This experiment was carried out to investigate the effect of plant growth regulator on essential oil production and its composition, and to find out the optimum level of plant growth regulator in Mentha piperita L. in vitro culture. The optimum level for oil content and menthol and menthone contents in 2, 4-D alone was 4mg/l. Proper level for menthone in NAA alone was 4mg/l, and difference for fresh weight and oil contents between NAA and 2,4-D alone was not observed. The combination of NAA 2mg/l and IAA were better than that of NAA alone for menthol (IAA 6mg/l) and menthylacetate (IAA 2mg/l) contents while any combination of these hormones did not affect on menthone. BA alone was inferior to auxin in fresh weight, oil content, menthol and menthone contents. In combination of NAA 1mg/l and BA 2mg/l, menthone(13.3%) was produced eleven times more than pulegone(1.2%).
