        Developmental changes in GABAA tonic inhibition are compromised by multiple mechanisms in preadolescent dentate gyrus granule cells

        The sustained tonic currents (Itonic) generated by γ-aminobutyric acid A receptors (GABAARs) are implicated in diverse age-dependent brain functions. While various mechanisms regulating Itonic in the hippocampus are known, their combined role in Itonic regulation is not well understood in different age groups. In this study, we demonstrated that a developmental increase in GABA transporter (GAT) expression, combined with gradual decrease in GABAAR a5 subunit, resulted in various Itonic in the dentate gyrus granule cells (DGGCs) of preadolescent rats. Both GAT-1 and GAT- 3 expression gradually increased at infantile (P6-8 and P13-15) and juvenile (P20-22 and P27-29) stages, with stabilization observed thereafter in adolescents (P34-36) and young adults (P41-43). Itonic facilitation of a selective GAT-1 blocker (NO-711) was significantly less at P6-8 than after P13-15. The facilitation of Itonic by SNAP-5114, a GAT-3 inhibitor, was negligible in the absence of exogenous GABA at all tested ages. In contrast, Itonic in the presence of a nonselective GAT blocker (nipecotic acid, NPA) gradually decreased with age during the preadolescent period, which was mimicked by Itonic changes in the presence of exogenous GABA. Itonic sensitivity to L-655,708, a GABAAR a5 subunit inverse agonist, gradually decreased during the preadolescent period in the presence of NPA or exogenous GABA. Finally, Western blot analysis showed that the expression of the GABAAR a5 subunit in the dentate gyrus gradually decreased with age. Collectively, our results suggested that the Itonic regulation of altered GATs is under the final tune of GABAAR a5 subunit activation in DGGCs at different ages.

        On New Centroid Based Techniques for Solving Fuzzy Linear Programming Problems

        In this paper we have proposed some new techniques based on the centroid of triangular and trapezoidal fuzzy numbers to solve a fuzzy linear programming problem. These proposed methods are easy to implement with less number of arithmetic operations required compared to the existing methods. To illustrate the proposed method, numerical examples are given. A comparative study between the proposed methods and existing methods are given at the end. The superiority of these centroid based methods over known ranking function method has also been shown with an example.

        A comparative study on free vibration analysis of delaminated torsion stiff and bending stiff composite shells

        This paper presents a finite element method to compare the effects of delamination on free vibration of graphite-epoxy bending stiff and torsion stiff composite pretwisted shallow conical shells. The generalized dynamic equilibrium equation is derived from Lagrange’s equation of motion neglecting the Coriolis effect for moderate rotational speeds. An eight noded isoparametric plate bending element is employed incorporating rotary inertia and effects of transverse shear deformation based on Mindlin’s theory. The multipoint constraint algorithm is utilized to ensure the compatibility of deformation and equilibrium of resultant forces and moments at the delamination crack front. The standard eigen value problem is solved by applying the QR iteration algorithm. Mode shapes for typical configurations are also depicted. Numerical results obtained are the first known non-dimensional frequencies which could serve as reference solutions for the future investigators.

        Endoglucanase Produced by Bacillus subtilis Strain CBS31: Biochemical Characterization, Thermodynamic Study, Enzymatic Hydrolysis, and Bio-industrial Applications

        Microbial cellulases have become the mainstream biocatalysts due to their complex nature and widespread industrial applications. Here, homogeneous endoglucanase GluCB31 from Bacillus subtilis subsp. inaquosorum CBS31 was studied. GluCB31 was purified to 17.68-fold with an 8.33% yield and a specific activity of 1066.37 U/mg. Biochemical properties of GluCB31 were performed and the results are as follows; molecular mass of 35 kDa with an optimum pH at 7.5 and temperature at 50oC. GluCB31 was immobilized in calcium alginate gel and it exhibited the highest activity at 10oC higher temperature than soluble enzyme, as the entrapment in alginate gel made GluCB31 more stable. Kinetic studies showed the Vmax of 1293.33 ± 2.51 U/mg and Km of 0.0183 mg/mL. Enzymatic activity was activated by Tween-20 (106.7%), Tween-80 (111.6%), Triton X-100 (142.3%), SDS (135.5%), Mg++ (185.7%), Cu++ (167.6%), Zn++ (153.7%), Mn++ (106.3%), Ba++ (181.9%), Ni++ (107.2%) while inhibited by Fe++ (15.8%), β-mercaptoethanol (46.8%), EDTA (54.5%). Enthalpy, free energy, and entropy of activation were calculated to be 38.526 kJmol-1, 44.187 kJmol-1, and -17.518 Jmol-1K-1 respectively. Also, ΔGE-S and ΔGE-T were found to be -10.75 kJmol-1 and -45.92 kJmol-1 respectively. A low ΔS, ΔGE-S, and ΔGE-T values were signified enzyme-catalyzed reaction occurs at a fast rate and the existence of the enzyme in its stable state. Cellobiose was the major end product of hydrolysis. These attributes of GluCB31 demonstrated the diversity of catalytic activities and serve in various biotechnological processes, thus deserve to be developed as a bio-industrial agent.

      • Low temperature wet-chemical synthesis of spherical hydroxyapatite nanoparticles and their in situ cytotoxicity study

        The present research work reports a low temperature ($40^{\circ}C$) chemical precipitation technique for synthesizing hydroxyapatite (HAp) nanoparticles of spherical morphology through a simple reaction of calcium nitrate tetrahydrate and di-ammonium hydrogen phosphate at pH 11. The crystallinity of the single-phase nanoparticles could be improved by calcinating at $600^{\circ}C$ in air. Thermogravimetric and differential thermal analysis (TG-DTA) revealed the synthesized HAp is stable up to $1200^{\circ}C$. Scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) studies confirmed the formation of spherical nanoparticles with average size of $23.15{\pm}2.56nm$ and Ca/P ratio of 1.70. Brunauer-Emmett-Teller (BET) isotherm of the nanoparticles revealed their porous structure with average pore size of about 24.47 nm and average surface area of $78.4m2g^{-1}$. Fourier transform infrared spectroscopy (FTIR) was used to confirm the formation of P-O, OH, C-O chemical bonds. Cytotoxicity and MTT assay on MG63 osteogenic cell lines revealed nontoxic bioactive nature of the synthesized HAp nanoparticles.

      • Comparative characterization of biogenic and chemical synthesized hydroxyapatite biomaterials for potential biomedical application

        <P><B>Abstract</B></P> <P>This present study illustrates the processing and comprehensive characterization of biogenic hydroxyapatite (FB-HAp) biomaterial from fish bone (Tuna fish) in a facile single step thermal decomposition procedure. Fish bones were calcined at 1000 °C in an air atmosphere for 1 h. The obtained FB-HAp flakes were ball milled for 30 h at a unidirectional milling speed of 300 rpm. A well-established wet chemical process was obtained to synthesize HAp nanoparticles (C-HAp). The synthesized non-stoichiometric (Ca/P = 1.87) FB-HAp phases were identified by X-ray diffraction study and compared with C-HAp. The thermal properties were studied by thermogravimetric and differential thermal analysis. Raman spectroscopy and Fourier transform infrared spectroscopy confirmed the structural and functional groups of FB-HAp and C-HAp. The morphological and elemental analysis was performed by transmission electron microscopy, energy dispersive X-ray spectroscopy and Inductive coupled plasma (ICP) spectroscopy. The synthesized FB-HAp and C-HAp were mixed with 2% starch porogen and compacted with ∼200 MPa unidirectional pressure to obtained small pellet scaffolds. The obtained scaffolds were sintered at 1000 °C in an air atmosphere for 2 h and further used for cell attachment and proliferation study. The FB-HAp biomaterials showed non-toxic behavior with MG-63 osteoblast cell lines similar to C-HAp. Additionally, FB-HAp scaffold showed enhanced cellular attachment and proliferation on its surface. The in-depth chemical, structural, and morphological analysis suggest that fish bone could be a promising sustainable biogenic source of HAp for biomedical applications.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Facile synthesis of biogenic hydroxyapatite from waste tuna bones. </LI> <LI> Comparative characterization of fish bone and chemically synthesized hydroxyapatite. </LI> <LI> Possible mechanism for enhanced bioactivity of biogenic hydroxyapatite. </LI> <LI> Scaffold fabrication and promising application for tissue engineering. </LI> </UL> </P>
