        Identification of genes involved in inbreeding depression of reproduction in Langshan chickens

        Objective: Inbreeding depression of reproduction is a major concern in the conservation of native chicken genetic resources. Here, based on the successful development of strongly inbred (Sinb) and weakly inbred (Winb) Langshan chickens, we aimed to evaluate inbreeding effects on reproductive traits and identify candidate genes involved in inbreeding depression of reproduction in Langshan chickens. Methods: A two-sample t-test was performed to estimate the differences in phenotypic values of reproductive traits between Sinb and Winb chicken groups. Three healthy chickens with reproductive trait values around the group mean values were selected from each of the groups. Differences in ovarian and hypothalamus transcriptomes between the two groups of chickens were analyzed by RNA sequencing (RNA-Seq). Results: The Sinb chicken group showed an obvious inbreeding depression in reproduction, especially for traits of age at the first egg and egg number at 300 days (p<0.01). Furthermore, 68 and 618 differentially expressed genes (DEGs) were obtained in the hypothalamus and ovary between the two chicken groups, respectively. In the hypothalamus, DEGs were mainly enriched in the pathways related to vitamin metabolism, signal transduction and development of the reproductive system, such as the riboflavin metabolism, Wnt signaling pathway, extracellular matrix-receptor interaction and focal adhesion pathways, including stimulated by retinoic acid 6, serpin family F member 1, secreted frizzled related protein 2, Wnt family member 6, and frizzled class receptor 4 genes. In the ovary, DEGs were significantly enriched in pathways associated with basic metabolism, including amino acid metabolism, oxidative phosphorylation, and glycosaminoglycan degradation. A series of key DEGs involved in folate biosynthesis (gamma-glutamyl hydrolase, guanosine triphosphate cyclohydrolase 1), oocyte meiosis and ovarian function (cytoplasmic polyadenylation element binding protein 1, structural maintenance of chromosomes 1B, and speedy/RINGO cell cycle regulator family member A), spermatogenesis and male fertility (prostaglandin D2 synthase 21 kDa), Mov10 RISC complex RNA helicase like 1, and deuterosome assembly protein 1) were identified, and these may play important roles in inbreeding depression in reproduction. Conclusion: The results improve our understanding of the regulatory mechanisms underlying inbreeding depression in chicken reproduction and provide a theoretical basis for the conservation of species resources.         Varilactones and wortmannilactones produced by Penicillium variabile cultured with histone deacetylase inhibitor

        Seven polyketides, including four new onesnamed as varilactones A–B (1–2) and wortmannilactonesM–N (3–4), as well as three biogenetically related knownwortmannilactones E, F and H (5–7), were isolated fromthe fungus Penicillium variabile HXQ-H-1, cultivated inpotato-based medium with the histone deacetylase inhibitorsuberoylanilide hydroxamic acid (SAHA). Their structures(1–7) were established by analysis of NMR and MS data,and the absolute configuration of oxabicyclo[2.2.1]heptaneunit was deduced from 1H-NMR analyses on MPA estersafter chemical reduction reaction. Among them, varilactonesA and B (1 and 2), features a novel skeleton with atriene unit linking an oxabicyclo[2.2.1]heptane and anoxabicyclo[3.3.0]octane rings.

        Antibacterial and Antioxidant Composite Fiber Prepared from Polyurethane and Polyacrylonitrile Containing Tea Polyphenols

        Antibacterial and antioxidant properties exert an indispensable effect on biomedical and packaging materialswhich are mainly made of fiber-based fabrics. The current research illustrates a novel type of composite fiber which wasprepared from thermoplastic polyurethane (PU), polyacrylonitrile (PAN) and tea polyphenols (TP) through wet-spinningtechnology. The PU-PAN mass ratio was optimized upon the mechanical property; TP was incorporated into the compositefiber with different contents. The obtained fibers were fully characterized in terms of mechanical, thermal, morphologicalproperties as well as surface wettability. Additionally, in vitro TP release, antioxidant and antibacterial activity of suchcomposite fibers before and after washing were under investigation systematically and orderly. The findings showed that TPloadedcomposite fibers with an average diameter around 120 μm performed a sustained TP release behavior and desirablecytocompatibility. According to the capacity on scavenging 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radicals, theobservation of scavenging ratio reaching to 97 % proved the excellent antioxidant performance. Furthermore, the idealantibacterial activity against Gram-negative E. coli and Gram-positive S. aureus was also demonstrated. Collectively, thisstudy provides a facile strategy to design and prepare a novel type of composite fibers and fabrics, which may be put intoapplication as wound dressing and tissue conduit.

        Probabilistic Power Flow Studies Incorporating Correlations of PV Generation for Distribution Networks

        This paper presents a probabilistic power flow (PPF) analysis method for distribution network incorporating the randomness and correlation of photovoltaic (PV) generation. Based on the multivariate kernel density estimation theory, the probabilistic model of PV generation is proposed without any assumption of theoretical parametric distribution, which can accurately capture not only the randomness but also the correlation of PV resources at adjacent locations. The PPF method is developed by combining the proposed PV model and Monte Carlo technique to evaluate the influence of the randomness and correlation of PV generation on the performance of distribution networks. The historical power output data of three neighboring PV generators in Oregon, USA, and 34-bus/69-bus radial distribution networks are used to demonstrate the correctness, effectiveness, and application of the proposed PV model and PPF method.

        Establishment of hybridization breeding of Ganoderma lucidum by protoplast monokaryogenesis method

        Four Ganoderma lucidum strains, Chizhi 05, Jingda, Huizhou and Xinzhou, were screened out as hybrid parent in order to establish G. lucidum cross breeding system that based on protoplast monokaryogenesis method. Monokaryotic strains of each parental strains were obtained and mating type of each monokaryotic strains were determined. One to three monokaryotic strains that have different mating types were mated, and hybrids were identified by clamp connection observation and antagonist response. The results showed that the number of monokaryon came from Chizhi 05, Jingda, Huizhou and Xinzhou was 9, 14, 40 and 38, respectively. Only one mating type was obtained from Jingda, and two mating types were obtained from the other three strains, Chizhi 05, Huizhou and Xinzhou, respectively. Chi-square test showed that the ratio of two mating types of the three strains was 1:1. Fourteen monokaryotic strains of different mating types from 4 parental strains were select as a cross- breeding materia, and 17 hybrids were obtained, which were identified by clamp connection observation and antagonist response. This study proclaimed that the practicality of the hybridization breeding of G. lucidum by protoplast monokaryogenesis method.

        Analysis of genetic diversity of Ganoderma strains based on rDNA ITS sequencing

        The genetic diversity of 16 Ganoderma strains was investigated by rDNA-ITS sequencing. Alignment analysis showed that whole length of internal transcribed spacer(ITS1+ITS2) was 500bp and with 139 variation sites (accounting for 23.3%, ITS1 was 66 and ITS2 was 73), 337 conserved sites (accounting for 72.2%), 59 informative sites (accounting for 9.88%), 86 conversion sites (G-A, C-T), 13 transversion site(C-G, T-A). The ratio of transition and transversion in ITS1 was higher than that in ITS2, and the variable sites of ITS2 were more than those of ITS1. The genetic distance among 16 Ganoderma strains is from 0 to 0.121. The genetic distance between G. lipsiense and F-1 was 0, and the genetic distance between Heizhi 02 and Huizhou, Jingda, G. capense was 0.121, 0.117 and 0.120, respectively. The 16 Ganoderma strains were classed into 4 groups. The biggest group is comprised of 12 strains, including Xinzhou, Huizhou, Jingda, 902, F-1, Xianzhi, Meiluo, Taishan, G. applanatum, 05, G. luteomarginatum. The G. atrum and G. sinense were clustered into one group. The G. capense and Zhongzhi was independent group, respectively. These results showed that there were some genetic difference among groups, and there was lower genetic diversity among strains in same groups.
