      • Effect of Ovarian Extract on Oocyte Maturation and Early Embryonic Development in Pigs

        Various factors in the ovary are known to regulate oocyte maturation and hormone secretory functions; however, the effect of ovarian extract (OE) on oocyte maturation and embryonic development in pigs remains unknown. In this study, we first evaluated whether OE supplementation in the in vitro maturation (IVM) medium alters the oocyte maturation capacity by affecting glucose/amino acid metabolites, meiotic maturation, cumulus cell (CC) expansion, and antioxidants. Various OE concentrations (50, 100, 200, 500, and 5000 μg/mL) were included in the IVM medium. Only the oocytes treated with 100 μg/mL OE exhibited an improved meiotic maturation rate when compared with that of the other groups (non-treated group, 78.6 ± 3.0% vs. 100 μg/mL OE-treated group, 81.6 ± 4.3%); however, the difference was not significant. To observe the changes in glucose and amino acid metabolism in the OE-treated oocytes, we measured the amounts of diverse constituents (glucose, lactate, glutamine, and ammonia) in the IVM medium containing OE. Lactate and ammonia levels in the OE-treated group after 44 h of IVM were higher (p < 0.01) than those in the non-treated group. In addition, the expression of the CC expansion factors (Has2 and Tnfaip6) significantly increased (p < 0.05), whereas the mRNA expression levels of antioxidative enzymes (Sod1, Cat, and Gpx1) significantly diminished (p < 0.05) in the OE-treated group. Moreover, mature oocytes treated with 100 μg/mL OE demonstrated increased subsequent embryonic development rates after 144 h of IVM. Thus, the addition of OE in IVM mediums may improve oocyte maturation capacity which could enhance antioxidative enzyme activation, energy metabolism, and expression of the CC expansion factors in porcine oocytes.

        Motivational Interviewing Training Using Role Play in Dental Hygiene Students

        The purpose of this study was to develop a motivational interviewing (MI) training program to improve competency in communication and to effect the evaluated changes that would occur in dental students’ counseling techniques as result of the training in the program. The study proceeded through the program via role-play practice, which was explained effectively during the MI program training process. A convenience sample of 43 fourth-year dental hygiene students was recruited. Twenty-two students were randomly assigned to the intervention group and 21 to the control group in the order of recruitment. The theoretical lecture was delivered over 3 hours and the practice was done in parallel by applying cases occurring in clinic settings. The practice was technical training, partner practice, and small group practice. The intervention group received three hour MI lecture while the control group received no lecture. Evaluation through role-play practice was separated according to “researcher’s viewpoint,” “counselor’s role viewpoint,” and “patient’s role viewpoint.” The results of the analysis showed that the intervention group had higher MI skills and ability than the control group. Furthermore, program participants showed positive impressions to the MI training program. The use of coaching sessions improved the MI techniques and counseling skills of dental hygiene students learning MI. The effect of the application of the MI training program was that the MI training improved counseling skills and interviewing abilities. Moreover, continuous training and feedback enhanced MI techniques and core skills. Training using lectures in parallel with practice rather than education through theory alone improved students’ techniques. Application of role play through the combined method of the MI program was confirmed by an effective training method.

      • Stress and Fatigue Analysis of the Power Transmission Sprocket Gears for Design of a Self-propelled and Small-sized Chinese Cabbage Harvester

        Harvesting operation of Chinese cabbage still relies on manual labor force, therefore harvesters need to be developed. When developing machines, computer simulations would improve the optimum design reliability and reduce the development time and costs. Objective of this study was to conduct stress and fatigue analysis for optimum design of sprocket gear in Chinese cabbage harvester power transmission system. Using software (midas NFX), stress and fatigue analysis were performed for material and gear pressure angles. The gear materials were chosen structural chrome steel SCR420H and mechanical structural carbon steel SM45C. The gear pressure angle was 16 ° to 24 ° at 2° intervals. As a result of stress and fatigue analysis, the maximum displacement and fatigue life did not change significantly with the material and pressure angle, but the maximum stress was the lowest at 20° in both materials.

        Synergistic Antimicrobial Effect of Lonicera japonica and Magnolia obovata Extracts and Potential as a Plant-Derived Natural Preservative

        Most people use cosmetics to protect their skin. Preservatives are often used to prevent their contamination upon use. There has been a great demand for natural preservatives due to recent reports on the side effects of parabens. Therefore, we evaluated the antimicrobial activities of Lonicera japonica and Magnolia obovata extracts and determined their potential as natural preservatives. We found that the 50% ethanol extract from L. japonica had antibacterial activity only against S. aureus and P. aeruginosa, while the ethyl acetate fraction showed antimicrobial activity against all six microbial strains tested. On the other hand, the 70% ethanol extract and the ethyl acetate fraction from M. obovata showed antimicrobial activity against all six strains. A synergistic effect against S. aureus, B. subtilis, and C. albicans was confirmed when two ethyl acetate fractions having antimicrobial activity against all six strains were used in combination. Synergistic activity against B. subtilis was also confirmed through kill-time analysis. Highperformance liquid chromatography was performed to identify the components of each extract. Based on the minimum inhibitory concentration and the results of a disc diffusion assay, we confirmed that caffeic acid and luteolin influenced the antimicrobial activity of L. japonica and that the antimicrobial activity of M. obovata was influenced by the interaction of magnolol and honokiol with other components. Therefore, this study suggests that the combination of L. japonica and M. obovata extracts may be used as a plant-derived natural preservative.

      • Ustekinumab, an effective therapeutic option for erythrodermic psoriasis

        Erythrodermic psoriasis is a severe form of psoriasis, presenting as prominent erythema, affecting the whole body. Ustekinumab (Stelara®) is a human monoclonal antibody targeting the p40 subunit shared by interleukin IL-12 and IL-23. Erythrodermic psoriasis has been an exclusion criteria in studies looking at the efficacy of ustekinumab against psoriasis, but recently, a few reports have proven its efficacy on erythrodermic psoriasis. Herein, we report 2 cases of erythrodermic psoriasis that successfully improved with ustekinumab. A 42-year-old man presented with a 7-year history of erythrodermic psoriasis and psoriatic arthritis. The patient was treated with ustekinumab 45 mg subcutaneously at 0, week 4, and every 12 weeks thereafter. At week 28, his Psoriasis Areaand Severity Index (PASI) score decreased from 64.8 to 9.6. The second case is a 26-year-old man suffering from erythrodermic psoriasis for 10 years. Four months after the first administration of ustekinumab, the patient achieved 75% improvement of the PASI score and 90% improvement after 112 weeks. Although further studies are required, it is likely that ustekinumab can be an effective therapeutic option for patients affected by erythrodermic psoriasis.

        국내 과학관 수는 지난 10여년 동안 양적으로는 팽창하고 있으나, 대부분이 중소규모의 과학관으로 인력부족 및 과학전시 컨텐츠 부족에 따라 운영에 어려움을 겪고 있다. 이에 따라 과학관이 보유하고 있는 전시 컨텐츠의 DB구축 및 정보 공유를 통해 과학관간 상호 교환 및 교류를 촉진할 수 있는 과학관 전시 플랫폼 구축이 요구되고 있는 실정이다. 국외 선진 과학관의 경우 자연사 컨텐츠의 전산화 및 DB 구축을 통해 대국민 검색서비스를 제공하여 전시품 등을 다양하게 활용하고 있는 추세이다. 따라서 본 연구에서는 과학관에서 활용가능한 자연사 컨텐츠 중, 곤충을 중심으로 이들의 전시 컨텐츠 발굴 및 DB확보 현황과 향후 플랫폼 구축 계획에 대해 소개하고자 한다.

        Population Genetic Structure and Evidence of Demographic Expansion of the Ayu (Plecoglossus altivelis) in East Asia

        Plecoglossus altivelis (ayu) is an amphidromous fish widely distributed in Northeastern Asia from the East China Sea to the northern Japanese coastal waters, encompassing the Korean Peninsula within its range. The shore lines of northeastern region in Asia have severely fluctuated following glaciations in the Quaternary. In the present study, we investigate the population genetic structure and historical demographic change of P. altivelis at a population level in East Asia. Analysis of molecular variance (AMOVA) based on 244 mitochondrial control region DNA sequences clearly showed that as the sampling scope extended to a larger geographic area, genetic differentiation began to become significant, particularly among Northeastern populations. A series of hierarchical AMOVA could detect the genetic relationship of three closely located islands between Korea and Japan that might have been tightly connected by the regional Tsushima current. Neutrality and mismatch distribution analyses revealed a strong signature of a recent population expansion of P. altivelis in East Asia, estimated at 126 to 391 thousand years ago during the late Pleistocene. Therefore it suggests that the present population of P. altivelis traces back to its approximate demographic change long before the last glacial maximum. This contrasts our a priori expectation that the most recent glacial event might have the most crucial effect on the present day demography of marine organisms through bottleneck and subsequent increase of effective population size in this region.

        The Effect of Clinical Dental Hygienist’s Moral Sensitivity and Psychological Well-Being on Job Engagement

        Background: The present study aimed to provide basic data to establish a foundation for efficient operation of the organization by identifying the effects of factors related to moral sensitivity and psychological well-being on job engagement of clinical dental hygienists. Methods: This cross-sectional survey consisted of self-administration questionnaires. After the approval by the Institutional Review Board, a survey was conducted from September 2019 to November 2019, targeting dental hygienists with clinical experience of 1 year or more. A total of 189 questionnaires were analyzed using SPSS 18.0 for Windows. The data were analyzed using average standard deviation, t-test, one-way analysis of variance, Scheffe test, Pearson's correlation analysis, and multiple regression analysis. Results: According to job-related characteristics, higher job engagement was associated with more amount of clinical experience, less turnover intention, higher positions, and occasional job fatigue. Job engagement was improved by increasing moral sensitivity and psychological well-being of clinical dental hygienists. The factors affecting job engagement revealed that along with clinical career, the sub-areas of job engagement, namely turnover intention, moral sensitivity, and psychological well-being had statistically significant influence on job engagement and the explanatory power of the model was approximately 44.8%. Conclusion: Joy of working as a clinical dental hygienist, clinical career, turnover intention, moral sensitivity, and psychological well-being were confirmed as influencing factors that affected job engagement of clinical dental hygienists. It is necessary to establish an effective plan to improve job engagement by increasing moral sensitivity and psychological well-being of clinical dental hygienists.
