Objective: To investigate possible signal pathway involvement in multi-drug resistant P-glycoprotein (P-gp) expression induced by cyclooxygenase-2 (COX-2) in a human gastric adenocarcinoma cell line stimulated with pacliaxel (TAX). Methods: The effects of TAX on SGC7901 cell growth with different doses was assessed by MTT assay, along with the effects of the COX-2 selective inhibitor NS-398 and the nuclear factor-KB (NF-KB) pathway inhibitor pyrrolidine dithiocarbamate (PDTC). Influence on COX-2, NF-KB p65 and P-gp expression was determined by Western blotting. Results: TAX, NS-398 and PDTC all reduced SGC7901 growth, with dosedependence. With increasing dose of TAX, the expression of COX-2, p65 and P-gp showed rising trends, this being reversed by NS-398. PDTC also caused decrease in expression of p65 and P-gp over time. Conclusion: COX-2 may induce the expression of P-gp in SGC7901 cell line via the NF-kappa B pathway with pacliaxel stimulation.

Ethic and value theory of new contemporary Taoism, should be the development and inheritance of excellent tradition of ethical thoughts of ancient Taoism, and it also should be a new annotation of Taoism ethical value on actual requirements of current society. Only if contemporary new Taoism carries forward traditional Taoism thoughts of advocating simply and honestly, getting along with subordinates humbly, having few desires and selfishnesses, and combines Ethical and Confucianism thoughts together to build the Confucianism thoughts of humanity and courtesy on Taoism thought that to be honest and sincere, having few desires and selfishnesses, we can overcome and avoid the 'dissimilation' danger of Confucianism thoughts of humanity and courtesy.

Objective Despite a high prevalence of dementia in older adults hospitalized with severe acute respiratory syndrome coronavirus 2 infection (SARS-CoV-2), or so called COVID-19, research investigating association between preexisting diagnoses of dementia and prognosis of COVID-19 is scarce. We aimed to investigate treatment outcome of patients with dementia after COVID-19. Methods We explored a nationwide cohort with a total of 2,800 subjects older than 50 years who were diagnosed with COVID-19 between January and April 2020. Among them, 223 patients had underlying dementia (dementia group). We matched 1:1 for each dementia- non-dementia group pair yielding 223 patients without dementia (no dementia group) using propensity score matching. Results Mortality rate after COVID-19 was higher in dementia group than in no dementia group (33.6% vs. 20.2%, p=0.002). Dementia group had higher proportion of patients requiring invasive ventilatory support than no dementia group (34.1% vs. 22.0%, p=0.006). Multivariable analysis showed that dementia group had a higher risk of mortality than no dementia group (odds ratio=3.05, p<0.001). We also found that patients in dementia group had a higher risk of needing invasive ventilatory support than those in no dementia group. Conclusion Our results suggest that system including strengthen quarantines are required for patients with dementia during the COVID- 19 pandemic.

Background/Aims: Coenzyme Q<sub>10</sub> (CoQ<sub>10</sub>), is a promising antioxidant; however, low bioavailability owing to lipid-solubility is a limiting factor. We developed water-soluble CoQ<sub>10</sub> (CoQ<sub>10</sub>-W) and compared its effects with conventional lipid-soluble CoQ<sub>10</sub> (CoQ<sub>10</sub>-L) in an experimental model of chronic tacrolimus (Tac) nephropathy. Methods: CoQ<sub>10</sub>-W was developed from a glycyrrhizic-carnitine mixed layer CoQ<sub>10</sub> micelle based on acyltransferases. Chronic nephropathy was induced in rats with 28-day Tac treatment; they were concomitantly treated with CoQ<sub>10</sub>-L or CoQ<sub>10</sub>-W. CoQ<sub>10</sub> level in plasma and kidney were measured using liquid chromatography-mass spectrometry. CoQ<sub>10</sub>-W and CoQ<sub>10</sub>-L effects on Tac-induced nephropathy were assessed in terms of renal function, histopathology, oxidative stress, and apoptotic cell death. Their effects on cell viability and reactive oxygen species (ROS) production were assessed in cultured proximal tubular cells, human kidney 2 (HK-2) cells. Results: The plasma CoQ<sub>10</sub> level was significantly higher in the CoQ<sub>10</sub>-W group than in the CoQ<sub>10</sub>-L group. Tac treatment caused renal dysfunction, typical pathologic lesions, and oxidative stress markers. Serum creatinine was restored in the Tac + CoQ<sub>10</sub>-L or CoQ<sub>10</sub>-W groups compared with that in the Tac group. CoQ<sub>10</sub>-W administration reduced oxidative stress and apoptosis markers. Mitochondrial ultrastructure assessment revealed that the addition of CoQ<sub>10</sub>-L or CoQ<sub>10</sub>-W with Tac increased mitochondrial size and number than Tac treatment alone. In vitro investigations revealed that both CoQ<sub>10</sub>-L and CoQ<sub>10</sub>-W improved cell viability and reduced ROS production in the Tac-induced HK-2 cell injury. Conclusions: CoQ<sub>10</sub>-W has a better therapeutic effect in Tac-induced renal injury than conventional CoQ<sub>10</sub>-L, possibly associated with improved CoQ<sub>10</sub> bioavailability.

Objective: This study compared the efficacy and tolerability of clonazepam with other benzodiazepines in patients with anxiety disorders. Methods: Inclusion criteria were as follows: age >20 years, diagnosis of anxiety disorder according to the Diagnostic and Statistical Manual of Mental Disorders 4th edition, text revision (DSM-IV-TR) criteria, taking only one type of antidepressant, and prescribed one of three oral benzodiazepines (alprazolam, clonazepam, or lorazepam). At baseline and week 6, clinical benefit was evaluated using the Clinical Global Impression-Severity Scale (CGI-S), Clinical Global Impression-Anxiety Scale (CGI-anxiety), and Clinical Global Impression-Sleep Scale (CGI-sleep). Results: Among 180 patients, no differences in demographic characteristics among the three benzodiazepine groups were noted. After six weeks of treatment, all benzodiazepine groups showed significant improvements in CGI-S, CGI-anxiety, and CGI-sleep scores ( p <0.001). There were no differences in mean changes in CGI-S, CGI-anxiety and CGI-sleep among the three benzodia-zepine groups. The incidence of side effects was significantly lower in the clonazepam group than with the other benzodiazepines. The incidences of adverse events for the clonazepam, alprazolam, and lorazepam groups were 26.7% (n=20), 48.4% (n=31), and 43.9% (n=18), respectively. Conclusion: The present study suggests that clonazepam is as efficacious as other benzodiazepines for the treatment of various anxiety disorders. Furthermore, the safety profile of clonazepam was superior to the other benzodiazepines in this study.
Tumor-Suppression Mechanisms of Protein Tyrosine Phosphatase O and Clinical Applications

Tyrosine phosphorylation plays an important role in regulating human physiological and pathological processes. Functional stabilization of tyrosine phosphorylation largely contributes to the balanced, coordinated regulation of protein tyrosine kinases (PTKs) and protein tyrosine phosphatases (PTPs). Research has revealed PTPs play an important suppressive role in carcinogenesis and progression by reversing oncoprotein functions. Receptor-type protein tyrosine phosphatase O (PTPRO) as one member of the PTPs family has also been identified to have some roles in tumor development. Some reports have shown PTPRO over-expression in tumors can not only inhibit the frequency of tumor cell division and induce tumor cell death, but also suppress migration. However, the tumor-suppression mechanisms are very complex and understanding is incomplete, which in some degree blocks the further development of PTPRO. Hence, in order to resolve this problem, we here have summarized research findings to draw meaningful conclusions. We found tumor-suppression mechanisms of PTPRO to be diverse, such as controlling G0/G1 of the tumor cell proliferation cycle, inhibiting substrate phosphorylation, down-regulating transcription activators and other activities. In clinical anticancer efforts, expression level of PTPRO in tumors can not only serve as a biomarker to monitor the prognosis of patients, but act as an epigenetic biomarker for noninvasive diagnosis. In addition, the re-activation of PTPRO in tumor tissues, not only can induce tumor volume reduction, but also enhance the susceptibility to chemotherapy drugs. So, we can propose that these research findings of PTPRO will not only support new study ideas and directions for other tumor-suppressors, importantly, but also supply a theoretical basis for researching new molecular targeting agents in the future.
Inorganic-organic Hybrid Proton Conductive Membranes Doped with Phosphoric Acid

A new proton conductive inorganic-organic hybrid membrane doped with H₃P0₄ was fabricated via sol-gel process wit 3-glycidoxypropyltrimethoxysilane(GPTMS), 3-aminopropyltriethoxysilane(APIES)andtetraethoxysilane(TEOS)asprecursors. Theproto conductivity of about 3.0 x 10¯³ S/cm was obtained at 120°C under 50% relative humidity (R.H). DTA curves showed that the thermal stability of the membrane is significantly enhanced by the presence of SiO₂ framework up to 250°C. SEM and XRD revealed that the gel is microporou and amorphous. The addition of APTES improved the conductivity of the membranes and the effect of the APIES on the conductivity was also discussed in this paper.
