        Differential Manifestation of Ocular Phenotypes in TALEN-mediated p19arf Knockout FVB/N and C57BL/6J mouse lines

        Background p19arf, primarily known as a tumor suppressor, has also been reported to play an essential role in normal development of mouse eyes. Consistently, lack of p19arf has been associated with ocular defects, but the mixed background of the knockout (KO) mouse strain used raised a concern on the accuracy of the phenotypes observed in association with the targeted gene due to genetic heterogeneity. Object We carried out a study to investigate into the efect of genetic background on the manifestation of p19arf KO associated phenotypes. Methods We characterized the phenotypes of novel p19arf KO mouse lines generated in FVB/N and C57BL/6J using a transcription activator-like efector nuclease (TALEN) system in comparison to the reported phenotypes of three other p19arf-defcient mouse lines generated using homologous recombination. Results Ninety-fve percent of FVB/N-p19arf KO mice showed ocular opacity from week 4 after birth which worsened rapidly until week 6, while such abnormality was absent in C57BL/6J-p19arf KO mice up to the age of 26 weeks. Histopathological analysis revealed retrolental masses and dysplasia in the retinal layer in FVB/N-p19arf KO mice from week 4. Besides these, both strains developed normally from birth to week 26 without increased tumorigenesis except for a subcutaneous tumor found in a C57BL/6J-p19arf KO mouse. Conclusion Our fndings demonstrated surprisingly variable manifestation of p19arf-linked phenotypes between FVB/N and C57BL/6J mice, and furthermore between our mouse lines and the established lines, indicating a critical impact of genetic background on functional study of genes using gene targeting strategies in mice.

      • 굴착 단계를 고려한 쉴드 터널의 점소성 유한요소 해석

        쉴드 터널공법은 연약지반이나 붕괴성 지반에 매우 유리한 터널공법이므로 날로 그 사용이 증대되고 있다. 그러나 이러한 쉴드 공법을 사용할 경우에는 대상 지반에 대한 충분한 지반 조사가 먼저 이루어져야만 한다. 본 연구에서는 쉴드 터널링을 모사 할 수 있는 점소성 유한요소를 통해 STAVIS라는 프로그램을 개발하였다. 이 프로그램은 Mohr-Coulomb 항복 규준을 사용하였고 지반의 변위 이력을 통한 유동계수 값을 사용하였다. 해석 대상은 부산 지하철 230공구로 하였고, 계측 라인에 대한 해석치와 계측치를 비교하였다. 융기 및 테일보이드 폐합단계, 라이닝 설치 후 지반 거동에 대한 해석결과가 계측치와 유사한 경향을 보여주었다. 본 논문의 연구결과는 지하 구조물의 가까운 곳에 쉴드 터널을 건설할 경우 유용하게 적용할 수 있을 것으로 본다. The construction method by the shield tunnel is increasing day by day, because it is useful method to soft ground and to be structure at neighboring place. Incase of use of the method, it must be planed enough a geological survey of soil and ground condition. Viscoplastic FEM program was developed STAVES in this study. The program is used Mohr-Coulomb yield criterion and adequate ·fluidity parameter was selected as comparing displacement histories of soil. The object of analysis was Busan Subway 230 site. The analytical values were compared with the measured values. The results of analysis are similar to the measured behavior of ground for heaving, tail void closure and liner installation. By using the results of this study, it can be applicable to construction of shield tunnel when it is nearly constructed to the underground structures.

        To use concrete returned from job sites, a method to maintain the fresh properties of the returned concrete for a long time was studied by adding a retarder to stop cement hydration temporarily and adding an activator to the concrete to reuse next day in the same manner as an ordinary concrete.

        This is a case of bony ankylosis of right temporomandibular joint with mild facial asymmetry resulted from trauma, which bring about the right condylar fracture, and corrected by interpositional arthroplasty with temporal fascia flap and coronoidectomy. The postoperative results were functionally and esthetically good, but continued osbervation is necessary to evaluation of abnormal bony growth and reankylosis.

        The pancreatic endocrine cells of the tree frogs were observed under light microscope by cytochmical technique. The results obtained are as follow; 1. The pancreatic islets are not surrounded with connective tissue capsuls and lots of blood vessels are distributed in the pancreatic islet. 2. The pancreatic islet cells of tree frogs Hyla arborea japonica consist of B,A_1, and A_2 cells. 3. The cell cords are composed of various number of the B cells which show columnar type with ovar-shaped nucleous. 4. A_1, A_2cells are located in the periphery of the pancteatic islet and B cells in the central portion of the pancreatic islat. 5. A_1 cells have long cytoplasmic process. 6. A few endocrine cells are observed in the pancreatic exocrine portion.

      • 하이브리드 광경화를 이용한 소광 도료 연구

        We investigated the effects of photosynergist and LED curing on the kinetic and film properties of UV curable coatings containing micro-silica. Cure kinetics were studied by DPC and the degree of curing at the surfaces and the interfaces with substrate were investigated by FT-IR/ ATR. Coating properties such as hardness, gloss, chemical and stain resistances and adhesion of the UV cured films containing various concentration of micro-silica were investigated. The results showed that UV cured films containing micro-silica provide an excellent matting property while sacrificing chemical resistance due to the surface morphology and oxygen inhibition. The UV cured films containing photosynergist produced superior surface properties such as chemical resistance while sacrificing the adhesion and matting properties. LED-UV dual curing system was proposed to optimize the process for the system where both high matting and excellent coating properties are required.

        The Rigid Element Method(R.E.M) is a kind of the numerical discretization, techniques, such as the finite element method or the boundary element method, for getting the approximate solution of continuum mechanics problem. This method is originally developed for elasto-plastic analysis of wall-type precast reinforced concrete structures. In R.E.M., each discreted element is regarded as rigid body and the elements are interconnected with virtual springs with the stiffness of element. The springs transmit the axial force, the shear force and the normal force. The degree of freedom is placed in an arbitrary point within the element and the rigid displacements of this point affect other element through the springs being around the element. In this paper, it is derived the generalized adaptation matrix of shell elements which consider the effect of bending moment in thickness direction and developed a computer program using the present theory. Also, it is analyzed the behaviors of structure, that is, the distribution of stresses, the progress of cracks, the shape of deformation and the mode of fractures, for the hemispherical shell supported on cylinder under internal pressure.

        The numerical methods have developed simultaneously with the increasing use of high-speed electronic digital computers, but most of these methods require a lot of computer time under elasto-plastic analysis. The Rigid Element Method (R.E.M.) is one of the very interesting method in order to reduce the computer time. The method is based on the assumptions that each of the divided elements of continuum is regarded as a rigid body and every element is jointed with virtual springs each other. In this study, the rigid element method concerning partial cylindrical shell is established and adapted to the partial cylindrical shear-wall in order to examin closely the possibility for application of this method to the continuum.

        The base mats for several types of reactor containment structures which rest directly on the surface a uniform rock foundation are nearly circular. In this paper these base-mats idealized thick circular plates on elastic foundation in engineering design and analysis. As the base mat is thick circular plate it must be consider the transverse shear deformation, then the basic equation of isotropc circular plate were derived by Reissner's theory. Based on the analytical studies performed, the following conclusions are obtained. 1) Analytical solutions for the thick circular plate are shown by Fig. 4.2 and Fig 4. 3. 2) In theory of thick plate, the thickness of plate is the thicker and the influence of w due to shear deformation is the larger, but M_r and M_rθdo not change.

        오늘날 전자무역은 EDI와 인터넷 등 통신네트워크를 통하여 거래가 이루어지기 때문에 시간과 공간적 제약을 극복하고, 신속하고 경제적인 거래를 수행할 수 있다. 이런 이점에도 불구하고 전자무역에 대한 기업의 정보화 마인드부족, 전문인력의 부족, 무역거래알선사이트의 역할미비, 무역관련 각 시스템간의 단절 및 통합 전자무역네트워크의 부재, 전자무역관련 법제도적 환경구축의 미비 등으로 인하여 전자무역의 활용도는 매우 저조한 실정이다. 따라서 전자무역 활성화방안으로 먼저 전자무역 친화적인 환경조성을 위하여 전자무역관련 법제도외 개선이 이루어져야 할 것이다. 둘째, 전자무역관련 전반적인 업무단절현상을 방지하고, 전자무역을 원활하게 수행하기 위해서는 국내외 관련 네트워크를 연결하는 통합 전자무역네트워크가 구축되어야 한다. 셋째, 무역유관기관에서 운영하고 있는 무역거래알선사이트의 효율성 제고와 더불어 중소기업의 전자무역활성화를 위하여 무역업무처리를 통합·관리할 수 있는 솔루션의 개발, 보급 및 e-무역상사의 육성이 요구된다. 그리고 마지막으로 전자무역관련 전문인력양성체제를 구축하고, 인재뱅크나 헤드헌터 시스템 등의 활성화가 요구된다. Electronic trade process includes all trade-related business, from foreign exchange, to customs clearance and logistics through EDI and, especially, processing various documents related to export and import using computer system realizing paperless trade business. All related government agencies and trade companies including the Customs, customs brokers, financial institutions, agencies, carriers and insurance companies are now connected to the electronic trade process. In consideration of these circwnstances, this paper is aimed to examine the practical uses of electronic trade and propose several suggestions for developing electronic trade in Korea. In implementing electronic trade, there are some obstacles as follows; absence of global electronic trade network, inefficiency of trade leads sites, and lack of reliable certificate authority and payment system, shortage of specialist for electronic trade, insufficiency of policy support, and so on. Therefore, for developing electronic trade, it is required to repeal or revise the current laws and regulations concerning electronic trade, promote the use of the reliable trade leads sites, establish global certificate authority, electronic trade network, and electronic payment systems, and develop training expert programing related to Electronic Trade.
