      • 콩에서 발생하는 강낭콩일반모자이크바이러스(BCMV)와 땅콩위축바이러스(PSV)의 저항성 평가

        It has been reported several viral diseases in soybean. The occurrence of Bean common mosaic virus and Peanut stunt virus are able to induce severe problem because the main breeding target for viral disease was focused on Soybean mosaic virus. BCMV is a spcies of the genus Potyvirus of the family Potyviridae. BCMV reduces yield by as much as 80% and also adversely affects seed quality in common bean. BCMV has a worldwide distribution because of its high rates 35% of transmission via seeds produced by plants systemically infected prior to bloom. PSV is a species of the genus Cucumovirus of the family Bromoviridae. PSV has been reported to be economically damaging on beans worldwide. Although the virus is reportedly seed transmissible at very low rates, infected plants rarely produce pods with normal viable seeds. The most satisfactory management method of viral diseases such as BCMV and PSV is the use of resistant cultivars and healthy seed. For the overcome of genetic resistance by new virus such as BCMV and PSV, virus diseases monitoring and researches need to study deeply and continually. In this study, we conducted the resistance screen on BCMV and PSV for selection of resistance cultivars. We inoculated on 128 cultivars with PSV and BCMV respectively. These cultivars were classified through pathogenic response such as resistance, necrosis and susceptibility. In addition, 99 and 66 resistant cultivars were confirmed against PSV and BCMV respectively.

      • RT-LAMP를 이용한 콩황화일반모자이크바이러스의 진단

        Soybean yellow common mosaic virus (SYCMV) has been recently reported, it has been occurred a lot with Soybean mosaic virus (SMV) and Soybean yellow mottle mosaic virus (SYMMV) in soybean field. SYCMV belongs to genus of Sobemovirus and induced viral symptoms with yellowing, mottle and mosaic. A reverse transcription loop-mediated isothermal amplification (RT-LAMP) method allowed one-step detection of gene amplification by simple procedure and needed only a simple incubator for isothermal template. This RT-LAMP method allowed direct detection of RNA from virus-infected plants without thermal cycling and gel electrophoresis. In this study, we designed RT-LAMP primers named SYCML-F3/B3/FIP/BIP from coat protein gene sequence of SYCMV. After the reaction of RTLAMP, products were identified by electrophoresis and with the detective fluorescent dye, SYBR Green I. under daylight and UV light. Optimal reaction condition was at 63 for 60min and the primers of RTLAMP showed the specificity for only SYCMV tested in this study.

        특수건강진단 자료를 이용한 소음성난청 판정기준의 비교

        Although noise-induced hearing loss (NIHL) is the most prevalent occupational disease in Korea, only 10% of the diagnosed cases are compensated. Old (1989-94) and current diagnostic criteria, criteria for workers' compensation of NIHL in Korea, compensation formulas of American Medical Association/American association of Ophthalmology and Otolaryngology (AMA/AAOO), the Committee on Hearing, Bio-Acoustics, Biomechanics (CHABA), American Academy of Otolaryngology (AAO) recommendation were compared. Each criterion was applied on the audiomety data of 4044 workers (8023 ears), who had received the second line screening test of Special Periodic Health Examination Program for noise-exposed workers during 1991-2. First, the resulting proportions of NIHL cases by employing each criterion were compared and strength of agreement was measured using kappa value. Temporary Threshold Shift (TTS) was corrected by noise free interval, and the reduction ratio of NIHL cases was calculated. Theoretical progression model of NIHL was reconstructed from previous studies on the natural course of NIHL to evaluate the change of diagnosis result of each criterion in the model. The kappa value between old and current criteria was 0.19, 0.55 for current criteria and workers' com. criteria, ranging from 0.08 to 0.78, highest coincidence was observed between current criteria and CHABA formula. The current criteria produce most similar results with CHABA formula. If TTS is corrected for NFI, there is about 14% reduction of NIHL cases. The results of applying on NIHL progression model divided the formulas roughly into 3 groups, of which compensation criteria was the most, old criteria and AAO the least conservative. In conclusion, the result of 4 ㎑ audiometry should be excluded in evaluation of hearing level and a new hearing conservation program should be set out. Current diagnostic criteria has an ambiguity in that managerial concept for prevention and purpose of compensation is mixed up. The current diagnostic criteria and compensation criteria could be incorporated into a new formula which is based on the state of the art test for estimating everyday hearing disability.

        VDT작업자에서 업무로 인한 정신사회적 스트레스에 대한 인지가 근골격계장애에 미치는 영향

        업무에 의한 정신사회적 스트레스가 근골격계 질환의 유병에 미치는 영향을 파악하기 위하여 전화안내원을 대상으로 단면적 조사연구를 수행하였다. 연구 결과 나이, 교육정도, 근무연한, 가사부담 등의 인구학적 변수는 근골격계질환의 유병에 영향을 미치지 않는 것으로 분석되었다. 정신사회적 스트레스 중 업무요구도가 높다고 느끼는 집단에서 그렇지 않은 집단에 비해 근골격계 질환의 유병률이 통계적으로 유의하게 높게 나타났으나 업무재량도나 사회적지지도는 근골격계질환과 유의한 상관관계를 발견할 수 없었다. 그러나 업무요구도는 높고 업무재량도는 낮다고 느끼는 '근무긴장도가 가장 높은 군'은 업무요구도는 낮고 업무재량도는 높다고 느끼는 '근무긴장도가 낮은 군'에 비해 근골격계 질환의 발생위험이 통계적으로 유의하게 높은 것으로 나타났다. 본 연구의 결과는 업무로 인한 정신사회적 스트레스에 대한 인지 정도에 따라 근골격계질환의 유병률이 달라짐을 시사하고 있다. 따라서 작업관련성 근골격계질환의 예방을 위해서는 기존의 인간공학적 접근 이외에 스트레스를 감소시키려는 노력이 동시에 진행되어야 할 것으로 생각한다. A cross-sectional study was conducted to assess the association between perceived occupational psychosocial stress at work and the work-related musculoskeletal disorder(WRMSD) among employees using video display terminals. The study included 111 female telecommunication employees from three companies at ChungBuk province. Cases of WRMSD were defined using symptom questionnaire and physical examination conducted by rehabilitation specialist. Information on demographics, individual factors, and perceived psychosocial stress, were obtained by self-administered questionnaire. For assessing perceived psychosocial stress, we used variables from 'job strain model' proposed by Karasek. Associations between perceived psychosocial stress and WRMSD were assessed by multiple logistic regression models. Forty nine(44% to the total) subjects met our operational definition for WRMSD. Age, seniority, housing load were not associated with WRMSD. Perceived psychosocial stress was associated with WRMSD[odds ratio=3.28, 95% confidence interval: 1.05-10.19]. This study suggests that perceived occupational psychosocial stress is related to the prevalence of WRMSD.

        혈압 조절 후 신기능의 호전을 보였던 중증 고혈압 환자들의 임상 특징 분석

        배 경 : 본태성 고혈압 환자의 10-40%에서 경도의 신기능 저하를 동반하는 것으로 보고되어 있으며, 고혈압 환자에서 신부전이 동반된 경우 심혈관 사망률이 높은 것으로 알려져 있다. 본 연구에서는 내원 당시 중증 고혈압과 동반된 신부전 소견을 보였으나, 적극적인 혈압 조절 후 신기능의 호진을 보인 환자들의 임상 특징을 분석해 보고자 하였다. 방 법 : 1993년 1월부터 2002년 12월까지 10년간 가톨릭의대 강남성모병원 및 성바오로병원에서 중증 고혈압 및 신부전증 (혈청 크레아티닌치 2.0 ㎎/dL 이상)으로 진단된 환자들 중에서 혈압 조절 후 신기능의 호전 (혈청 크레아티닌의 감소)을 보인 환자들을 대상으로 이들의 평균 동맥압 및 혈청 생화학 검사치 변화를 분석하고, 신생검 소견 및 투어 혈압 약제를 분석하였다. 결 과 : 전체 14명의 환자 중 남자는 8명 여자는 6명이었고, 이들의 평균 연령은 38±8세 였다. 평균 동맥압의 유의한 감소와 더불어 혈청 크레아티닌치의 감소 (p=0.001)와 헤마토크리트치의 유의한 증가 (p=0.001) 소견이 관찰되었다. 그러나 혈청 요산, 총콜레스테롤, 중성지방 농도의 유의한 변화는 없었다. 신초음파 검사상 정상 신장 크기를 보인 경우는 12명 (80%)이었고, 대상 환자군은 대부분 3가지 이상의 항고혈압제를 병합 투여하여 적극적인 혈압 조절을 하였으며, 전환효소차단제 및 안지오텐신 Ⅱ 수용체 길항제 (22%), 칼슘통로차단제 (21%), 베타차단제 (21%)를 주로 투여하였다. 신생검을 시행한 6예를 분석한 결과, 4예는 고혈압성 신경화증, 2예는 IgA 신증으로 진단되었다. 6예 모두 신세동맥의 초자질 침착과 혈관내피의 점액양 변성을 동반한 신세동맥의 비후 소견이 존재하였고, 경도에서 중등도의 세뇨관 위축 소견과 간질성 섬유화 소견이 관찰되었다. 결 론 : 신부전을 동반한 중증 고혈압 환자들 중, 신장 크기가 정상이고 나이가 젊은 환자의 경우, 혈압 조절을 통해 신기능을 호전시킬 수 있으므로, 이들에 대한 적극적인 혈압 조절이 필수적이라 하겠다. Background : Mild renal dysfunction is relatively common in patients with long standing primary hypertension, ranging from 10% to 40% in various studies. The presence of renal dysfunction is associated with high cardiovascular mortality and morbidity rates of patients with primary hypertension. The purpose of this study is to analyze the clinical characteristics of patients with severe hypertensin and reversible renal dysfunction after blood pressure control. Methods : This retrospective study enrolled 14 patients with severe hypertension and reversible renal dysfunction after blood pressure control, between January 1993 and December 2002 at Kangnam St. Mary`s Hospital and St. Paul`s Hospital. We investigated the laboratory data using Wilcoxon signed rank test, and analysed renal biopsy findings and antihypertensive drugs. Results : The mean age of the patients was 38±9 years and the number of male patients was 8. During 33.5±28.8 months of mean follow-up period, there was a significant decrease in mean arterial pressure and serum creatinine level, and significnat increase in hematocrit level. But there was no significant changes in the level of uric acid, total cholesterol, and triglyceride. Each patient took more than 3 antihypertensive drugs consisting angiotensin converting enzyme inhibitor or angiotensin Ⅱ recep색 blocker (22%), calcium channel blocker (21%), and beta blocker (21%). Renal biopsy was done in 6 cases, and histologic diagnosis resulted in 4 cases of benign hypertensive nephrosclerosis and 2 cases of IgA nephropathy. The typical morphological features of hypertensive nephrosclerosis were seen in all cases, and there were varying degrees of glomerular sclerosis from 0% to 92%. But the percent of glomerular sclerosis was not related to the level of initial serum creatinine, mean arterial pressure, and amount of proteinuria per day. Conclusion : Careful monitoring of renal function and effective treatment of blood pressure are therefore mandatory in treating young patient with severe gypertension with renal dysfunction.       • Introduction of UGT72E3/2 gene into soybean increases higher content of phenylpropanoids

        UGT72E3/2 gene encodes UDP-glycosyltransferase shown to glucosylate several phenylpropanoids such as syringin and coniferin. Syringin has effect of anti-stress and anti-fatigue. Korean soybean variety Kwangan was transformed with UGT72E3/2 gene. This gene was transformed into Kwangan using highly efficient soybean transformation system. This study used two promoters, beta-conglycinin promoter for seed-specific expression and 35s promoter for total expression. Transgenic plants were confirmed for gene introduction and their expression using PCR and RT-PCR. The analysis of syringin in transgenic plants was performed using HPLC. Currently, the confirmation of stable gene introduction with UGT72E3/2 gene is also performing by Southern blot analysis.

        Insect resistant genes encode insecticidal δ-endotoxins that are widely used for the development of insect-resistant crops. Common soybean is a crop of economic and nutritious importance in many parts of the world. Korean soybean variety Kwangan was transformed with Insect resistant genes. These genes were transformed into Kwangan using highly efficient soybean transformation system. Transgenic plants harboring Insect resistant genes were confirmed for gene introduction and their expression using PCR, real-time PCR and RT-PCR. The confirmation of stable gene introduction with Insect resistant genes was also performing by Southern blot analysis. In addition, Flanking sequence analysis and agronomic characters were also investigated
