        Synthesis of Multifunctional Organic Nanoparticles Combining Photodynamic Therapy and Chemotherapeutic Drug Release

        Cancer is a group of diseases that are caused by uncontrolled proliferation of cells in various parts of the body and it is one of the most studied diseases worldwide. Photodynamic therapy (PDT) is a treatment that uses photosensitizers called photosensitizing agents in addition to light to kill cancer cells. Photosensitizers work only after they have been activated by certain types of light. PDT contains three basic components, these are a photosensitizer, visible light and molecular oxygen (3O2). The efficiency of the therapeutic action is directly related to the property of the photosensitizer. In this study, chitosan and BODIPY based nanoparticles that were capable of carrying out drug delivery and producing singlet oxygen (1O2) were synthesized for the first time. For this purpose, organic nanoparticles showed PDT feature were synthesized via the formation of ionic complexes formed with opposite charged ionic interactions between the synthesized BODIPY derivative (PDT agent) and chitosan hydrochloride at appropriate pH (pH=6). Later, during the formation of this ionic complex, a chemotherapeutic model drug (Doxorubicin) was added to the medium and chemotherapeutic drug-loaded chitosan and BODIPY-based nanoparticles were synthesized. Finally, while drug-free (Y1-Chitosan nanoparticles) and drug-loaded organic nanoparticles (Y1-Chitosan-Dox nanoparticles) showed very good PDT properties, they were found to be effective on MCF7 cancer cells and less toxic to L929 cells.

        Protective effects of silymarin on fumonisin B1-induced hepatotoxicity in mice

        The present study was conducted to investigate the effect ofsilymarin on experimental liver toxication induced byFumonisin B1 (FB1) in BALB/c mice. The mice were dividedinto six groups (n = 15). Group 1 served as the control. Group2 was the silymarin control (100 mg/kg by gavage). Groups 3and 4 were treated with FB1 (Group 3, 1.5 mg/kg FB1,intraperitoneally; and Group 4, 4.5 mg/kg FB1). Group 5received FB1 (1.5 mg/kg) and silymarin (100 mg/kg), andGroup 6 was given a higher dose of FB1 (4.5 mg/kg FB1) withsilymarin (100 mg/kg). Silymarin treatment significantlydecreased (p < 0.0001) the apoptotic rate. FB1 administrationsignificantly increased (p < 0.0001) proliferating cell nuclearantigen and Ki-67 expression. Furthermore, FB1 elevated thelevels of caspase-8 and tumor necrosis factor-alpha mediatorswhile silymarin significantly reduced (p < 0.0001) theexpression of these factors. Vascular endothelial growthfactor (VEGF) and fibroblast growth factor-2 (FGF-2)expressions were significantly elevated in Group 4 (p <0.0001). Silymarin administration alleviated increased VEGFand FGF-2 expression levels (p < 0.0001). In conclusion,silymarin ameliorated toxic liver damage caused by FB1 inBALB/c mice.

        Pomegranate Wine Has Greater Protection Capacity Than Red Wine on Low-Density Lipoprotein Oxidation

        Although there is a large body of evidence on the main role of red wine in protection of low-density lipopro-tein (LDL) against oxidation, there are few data on the role of pomegranate juice, which has high phenolic content. We con-ducted this study considering the possible importance of pomegranate wine as an antioxidant and in order to make a com-parison between red and pomegranate wines. The phenol levels of pomegranate and red wines (4,850 mg/L gallic acidequivalents and 815 mg/L gallic acid equivalents, respectively) were in accordance with their total antioxidant activity (39.5%and 33.7%, respectively). Both wines decreased LDL-diene levels following a 30-minute incubation period compared withcontrols (145. 3.2 .mol/mg of LDL protein). However, pure pomegranate wine demonstrated a greater antioxidant effect(P. .01) on diene level (110. 4.6 .mol/mg of LDL protein) than pure red wine (124. 3.2 .mol/mg of LDL protein). Inconclusion, we suggest that pomegranate wine has potential protective effects toward LDL oxidation, and it may be a dietarychoice for people who prefer fruit wines.

        Alpha basic crystallin expression in canine mammary tumors

        The aim of this study was to evaluate prognostic and/or diagnostic factors of canine mammary tumors by immunohistochemically analyzing the expression of alpha basic crystallin (aB-c). For this, formalin-fixed, paraffin-embedded blocks of 51 naturally-occurring canine mammary tumors (11 benign and 40 malignant) were used. Tissue from eight normal canine mammary glands were served as a control. Immunohistochemically, in the control mammary tissues, a few luminal epithelial cells were aB-c positive but myoepithelial cells were negative. In benign or simple type malignant tumors, aB-c expression was observed in luminal epithelial cells while the myoepithelial basal cells were negative. In benign or complex type malign tumors, positive staining was predominantly found in the cytoplasm of epithelial cells. Immunoreactivity of aB-c was also observed in neoplastic myoepithelial cells. Statistically, the number of cells immunolabeled with aB-c was found to be significantly different among tissues from normal canine mammary glands, benign lesions, and malignant tumors (p < 0.05). aB-c immunoreactivity was higher in malignant tumors than the control mammary tissues (p < 0.001). Data obtained in the current study revealed a strong association between high expression levels af aB-c and primary mammary gland tumors in canines.
