Cytokines involve in both inflammatory and healing mechanism. The aim of this study to evaluate the changing nature of cytokines and thereby appropriate time for using bone marrow derived monuclear cells(BMMNC) to acquire best effect. We chose 8 patients whose age is ranged from 38 to 51(mean=49). Bone marrow aspirate was obtained from iliac bone of patients with consented to the procedure. Cytokines from plasma layer acquired after 30 minutes of centrifugation at 1,820rpm were analyzed. Cytokines were evaluated after abstraction of mononuclear cells using ficoll method. The cytokines evaluated were Eotaxin, GM-CSF, IFN r, IL-1 a, IL-1 bIL-10, IL- 12(p40), IL-12(p70),IL-13, IL-15, IL-2, IL-3, IL-4, IL-5, IL-6, IL-7, IL-8, IP-10, MCP-1, MIP-1a, RANTES and TNF-a. High level of IP-10, IL-12 p40, RANTES were detected after centrifugation. Additionally, high level of IL- 6, IL-7, IL-8, MCP-1, MIP-1a were noticed in 1 and 7 days following culture. Mononuclear cells derivedfrom bone marrow have high activity character and the change of cytokine was observed after simple culture. Our results show that bone marrow derived mononuclear cells immediately after centrifugation of bone marrow is most effective.

Porcine pluripotent stem cells have important in modeling embryonic development and disease processes in biomedical research, especially to transplantation medicine, immunology and circulatory system. However, despite years of effort, the pluripotency machinery of porcine PSCs has not been demonstrated clearly. Porcine induced plu-ripotent stem cells (piPSCs) were generated using combination of six human tran-scriptional factors under stem cell culture condition. To confirm dependency of these piPSCs, they were separately cultured in stem cell media supplemented with i) 20 ng/ml LIF, ii) 20 ng/ml bFGF, iii) 20 ng/ml BMP4, iv) 20 ng/ml LIF and Su5402, or v) 20 ng/ml bFGF and Jak inhibitor I, for 48 h, respectively. Upon bFGF treated groups, colonies show signs of differentiation with dispersing colony morphology and losing alkaline phosphatase activity. In contrast, colonies with tightly expanded and strong AP activity are shown in LIF or BMP4 treated groups. Western blotting shows phospho- Stat3 and phospho-Smad1/5 are significantly suppressed by Jak inhibitor I. We con-clude that piPSCs generated by six human pluripotent factors, are dependent on LIF or BMP4 to sustain self-renewal and pluripotency.

<P>Conclusions: RNA-seq revealed transcriptomic differences between normal mucosa, noninflamed CD mucosa, and inflamed CD mucosa. Intestinal and serum CXCL1 was substantially increased with CD activity and can be used as a potential biomarker of CD.</P>
