      • Tomato germplasm with resistance to multiple species of Xanthomonas causing bacterial spot

        Sung-Chur Sim,David M. Francis 한국육종학회 2015 한국육종학회 심포지엄 Vol.2015 No.07

Bacterial spot of tomato is a disease complex caused by at least four species of Xanthomonas and leads to severe yield and quality losses in humid growing conditions in the world. Five physiological species (T1-T5) have been defined by their virulence on tomato varieties. These races are associated with three species: X. euvesicatoria (T1), X. vesicatoria (T2), and X. perforans (T3-T5). Recent epidemics of X. gardneri has occurred in the Midwest United States. In this study, we developed germplasm with resistance to multiple species of bacterial spot. Six advanced breeding lines with at least three different source of resistance were crossed and their F1 hybrids were inter-mated to produce a complex breeding population consisting over 1,100 progeny. Three lines (OH7663, OH7667 and 2k7-6117-S2) were selected by field evaluations of the population against T1, T2, T3, and X. gardneri. Graphical genotypes demonstrated that these breeding lines contain a QTL and Rx-4/Xv3 in coupling phase on chromosome 11 as well as Rx-3 on chromosome 5. In order to test the combining ability of the lines, we developed hybrids from multiple crosses and conducted replicated field trials to evaluate bacterial spot resistance and yield. As a male parent, OH7663 showed acceptable combining ability for yield and for resistance against multiple species of Xanthomonas. Several hybrids produced yields that were not significantly different from yields of commercial varieties.

      • Association mapping for novel resistance loci against bacterial spot using genome-wide SNPs in tomato

        Sung-Chur Sim,David M. Francis 한국육종학회 2014 한국육종학회 심포지엄 Vol.2014 No.07

Bacterial spot of tomato (Solanum lycopersicum L.) is caused by at least four species of Xanthomonas with multiple physiological races. In this study, we developed a mapping population for association analysis of bacterial spot resistance. For this population, six advanced breeding lines with distinct sources of resistance were first crossed in all combinations and their F1 hybrids were intercrossed. The 1,100 segregating progeny from these crosses were evaluated in the field against T1 strains. Based on this individual evaluation, we selected 5% of the most resistant and 5% of the most susceptible progeny for evaluation as plots in two subsequent replicate field trials inoculated with T1 and T3 strains. A total of 461 markers across 12 chromosomes were used for genotyping these selections. Of these markers, an optimized subset of 384 SNPs was derived from the 7,720 SNP Infinium array developed by the Solanaceae Coordinated Agricultural Project (SolCAP). For association analysis to detect known resistance loci and additional novel loci, we used the mixed models with correction for population structure, and found that accounting for kinship appeared to be sufficient. Detection of known loci was not improved by adding a correction for structure using either a Q matrix from model-based clustering or covariate matrix from Principal Component Analysis. Both single-point and haplotype analyses identified strong associations in the region of the genome known to carry Rx-3 (chromosome 5) and Rx-4/Xv3 (chromosome 11). Additional QTL associated with resistance were detected on chromosomes 1, 3, 4, 6 and 7 for T1 resistance and chromosomes 2, 4, and 6 for T3 resistance. Haplotype analysis improved our ability to trace the origin of positive alleles. These results demonstrate that both known and novel associations can be identified using complex breeding populations that have experienced directional selection.

Subgroup analysis of a phase 2/3 study of rurioctocog alfa pegol in patients with severe hemophilia A: efficacy and safety in previously treated Korean patients

        Chur Woo You,백희조,Sang Kyu Park,박영실,Ho-Jin Shin,Werner Engl,Srilatha Tangada 대한혈액학회 2019 Blood Research Vol.54 No.3

BackgroundThe efficacy and safety of extended half-life, full-length, pegylated recombinant factor VIII rurioctocog alfa pegol [BAX 855, ADYNOVATE (USA)/ADYNOVI (Europe); Baxalta US Inc., a Takeda company, Lexington, MA, USA] was investigated in previously treated Korean patients with severe hemophilia A (HA).MethodsA post hoc data analysis from the international, multicenter, phase 2/3 PROLONG-ATE study of rurioctocog alfa pegol in patients with severe HA (NCT01736475) determined annualized bleeding rates (ABRs) and rates of adverse events (AEs) in Korean patients treated in this study.ResultsAll 10 enrolled Korean patients receiving rurioctocog alfa pegol (9 prophylaxis, 1 on-de-mand) completed the study [median (range) age, 28.0 (12‒50) yr; weight, 64.8 (45‒90) kg; 8 patients had ≥1 target joint at screening]. Median (range) ABR was 1.9 (0.0‒14.5) for patients on prophylaxis and 62.2 for the patient receiving on-demand treatment. The hemostatic efficacy of rurioctocog alfa pegol was rated “excellent” or “good” and only single infusions were required per bleeding episode. ABRs improved in most patients compared with prestudy values. No dose adjustments were required for prophylaxis, and the dosing frequency was reduced in 8 patients, compared with their previous prophy-laxis regimen. No serious AEs were reported; all 9 nonserious AEs (in 3 patients) were mild in severity and unrelated to the study treatment.ConclusionThis post hoc analysis of a small group of Korean patients with severe HA indicated that rurioctocog alfa pegol was effective, and no serious AEs were observed. For most patients, the dosing frequency was also reduced compared with their previous regimen.

Comparison of Surgically Induced Astigmatisms after Clear Corneal Incisions of Different Sizes

( Sung Chur Moon ),( Tarek Mohamed ),( I Howard Fine ) 대한안과학회 2007 Korean Journal of Ophthalmology Vol.21 No.1

Purpose: This study was performed to assess efficiency and stability of astigmatic change by incision size after cataract surgery. Methods: This work was designed as a retrospective, comparative, nonrandomized interventional study. A total of 121 cases of cataract surgery were reviewed in 98 patients performed by one surgeon at the Oregon Eye Institute in Eugene, OR, USA with 3-year follow-ups. All procedures were performed with the temporal approach of self-sealing incisions. The serial change in surgically induced astigmatisms were examined in all cases of three groups: Group A, cartridge injection of a foldable IOL through a 2.5 mm self-sealing incision; Group B, cartridge injection of a foldable IOL through a 3.0 mm self-sealing incision; Group C, cartridge injection of a foldable IOL through a 3.5 mm self-sealing incision. Keratometric data were obtained preoperatively, and 3 weeks, 3 months, 6 months, 9 months, 12 months, 24 months and 36 months postoperatively. Polar value analysis was performed to calculate the surgically induced astigmatism. Results: The astigmatic change decreased over time in Group B (P<0.05). The other groups tended to remain in induced astigmatism. All groups showed anticlockwise torque at 3 weeks following surgery. Group B showed a decrease in deviation, but the other groups showed increases in their torque value at postoperative 12 months (P<0.05). Conclusions: The 3.0 mm incision size correlated with the least surgically induced astigmatism.
