Growth characteristics Leuconostoc mesenteroides 1B12 isolated from chonggak kimchi was studied to determine its growth ability at a high temperature. Leuc. mesenteroides 1B12 was able to grow very well at 36C~40C which implied a human body temperature. Thus the strain can survive after intake with diet into human intestine. In order to understand a reason of survival at such a high temperature, fermentation test done with L-arabinose, glucose and sucrose. Cell growth of Leuc. mesenteroides 1B12 measured by O. D. at 600 nm was better than that of Leuc. mesenteroides subsp. mesenteroides KCTC 3722 in L-arabinose treatment throughout cultured temperatures from 30C to 38C. This tendency was also found in treatments of glucose and sucrose. From the results of SDS-PAGE, it was found that some distinctive protein bands appeared between size of 24 kDa and size of 15 kDa only in Leuc. mesenteroides 1B12 and Leuc. Mesenteroides 2A9. The protein band density was darker in samples of Leuc. mesenteroides 2A9. Because the protein bands were not appeared in standard strains i. e., Leuc. mesenteroides subsp. dextranicum KCTC 3530 and Leuc. mesenteroides subsp. mesenteroides KCTC 3722, it is assumed that the low molecular weight proteins may be responsible for growth ability at the high temperature. Since it is desirable to investigate antibiotics susceptibility of the Leuc. mesenteroides 1B12 as probiotics, antibiotics inhibition test was done. Among tested antibiotics, Leuc. mesenteroides 1B12 was able to grow in media containing kanamycin, streptomycin and vancomycin. Typically all tested strains were fail to grow on ampicillin containing media. In addition, from the microscopic observation for live cells, cell shape of Leuc. mesenteroides 1B12 was appeared as a small coccoid form in a MRS medium (liquid) however it was observed as a very elongated rod-like form on a solid MRS medium.
본 연구는 성인 남자의 식생활 형태가 혈중지질 농도에 미치는 영향을 연구하기 위한 기초적인 연구로 영남의료원의 건강피검진자를 대상으로 식습관, 영양소 섭취량, 혈중지질농도를 조사하였다. 전체 조사대상자 220명중에 정상인 133명(60.5%), 경계수준군과 고콜레스테롤환자군은 각각 62명(28.2%)과 25(11.3%)를 차지하였다. 조사대상의 평균연령은 48.4±9.5세이었고, 체중은 67.7±10.2㎏이었다. 평균 Broca’s 지수(RBM)와 체질량지수(BMI)는 각각 109±14.3, 23.6±3.0이었다. 식습관이 좋은 사람이 12명(5.4%)이고 보통인 사람이 130명(59.1%)에 해당하였으며 나쁜 사람이 78명(35.5%)으로 나타났으며, 식사형태와 식품의 배합에 대한 인식이 부족한 것으로 나타났다. 조사 대상자의 혈중 중성지질의 평균농도는 약 115.8㎎/dL이었고, 90~120㎎/dL인 사람이 53명(24.1%)로 가장 많았고 60~150㎎/dL인 사람이 대상자 220명중에 131명으로 전체의 59.9%를 차지하였다. 조사 대상자의 혈중 콜레스테롤 농도의 평균은 192.4㎎/dL이었고, 혈중 콜레스테롤의 농도에 따른 대상자의 분포는 180~210㎎/dL인 대상자가 73명으로 전체의 33.2%를 차지하여 가장 많았고 150~210㎎/dL사이 포함되는 사람이 전체의 60.5%로 133명인 것으로 나타났다.
Isolation of psychrotrophic lactic acid bacteria (LAB) from Chonggak kimchi was attempted in order to study characteristics of LAB growing at the low temperature. Among isolated strains, CL-1 showed exactly the same fermentation characteristics of Leuc. mesenteroides subsp. mesenteroides KCTC 3722 in tested carbohydrates except melezitose and the CL-1 showed very wide range temperature profiles (16C-28C) for growth comparing to other isolates. Its lowest temperature for growth was 16C which was much lower than other isolates. However all isolates couldn’t grow at 30C. In order to identify sudden inhibition of cell growth occurred at the transition temperature (16C → 14C), the CL-1 was tested for fermentation ability of hexoses and disaccharides at the lowered temperature. Among 5 tested hexoses, significant change was resulted from mannose fermentation. The CL-1 fermented mannose very well at 24C that gave a final pH 4.7 after 48 hr incubation but at 16C its final pH was only 5.7. This means that the CL-1 could not ferment mannose for its proper cell growth at 16C and inhibition of the cell growth was to begin. Similar tendency was found in fermentation of disaccharides at the lowered temperature. Among 6 tested disaccharides, inhibition of fermentation occurred only in melibiose. The CL-1 showed decreasing fermentation ability with the melibiose from pH 4.8 at 24C to pH 5.2 at 16 C. Fermentation of maltose, sucrose and trehalose was not affected by the lowered temperature at all. Thus, such growth inhibition of the psychrotrophs by lowered temperatures is related to occurrence of inability of fermenting the sugars and it leads to sudden growth inhibition of the psychrotrophic CL-1. From the test for bile salt tolerance, the CL-1 showed strong tolerance for glycocholate as much as Leuc. mesenteroides subsp. mesenteroides KCTC 3722 but both strains were strongly inhibited by 0.5% taurocholate. In case of taurocholate, the CL-1 could overcome the bile salt effect on cell growth with 0.2% taurocholate as incubation time goes by from 12 hr to 24 hr. The taurocholate tolerance of CL-1 was much better than that of Leuc. mesenteroides subsp. mesenteroides KCTC 3722.
