        Genomic and transcriptomic analysis of Korean colorectal cancer patients

        Background: Colorectal cancer (CRC) is the third most common type of diagnosed cancer in the world and has the second-highest mortality rate. Meanwhile, South Korea has the second-highest incidence rate for CRC in the world. Objective: To assess the possible influence of ethnicity on the molecular profile of colorectal cancer, we compared genomic and transcriptomic features of South Korean CRCs with European CRCs. Methods: We assembled a genomic and transcriptomic dataset of South Korean CRC patients (KOCRC; n = 126) from previous studies and European cases (EUCRC; n = 245) selected from The Cancer Genome Atlas (TCGA). Then, we compared the two datasets in terms of clinical data, driver genes, mutational signature, gene sets, consensus molecular subtype, and fusion genes. Results: These two cohorts showed similar profiles in driver mutations but differences in the mutation frequencies of some driver genes (including APC, TP53, PABPC1, FAT4, MUC7, HSPG2, GNAS, DENND5B, and BRAF). Analysis of hallmark pathways using genomic data sets revealed further differences between these populations in the WNT, TP53, and NOTCH signaling pathways. In consensus molecular subtype (CMS) analyses of the study cases, no BRAF mutations were found in the CMS1 subtype of KOCRC, which contrasts with previous findings. Fusion gene analysis identified oncogenic fusion of PTPRK-RSPO3 in a subset of KOCRC patients without APC mutations. Conclusions: This study presents insights into the genomic landscape of KOCRCs and reveals some similarities and differences with EUCRCs at the molecular level.

        Comparison of the MGISEQ-2000 and Illumina HiSeq 4000 sequencing platforms for RNA sequencing

        Currently, Illumina sequencers are the globally leading sequencing platform in the next-generation sequencing market. Recently, MGI Tech launched a series of new sequencers, including the MGISEQ-2000, which promise to deliver high-quality sequencing data faster and at lower prices than Illumina's sequencers. In this study, we compared the performance of two major sequencers (MGISEQ-2000 and HiSeq 4000) to test whether the MGISEQ-2000 sequencer delivers high-quality sequence data as suggested. We performed RNA sequencing of four human colon cancer samples with the two platforms, and compared the sequencing quality and expression values. The data produced from the MGISEQ-2000 and HiSeq 4000 showed high concordance, with Pearson correlation coefficients ranging from 0.98 to 0.99. Various quality control (QC) analyses showed that the MGISEQ-2000 data fulfilled the required QC measures. Our study suggests that the performance of the MGISEQ-2000 is comparable to that of the HiSeq 4000 and that the MGISEQ-2000 can be a useful platform for sequencing.

        The rapid development in Information and Communication technology has progressed society to evolve and transform into a paradigm as an IT based Information Knowledge Society. Such progression has made significant change in working environments. The concept of Smart Work is an idea that is starting to become a reality. Much research has been conducted on the concept, approached from operational and technological processes; in addition to further studies that are being conducted on improving physical space, in which results are slowly being applied and implemented. The following research aims to suggest prospects of improvement for physical spaces pertaining to the Smart Work Center. First, spatial aspects should be allocated according to the continuous and flexible flow of user traffic. It should also blur the boundaries for various spaces and organizations. Second, the Smart Work Center should also consider psychological aspects in order to provide seamless transitions during its application and encouraging creativity for its users. Lastly, a remote control system should be established. The integration of a remote control should consider the navigation of diverse programs, as well as their basic application in business, taking into consideration for various types of users, in order to make good use of the Smart Work Center.

      • 예산군 밭관개시설물 기후변화 영향평가를 위한 작물 필요수량 모의

        본 연구는 밭관개시설물의 취약성을 평가하기 위하여 밭작물이 필요로 하는 필요수량을 산정하였다. 밭작물의 필요 수량은 강수량 등과 같은 기상 조건이 매년 다르고 또 이에 의해 밭작물의 생장이 달라질 수 있기 때문에, 실측에 의한 필요수량 산정은 수년에 걸친 모니터링이 필요하다. 따라서 본 연구에서는 장기간에 걸친 수문 거동 모의와 기상 조건에 따른 다양한 작물의 생장을 모의할 수 있는 Soil and Water Assessment Tool(SWAT; Arnold and Srinivasan, 1994) 모형을 이용하였다. 밭작물 필요수량 산정을 위한 SWAT 모형의 대상 유역은 충청남도 예산군을 포함하는 유역으로 선정하였다. SWAT 모형의 구동에 필요한 입력 자료인 강수량 자료와 모형의 보정 및 검정에 필요한 유량 자료는 국가수자원관리종합정보시스템(http://wamis.go.kr)에서 제공하는 국토교통부 관측소 자료를 이용하였다. SWAT 모형은 작물의 생장에 따른 수문 및 수질 변동을 모의하지만 작물에 의해 소비되는 물의 양을 모형의 결과로 직접적으로 제공하지는 않기 때문에, 모형의 결과를 이용하여 작물에 의한 물 소비량을 산정해야 하며, 이 과정에 대한 모형의 소스코드를 수정할 필요가 있다. 본 연구에서는 작물이 재배되도록 지정된 수문학적 반응 단위에 대해서만 초기 함수량과 최종 함수량을 출력할 수 있도록 소스코드를 수정하였다. 본 연구에서는 배추, 사과에 대한 필요수량을 모의하였다. 2000년부터 2017년 동안의 모의에서 작물별 필요수량은, 배추의 경우 253.10 mm∼337.45 mm의 범위를 보이면서 평균 303.93 mm로 모의되었으며, 사과의 경우 347.00 mm∼425.16 mm의 범위를 보이면서 평균 386.24 mm로 모의되었다.

        대식세포에서 Nrf2/HO-1경로를 통한 청심연자음의 항산화효과

        활성산소종(reactive oxygen species, ROS)은 우리 몸의 항상성 유지에 있어 중요한 역할을 한다. 그러나 과도한 ROS의 생성은 단백질, 지질, 핵산과 같은 세포 구성성분을 손상시키고 피부노화를 촉진시킨다. 이에 본 연구에서는 과도한 산화 스트레스를 예방하기 위해 Chungsimyeonja-um (CSYJE)의 항산화 효과를 확인하였다. 먼저 DPPH 및 ABTS assay를 실시하여 CSYJE의 항산화 효과를 확인한 결과 농도 의존적으로 radical 소거 활성을 확인하였다. 세포생존율 확인을 위해 MTT assay를 실시한 결과 1,000 μg/mL 농도에서 세포 독성이 없음을 확인하였다. 항산화 관련 단백질인 nuclear-E2-related factor 2 (Nrf2), Heme oxygenase-1 (HO-1)의 발현 수준을 확인하기 위해 western blotting을 실시한 결과 농도 의존적으로 발현이 증가하는 것을 확인하였다. 세포 내 ROS유발 물질인 lipopolysaccharide (LPS)로 ROS를 유도한 후, ROS생성억제효과를 확인하기 위해 DCF-DA 염색법을 실시한 결과 농도 의존적으로 ROS 생성 억제효과를 확인하였으며 ROS의 생성으로 인한 염증성 사이토카인과 염증인자의 mRNA발현 수준을 확인하기 위해 real-time RT-PCR을 실시한 결과 농도 의존적으로 염증성 사이토카인과 염증인자의 mRNA 발현을 억제시켰다. 따라서, 본 연구는 Nrf2/HO-1 신호 전달 경로 활성을 통해 CSYJE의 항산화효과를 확인했으며 이는 CSYJE가 활성산소를 억제하여 항산화 화장품의 재료로서 사용될 수 있음을 시사한다. Reactive oxygen species (ROS) plays an important role in maintaining homeostasis. However, excessive ROS production damages cellular components such as proteins, lipids, and nucleic acids and promotes skin aging. In this study, we confirmed the antioxidant effect of CSYJE to prevent excessive oxidative stress. First, DPPH and ABTS assays were performed to confirm the antioxidant effect of CSYJE and the radical scavenging activity was confirmed depending on the concentration. As a result of performing the MTT assay to confirm the cell viability, it was confirmed that there was no cytotoxicity at a concentration of 1,000 μg/mL. As a result of western blotting to confirm the expression levels of the antioxidant-related proteins nuclear-E2-related factor 2 (Nrf2) and Heme oxygenase-1 (HO-1), it was confirmed that the expression was increased in a concentration-dependent manner. After inducing ROS with lipopolysaccharide (LPS), an intracellular ROS-causing substance, DCF-DA was performed to confirm the inhibitory effect of ROS production, and the inhibition of ROS production was confirmed to concentration-dependent. Real-time RT-PCR was performed to confirm the mRNA expression level of inflammatory cytokines and inflammatory mediator caused by ROS generation, mRNA expression was reduced in a dose dependent manner. Therefore, this study confirmed the antioxidant effect of CSYJE through the Nrf2/HO-1 signaling pathway, which suggests that CSYJE can be used as an antioxidant cosmetic material by inhibiting free radicals.

        Early infectious complications of percutaneous metallic stent insertion for malignant biliary obstruction.

        <P>OBJECTIVE: This study was designed to evaluate predisposing factors for early infectious complications after percutaneous metallic biliary stent insertion in patients with malignant biliary obstruction. MATERIALS AND METHODS: From August 1999 to June 2008, 215 consecutively registered patients with inoperable malignant biliary obstruction were treated with percutaneous placement of a metallic stent. The incidence of early infectious complications was evaluated. Sex, age, level of obstruction, type and number of stents, and stent position (across or above the main duodenal papilla) were retrospectively reviewed. The findings in patients with early infectious complications were compared with those in patients without early infectious complications. RESULTS: Infectious complications occurred within 30 days after stent placement in 14 of 215 (6.5%) patients. Univariate analysis showed a significant difference between the two groups with respect to nontranspapillary stent placement (p = 0.003). In the multiple logistic regression analysis, age, sex, level of obstruction, and type and number of stents were negatively associated with the development of early infectious complications (p > 0.05). CONCLUSION: Our study data showed that stent placement across the main duodenal papilla seemed to prevent early infectious complications after percutaneous metallic biliary stent insertion in patients with malignant biliary obstruction.</P>

      • Polymorphic Phase Control Mechanism of Organic–Inorganic Hybrid Perovskite Engineered by Dual-Site Alloying

        <P>As a next-generation solar cell, perovskite solar cells (PSCs) have been attracting considerable attention. A FAPbI(3) is particularly considered as an optimal material with a proper band gap and thus has been employed as a base material for the PSCs with more than 20% efficiency; however, the competitive polymorphic growth of alpha- and delta-phases is a major hurdle in utilizing this material. To provide the theoretical model of the polymorphic phase competition of FAPbI(3) for the first time, we here investigate how compositional engineering can pave a route to control the polymorphic growth of FAPbI(3) using density functional theory combined with a statistical mechanical treatment of the configurational space. We find that dual-site alloying of both cations and halides is critically important to achieve the specific stabilization of the a-phase while maintaining the good miscibility, thermodynamic stability, and optimal band gap property. Based on our first successful theoretical modeling of the FAPbI(3) system and its polymorphic phase competition behavior during, dual-site alloying, we anticipate deriving new rational guidelines on compositional engineering of organic inorganic hybrid perovskite alloys for designing PSCs with high efficiencies and stabilities.</P>
