      • 인삼 Peroxidase(PgPrx3) 유전자의 분리 및 연초의 형질전환

        Peroxidase는 항산화에 관련된 효소이며 식물의 환경스트레스와 성장에 관련되는 중요한 유전자이다. 본 실험에서 사용된 Peroxidase Ⅲ의 cDNA는 Panax ginseng C.A. Meyer 잎에서 추출하였으며 PgPrx3라 명명하였다. 이 gene의 ORF영역은 1,065 bP이고 355개의 아미노산을 가지고 있다. BioEdit 프로그램을 이용하여 PgPrx3와 다른 식물의 peroxidase gene과 비교한 결과 상동성을 가지고 있었으며, Spinacia oleracea(70%), Vigna angulais(71%), Nicotiana tabacum(69%) and Linum usitatissimum(65%). 그중 Vigna angularis의 상동성이 가장 높았다. 담배에 본 유전자를 형질전환시켜 유전자의 특성을 검정하기 위하여 35S/35S/AMV/peroxidas/Tnos를 벡터 pRD400에 재조합 후 아그로박테리아를 이용하여 담배에 도입시켰다. 그리고 PCR를 이용하여 유전자 PgPrx3가 담배에 도입된 것을 확인하였으며, RT-PCR로 정상적으로 PgPrx3 유전자가 전사되어 RNA를 생성하고 있음을 확인하였다. A peroxidase (E.C. 1.11.1.7) is very important enzyme as antioxidants. The function of this gene is connected with growth and environmental stress of plant. A class Ⅲ peroxidase cDNA was isolated from the leaf of Panax ginseng C.A. Meyer and named as PgPrx3 which is consisted of an ORF (open reading frame) of 1,065 bp and an amino acid of 355 residues. BioEdit software was used to compare the PgPrx3 amino acid sequence with other plants that showed homologies with Spinacia oleracea (70%), Vigna angularis (71%), Nicotiana tabacum (69%) and Linum usitatissimum (65%). The peroxidase of Vigna angulatis was the most homologous with ginseng. The chimeric PgPrx3 gene, 35S/35S/AMV/peroxidas/ Tnos, was constructed in the binary vector pRD400. Agrobacterium tumefaciens strain LBA4404 harboring disarmed Ti-plasmid was used to transform Nicotiana tabacum L. using the leaf disc. Incorporation of the chimeric gene PgPrx3 into plants were confirmed by PCR analysis of genomic DNA and RT-PCR analysis of mRNA from transgenic tobacco. The PgPrx3 gene from ginseng was stably expressed in transgenic tobacco plants.

      • 더덕의 효율적인 재분화 및 뿌리발달

        더덕은 식용, 관상용, 생약재료로 쓰이는 방향성 식물이다. 본 연구는, 종자의 기내배양을 통하여 식물체 재분화와 뿌리발달을 실험했다. 종자의 발아는 GA_(3) 100 mg·L(-1)처리와 저온처리 시 매우 양호하였다. 더덕의 기내에서 shoot의 형성은 2 mg·L^(-1) BA, 2 mg·L(-1) NAA가 첨가된 MS 기본배지에서 가장 양호하였으며, 줄기의 절편체와 비교하여 잎 절편체에서 재분화가 잘되었다. 또한 뿌리의 발육은 계속적인 계대배양에 의해서 양호하였는데 shoot의 길이를 1 cm 정도 남긴 뿌리를 연속적으로 4회 계대배양에서 뿌리가 주근으로 발육하였으며 길이는 약 3 cm 및 직경은 0.5 cm의 정상적인 뿌리를 얻을 수 있었다. Codonopsis lanceolata is widely used as edibles, ornamental plant and crude drug material. This study was carried out to confirm the ratio of germination and efficient differentiation of shoot and root in vitro. The germination of Codonopsis lanceolata seed was excellent in the medium with GA_(3) 100 mg·L(-1) and chilling treatment. The shoot formation of Codonopsis lanceolata was good at the MS medium with 2 mg·L(-1) BA and 2 mg·L(-1) NAA. According to inoculum part, ratio of re-differentiation was higher from the leaf explant compared with the stem explant. For root development, successive subculture with 1 cm shoot of upper part from root was efficient. Roots of Codonopsis lanceolata cultured in vitro system was grown like normal root with length of 3 cm and thickness of 0.5 cm at the 4 successive subcultures.

      • 播種期, 苗令 및 土壤水分不足이 黃色種 담배의 生育 및 收量에 미치는 影響

        This experiment was conducted to study the influence of environment cultural method and their interaction on the growth and yield components of flue-cured tobacco. Applied variety was NC 2326, and was treated by sowing time(Mar. 11, Mar. 21 and Mar.31), seedling age (40,50 and 60 day) and soil moisture stress period (20 days from 10, 20 and 30 days after setting). The results are summarized as follows. 1.During growing period, stem height, length and width of the largest leaf and total leaves were affected remarkably by the treatments. 2.Soil moisture stress period affected more the characters of tobacco plants than other factors. 3.The tobacco plant treated by early stage soil moisture stress (period from 10 to 30 days after setting) made the early growth very poor, and that tobacco plant showed poor growth until 45 to 50 days after setting. 4.Early sowing, 40 day seedling and early stage soil moisture stress made the plant type sylindrical, and late sowing and 50 to 60 day seedling made the plant type cone shape. 5.Number of total leaves and harvested leaves, leaf area per plant percentage of dry matter of leaf and specific leaf weight decreased by the treatment of soil moisture stress during early and middle part of maximum growth stage. 6.There were little differences of yield among the treatment of different sowing time and seedling age. Early sowing increased the thickness of leaf and weight per unit leaf area, and decreased number of total leaves and specific leaf weight. Number of total and harvested leaves and specific leaf weight of cured leaves were high in the plot of 60-day seedling 40-day seedling showed heavy weight of unit leaf area and low specific leaf weight of cured leaves. 7.From the point of view of interaction between sowing time and seedling age, early sowing (Mar. 11) with 60-day seedling, middle sowing (Mar. 21) with 50-day seedling and late sowing (Mar. 31) with 40-day seedling are recommended. 8.Rapid growth of early growing stage was shown in the plot of adequate soil moisture and late sowing (Mar. 31) with 50-day to 60-day seedling. 9.From the point of standard quality of cured leaves, sowing time can be shifted until late March. And to fix the sowing time, It should be considered that the maximum growth stage and the harvesting time of cutter and leaf would not match with severe drought of May and rainy season of early summer respectively.

        북한 회색지대 전략에 대한 억제전략 발전방향

        이 연구의 목적은 북한 회색지대 전략을 다시 바라보고 이에 대비한 우리의 억제전략 발전방향을 제시하는 것이다. 논문에서 제시한 방안은 新통합억제전략을 통해 북한의 회색지대 전략 준비와 실행을 억제하는 것이다. 방법으로 평시 DIE 공세 및 억제실패 시 DIME 통합대응을, 수단으로 사이버역량 중심의 DIME 역량을 제시한다. 우리의 新통합억제전략을 구상하기 위한 논리 전개는 다음과 같다. 첫째, 북한 회색지대 전략은 다른 나라의 회색지대 전략과는 차이가 있다. 두드러지는 특징은 북한 전략문화 속성의 지배, 외부와의 정보차단으로 인한 폐쇄성, 대한민국의 취약성을 이용, 사이버전과 같은 비대칭수단에 집중하는 것이다. 둘째, 이에 대응한 우리의 억제전략은 실효성이 떨어지며 문제가 있다고 평가한다. 특히, 일관되지 못한 정부정책과 국민적 공감대 미형성, 통합적 · 장기적 사고 결여, 정병(正兵) 측면의 경직된 전략, 군사력 중심의 대응, 수동적인 전략의 한계가 있다. 셋째, 이 문제를 보완하기 위한 키워드는 “통합”이다. 통합 측면에서 억제전략을 살펴보기 위해 우리나라의 통합억제전략 수립 사례와 미국의 3차 상쇄전략과 그 논리를 계승한 통합억제전략을 살펴본다. 넷째, 이를 기초로 우리만의 新통합적 억제전략을 제안한다. 핵심 방향은 통합적ㆍ장기적 사고 기반 위에서 국가의 모든 DIME 역량이 통합된 유연하고 적극적인 전략, 더 나아가 평시에 DIE 공세전략을 구사하는 것이다. The purpose of this study is to re-examine the North Korean gray zone strategy and present the direction of our deterrence strategy development in preparation for it. The deterrence strategy presented in the paper is to deter the preparation and provocation of North Korea’s gray zone strategy through a New Integrated Deterrence Strategy. The method is a DIE offensive in peacetime and DIME integrated response in the event of deterrence failure. The means presents DIME capabilities centered on Cyber competencies. The logic to devise a New Integrated Deterrence Strategy is as follows. First, it was considered that the North Korean gray zone strategy was different from other countries gray zone strategies. Its characteristics are to focus on domination of North Korea’s strategic culture attributes, closure through information blocking from the outside, strategies that penetrate the vulnerability of the Republic of Korea, and asymmetric means such as Cyber. Second, Republic of Korea deterrence strategy in response to this is evaluated as ineffective and problematic. The problems are inconsistent government policies, lack of public consensus, lack of integrated and long-term thinking, rigid strategies in terms of symmetric and asymmetric, and limitations of passive strategies. Third, the keyword to solve this problem is Integration. In order to examine the deterrence strategy in terms of integration, we examine the case of establishing an Integrated Deterrence Strategy in Korea and the Integrated Deterrence Strategy in connection with the 3rd Offset Strategy in the United States. Fourth, based on this, we present own direction of a New Integrated Deterrence Strategy. The key direction is to use a flexible and active strategy that integrates all the country’s DIME capabilities on long-term thinking basis, and furthermore, a DIE offensive strategy in peacetime.

        Arterial pseudoaneurysm is rare but potentially a catastrophic complication of pancreatitis because it can cause massive gastrointestinal bleeding. Since surgical treatment of arterial pseudoaneurysm has a high mortality, percutaneous angiographic embolization of bleeding artery has recently been advocated as the alternative therapy. Acute hemorrhage into the peritoneal cavity or pseudocyst complicating chronic pancreatitis was frequently reported. However, multiple pseudoaneurysmal hemorrhage without pseudocyst complicating acute pancreatitis are extremely rare. Here, we present a case of multiple pseudoaneurysmal hemorrhage secondary to acute pancreatitis, which were managed successfully by percutaneous angiographic embolization.         Biological Control of Gray Mold Rot of Perilla Caused by Boftis cinerea 1. Resistance of Perilla Cultivars and Selection of Antagonistic Bacteria

        Resistance of perilla varieties to Botrytis cinerea LVF12 was evaluated, while antagonistic bacteria were selected and tested for their efficacy towards biological control of gray mold rot caused by B. cinerea. Among 11 perilla varieties tested for disease resistance, Milyang variety showed some degree of resistance, while the rest of varieties showed no resistance. Among 250 bacterial isolates collected from perilla loaves and rhizosphere of perilla plants, six isolates showed high levels of inhibitory effect on mycelial growth and conidial germination of B. cinerea in in vitro test. Using the pot test in growth chambers these isolates showed high levels of disease suppression, with Nl isolate showing 95.3% of control value and N4 isolate showing 90.8% of control value. Further test was performed to evaluate the two isolates ability for disease prevention and/or disease therapy, and results showed almost 100% of control vague. Isolates Nl and N4 were identified as Bacillus licheniformis and 5. megatepium, respectively, according to Bergey's manual, API 20E and 50CHB test kit, and Transmission electron microscope.

        혈압 조절이 당뇨병성 만성 신부전증의 진행속도에 미치는 영향

        연구 배경 : 당뇨병성 신증은 신 대치요법을 필요로 하는 주요 원인이며, 그 빈도가 증가추세에 있고, 말기 신부전으로의 진행이 필연적이므로 사구체의 손상에 대한 위험 요소들을 조절하여 신부전의 진행속도를 저하시키려는 노력이 필요하다. 그 중 고혈압의 조절이 그 진행속도를 늦출 수 있다는 여러 임상보고 및 동물실험의 증거가 있으나, 적정 혈압의 목표에 대해서는 알려져 있지 않다. 이에 저자들은 혈압 조절이 당뇨병성 만성 신부전증의 진행속도에 미치는 영향을 관찰하였다. 방법 : 가톨릭 의과대학에서 당뇨병성 만성 신부전증으로 진단받고 보존적인 치료를 받고 있는 6개월 이상 추적이 가능했던 환자 30명을 대상으로 내원시마다 측정된 혈압과 혈청 크레아티닌을 조사하였다. 또한 추적기간을 평균동맥압에 따라 3개의 period로 나누고 각 peirod에서의 신기능의 진행속도를 혈청 크레아티닌의 역수와 시간과의 상관계수로 계산하였다. 결과 : 혈청 크레아티닌의 증가치와 그 역수의 감소로 본 평균 동맥압에 따른 신부전증의 진행속도는 각각 A period(평균동맥압?105mmHg)에서 0.0797±0.0286mg/dl/month, -0.0079±0.0018dl/mg/month, B period(105<평균동맥압<115mmHg)에서0.2224±0.0976mg/dl/month, -0.0430±0.0093dl/mg/month, C period(평균동맥압?115mmHg)에서 0.2731±0.0452mg/dl/month, -0.0434±0.0073dl/mg/month였으며, 모두 평균동맥압이 105mmHg 이하일때 의미있게 신부전의 진행속도가 감소되었다(p<0.0). 결론 : 이상의 성적으로 보아 당뇨병성 만성 신부전증환자에서 평균 동맥압을 적어도 105mmHg 이하로 유지하는 것이 신부전의 진행속도를 감소시키는데 중요하다고 사료된다. Background : Dabetic nephropathy is considered as the main cause of end stage renal disease which needs the renal replacement therapy. The adequate control of blood pressure is known important to reduce the progression rate of renal function in patients with diabetic renal insufficiency. We investigate the effect of blood pressure control on the progression rate of renal function in 30 patients with diabetic renal insufficiency. Method : We divided the observation time to 3 periods arbitarily according to degree of blood pressure control. The progression rate of renal function was measured by the monthly changs of serum creatinine concentration and reciprocal serum creatinine concentration. Result : The monthly changes in serum creatinine concentration in A period (MBP?105mmHg), B period (105mmHg<MBP?115mmHg) and C period (MBP?115mmHg) were 0.0797±0.0286, 0.2224±0.0976 and 0.2731±0.0452mg/dl/month, respectively. The monthly changes in reciprocal serum creatinine concentrations in A, B, C period were -0.0079±0.0018, -0.0430±0.0093 and -0.0434±0.0073dl/mg/month respectively. The monthly changes in serum creatinine concentrations and reciprocal serum creatinine concentrations in A period were significantly low compared with B and C periods. Conclusion : The results may suggest that adequate control of mean blood pressure below 105mmHg is important to reduce the progression rate of renal function in patients with diabetic renal insufficiency.

      • Inhibitory Effects on Osteoclastogenesis of Tenebrio Molitor Larva Fermentation Extract in vitro and in vivo

        Edible insects are attracting attention as a sustainable food resource in future food. Recently, researches on the relationship between bone health and chronic diseases are increasing. Therefore, this study attempted to determine the anti-osteoporosis effect of Tenebrio Molitor larva fermentation extract (TM). The TRAP staining of bone marrow macrophage cells indicated that TM dose-dependently inhibited osteoclastogenesis compared with the vehicle. TM treatment down-regulated NFATc1, DC-STAMP, OSCAR, Ctsk, and Trap mRNA expression. NFATc1, a major protein in osteoporosis, level was decreased by the TM treatment. The ALP staining of C2C12 cells indicated that TM dose-dependently increased osteoblastogenesis. In high-fat fed mice, TM supplementation significantly increased femoral and tibial bone mineral density. These results demonstrate the anti-osteoporotic effects of TM by regulating osteocalstogenesis and osteoblastogenesis, which provided useful information for its application and development against obesity-induced osteoporosis.
