        Gintonin facilitates catecholamine secretion from the perfused adrenal medulla

        The present study was designed to investigate the characteristics of gintonin, one of components isolated from Korean Ginseng on secretion of catecholamines (CA) from the isolated perfused model of rat adrenal gland and to clarify its mechanism of action. Gintonin (1 to 30 μg/ml), perfused into an adrenal vein, markedly increased the CA secretion from the perfused rat adrenal medulla in a dose-dependent fashion. The gintonin-evoked CA secretion was greatly inhibited in the presence of chlorisondamine (1 μM, an autonomic ganglionic bloker), pirenzepine (2 μM, a muscarinic M<sub>1</sub> receptor antagonist), Ki14625 (10 μM, an LPA<sub>1/3</sub> receptor antagonist), amiloride (1 mM, an inhibitor of Na<sup>+</sup>/Ca<sup>2+</sup> exchanger), a nicardipine (1 μM, a voltage-dependent Ca<sup>2+</sup> channel blocker), TMB-8 (1 μM, an intracellular Ca<sup>2+ </sup>antagonist), and perfusion of Ca<sup>2+</sup>-free Krebs solution with 5mM EGTA (a Ca<sup>2+</sup>chelater), while was not affected by sodium nitroprusside (100 μM, a nitrosovasodialtor). Interestingly, LPA (0.3~3 μM, an LPA receptor agonist) also dose-dependently enhanced the CA secretion from the adrenal medulla, but this facilitatory effect of LPA was greatly inhibited in the presence of Ki 14625 (10 μM). Moreover, acetylcholine (AC)-evoked CA secretion was greatly potentiated during the perfusion of gintonin (3 μg/ml). Taken together, these results demonstrate the first evidence that gintonin increases the CA secretion from the perfused rat adrenal medulla in a dose-dependent fashion. This facilitatory effect of gintonin seems to be associated with activation of LPA- and cholinergic-receptors, which are relevant to the cytoplasmic Ca<sup>2+</sup> increase by stimulation of the Ca<sup>2+</sup> influx as well as by the inhibition of Ca<sup>2+</sup> uptake into the cytoplasmic Ca<sup>2+</sup> stores, without the increased nitric oxide (NO). Based on these results, it is thought that gintonin, one of ginseng components, can elevate the CA secretion from adrenal medulla by regulating the Ca<sup>2+</sup> mobilization for exocytosis, suggesting facilitation of cardiovascular system. Also, these findings show that gintonin might be at least one of ginseng-induced hypertensive components.

        A 33-year-old female was admitted to our hospital due to removal of lung mass which indicentally detected Chest radiography. Chest computerized tomography revealed lung parenchymal cystic lesion. Open thoracotomy was done for diagnosis and therapy. Cystic lesion was communicated to bronchus and wedge resection including cystic lesion was done. Bronchogenic cyst is congenital disease and mainly locate in mediastinum. It is rare in lung parenchyme. Herein we report a case of intrapulmonary bronchogenic cyst.

      • Prognostic factors for classifying extranodal NK/T cell lymphoma, nasal type, as lymphoid neoplasia

        <P>Abstract</P><P>This study evaluated the applicability of prognostic factors commonly used for diagnosis of classical lymphoma outcomes to extranodal NK/T cell lymphoma, nasal type (NTCL). Clinical features and their associations with lactate dehydrogenase (LDH) were evaluated in 70 patients. RLDH was defined as the ratio of LDH to the upper normal limit. RLDH was associated with stage (I–II vs. III–IV), lymph node involvement (LNI), and International Prognostic Index score (<2 vs. ≥2). Poor performance status and advanced stage were common in patients with local tumor invasiveness (LTI). LDH level, classified into three levels (low, high, and very high) was associated with survival (<I>P</I> < 0.001). In multivariate analysis, the predictive values of LDH level, B symptom, performance status, and stage remained significant whereas those of LTI and LNI did not. Scoring was performed by weighting each factor with 0.5 or 1.0 according to its hazard ratio. Scores were classified into four groups. Groups with high scores were associated with unfavorable outcomes (<I>P</I> < 0.001). Current study suggests that prognostic factors for NHL may be useful to predict the outcome of NTCL but the model should take LDH level and the prognostic weight of each factor into account.</P>

        선회 및 기하학적 형상 변화에 따른 중잔유 가스화 반응 특성 연구

        본 연구에서는 원유 정제후 남는 고유황 함유 물질인 중잔유를 가지고 전산해석방법론을 적용하여 고온/고압 가스화기내 에서의 가스화 반응 특성을 파악하고자 하였다. 특히, 가스화기 설계의 주요 인자인 이차주입노즐에서의 선회 강도(선회수)와 원통형 반응로의 형상(L/D)을 해석 변수로 선정하여 반응 유동장의 패턴 변화를 고찰해 봄으로써 중잔유 가스화기의 적절한 설계 및 운전 조건의 설정에 중요한 역할을 하는 변수들의 영향을 파악하고자 하였다. 전산해석 결과, 선회수가 커질수록 화염 안정화에 도움을 주는 가스화기 중심축 재순환영역의 크기가 증가하고, 연료와 산화제의 혼합을 향상시켜 가스화기내 유동을 균일화시킴으로써 전체적인 반응 완료 시간이 빨라지는 경향을 확인할 수 있었다. 그리고, L/D비가 작을수록 반응로내 연료의 반응 완료 시간이 길어지고, 선회에 의한 영향력이 상대적으로 약화됨을 확인할 수 있었다. 따라서, 중잔유 가스화기 설계시 화염안정화 및 산화제와 연료의 적절한 혼합을 유지하고 내화재 손상을 최소화하기 위해서는 L/D비와 선회강도의 조합을 최적화하는 것이 필수적이라고 판단되었다. Numerical study of the turbulent reacting flow in a high-temperature and high-pressure gasifier has been carried out to analyze the characteristics of gasification reaction of heavy residual oil, which has high sulfur content. In this study, the effect of swirl strength of the secondary nozzle and reactor geometry(L/D), known to be the crucial design parameters of gasifier, has been extensively studied. Especially, we intended to find the effect of operating and design parameters by observing the reacting flow pattern as the key parameters have changed. It is shown that there is progressive increase in the length of central toroidal recirculation zone which plays an important role in flame stabilization and the increase in the reactivity of heavy residual oil as the swirl number increases. In addition, required time for the complete reaction increases and the swirl effect is relatively weakened as the L/D ratio decreases. Most of all, it is found that the optimal combination of L/D ratio and swirl strength is essential to maintain the flame stabilization and efficient mixing ratio of fuel and oxidizer, and to minimize the damage of wall refractory.

        Protective effect of a novel herbmedicine, Hepad, on apoptosis of SH-SY5Y cells and a rat model of Parkinson’s disease

        Parkinson’s disease (PD) is a neurodegenerative disorder characterized by loss of dopaminergic neurons in the substantia nigra pars compacta. In this study, we investigated the effects of a novel herb formula, Hepad, on PD. Dose-dependent treatment with 1-methyl-4-phenylpyridinium (MPP+) decreased the viability of SH-SY5Y cells, and Hepad inhibited the toxic effect of MPP+. Hepad blocked the production of reactive oxygen species (ROS) induced by MPP+ in SH-SY5Y cells, and suppressed the activation of caspase 9 and caspase 3 due to MPP+. A rat model of PD was generated by 6-hydroxydopamine (6-OHDA) injection into the left medial forebrain bundle (MFB) of SD rats. In D-amphetamine sulfate-induced rotational behavioral tests, Hepad administration attenuated circling behavior relative to the 6-OHDA-treated disease group. In addition, Hepad treatment significantly increased the tyrosine hydroxylase (TH)-positive cells in the substantia nigra pars compacta (SNpc) that had decreased in response to 6-OHDA treatment (P<0.05). OX-6 expression, which indicates the presence of microglial cells, decreased significantly after treatment of Hepad in contrast to the 6-OHDA-treated disease group (P<0.05). These results indicate that Hepad may be a useful neuroprotective material for the treatment of neurodegenerative disorders such as PD.

        Effects of a gintonin-enriched fraction on hair growth: an in vitro and in vivo study

        Background: Ginseng has been widely used as a health-promoting tonic. Gintonin present in ginseng acts as a lysophosphatidic acid (LPA) receptor ligand that activates six LPA receptor subtypes. The LPA6 subtype plays a key role in normal hair growth, and mutations in the LPA6 receptor impair normal human hair growth. Currently, human hair loss and alopecia are concerning issues that affect peoples' social and day-to-day lives. Objective: We investigated the in vitro and in vivo effects of a gintonin-enriched fraction (GEF) on mouse hair growth. Methods: Human hair follicle dermal papilla cells (HFDPCs) and six-week-old male C57BL/6 mice were used. The mice were divided into the four groups: control, 1% minoxidil, 0.75% GEF, and 1.5% GEF. The dorsal hair was removed to synchronize the telogen phase. Each group was treated topically, once a day, for 15 days. We analyzed hair growth activity and histological changes. Results: GEF induced transient [Ca<SUP>2+</SUP>]<SUB>i</SUB>, which stimulated HFDPC proliferation and caused 5-bromo-2'-deoxyuridine (BrdU) incorporation in a concentration-dependent manner. GEF-mediated HFDPC proliferation was blocked by the LPA receptor antagonist and Ca<SUP>2+</SUP> chelator. HFDPC treatment with GEF stimulated vascular endothelial growth factor release. Topical application of GEF and minoxidil promoted hair growth in a dose-dependent manner. Histological analysis showed that GEF and minoxidil increased the number of hair follicles and hair weight. Conclusion: Topical application of GEF promotes mouse hair growth through HFDPC proliferation. GEF could be one of the main components of ginseng that promote hair growth and could be used to treat human alopecia.
