        Unambiguous Identification of Fugitive Pollutants and Determination of Annual Emission Flux as Diurnal Monitoring Mode

        Toxic air pollutants are investigated in a petrochemical industrial park in Taiwan by using a movable open path FTIR. The results show the qualitative and quantitative analysis of emission gases from plants, and also provide the emission rate of different compounds. More than twenty compounds under usual operation are found from this industrial park. The concentration variation with time can be correlated exactly with wind direction. It means that by changing the measuring points, the source of emission can be unambiguously identified. An EPA proved PAL model is applied to estimate the emission rate of either a point or an area source. Local atmospheric stability is determined by releasing the $SF_6$ tracer. The origins of errors come mainly from the uncertainty of source's configuration and the variation of meteorological condition. Through the continuous measurement (half an hour base in this study) of OP-FTIR sensor, the maximum value of emission rate and the annual amount of emission can be derived. The emission rate of the measured toxic gases are derived by the model technique and the results show that the emission amount are in the order of ten to hundred tons per year.

        Characterization of the dynamic behavior of a linear guideway mechanism

        Dynamic behaviors of the contact surface between ball and raceway in a guideway mechanism vary with the applied loads and hence affect the mechanical responses of machine tools. The study aims to investigate the nonlinear characteristics of dynamic behaviors at the rolling contact interface in linear guideway mechanisms. Firstly, analytical method was introduced to understand the contact behaviors based on Hertz contact theory in a point-to-point way. Then, the finite element approach with a three-dimensional surface-to-surface contact model and appropriate contact stiffness was developed to study the dynamic characteristics of such linear guideways. Finally, experiments with modal test were conducted to verify the significance of both the analytical and the numerical results. Results told that the finite element approach may provide significant predictions. The study results also concluded that the current nonlinear models based on Hertz's contact theory may accurately describe the contact characteristic of a linear guideway mechanism. In the modal analysis, it was told that the natural frequencies vary a little with different loading conditions; however, the mode shapes are changed obviously with the magnitude of applied loads. Therefore, the stiffness of contact interface needs to be properly adjusted during simulation which may affect the dynamic characteristics of the machine tools.

        Increase in Plasma Glucose Lowering Action of Rosiglitazone by Electroacupuncture at Bilateral Zusanli Acupoints (ST.36) in Rats

        Objectives: Hypoglycemia induced by electroacupuncture (EA) is due to an increase of insulin secretion and/or mediation of β-endorphin. We applied EA at the Zusanli (ST.36) acupuncture point (acupoint) in combination with rosiglitazone (TZD) administration to evaluate their effect on plasma glucose and to explore possible mechanisms of action. Methods: Thirty six normal adult Wistar rats were randomly divided into four groups: the 0.1 mg/kg TZD group (0.1TZD), 0.1 mg/kg TZD and EA group (0.1TZD + EA), EA group, and control group. In other experiments, streptozotocin was used to induce type 2 diabetes mellitus in neonatal rats; these were then randomly divided into a 0.1TZD group, 0.1TZD + EA group, and EA group and changes in plasma glucose and insulin concentrations evaluated. Results: A marked hypoglycemic response was observed in the normal rat 0.1TZD, 0.1TZD + EA and EA groups, with the response more significant in the 0.1TZD + EA group than in the 0.1TZD group. Among the diabetic animals, the hypoglycemic responses in the 0.1TZD + EA and EA groups were greater than in the 0.1TZD group. In both the normal and diabetic rats, insulin secretion was increased by EA or 0.1TZD + EA treatment, but not by 0.1TZD. Conclusions: The plasma glucose lowering action of rosiglitazone was increased by EA in both normal and diabetic rats, indicating that the application of EA may enhance the hypoglycemic action of this insulin sensitizer.         BRCA1/2 mutation status in patients with metachronous breast and ovarian malignancies: clues towards the implementation of genetic counseling

        Objective: The characteristics of patients with metachronous breast and ovarian malignancies and the pathogenic role of BRCA1/2 mutations remain poorly understood. We investigated these issues through a review of hospital records and nationwide Taiwanese registry data, followed by BRCA1/2 mutation analysis in hospital-based cases. Methods: We retrospectively retrieved consecutive clinical records of Taiwanese patients who presented with these malignancies to our hospital between 2001 and 2017. We also collected information from the Data Science Center of the Taiwan Cancer Registry (TCR) between 2007 and 2015. Next-generation sequencing and multiplex ligation-dependent probe amplification were used to identify BRCA1/2 mutations and large genomic rearrangements, respectively. When BRCA1/2 mutations were identified in index cases, pedigrees were reconstructed and genetic testing was offered to family members. Results: A total of 12,769 patients with breast cancer and 1,537 with ovarian cancer were retrieved from our hospital records. Of them, 28 had metachronous breast and ovarian malignancies. We also identified 113 cases from the TCR dataset. Eighteen hospital-based cases underwent BRCA1/2 sequencing and germline pathogenic mutations were detected in 7 patients (38.9%, 5 in BRCA1 and 2 in BRCA2). All BRCA1/2 mutation carriers had ovarian high-grade serous carcinomas. Of the 12 patients who were alive at the time of analysis, 5 were BRCA1/2 mutation carriers. All of them had family members with BRCA1/2-associated malignancies. Conclusions: Our results provide pilot evidence that BRCA1/2 mutations are common in Taiwanese patients with metachronous breast and ovarian malignancies, supporting the clinical utility of genetic counseling.

      • The Protective Effect of Black Garlic Extracts on tBHP-induced Injury in Hepatocytes in vitro and in vivo

        Black garlic has been reported to show multiple bioactivities against the development of different diseases. In this study, we investigated the hepatoprotective effect of black garlic on injured liver cells. Rat clone-9 hepatocytes were used for all the experiments. We used tert-Butyl hydroperoxide (tBHP) to induce injury of rat clone-9 hepatocytes. The contents of malondialdehyde (MDA) and glutathione (GSH), antioxidative enzyme activities of catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), and mRNA expressions of IL-6 and IL-8 in rat clone-9 hepatocytes were determined to evaluate the level of cell damage. We found that black garlic extracts significantly recovered tBHP-induced cell death of rat clone-9 hepatocytes. Pretreatment with black garlic extracts antagonized GSH depletion, tBHP-increased MDA accumulation, IL-6/IL-8 mRNA expressions, and tBHP-decreased antioxidative enzyme activities. Moreover, we further found that JNK signaling regulated black-garlic-inhibited tBHP effects in rat clone-9 hepatocytes. Our findings demonstrate that black garlic has great hepatoprotective potential to block tBHP-damaged effects on cell death, lipid peroxidation, oxidative stress, and inflammation in rat clone-9 hepatocytes. Thus, this study suggests that black garlic may be an excellent natural candidate in the development of adjuvant therapy and healthy foods for liver protection.

        An update of preimplantation genetic diagnosis in gene diseases, chromosomal translocation, and aneuploidy screening

        Preimplantation genetic diagnosis (PGD) is gradually widely used in prevention of gene diseases and chromosomal abnormalities. Much improvement has been achieved in biopsy technique and molecular diagnosis. Blastocyst biopsy can increase diagnostic accuracy and reduce allele dropout. It is cost-effective and currently plays an important role. Whole genome amplification permits subsequent individual detection of multiple gene loci and screening all 23 pairs of chromosomes. For PGD of chromosomal translocation, fluorescence $in-situ$ hybridization (FISH) is traditionally used, but with technical difficulty. Array comparative genomic hybridization (CGH) can detect translocation and 23 pairs of chromosomes that may replace FISH. Single nucleotide polymorphisms array with haplotyping can further distinguish between normal chromosomes and balanced translocation. PGD may shorten time to conceive and reduce miscarriage for patients with chromosomal translocation. PGD has a potential value for mitochondrial diseases. Preimplantation genetic haplotyping has been applied for unknown mutation sites of single gene disease. Preimplantation genetic screening (PGS) using limited FISH probes in the cleavage-stage embryo did not increase live birth rates for patients with advanced maternal age, unexplained recurrent abortions, and repeated implantation failure. Polar body and blastocyst biopsy may circumvent the problem of mosaicism. PGS using blastocyst biopsy and array CGH is encouraging and merit further studies. Cryopreservation of biopsied blastocysts instead of fresh transfer permits sufficient time for transportation and genetic analysis. Cryopreservation of embryos may avoid ovarian hyperstimulation syndrome and possible suboptimal endometrium.

        Formation of Solid-Solution Cu-to-Cu Joints Using Ga Solder and Pt under Bump Metallurgy for Three-Dimensional Integrated Circuits

        Three-dimensional (3D) integrated circuits (ICs) are the most important packaging technology for next-generation semiconductors. Cu-to-Cu throughsilicon via interconnections with micro-bumps are key components in the fabrication of 3D ICs. However, significant reliability concerns have been raised due to the formation of brittle intermetallic compounds in the entire 3D IC joints. This study proposes a Ga-based Cu-to-Cu bonding technology with Pt under bump metallurgy (UBM). A systematic analysis of reactive wetting between Ga solders and polycrystalline, single-crystalline, and Ptcoated Cu substrates was conducted. Pt UBM as a wetting layer was identified to be a key component for Ga-based Cu-to-Cu bonding. Pt-coated Cu substrates were bonded using Ga solders with various Ga-to-Pt ratios (n) at 300°C. When n ≥ 4, the Cu/Pt/Ga/Pt/Cu interface evolves to Cu/facecentered cubic (fcc)/γ1-Cu9Ga4/fcc/Cu, Cu/fcc/γ1-Cu9Ga4 + Ga7Pt3/fcc/Cu, and finally Cu/fcc + Ga7Pt3/Cu structures. The desired ductile solid solution joint formed with discrete Ga7Pt3 precipitates. When n ≤ 1, a Cu/Ga7Pt3/Cu joint formed without Cu actively participating in the reactions. The reaction mechanism and microstructure evolution were elaborated with the aid of CALPHAD thermodynamic modeling.

        Applying Interactive Genetic Algorithms to Disassembly Sequence Planning

        In addition to garbage sorting and resource recycling, green design should be a fundamental method for solving environmentalproblems, and design for disassembly is an important foundation of green design. This study focuses on providingquantitative assessment methods for designers’ reference. This study proposes interactive genetic algorithms to solve theproblem of disassembly sequence planning. First, the disassembly factor is measured by the fuzzy scoring procedure method,and then the genetic algorithm is used to select the optimal sequence. With the penalty value provided from the process, areference is provided for the revised design. Finally, examples are discussed to demonstrate that the proposed approach isa feasible method.
