      • Analysis of Indoleamine 2-3 Dioxygenase (IDO) and EGFR Co-expression in Breast Cancer Tissue by Immunohistochemistry

        Background: To determine the amount of co-expression of IDO and EGFR in breast cancer patients. Materials and Methods:In order to obtain the distribution of co-expression of IDO and EGFR in breast cancer, we tested 110 breast cancer paraffin tissue blocks with immunohistochemical methods. Then we investigated the relationship between the diagnostic and pathologic characteristics (tumor size, lymph node status, histologic grade, the gene expression of ER, PR, HER2, p53, Ki67 and PCNA) with the situation of co-expression of IDO and EGFR by reviewing the medical records of 32 breast cancer patients. Results: Among 110 breast cancers, 32 cases demonstrated IDO and EGFR co-expression (29.1%), IDO and EGFR synchronous co-expression being found in 19.1% and asynchronous in 10.0%. Conclusions: IDO and EGFR were co-expressed in breast cancer, including synchronous and asynchronous co-expression. The results suggest that considering IDO and EGFR as two indicators for breast cancer treatment or prognosis analysis provides a potential option of individual treatment for the portion of breast cancer patients with co-expression of IDO and EGFR.

        Nonlinear identification of Bouc–Wen hysteretic parameters using improved experience-based learning algorithm

        In this paper, an improved experience-based learning algorithm (EBL), termed as IEBL, is proposed to solve the nonlinear hysteretic parameter identification problem with Bouc-Wen model. A quasi-opposition-based learning mechanism and new updating equations are introduced to improve both the exploration and exploitation abilities of the algorithm. Numerical studies on a single-degree-of-freedom system without/with viscous damping are conducted to investigate the efficiency and robustness of the proposed algorithm. A laboratory test of seven lead-filled steel tube dampers is presented and their hysteretic parameters are also successfully identified with normalized mean square error values less than 2.97%. Both numerical and laboratory results confirm that, in comparison with EBL, CMFOA, SSA, and Jaya, the IEBL is superior in nonlinear hysteretic parameter identification in terms of convergence and accuracy even under measurement noise.

        What are the Individual`s Real Cares to Switch Personal Cloud Services?

        As the fast development of information technology (IT), abundant attractive alternatives have showed up and challenged to the traditional information technology (IT). Cloud services, the hottest representative among these alternatives, has attracted the attention from all walks of life. Considering the diversity of cloud services, this study attempts to find out the important factors affected on individual`s switching intention from incumbent IT to cloud services mainly based on two-factor theory (switching enablers and switching inhibitors) and explore the direct influences of these factors on individual`s switching intention. According to our findings, individual`s switching intention to cloud services is not only positively influenced by switching enablers but also negatively influenced by switching inhibitors. All the switching enablers in this study (perceived omnipresence, perceived collaboration support and perceived compatibility of cloud services) have positively significant influences on individual`s switching intention as well as the switching inhibitors (usage habit of incumbent IT and perceived risk of cloud services) have negatively significant influences on individual`s switching intention. It`s noteworthy that satisfaction of incumbent IT has insignificant influence on individual`s switching intention in this study. Moreover, as we expected, both social influence and personal innovativeness have significant influences on the generation of individual`s switching intention.

      • Microbial production of methyl anthranilate, a grape flavor compound

        <P><B>Significance</B></P><P>Methyl anthranilate (MANT) is widely used in the flavoring and cosmetics industry to give grape scent and flavor. In an effort to replace the conventional petroleum-based synthesis of MANT, we report the direct fermentative production of MANT from glucose in metabolically engineered <I>Escherichia coli</I> and <I>Corynebacterium glutamicum</I> strains. A synthetic plant-derived metabolic pathway was introduced and extensive metabolic engineering was performed including fine-tuning key enzyme levels and increasing the availability of precursor and cofactor metabolites. A two-phase extractive cultivation was developed using an extractant solvent to recover MANT in situ, which led to high levels of MANT production. This work demonstrates a promising sustainable alternative to MANT production and presents strategies applicable toward production of other valuable natural compounds.</P><P>Methyl anthranilate (MANT) is a widely used compound to give grape scent and flavor, but is currently produced by petroleum-based processes. Here, we report the direct fermentative production of MANT from glucose by metabolically engineered <I>Escherichia coli</I> and <I>Corynebacterium glutamicum</I> strains harboring a synthetic plant-derived metabolic pathway. Optimizing the key enzyme anthranilic acid (ANT) methyltransferase1 (AAMT1) expression, increasing the direct precursor ANT supply, and enhancing the intracellular availability and salvage of the cofactor <I>S</I>-adenosyl-<SMALL>L</SMALL>-methionine required by AAMT1, results in improved MANT production in both engineered microorganisms. Furthermore, in situ two-phase extractive fermentation using tributyrin as an extractant is developed to overcome MANT toxicity. Fed-batch cultures of the final engineered <I>E. coli</I> and <I>C. glutamicum</I> strains in two-phase cultivation mode led to the production of 4.47 and 5.74 g/L MANT, respectively, in minimal media containing glucose. The metabolic engineering strategies developed here will be useful for the production of volatile aromatic esters including MANT.</P>

        A general closed-form solution to a Timoshenko beam on elastic foundation under moving harmonic line load

        In this paper, a general closed-form solution for evaluating the dynamic behavior of a Timoshenko beam on elastic foundation under a moving harmonic line load is formulated in the frequency-wavenumber domain and in a moving coordinate system. It is found that the characteristic equation is quartic with real coefficients only, and its poles can be presented explicitly. This enables the substitution of these poles into Cauchy’s residue theorem, leading to the general closed-form solution. The solution can be reduced to seven existing closed-form solutions to different sub-problems and a new closed-form solution to the subproblem of a Timoshenko beam on an elastic foundation subjected to a moving quasi-static line load. Two examples are included to verify the solution.

        Enhanced Production of L-Tryptophan with Glucose Feeding and Surfactant Addition and Related Metabolic Flux Redistribution in the Recombinant Escherichia coli

        The production of L-tryptophan was investigated in a recombinant strain Escherichia coli W3110-ZDrr. It was observed that phosphate and feeding strategies are key factors to ensure the good cell growth and high production of L-tryptophan. The simple exponential feeding strategy could only produce 10.6 g/L L-tryptophan due to the improper feeding rate; while the manual glucose-feedback feeding approach could effectively control the substrate and inhibit the formation of acetate, and thus improvemed the Ltryptophan production to 25.5 g/L. The modified exponential feeding approach avoided overfeeding or underfeeding and achieved high production of L-tryptophan. Moreover, the addition of Tween 60 or PL61 could enhance the cell growth and the production of L-tryptophan in the fed-batch mode. Subsequent metabolic flux analysis showed that more carbon flux was distributed into the biosynthesis of Ltryptophan when Tween 60 or PL61 was supplied. The present work presents one base for further large-scale production of this important amino acid.

        Enhanced interfacial adhesion between polypropylene and carbon fiber by graphene oxide/polyethyleneimine coating

        In order to improve the interfacial properties in polypropylene/carbonfiber (PP/CF) composites,graphene oxide and brached polyethyleneimine were coated onto the surface of carbonfiber by layer-by-layer assembly in this work. Compared with the origin PP/CF composite, the composites reinforced by PP/CF–GO showed significant enhancement not only in tensile strength but also in elongation at break. Theimprovedfiber–matrix adhesion was proved by fracture morphology observation of scanning electronmicroscopy and almost unaffected mechanical properties of thefiber itself during the coating process. The optimal assembly time was found to be 10 for enhancing the overall composite mechanicalperformance.

        Metagenomics analysis of the gut microbiome in healthy and bacterial pneumonia forest musk deer

        Background The forest musk deer (FMD, Moschus berezovskii) is an threatened species in China. Bacterial pneumonia was found to seriously restrict the development of FMD captive breeding. Historical evidence has demonstrated the relationship between immune system and intestinal Lactobacillus in FMD. Objective We sought to elucidate the diferences in the gut microbiota of healthy and bacterial pneumonia FMD. Methods The bacterial pneumonia FMD was demonstrated by bacterial and pathological diagnosis, and the gut microbiome of healthy and bacterial pneumonia FMD was sequenced and analysed. Results There are three pathogens (Pseudomonas aeruginosa, Streptococcus equinus and Trueperella pyogenes) isolated from the bacterial pneumonia FMD individuals. Compared with the healthy group, the abundance of Firmicutes and Proteobacteria in the pneumonia group was changed, and a high level of Proteobacteria was found in the pneumonia group. In addition, a higher abundance of Acinetobacter (p=0.01) was observed in the population of the pneumonia group compared with the healthy group. Several potentially harmful bacteria and disease-related KEGG subsystems were only found in the gut of the bacterial pneumonia group. Analysis of KEGG revealed that many genes related to type IV secretion system, type IV pilus, lipopolysaccharide export system, HTH-type transcriptional regulator/antitoxin MqsA, and ArsR family transcriptional regulator were signifcantly enriched in the metagenome of the bacterial pneumonia FMD. Conclusion Our results demonstrated that the gut microbiome was signifcantly altered in the bacterial pneumonia group. Overall, our research improves the understanding of the potential role of the gut microbiota in the FMD bacterial pneumonia.
