        Correlation between DNA methylation and Thymic Stromal Lymphopoietin expression in asthmatic airway epithelial cells

        Background: The overexpression of TSLP and DNA methylation in asthma were both risk factors the relationship was not clear. Objective: This study aimed to investigate the relationship between methylation status of TSLP promoter and mRNA/protein expression in asthmatic airway epithelial cells. Methods: Human bronchial epithelial cells were cultured in vitro and divided into: Control group, treated with PBS, model group, sensitized with LPS (10 μg/mL) for 12 h (37 °C, 5% CO2). Other groups were cultured with the pCMV3 plasmid (M + NC/pCMV), pGPH1 plasmid (M + NC/pGPH), DNMT1/pCMV3 plasmid (M + DNMT1/pCMV), and DNMT1/pGPH1 plasmid (M + DNMT1/pGPH) for 48 h. The expression of DNA methyltransferase 1 and TSLP were measured using real-time PCR and western blotting. Results: Compared with the control group, TSLP mRNA (1.00 ± 0.00 vs. 2.82 ± 0.81 vs. 1, P < 0.001) and protein (1.07 ± 0.04 vs. 1.46 ± 0.11, P < 0.01) were significantly greater, and the methylation of promoter was lower (92.75 ± 1.26 vs. 58.57 ± 3.34, P < 0.05) in the model group. Compared with the model group, TSLP mRNA (2.82 ± 0.81 vs. 1.17 ± 0.10, P < 0.001) decreased, but TSLP promoter methylation increased (58.57 ± 3.34 vs. 92.58 ± 7.30, P < 0.05) in M + DNMT1/pCMV. TSLP mRNA and protein were higher (2.82 ± 0.81 vs. 5.32 ± 0.21, P < 0.001; 1.46 ± 0.11 vs. 1.94 ± 0.11, respectively, P < 0.01), TSLP promoter methylation was lower (58.57 ± 3.34 vs. 33.57 ± 4.29, P < 0.05) in M + DNMT1/pGPH. Conclusions: Overexpression of TSLP in asthmatic airway epithelial cells may be regulated by DNA demethylation.

        Nodule Rich Protein 2 modulates nodule number in Medicago truncatula

        Symbiotic nitrogen fxation is beneft to sustainable agriculture and global nitrogen cycle. Many small peptides were identifed as regulators involving in the interaction between rhizobia and legume. Here we reported Nodule Rich Protein 2 (MtNRP2) encoding a small peptide in Medicago truncatula, belonged to a group of nodule rich protein restricted in legume species. MtNRP2 expressed highly in root nodule and its promoter was active during the initiation and development of root nodule and lateral root. To investigate the function of MtNRP2 in nodulation, we generated MtNRP2-overexpression and MtNRP2- knockdown transgenic Medicago. MtNRP2-overexpression transgenic lines performed normal nodulation phenotype compared with vector control. However, in the MtNRP2-RNAi transgenic plants, the decrease of MtNRP2 expression lead to the increase of infection threads number (7 day post inoculation) and nodules number (3 week post inoculation); meanwhile, the expression of MtRGF3 and MtPUB1 was inhibited. These results suggested that MtNRP2 negatively regulated nodulation in Medicago truncatula.

        Transcriptional activation of insulin-like growth factor binding protein 6 by 17β-estradiol in SaOS-2 cells

        Osteoblasts can synthesize the insulin-like growth factors (IGFs) and the IGF-binding proteins (IGFBPs), which may either enhance or attenuate IGF-stimulated bone cell proliferation. Since estrogen induced osteoblastic differentiation and proliferation through an estrogen- responsive gene in target cells, we investigated the effects of estrogen on IGFBP-6 expression in the human osteoblastic-like cell line SaOS-2. Expressions of IGFBP-6 protein and mRNA increased 2.8 and 2-fold, respectively, in the presence of 17-β-estradiol (E2) (0.01 to 1 μM) and estrogen receptor (ER) in SaOS-2 cells. On the other hand, E2 induced a 2-fold increase in SaOS-2 cell proliferation. To identify genomic sequences associated with estrogen responsiveness, the 5'-promoter region (-44 to +118) of the IGFBP-6 gene was cloned into a chloramphenicol acetyltransferase (CAT) reporter vector. E2 induced a 3-fold increase in CAT activity in SaOS-2 cells transiently transfected with this construct. Identification of the estrogen- responsive element (ERE) [5'-CCTTCA CCTG-3'] (-9 to +1) in this IGFBP-6 gene promoter region was confirmed using electromobility shift assays and deletion analysis. This functional ERE was important for E2-induced trans-activation of the IGFBP-6 gene. These results demonstrate that E2 exhibits a positive effect on IGFBP-6 gene transcription through estrogen- liganded ER binding to the functional ERE in the IGFBP-6 gene promoter in SaOS-2 cells.         Reporting quality of economic evaluations of the negotiated Traditional Chinese Medicines in national reimbursement drug list of China: A systematic review

        Background: Traditional Medicine (TM) has a wide uptake in most countries. In China, Traditional Chinese Medicine (TCM) is a common kind of primary health because of its beneficial effects. This review aimed to appraise the publication reporting quality of economic evaluations for selective TCM in the National Reimbursement Drug List (NRDL), Version 2020, based on the Consolidated Health Economic Evaluation Reporting Standards (CHEERS) statement. Methods: Electronic databases were searched for economic evaluation that supported the TCM negotia- tions in NRDL (2020 version) published from 2001 to 2021, including PubMed, Web of Science, Embase, CNKI, WanFang, and SinoMed. The CHEERS statement was used to appraise the reporting quality of in- cluded TCM economic evaluations. Results: A total of 360 articles were retrieved, but only 38 economic evaluations met the inclusion crite- ria. None of the articles reported all items in the CHEERS checklist. The mean score of included articles is low at 10.93 ±2.62, with an average scoring rate of 51.31 ±10.53%. The least reported items included: “Characterizing heterogeneity,”“Conflicts of interest”, “Discount rate,”and “Study perspective,”with a re- porting rate of 0.00%, 5.26%, 7.89%, and 15.79%, respectively. Conclusion: An upward trend occurred in the quantity and quality of the economic evaluation publica- tions of TCM in China. TCM economic evaluations are still at an early stage, with an urgent need for improving reporting quality. It may result from research experiences or different ideas between TCM and Western Medicine. Adhering to reporting guidelines like CHEERS and educating economic evaluation in- vestigators can improve TCM economic evaluations’ reporting quality.

        Molecular Dynamics Simulation on Microstructure Evolution During Solidification of Copper Nanoparticles

        The effect of cooling rate on the microstructure evolution of liquid Cu nanoparticles during their solidification process is investigated by using a molecular dynamics simulation based on the embedded atom method (EAM) potential developed by Foiles et al.. The potential energy analysis,the pair distribution function and the common neighbor analysis have been used. The results show that the solidification point increases with decreasing cooling rate and that the solidification of the microstructure of Cu nanoparticles varies with the cooling rate. The microstructure consists of fcc,hcp and bcc crystals or mixtures, though the fcc structure dominates, except in the amorphous state. An amorphous structure was obtained when the cooling rate reached 1.0 × 1013 K/s or higher while crystallization degree increased with decreasing cooling rate, and the total content of crystal structures reached to 95% when the cooling rate dropped to 4.0 × 1011 K/s, which was nearly a perfect crystal structure. The results also indicate that a single-crystal nanoparticle will not be obtained by quenching the liquid metal under various cooling rates.

        A Study on the Civil Liability of Freight Forwarders under Chinese Law - Focus on the Regulations on the Trial of Disputes over Ocean Freight Forwarding - 1

        오늘날, 해상화물 운송주선인은 해상운송 시장 내 경쟁이 심화됨에 따라 그 의 업무 범위도 많이 변화되어, 화주와 운송업자 간의 연결 뿐만 아니라 많은 업무를 수행하고 있는데, mere agent, contracting carrier 및 performing carrier 등의 업무를 수행하면서 여러 가지 법적 문제가 발생 할 수 있다. 이러한 배경 을 바탕으로 중국최고법원은 해상화물 운송주선인의 분쟁의 재판에 관한 법규 를 제정하였다. 본 연구의 목적은 이 법규를 소개하고 해상화물 운송주선인의 정의 및 법적 책임, 운송 서류의 송달 지연, FOB 조건 하에서의 운송 서류의 송달, 화물의 멸실 및 훼손의 고지 및 해상화물 운송주선에 관한 분쟁의 시효 및 관할권에 대하여 논하고자 한다. With the highly competitive market, the functions of the freight forwarders have changed a lot. They are not only the link between the owner of the goods and the carrier, but also undertake many duties and can act as the mere agent, the contracting carrier or the performing carrier, while these may led to the arguments in judicial procedure with economic disputes. On such basis, the Supreme People’s Court of China had promulgated the Regulations concerning the trial of disputes over ocean freight forwarding. This article aims to outline the highlights of the Regulation on the identification of freight forwarders and their civil liability, and hope to answer and explain the detention of documents, the delivery of transport documents under FOB Terms, notice of loss or damage, time bar and jurisdiction over disputes of freight forwarding service.

        Prescribed Performance-tangent Barrier Lyapunov Function for Adaptive Neural Backstepping Control of Variable Stiffness Actuator with Input and Output Constraints

        Due to the complexity of modeling and the strong transmission coupling, the rich background of rigid actuator control has not been transferred to variable stiffness actuator (VSA). Therefore, most model-based control techniques developed for VSA require feedback linearization first. Alternatively, VSA can use non-model-based control techniques such as PD control, but it does not show strong robustness under disturbances. This paper is concerned with designing a novel adaptive neural network backstepping control scheme without using feedback linearization for a special VSA with saturation inputs, output constraints, and disturbances. Firstly, for ensuring the VSA with lower tracking error and higher security, the prescribed performance-tangent barrier Lyapunov function (PP-TBLF) is introduced to handle the prescribed output performance constraints. Subsequently, the Chebyshev neural network and the Nussbaum-type function are exploited to approximate the unknown nonlinearities and unknown gains. Meanwhile, the inverse hyperbolic sine function tracking differentiator is utilized to solve the “explosion of complexity” caused by the differentiation of virtual inputs and also approximate the complex partial derivatives caused by the auxiliary control signals. Finally, the stability of the whole scheme is proved by the Lyapunov criterion. The simulation results illustrate the raised control scheme’s feasibility and show a better closed-loop behavior relative to that obtained using a classic PD controller.
