        Inhibition of MicroRNA-15a/16 Expression Alleviates Neuropathic Pain Development through Upregulation of G Protein-Coupled Receptor Kinase 2

        There is accumulating evidence that microRNAs are emerging as pivotal regulators in the development and progression of neuropathic pain. MicroRNA-15a/16 (miR-15a/16) have been reported to play an important role in various diseases and inflammation response processes. However, whether miR-15a/16 participates in the regulation of neuroinflammation and neuropathic pain development remains unknown. In this study, we established a mouse model of neuropathic pain by chronic constriction injury (CCI) of the sciatic nerves. Our results showed that both miR-15a and miR-16 expression was significantly upregulated in the spinal cord of CCI rats. Downregulation of the expression of miR-15a and miR-16 by intrathecal injection of a specific inhibitor significantly attenuated the mechanical allodynia and thermal hyperalgesia of CCI rats. Furthermore, inhibition of miR-15a and miR-16 downregulated the expression of interleukin-1β and tumor-necrosis factor-αin the spinal cord of CCI rats. Bioinformatic analysis predicted that G protein-coupled receptor kinase 2 (GRK2), an important regulator in neuropathic pain and inflammation, was a potential target gene of miR-15a and miR-16. Inhibition of miR-15a and miR-16 markedly increased the expression of GRK2 while downregulating the activation of p38 mitogen-activated protein kinase and NF-κB in CCI rats. Notably, the silencing of GRK2 significantly reversed the inhibitory effects of miR-15a/16 inhibition in neuropathic pain. In conclusion, our results suggest that inhibition of miR-15a/16 expression alleviates neuropathic pain development by targeting GRK2. These findings provide novel insights into the molecular pathogenesis of neuropathic pain and suggest potential therapeutic targets for preventing neuropathic pain development.

      • Inhibitory Effects of Syk Transfection on Lung Cancer Cell Invasion

        Objective: Spleen tyrosine kinase (Syk) is closely related to tumor invasion and metastasis, and has been shown to have potential inhibitory effects in tumors. In this study, we constructed a eukaryotic expression vector for Syk and analyzed its effects on invasive ability of the A549 non-small cell lung cancer cell line in vitro. Methods: A fragment of Syk was obtained by RT-PCR from human lung cancer cells and cloned into the expression vector pLNCXSyk. After restriction endonuclease digestion, PCR and DNA sequencing confirmation, the recombinant Syk expression plasmid was transfected into A549 human lung cancer cells using lipofectamine protocols. After selection, the cells stably expressed Syk. Detection of Syk expression of the cells by RT-PCR, and invasive ability were examined. Results: The eukaryotic expression plamid pLNCXSyk was constructed and expressed stably in the A549 human lung cancer cells. The RT-PCR results showed that Syk mRNA expression was upregulated significantly (P<0.05). Lower invasion through a basal membrane were apparent after transfection (P<0.05). Conclusions: A eukaryotic expression plasmid to cause Syk expression in lung cancer cells can obviously inhibit their invasive ability in vitro.

        Influence of Energy Density on Energy Demand and Porosity of 316L Stainless Steel Fabricated by Selective Laser Melting

        Selective laser melting (SLM) is one of the most widely used metal additive manufacturing technologies in producing high density parts. Energy density, a key-parameter combination, has been recognized to have a relationship with part formation, but such a relationship is extremely complex. This work aims to investigate energy density as a measure to evaluate energy demand in fabricating pore-free 316L stainless steel SLM parts. Key parameters in energy density were considered in the developed energy demand model. The impact of energy density on the porosity was analyzed with the data from experiments and existing works. Either low or high energy density can result in larger and more pore formation, and the influencing parameter was laser power, followed by layer thickness, scan speed, and hatch space. An effective energy-optimal (E2O) zone was proposed, where a relationship between energy density and porosity was developed. It is suggested that high laser power with high scan speeds can deliver energy to a thicker layer with relatively stable melt pool, fabricating high density parts. Hatch space can be decided accordingly to actual melt pool formation. This combination can effectively reduce energy density, and corresponding energy demand.

        Synthesis and Characterization of pH-Responsive Poly(2-hydroxyethyl aspartamide)-g-Poly(β-amino ester) Graft Copolymer Micelles as Potential Drug Carriers

        Novel pH-responsive graft copolymers consisting of poly(2-hydroxyethyl aspartamide)-g-poly(β-amino ester) (PHEA-g-PAE) were prepared from polysuccinimide, which is a thermal polycondensation product of aspartic acid, via the Michael-addition reaction with β-amino ester moieties. The chemical structures of the prepared graft copolymers were characterized by 1H NMR. Their micelle properties were examined by dynamic light scattering and fluorescence spectrometry. The transmission electron microscopy images demonstrated that these micelles were regularly spherical in shape. For cancer therapy, doxorubicin (DOX) was efficiently encapsulated into the micelles. The DOX loaded micelles exhibited relatively sharp pH-dependent DOX release behavior.

        Molecular and Cellular Microbiology / Biomedical Sciences : Surface-Displayed IL-10 by Recombinant Lactobacillus plantarum Reduces Th1 Responses of RAW264.7 Cells Stimulated with Poly(I:C) or LPS

        Recently, poly-γ-glutamic acid synthetase A (pgsA) has been applied to display exogenous proteins on the surface of Lactobacillus casei or Lactococcus lactis, which results in a surfacedisplayed component of bacteria. However, the ability of carrying genes encoded by plasmids and the expression efficiency of recombinant bacteria can be somewhat affected by the longer gene length of pgsA (1,143 bp); therefore, a truncated gene, pgsA, was generated based on the characteristics of pgsA by computational analysis. Using murine IL-10 as an exogenous gene, recombinant Lactobacillus plantarum was constructed and the capacity of the surface-displayed protein and functional differences between exogenous proteins expressed by these strains were evaluated. Surface expression of IL-10 on both recombinant bacteria with anchorins and the higher expression levels in L. plantarum-pgsA’-IL-10 were confirmed by western blot assay. Most importantly, up-regulation of IL-1β, IL-6, TNF-α, IFN-γ, and the nuclear transcription factor NF-κB p65 in RAW264.7 cells after stimulation with Poly(I:C) or LPS was exacerbated after co-culture with L. plantarum-pgsA. By contrast, IL-10 expressed by these recombinant strains could reduce these factors, and the expression of these factors was associated with recombinant strains that expressed anchorin (especially in L. plantarum-pgsA’-IL-10) and was significantly lower compared with the anchorin-free strains. These findings indicated that exogenous proteins could be successfully displayed on the surface of L. plantarum by pgsA or pgsA’, and the expression of recombinant bacteria with pgsA’ was superior compared with bacteria with pgsA.

      • Blind Separation of Permuted Alias Image with Motion Blurred Using Image Enhancement in NSCT Domain

        Focused on the issue that motion blurred permuted alias image blind separation, an algorithm using image enhancement based on nonsubsampled contourlet transform (NSCT) domain was proposed. Firstly, permuted alias image was decomposed into low-frequency sub-band and high-frequency sub-bands, which were obtained by spare decomposition based on NSCT domain. Coefficients of high-frequency sub-bands were enhanced according to Bayesian shrinkage threshold and nonlinear gain function, and the enhanced version was got by this method. Then the permuted alias image and the enhanced version were blocked, the correlation coefficients were estimated by each corresponding sub-block, because the permuting image was changed larger, the permuting image could be separated by using threshold method. Experimental results show that the proposed algorithm can separate the permuting image effectively from the permuted alias image in spite of the motion blurred direction, blur degree, size, location and the number of permuting image.

      • Study of One Kind of Extended Proportional Guidance Law

        One kind of extended proportional guidance law is presented. This guidance law can be used without restriction about the motion speed of missile and target, and this is a different point with the classical proportional guidance law. In the case of random alter the tracking target curvature radius, compared with classical proportional guidance law under the proper random quantity, the normal overload of trajectory is relatively small, and has certain advantage, so the study has significant reference value in actual application. And interception time and normal overload have good performance. Tracking performance for high maneuverability target is better than the classical proportional guidance law. A target intercepting simulation based on MATLAB of the proportional coefficients and line-of-sight angular velocities is generated to analyze the trajectory properties. The effect of guidance coefficients on trajectory properties is studied. Through simulation, we simplify the problem by ignoring the actual factors so that a certain level of error is expected. The situation in practical requires more in-depth analysis and research.

        A20 ameliorates disc degeneration by suppressing mTOR/BNIP3 axis-mediated mitophagy

        Background The pathological mechanism of intervertebral disc degeneration (IDD) is an unanswered question that we are committed to exploring. A20 is an anti-inflammatory protein of nucleus pulposus (NP) cells and plays a protective role in intervertebral disc degeneration. Objective This study aims to investigate the molecular mechanism by which A20 attenuates disc degeneration. Methods The proteins of interest were measured by immunoblotting, immunofluorescence, ELISA assay, and immunohistochemical technique to conduct related experiments. Immunofluorescence assays and mitochondrial membrane potential (JC-1) were used to assess mitophagy and mitochondrial fitness, respectively. Results Here, we demonstrated that A20 promoted mitophagy, attenuated pyroptosis, and inhibited the degradation of the extracellular matrix, consequently significantly ameliorating disc degeneration. Mechanistically, A20 reduces pyroptosis and further suppresses cellular mTOR activity. On the one hand, A20-induced mTOR inhibition triggers BNIP3-mediated mitophagy to ensure mitochondrial fitness under LPS stimulation, as a result of mitigating mitochondrial dysfunction induced by LPS. On the other hand, A20-induced mTOR inhibition reduces the loss of mitochondrial membrane potential and the generation of Mitochondrial ROS. Conclusion The study revealed that A20 promotes BNIP3-mediated mitophagy by suppressing mTOR pathway activation against LPS-induced pyroptosis.

      • A Nation Scale Seismic Landslide Hazard Assessment of China

        Mountainous area accounts for about 69% of Chinese land area. Most of these mountainous areas are classified as high seismic intensity zones. This specific geological background makes China one of the countries encountered most serious seismic landslide losses. However, there is not such a seismic landslide hazard assessment result in nationwide scale available. For potential seismic landslide risk prevention and reduction, authors conduct a preliminary seismic landslide hazard assessment on a countrywide scale based on the Newmark d isplacement a nd i nformation v alue m odels. Firstly, t he P robabilistic S eismic H azard A nalysis (PSHA) method is applied to obtain peak ground acceleration (PGA) with a standard probability of exceedance (10% in 50 years). On the one hand, considering PGA results and engineering geological factors, the occurrence probability of seismic landslides under corresponding potential earthquake condition is analyzed based on Newmark displacement model. On the other hand, with the information value model, existing landslides and geospatial factors are considered, such as slope angle, relief, engineering geological lithology groups, distances to faults, distances to rivers. The landslide susceptibility is calculated based on information model. Furtherly, the seismic landslide hazard levels are obtained through matrix classification with susceptibility levels and PGA results. Finally, comprehensive seismic landslide hazard maps on a nationwide scale are zoned and the major cities of potentially highly hazard level are identified based on the predicted Newmark displacements and information values. The results represent a preliminary assessment which can provide reference for urban development, spatial plan and detailed seismic landslide risk management in national scale.
