      • Oil Contents and Fatty Acid Composition of Korean Perilla (Perilla ocimoides L.) Collections

        This study was carried out to obtain informations on the variations of oil content and fatty acid composition among 90 Korean perilla collections. Average oil content of 90 perilla collections was 44.2% with a range from 29.7% to 61.9%. Perilla collections with late-maturing, super-large seed and gray seed coat showed higher oil content than other types in general. Average saturated fatty acid content in perilla oil was 9.0% with a range from 8.2% to 10.7%, while average unsaturated fatty acid content varied from 89.3% to 91.8% with a mean of 91.0%. Contents of saturated and unsaturated fatty acids were not related to maturity. There were no differences in the contents of saturated and unsaturated fatty acids according to maturity. Linoleic acid and linolenic acid contents were the highest in the super large-sized group(15.5%) and in the large-sized seed group(61.4%), respectively, while contents of fatty acids among the perilla collections were variable with different seed coat colors. Most of the traits studied were not significantly correlated with oil content, but linoleic acid($\omega$ 수식 이미지-6) content was negatively correlated (r=-0.217*) with linolenic acid(ω-3) content.

      • 인체 대장암 및 자궁경부암에서 PCR-SSCP법을 이용한 Ki-ras 암유전자의 점돌연변이에 관한 연구

        인체 대장암 20례와 자궁경부암 10례의 암조직을 대상으로 c-Ki-ras 유전자 codon 12와 13에서의 점돌연변이를 알아보고자 polymerase chain reaction(PCR)과 single-strand conformation polymorphism(SSCP) 검사법을 시행하였다. 대장암의 경우 20례중 9례(45%)에서, 자궁경부암의 경우 10례중 1례(10%)에서 양성으로 나왔다. 대장암에서는 codon 12에서 GGT가 TGT로의 치환이 4례로서 가장 많았으며, AGT, CGT로의 치환이 각 1례였고 codon 13에서는 GGC가 GAC로의 치환이 2례, TGC로의 치환이 1례인 것으로 나타났다. 자궁경부암의 경우 codon 12에서 GGT가 AGT로의 치환이 1례인 것으로 나타났다. In an attempt to clarify the role of genetic alteration in the genesis of human colorectal and cervical cancers, tissue specimens from 20 patients with colorectal cancer and 10 patients with cervical cancer were examined for the presence of point mutation in K-ras2 exon 1 by single strand conformation polymorphism analysis of PCR product. Exon I of c-Ki-ras2 was amplified by polymerase chain reaction(PCR) and comparison was made between the normal and mutated genes by nondenaturing polyacrylamide gel electrophoresis(PAGE) of PCR product and nucleotide sequence analysis using asymmetric PCR with direct sequencing. Genomic DNA from white blood cells were used as normal control and those from A427 cell line were used as mutated control. 1.PCR product from A427 cell line showed a distinct migration shift pattern compared to the normal control in PAGE and the direct sequencing indicated that nucleotide sequence of codon 12 was mutated from GGT to GAT. In addition, there was a loss of normal allele in A427 cell line. 2.PCR product from 9 cases(45%) out of a 20 colon cancer patients showed migration shifts in PAGE, and all of these 9 patients invariably demonstrated mutation of ras gene, either in codon 12(6 cases:30%) or 13(3 cases:15%). The base substitutions in codon 12 were: from GGT to AGT(1), CGT(1), or TGT(4). Base changes in codon 13 were from GGC to TGC(1) or GAC(2). 3.PCR product from 1 cases(10%) out of a 10 cervical cancer patients showed migration shrifts in PAGE, Invariably demonstrated mutation of ras gene in codon 12. The base substitutions was from GGT to AGT(1) TQE incidence of point mutation of c-Ki-ras in colon cancer was high, however, it was low in cervical cancer, which showed the relation of ruts gene mutation with colon cancer. PCR -SSCP analysis is a simple, rapid and efficient method of detection of point mutation, especially when dealing with multiple samples.

        In the informative society the classroom is not the only place for education and the given curriculum and texts are not the only educational contents. Education possible wherever the computer is installed. In the existing web-based cyber university, however, the contents of lecture are distributed at a time in advance, so level-based learning and education are impossible. Therefore, this study tries to solve problems through the feedback between the professors and the students by the interactive learning on the web, and suggests a system model which makes level-based learning possible. The suggested system model has the learning session and the evaluation session in each unit. After learning each unit, the learner evaluates him learning with the extracted questions from the database. When he does not reach the professor's expected scores, he repeats the same process. So, the learner can control the learning progress and thus level-based learning and evaluation become possible.

      • 영도 토양환경의 중금속오염에 대한 기초연구

        Youngdo Island is suitable to be developed as a tourist town due to its beautiful natural surroundings. In connection with the above purpose, various harmful heavy metals were analyzed to investigate the level of pollution of Youngdo soil. The elements tested were Cd, As, Sn, Cr, Cu, Pb, Co, Ni, Mn and Zn. The results from Youngdo were compared with those of the Busan and the whole country in the investigation items which were decided by the Korean Ministry of Environment. The average contents of heavy metals in Youngdo soil were comparatively lower than those of Busan, and close to those of the whole country including uncontaminated natural soil. It was found that Youngdo-Gu remains clean soil environment except a few investigated sites at the industrial areas even though this district belongs to the Metropolitan Busan. This fact implies that Youngdo is to get well ready for a tourist town.

      • 수면 부유 방법에 의한 연초 육묘

        Carbonized rice hull, perlite, and vermiculite were mixed and investigated for the probability of tobacco transplant production by means of plug and transfer float system. These seedling bed materials in condition of float system have gaseous phase ranged from 6% to 13% showing a proper growth of tobacco seedling. However compost media mixed with soil, sand, and compost in condition of float system led to the shortage of respiration in root system by very low proportion of gaseous phase skewing a wicked growth of seedling. Absorption of heat by floating water for the daytime increased rhizosphere temperature in condition of float system during the night and low temperature season resulting in better growth of tobacco seedling. Consequently transplant production of tobacco seedling by float system with mixed materials of carbonized rice hull, perlite, and vermiculite was thought to be desirable method for manufacturing production system of transplant considering the effective management of water, nutrients, and temperature of seedling bed materials.

      • 콩의 식품적 의의 및 생산수급과 식용콩의 자급 향상

        콩은 단백질 공급을 비롯하여 영양가가 높아 예로부터 우리의 중요 식품으로 널리 다양하게 이용하여 왔을 뿐 아니라 지속적인 밭작물 재배를 가능케 하는 중요한 역할을 하여 왔으며, 최근에는 건강식품으로 또 친환경 농업 작물로 주목을 받고 있다. 콩은 세계적으로 99,501천ha에서 223,185천톤(2,243kg/ha)이 생산되고 그의 35.5% (35,130백만불)가 수출되고 그 중 43%는 미국 콩이, 96.5%는 미대륙 생산 콩이 수출되고 있으며, 수입의 반 정도는 중국이, 37.6% 정도는 유럽의 선진국과 일본 등 10여 개국에서 각각 수입하고 있고, 우리나라도 10번째의 수입국이며 세계적 소비수요는 계속 증가되고 있다. 우리나라는 1968년의 313,501ha의 재배에서 계속 감소하여 현재는 75,242ha에서 132,674톤이 생산되고 1,261천톤이 수입되었으며, 전체 자급률은 수요량 1,333천톤의 8.6%, 식용 및 기타는 수요량 387천톤의 29.5%에 불과하다. 콩, 특히 식용콩의 증산에 의한 자급율 향상은 경지정리 및 주산단지 조성을 통한 생력기계화 재배를 전제로 첫째, 밭의 작부체계 개선 둘째, 논의 밭 전환재배 셋째, 북한의 동해안 지대의 밭을 장기임대하여 재배하고 그 대가를 쌀 지원으로 하는 것이며 넷째는 인접국에서 밭을 매입 또는 장기 임대하여 생산 공급하는 방법 등을 생각할 수 있을 것이다.

      • 콩에서 발생하는 강낭콩일반모자이크바이러스(BCMV)와 땅콩위축바이러스(PSV)의 저항성 평가

        It has been reported several viral diseases in soybean. The occurrence of Bean common mosaic virus and Peanut stunt virus are able to induce severe problem because the main breeding target for viral disease was focused on Soybean mosaic virus. BCMV is a spcies of the genus Potyvirus of the family Potyviridae. BCMV reduces yield by as much as 80% and also adversely affects seed quality in common bean. BCMV has a worldwide distribution because of its high rates 35% of transmission via seeds produced by plants systemically infected prior to bloom. PSV is a species of the genus Cucumovirus of the family Bromoviridae. PSV has been reported to be economically damaging on beans worldwide. Although the virus is reportedly seed transmissible at very low rates, infected plants rarely produce pods with normal viable seeds. The most satisfactory management method of viral diseases such as BCMV and PSV is the use of resistant cultivars and healthy seed. For the overcome of genetic resistance by new virus such as BCMV and PSV, virus diseases monitoring and researches need to study deeply and continually. In this study, we conducted the resistance screen on BCMV and PSV for selection of resistance cultivars. We inoculated on 128 cultivars with PSV and BCMV respectively. These cultivars were classified through pathogenic response such as resistance, necrosis and susceptibility. In addition, 99 and 66 resistant cultivars were confirmed against PSV and BCMV respectively.

        외래어종 베스, Micropterus salmoides를 포함하는 군집의 종간 Association에 관한 연구

        1970년부터 2002년까지 전국을 조사한 자료 중 외래도입어중인 M. salmoides가 포함된 군집을 대상으로 공존성 및 종간상관성에 대하여 논의하였다. 총 7,192개의 조사지역에서 M. salmoides가 포함된 군집은 46군집, 공존하는 어종은 72종으로 나타났다. 공존성이 높은 종은 Z. platypus, O, bidens, H. eigenmanin, S. japonicus coreanus의 순이었다. 종집단간 공존도는 M. salmoides - Z. platypus 집단, M. salmoides - C. auratus 집단, M. salmoides 집단을 포함하고 있는 군집내에서 7종이 그룹화되었다. M. salmoides가 포함된 군집내에서 O. bidens와 Z. platypuswlqeks 등 7개 종집단은 종간 상관성이 매우 높았으며(p<0.01), L. macrochirus와 C. cuvieri 집단 등 11개 종집단은 종상관성이 다소 높은 것으로 나타났다(p<0.05). 한국의 하천이 점차 호수성하천으로 변화된다고 볼 때, M. salmoides 집단의 조절대책이 필요하다. On the fish community including Micropterus salmoides population, introduced species in Korea, interspecific association through coexistence by presence-absence among species populations were discussed from the year of 1970 to 2002. In the total of 7,192 sampling stations in Korea, the community including Micropterus salmoides population were confirmed at 46 sampling sites, and coexistent fishes were 72 species. Fish species having high coexistence rate were Zacco platypus, Obsarlichthys bidens, Hemiculter eigenmanni and Squalidus japonicus coreanus populations in the order of their number. A association indices values between population were in the order of Micropterus salmoides-Zacco platypus, Micropterus salmoides-Carassius auratus, and Micropterus salmoides-Hemiculter eigenmanni population. Judging from the dendrogram according to association indices between species population, 7 species populations were clustered in the community including M. salmoides population. Being based on association values among species population in the community including M. salmoides population, 11 species population pairs including Lepomis macrochirus -Carassus cubieri etc. showed somewhat high significance (p < 0.05), and 7 species population pair including Zacco platypus-Obsarlichthys bidens etc. represented higher association among species population (p < 0.01). With the increasing of lake- river in Korea, it keenly need to take a regulation program of the Micropterus salmoides population.

      • 超微量 銅의 螢光光度定量法

        A spectrofluorometric method for the ultra microdetermination of copper was established. An excess amount of 2-(o-hydroxypheny1)benzoxazole(HPB) is added to precipitate cu(HPB)₂ with copper ion. Copper ion forms a chelate with HPB and quenches the fluorescence of HPB. The amount of copper is determined by measuring the fluorescence of the remaining HPB. The optimum conditions for these reactions were examined at a very low concentration range of the reactants. By the proposed method, 0.025~5 nmol copper content was determined with a accuracy of 2.1_(7)% relative standard deviation at 2 nmol Cu level.

      • 이동호스트를 위한 적응성 멀티캐스트 알고리즘

        To Provide multicast service for mobile hosts must deal with dynamic member location and its group membership in mobile networks. There are two old approaches as hi -directional tunneling and remote subscription for mobile multicast. The deployment of existing multicast protocols(home agent-based multicast, Bidirectional packet tunneling) is very inefficiently to use network resources such as duplicate copies of multicast datagrams, tunnel convergence, and unicast by long delivery path. The remote subscription scheme may be very expensive because muticast packet delivering is delayed and discontinued frequently by rebuilding a multicast tree to serve multicast when the host move highly. In this paper, we analyse these problems of multicast routing protocols and propose an efficient multicast protocol using cost function to consider the host mobility in wireless mobile networks, By cost function, a mobile host make a decision to receive a tunneled multicast datagram from home agent or to receive a rebuild path from foreign agent. The performance of the proposed protocol was evaluated by simulation under various environments and we got an improved performance.
