Pyrrolidine-2,3-dione derivative A was synthesized from oxalylation of chiral enamine Ba. The stereostructure of its major diastereomer (Amaj) was determined as octahydro-2,3-dioxo-6aS-hydroxy-1-(1S-methoxycarbonyl-2-phenylethyl)-cyclopenta[b]pyrrole-3aR-carboxylic acid ethyl ester Aa by means of NMR spectrum. This result implied that the asymmetric carbon-carbon bond forming reaction occurred preferentially at the beta-face of Ba.

In this study we investigated the effect of 3-methoxy-6-allylthiopyridazine on cell growth in BxPC3 and PANC1 human pancreatic cancer cells. The treatment with 3-methoxy-6-allylthiopyridazine for 48 h decreased cell viability and induced apoptotic cell death in a dose-dependent manner, assessed by using the MTT assay and the flow cytometry, respectively. These results suggest that 3-methoxy-6-allylthiopyridazine may be a good candidate for the therapeutic management of human pancreatic cancers.

In order to discover new useful NSAIDS, novel N-substituted 1,2-benzisothiazoline-3- one 1,1-dioxide derivatives, which can exhibit potentially antiinflammatory activity were synthesized.1,2-Benzisothiazoline-3-one-N-acetic acids 6a, b were obtained from monochloroacetic acid and sodium 1,2-benzisothiazoline-3-ones in DMF by N- al1kylation reaction. N-Substituted 1,2-benzisothiazoline-3-one 1,1-dioxice derivatives 7a-e were synthesized through the coupling of compound 6a, b and several amines (aniline, 2-aminopyridine, 2- aminothiazole, 2-aminotetrazole) with dicyclohexylcarbodiimide in methylene chloride.

Expression of xenobiotic-metabolizing enzymes can be altered by xenobiotics, which represents changes in the production of reactive metabolic intermediates as well as toxicities in tissues. Metabolic intermediates derived from xenobiotics are considered to produce the reactive oxygen species including drug free radicals and hydroxyl free radicals, which would be ultimately responsible for drug-induced toxicities. The effects of 1,2-benzothiazine anti-inflamniatory agents on the expression of xenobiotic-metabolizing enzymes including major cytochrome P450s, microsomal epoxide hydrolase (mEH) and glutathione S-transferase (GST) were studied in the liver with the aim of providing the part of information on potential production of reactive metabolites and hepatotoxicity by the agents. The synthetic compounds 24, 36 and 39 exhibited anti-inflammatory effects in rats as assessed by the Randall-Selitto method. The anti-inflammatory effect was detected as early as at 30 min after gavaging the agents with the ED50 being noted at 80mg/kg, which was comparable to that of ibuprofen. Treatment of rats with each compound (100mg/kg, 3d) resulted in no significant induction in the immunochemically-detectable cytochromes P450 1A1/2, P450 2B1/2, P450 2C11 and P450 2E1. Changes in the mEH expression were also minimal, as evidenced by both Western blot and Northern blot analyses. Hepatic GST expression was slightly increased by the agents: GST Ya protein and mRNA expression was ~1.5-fold increased after treatment with compounds 24 and 39, whereas GST Yb1/2 and Yc1/2 mRNA levels were elevated 2- to 3-fold. In summary, the effects of the synthetic 1,2-benzothiazines on the expression of major P450, mEH and GST were not significant, providing evidence that metabolic activation of the agents, potential drug interaction and hepatotoxicity would be minimal.

나노위성 HAUSAT-2 설계 검증의 일환으로 소형위성의 초기개념설계를 위한 시스템 엔지니어링 설계 도구를 개발하였다. 본 프로그램은 위성의 임무 및 사용자 요구조건을 바탕으로 탑재체와 각 서브시스템의 설계 과정을 거치면서 위성 전체 시스템의 질량 및 전력을 산출하고 설계된 시스템의 제작비용을 예측할 수 있다. 약 200여개의 소형위성 분석 데이터를 포함하고 있으며, 데이터베이스는 소형위성의 경향 분석을 제공할 뿐만 아니라 중요한 설계 요소로 쓰이고 있다. SEDT는 약 10개 이상의 소형위성 데이터를 경우에 따라 적용하면서 검증되었다. SEDT(System Engineering Design Tool) has been developed for small satellite conceptual design with an aim to verifying the nanosatellite HAUSAT-2 design. The program can calculate the mass and power of whole satellite system having specific mission and estimate the system cost based on mission and user requirements. It is containing various analysis data of more than 200 small satellites. The database will provide the trend analysis results of the small satellites which will become important design factors. This tool has also been verified by applying more than 10 small satellite data through case studies.

In order to investigate the content of total mercury in cattle, we measured the contents of total mercury in fur, hide and liver by mercury analyzer. The content of total mercury of fur in cattle was 59.10±7.97 ㎍/kg, and that of hide and liver was 2.46±0.33, 2.55±0.47 ㎍g/kg, respectively. The contents of total mercury of hide and liver in cattle were less than 20 ㎍/kg, but 49.1% of fur were over than 20 ㎍/kg, 32.7% 20-100 ㎍/kg, and 18.2% over than 100 ㎍/kg. There was higly significant between the content of mercury in fur and hide, but not between sex and age.

The cultural conditions in shake flasks were investigated under which maximum amounts of laccase produced by a strain of white-rot fungus Coriolus versicolor CV3. The enzyme yields on potato-malt extract medium by the fungus were higher than on other media consisted of onion infusion or malt extract, with maximum activity of 1.50 unit/㎖ culture or 119.5 unit/g mycelium at 11 days of incubation. Maximum yields of laccase and growth were obtained by supplementation of yeast extract or potassium nitrate to the potato-malt extract medium. Addition of 2.5-xylidine at 4×10 exp(-4) M concentration to the medium induced the lactase production 3.1-fold higher than the basal level, while the mycelial growth was somewhat repressed. The pH optimum. for the growth and lactase formation by the fungus was between pH 4 to 4.5.

Allylthio group of the sulfur compounds of garlic oil plays an important role for prevention and treatment of hepatic diseases induced by toxic substances or carbon tetrachloride. Thus allylthio group as pharmacologically active group was introduced into pyridazine heterocycle ring. Aryloxyallylthiopyridazine derivatives were synthesized and their hepatoprotective activities were screened in rat. The activities of these compounds were weaker than alkoxyallylthiopyridazine derivatives, which exhibit a superior hepatprotective effect.

α-Diethylaminoacetophenone을 50% alcohol 용액에서 potassium cyanide, ammonium carbonate와 반응시키면 5-phenylhydantoin이 생성된다. 이 반응의 반응 메카니즘, 특히 α-diethylaminoacetophenone group이 분리되는 step을 연구했다. GC/MS System을 이용하여 분석한 결과 ethylbenzoate가 생성되는 것으로 보아 α-diethylaminoacetophenone은 hydantoin-ring 형성전에 알칼리 분해되어 benzaldehyde로 될 것으로 사료된다.

상백피에서 추출한 단백질 분해효소의 효소적 성질을 분석하였다. 상백피 추출액으로부터 분리한 단백질 함량은 1.12mg/ml, protease는 비활성도로서 5.14U/mg 수준이었다. 효소의 기질에 대한 특이성은 casein을 100으로 하였을 때 albumin 63, collagen 58, hemoglobin 45, gelatin 36의 비율로 가수분해하여 육류 단백질에 주로 함유된 collagen, hemoglobin을 분해하는 능력이 비교적 큰 것으로 나타났다. 효소의 최적온도는 60℃, 열안정성은 50∼80℃로서 비교적 열에 안정하였다. 효소의 최적 pH는 6.0이었으며 pH 6.0∼7.0에서 안정하였고 pH 9.0 이상에서는 활성이 급격히 저하되었다. 따라서 옛날부터 가정에서 고기를 삶을 때 연회를 위하여 사용하였던 상백피 중에 단백질 분해효소력이 존재함을 확인하였다. Water extract of mulberry tree barks(Morus alba Linne) was studied for its proteolytic activity. Protein content of the extract was 1.12 ㎎/ml and its specific activity was 5.14 U/ml. The enzyme was active on various proteins : the relative acitities were 100 for casein, 63 for albumin, 58 for collagen, 45 for hemoglobin and 36 for gelatin, respectively. These suggested that the ability of the enzyme to hydrolyze meat was relatively high since those are major meat proteins. Optimum pH and temperature for proteolytic activity were : pH 6.0 and 60℃. And the enzyme was stable at the pH range of 6.0 to 7.0 and temperature between 50 and 80℃. Apparent proteolytic activities could support some scientific grounds of traditional application of mulberry tree barks to home cooking for meat tenderization.
