaction kinetic equation was obtained as: dα / dt =1022.01 (1−α )[−ln(1−α )]1/3 exp(−2.75×104 /T) . The standard mole specific heat capacity of the complex was determined and the standard molar heat capacity is 305.66 J·mol‒1·K‒1 at 298.15 K. The entropy of activation ( ΔS ≠ ), enthalpy of activation (ΔH ≠), and Gibbs free energy of activation ( ΔG≠) are calculated as 171.88 J·mol‒1·K‒1, 225.81 kJ·mol‒1 and 141.18 kJ·mol‒1, and the adiabatic time-to-explosion of the complex was obtained as 389.20 s.
