      • Function of MST1/2 in the Regulation of Breast Cancer Cell Proliferation

        Mammalian sterile 20-like kinase 1/2 (MST1/2)는 종양 형성에 중요한 역할을 하지만, 유방암 발병 시 MST1/2의 기능은 완전히 밝혀진 것은 아니다. 이 연구에서 MST1/2-siRNA를 이용한 MST1/2 억제를 통해 ER가 존재하는 MCF-7 유방암 세포주의 세포 증식이 효과적으로 억제됨을 확인하였고, ER, cyclin D1, pS2와 같은 유방암세포 성장에 중요한 인자들이 MST2억제로 인해 감소함을 확인할 수 있었다. 게다가 breast invasive carcinoma datasets (from Gene Expression Omnibus) 와 The Cancer Genome Atlas (from Gene set Enrichment Analysis)를 통하여 MST2의 발현과 세포주기 조절에 관련된 gene set의 발현 패턴이 양성적으로 관련이 있음을 확인 할 수 있었고, Kaplan Meier plotter를 통하여 MST2 발현에 따른 유방암 환자의 생존률을 확인한 결과 MST2 발현이 낮은 유방암 환자 그룹이 예후가 더 좋음을 확인할 수 있었다. 이 연구는 MST2와 유방암의 진행과의 관계를 이해하는데 도움을 줄 수 있을 것이라 생각된다. Mammalian sterile 20-like kinase 1/2 (MST1/2), upstream kinase of Hippo pathway, plays important roles in tumorigenesis, yet the function of this factor in breast cancer development is not fully characterized. In this study, I utilized distinct methods of checking cell viability by knock down of MST1/2 to show that the ablation of this kinase dramatically suppresses the proliferation of ER -positive breast cancer cell line (MCF7). I also identify some key proteins that regulate breast cancer cell growth, which are ER, cyclin D1, pS2, are downregulated specifically by MST2 elimination. In addition, breast invasive carcinoma datasets from Gene Expression Omnibus (GEO) (NCBI) and The Cancer Genome Atlas (TCGA), which were subjected to Gene Set Enrichment Analysis (GSEA), suggested that high expression values of STK3 (MST2) were positively coordinated with expression of cell cycle related gene sets. My observations are firmly supported by Kaplan Meier plotter which demonstrates that breast cancer patients with high expression of MST2 have a poorer prognosis than the low-expressed group. This investigation provides a new insight into the relationship between MST2 and the mediation of breast cancer progression.       • Classification of Microbial Community in Soil of Ginseng Field and Korean Fermented Food

        Ginseng soil is one of the best habitats for the soil borne microorganisms such as bacteria, fungi and yeast. Without any fertilization and direct sunlight, ginseng soil is a favourite environment for microbial colonization. On the other hand, rhizosphere microbial community does metabolites to supply nutrients for ginseng root which contains a lot of pharmaceutical compounds. Korea was known as a country which has many kinds of traditional fermented foods. Among them, it is impossible not to mention kimchi. Including a lots of spices, kimchi could be an ideal source to provide valuable microorganisms, especially lactic acid bacteria (LAB) play an important role in taste of kimchi, as in dairy products such ascheese and fermented milk. The soil samples were collected from ginseng field near the Deajon city, South Korea. The soil samples at the depth, near the rhizosphere region of the ginseng plant and kimchi sauce from Suwon market were collected to study the bacterial community structure and discover novel microorganisms for their taxonomic characterization. By dependent culture method, bacterial colonies were isolated by dependent culture method and then purified on new agar plates. The three novel strains were isolated in the currentstudy from samples of ginseng soil and kimchi sauce. At first the novelty of the strains were confirmed by complete sequencing of 16S rRNA gene. The strains which their genome have no more than 70% DNA-DNA relatedness or 98% 16S rRNA identity to their nearest relatives of another species were chosen to propose as novel species. Polyphasic taxonomic characterization was conducted for three novel strains based on genomic, phenotypic methods. The novel strains later were characterized in physiology including Gram straining, motility, spore formation, and growth pattern. The chemotaxonomic characteristics of the novel isolates were examined by quinone, fatty acidand amino acid analysis. In order to check utilization of carbohydrate, API kits including API 50CH, API 50CHL, API 32 GN and API 20NE were recruited. The production of enzymes was examined by using API ZYM microsystem analysis. Based on polyphasic taxanomic characterization two novel strains were reported from the ginseng field and one novel strain from Kimchi. Two novel strains namely Rhodanobacterium soli sp. nov., Methylobacterium ginsengisoli sp. nov., belonging to the phylum Proteobacteria and one novel strain namely Lactobacillus koreensis sp. nov.,belonging to the order Lactobacillales were isolated. This study help to understand further about microbial community in ginseng soil and Korean fermented food.

      • Non-enzymatic electrochemical sensing of pollutants using nanoporous gold

        Nanoporous gold thin films were prepared by electrochemical dealloying Si out of AuxSi1−x (0.1 < x < 0.9) thin films co-deposited on Si substrate. Structure of pores and gold ligaments of the resulting porous thin films were easily controlled via adjusting the initial alloy compositions. It was found that the nanoporous gold thin films with roughness factors of about 25 were able to show good linear amperometric responses towards phenol, catechol, aniline and hydroxylamine. Successful amperometric detections of phenol were recorded in different electrolytes, including phosphate buffered saline, sulfuric acid and sodium hydroxide solutions, with high sensitivities, relatively low detection limits and wide linear detection ranges. The repeatability was found to be affected by the type of electrolyte and the structural change upon heat treatment. Polymeric fouling of NPG resulted from polymerization of phenoxy radicals leaded to electrode deactivation after a number of using times in neutral pH solution but a mild annealing of NPG was found suppress such deactivation. Meanwhile, a stable sensing performance was observed in the sulfuric acid electrolyte as NPG surface was free from carbon contamination. NPG thin films also offered high sensitivity, good repeatability and relatively low detection limits towards hydroxylamine and aniline while a significantly large linear detection range with moderate sensitivity was observed for catechol in neutral PBS solution. In addition, based on cyclic voltammetry studies, we explored that NPG thin films could efficiently oxidize/reduce a number of benzene derivatives, thus offering great potentials for further applications in various electrochemical processes. The electrochemical pathways and effects of functional groups as well as substitution positions were also proposed.

      • Steam reforming of biogas over plate-type Nickel catalysts

        This study aims at investigating catalytic activity of plate catalysts based on bulk Ni towards steam reforming of simulated biogas in the temperature range of 600-700oC. Three types of catalysts including porous Ni, Ni-Cr plates, and commercial Ni fiber mat were employed, and the former two Ni catalysts were prepared by conventional tape casting method. Although all of these original materials exhibited insignificant catalytic activity owing to the low surface area of the bulk structure, it was found that surface reconstruction induced by oxidative pretreatment helped improve the activity. The oxidized surfaces are believed to provide a number of different active sites such as sites on active crystal planes, steps, and pores as well as other defect sites when exposing to reductive agent such as hydrogen or methane. On the other hand, the presence of promoters such as Cr2O3 and CeO2 appeared to stabilize the activity by preventing sintering of the catalytically active sites.

      • Development of musculoskeletal model and biomechanical analysis of lower limb during daily activities

        Biomechanical analysis combined with computer musculoskeletal models can provide significant insight into how the neuromuscular and musculoskeletal systems interact to produce movement. In this thesis, I built a musculoskeletal model of lower extremity. In addition, biomechanical analysis programs were also developed to study about joint kinetics, joint kinematics and muscle forces of lower extremity during daily activities. Kinematic data was investigated by kinematic analysis program, joint forces and moments were calculated from inverse dynamic analysis program whereas muscle forces were predicted using two procedures: inverse dynamics and forward dynamics. In inverse dynamics analysis procedure, both static and global optimizations were suggested. While the inverse dynamic with static optimization method has been proven useful in predicting muscle forces from the measured kinematics and kinetics of the segments, one major criticism is that this method does not properly predict co-contraction of antagonistic muscles. Global optimization that determines the knee joint center of rotation and constructs three knee moment equilibrium equations with respect to this rotation center was suggested to solve the elimination of ignoring co-contraction between agonist and antagonist muscles. These methodologies were used to study about biomechanics of lower extremity during daily activities (walking, sit to stand, squat).

      • When the forest became the enemy and the legacy of American herbicidal warfare in Vietnam

        This dissertation offers an environmental perspective on the Vietnam War by an intensive interdisciplinary and cross-cultural examination of the American use of herbicides for war purposes to destroy the jungles in the Vietnamese Central Highlands. The main issues explored are the cultural, political and military causes of U.S. ecocide in Vietnam at a time of rising domestic environmentalist concern about pollution and toxicity, and the search for extreme visibility in Vietnam that led to massive destruction of ecologically dependent human communities and local eco-systems with questionable military results. The thesis begins with an account of how the Kennedy Administration and the South Vietnamese Diem government started the program of defoliation and crop destruction and ends with the Agent Orange Controversy in the United States.
