        Functional genomics by integrated analysis of transcriptome of sweet potato (Ipomoea batatas (L.) Lam.) during root formation

        Background Sweet potato is easily propagated by cuttings. But the molecular biological mechanism of adventitious root formation are not yet clear. Objective To understand the molecular mechanisms of adventitious root formation from stem cuttings in sweet potato. Methods RNA-seq analysis was performed using un-rooted stem (0 day) and rooted stem (3 days). Gene Ontology (GO) enrichment analysis, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway, comparison with Arabidopsis transcription factors (TFs) of DEGs were conducted to investigate the characteristics of genes and TFs involved in root formation. In addition, qRT-PCR analysis using roots at 0, 3, 6, 9, and 12 days after planting was performed to confirm RNA-seq reliability and related genes expression. Results 42,459 representative transcripts and 2092 DEGs were obtained through the RNA-seq analysis. The DEGs indicated the GO terms related to the single-organism metabolic process and cell periphery, and involved in the biosynthesis of secondary metabolites, and phenylpropanoid biosynthesis in KEGG pathways. The comparison with Arabidopsis thaliana TF database showed that 3 TFs (WRKY, NAC, bHLH) involved in root formation of sweet potato. qRT-PCR analysis, which was conducted to confirm the reliability of RNA-seq analysis, indicated that some metabolisms including oxidative stress and wounding, transport, hormone may be involved in adventitious root formation. Conclusions The detected genes related to secondary metabolism, some hormone (auxin, gibberellin), transports, etc. and 3 TFs (WRKY, NAC, bHLH) may have functions in adventitious roots formation. This results provide valuable resources for future research on the adventitious root formation of sweet potato.

      • Drying of a Charge-Stabilized Colloidal Suspension in Situ Monitored by <i>Vertical</i> Small-Angle X-ray Scattering

        <P>We report a first application of <I>vertical</I> small-angle X-ray scattering to investigate the drying process of a colloidal suspension by overcoming gravity related restrictions. From the observation of the drying behavior of charge-stabilized colloidal silica in situ, we find the solidification of the colloidal particles exhibits an initial ordering, followed by a sudden aggregation when they overcome an electrostatic energy barrier. The aggregation can be driven not only by capillary pressure but also by thermal motion of the particles. The dominating contribution is determined by the magnitude of the energy barrier at the transition, which significantly decreases during drying due to an increased ionic strength.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/langd5/2013/langd5.2013.29.issue-32/la401897n/production/images/medium/la-2013-01897n_0009.gif'></P>

      • Structural Development of Nanoparticle Dispersion during Drying in Polymer Nanocomposite Films

        <P>Dispersions of nanoparticles (NPs) in a polymer matrix are a key element to set various properties of solution-cast polymer nanocomposite films. While the dispersion state of NPs in nanocomposite films has been extensively studied over the past decades, the structural development during drying and the relation of liquid and solid structure still remains poorly understood. In the present work, we study how NP dispersions develop during drying in polymer nanocomposite films, particularly focusing on the role of particle polymer interaction in the structural development. Aqueous dispersions of nanosilica and poly(vinyl alcohol) are used as model NP/polymer mixtures, where the particle polymer interaction can systemically be varied via the pH. A novel vertical small-angle X-ray scattering technique enables us to directly observe the development of the NP's dispersion state during drying. At a high pH 10, where silica particles have a poor affinity to PVA, SAXS intensity evolution shows phase separation during drying, resulting in the formation of dense aggregates of bare particles in the solid film. On the other hand, at a low pH 3, where silica particles have a good affinity with PVA, the SAXS data indicate a gradual densification of the NPs while maintaining a constant interparticle distance, which is originating from adsorbed PVA. The resulting solid film after drying exhibits an improved dispersion of NPs. The evaluation of the interparticle interaction suggests that the adsorbed polymer plays generally a key role in the uniform distribution of NPs in solid films, as it sterically stabilizes NPs over short ranges during all drying stages whereas depletion attraction dominates at longer ranges.</P>

      • A generality in stress development of silica/poly(vinyl alcohol) mixtures during drying process

        <P><B>Abstract</B></P> <P>Colloidal dispersions in polymer solution are important material systems in coating and printing applications. Understanding the role of polymers in drying process is important to obtain the desired performance of the solid film. In this study, we investigated the role of polymers in stress development during drying of aqueous silica and poly(vinyl alcohol) (PVA) suspensions, and the microstructure of solid film. Adsorbed polymers were found to affect the spatial organization of silica particles, which reduces the size of aggregates and the increase in stress development. The effect of adsorbed polymers on the stress development was hindered as the amount of non-adsorbed polymer increased. The role of adsorbed and non-adsorbed PVA in stress development was found to be generalized to yield a single master curve independent of polymer concentration.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We investigate the role of polymers in stress development of drying silica/PVA mixtures. </LI> <LI> Adsorbed PVA reduces the size of aggregates, thus increases the stress in the solid film. </LI> <LI> The effect of adsorbed PVA is hindered as non-adsorbed PVA increases. </LI> <LI> The role of PVA in stress development is generalized to yield a single master curve. </LI> </UL> </P>

        Depletion Stabilization in Nanoparticle–Polymer Suspensions: Multi-Length-Scale Analysis of Microstructure

        <P>We study the mechanism of depletion stabilization and the resultant microstructure of aqueous suspensions of nanosized silica and poly(vinyl alcohol) (PVA). Rheology, small-angle light scattering (SALS), and small-angle X-ray scattering (SAXS) techniques enable us to analyze the microstructure at broad length scale from single particle size to the size of a cluster of aggregated particles. As PVA concentration increases, the microstructure evolves from bridging flocculation, steric stabilization, depletion flocculation to depletion stabilization. To our surprise, when depletion stabilization occurs, the suspension shows the stabilization at the cluster length scale, while maintaining fractal aggregates at the particle length scale. This sharply contrasts previously reported studies on the depletion stabilization of microsized particle and polymer suspensions, which exhibits the stabilization at the particle length scale. On the basis of the evaluation of depletion interaction, we propose that the depletion energy barrier exists between clusters rather than particles due to the comparable size of silica particle and the radius gyration of PVA.</P>

        뉴미디어와 역사의 상품화 : <튜더스>의 사례

        This article is an attempt to examine the way The Tudors, a recent hit multinational TV mini-series, exploits and eventually reduces history to nothing but a pretext to project the fantasies and desires of the contemporary mass audience. The mechanism involves a few important trends in the current globalized mass culture: the post-modern condiditon called 'the end of temporality,' as closely related to generic traits and conventions of TV narratives, and the technical innovations creating various new venues in the name of 'new media'. In discussing TV narratives, it is important to note that they are essentially commercial products and that the material conditions of production such as funding, distribution and program formats heavily influence the narrative content itself. The Tudors adopted a marketing strategy different from the usual network TV shows; it was first premiered on a membership-based cable channel and then got distributed through new venues such as the Internet and IPTV, while reaping considerable revenue from DVD sales. Its funding and production also involved multinational capital and talents. By means of the new media, The Tudors manages to get around the censorship that defines the network TV programs, notoriously generous on violence but very strict on sexual content. Under the circumstances, it is noteworthy that The Tudors marked nostalgia and pornography as its major selling points to transend all localities and nationalities. The pornographic visual of The Tudors eventually reduces every complex thematic struggle of the early modern England into one general erotic power play. The Tudors represents history as a parallel universe devoid of time and change, charged only with replete sex drive, projecting the 21th century obsession of eternal youth and beauty. History in The Tudors is a spatialized matrix of the present, myth rather that history, all the more problematic for its naked fictionality.

        정보화 사회가 가속화됨에 따라 교육은 학교 교육 중심, 공급자 중심에서 평생교육 중심, 수요자 중심으로 바뀌고 있다. 또한, 평균 수명이 연장됨에 따라 생애에 걸친 인적 자원 개발, 권리라는 평생교육 개념에 새로운 방향을 요구하고 있다. 따라서 앞으로의 원격교육은 다양한 기능과 품질을 갖추고 어떠한 교수자나 학습자라도 쉽게 활용함으로써 가상학습 환경에서의 교수-학습을 지원할 수 있는 그러한 학습지원시스템의 출현이 필요하다.학점인정제도의 미흡, 뚜렷한 원격교육 추진목표의 부재, 체제적 원격교육의 부재, 경제적인 차이로 인한 계층간의 갈등 가속화, 학습자 관리의 문제, 학습 정보 관련 문제 등은 현 원격교육이 갖고 있는 고질적인 문제점들이다. 원격교육의 지속적인 발전을 위해서 기술적, 지적, 인적 요소의 균형 있는 개발과 육성에 힘써야 하며 이와 함께 행정적, 법적 제도화를 통한 지원이 수반되어야 할 것이다. 본 논문에서는 현재 윈격교육의 실태를 파악하고 여러 가지 측면에서 기존의 문제점을 개선할 수 있는 방안을 제시함으로써 더 나은 교육 환경을 통해 평생교육의 활성화를 촉진한다.
