      • Diatomaceous earth를 이용한 경제적이고 효율높은 homemade PCR purification column의 제조 및 평가

        In recent years the amount of sequencing has increased tremendously, especially due to the introduction of automated DNA sequencing. PCR is principal method for the synthesis of templates for DNA sequencing. Consequently, the need for PCR cleanup for template preparation has increased. Already, many PCR purification kits are marketed. This DNA purification kits uses most glass membrane or silica gel, So, can do rapidly efficient purification by simple miniprep. But, efficiency of purification kits is different according to manufacturer and is very expensive. Therefore, we compare the homemade Diatomaceous earth column with major manufacturer's spin column methods for DNA purification. As a result, we found that the homemade Diatomaceous earth column method resulted in equal or better yields relative to the commercial purification kits.

      • 마늘재배의 생력기계화 시스템 모형

        Garlic is one of major vegetables and grown throughout the country of Korea. But the average farm size of garlic production is about 0.3ha and most of the farming operation for garlic production are done by human labour. There are more than ten operation steps for garlic production but only two steps are mechanized, or land preparation and polyethylene covering. Thus production cost is very high and the price of garlic is about 2 to 4 times higher compared with that of international marketing price. We evaluated present production system and problems of garlic production, and suggest mechanized garlic production system for lowering the production cost of garlic.

      • 砂壤土에 있어서 모델테스트에 의한 車輪의 性能豫測에 관한 硏究

        A series of soil bin experiment was carried out on sandy loam to evaluate the cone index whether it represent the soil physical properties pertinent to soil-wheel system and to investigate if true model theory is applicable to powered rigid, wheel-soil system. Four different sized wheels having diameter of 45, 60, 75 and 90cm were used for the experiment. The following conclusions were derived from the study. (1) True model theory can be sufficiently utilized to study the wheel traction and sinkage on sandy loam. (2) Cone index was found to he good measure of soil physical properties which arc pertinent to predict the performance of the powered rigid wheel-soil system.

      • 動力耕耘機의 牽引 및 牽引補助裝置에 관한 硏究

        The objectives of this study were i) to find the traction aids of power tiller, which can be used more efficiently for grassland, slope land and other soil surfaces on which power tiller have traction problems, and ii) to evaluate the comparative traction performance of two different. types of traction devices of rower tiller; tire and tire with traction aids (track). This experiment was carried out on the four different soil surfaces : hard soil surfaces, plowed land, soil covered with weeds and asphalt road. The results of this experiment were summarized as follows. 1) Power tiller with tires show maximum draft of 200kg at slip range of 40∼50% on four different soil surfaces. Power tiller with traction aids (track type rower tiller) reaches maximum draft of 300kg at about 50% slip on the soil covered with weeds and at 70% slip on the hard soil surface and plowed land. 2) Track type power tiller showed 1.6∼1.8 times as much maximum draft as fewer tiller with tire showed on the all soil surfaces on which experiment were carried out except asphalt road. 3) Track type power tiller showed maximum drawbar horse-power of 1.0∼1.5HP at 28∼35% slip for four different soil surfaces, when the draft was about 300Kg. Power tiller with tire reaches maximum drawl ar horsepower of 0.5∼1.0HP at 20∼25% slip with 200Kg of draft for four different soil surfaces. 4) Track type power tiller showed 1.6 times as much maximum drawbar horsepower as Lower tiller with tire showed. This means that track type rower tiller shows much better tractive efficiency compared with that of power tiller with tire.

      • Group B Streptococci 감염의 임상양상과 Pulsed-Field Gel Electrophoresis를 이용한 유전자형 분석

        Pulsed-field gel electrophoresis (PPGE) has been employed as a tool to study correlation between apparent clinical features and genotyping of Group B Streptococci (GBS) infection. The result of serotyping among 45 isolates showed that serotype la, lb, Ⅲ, Ⅳ, Ⅴ, NT (nontypeable) were 15.5%, 11.1%, 35.5%, 2.22%, 20%, 15.5%, respectively. The result of serotyping for infection group of 26 isolates showed that serotype la, lb, Ⅲ, Ⅳ, Ⅴ, NT were 11.5%, 15.4%, 34.6%, 3.8%, 11.5%, 23%, respectively. The result of serotyping for carrier group of 19 isolates showed that serotype la, lb, Ⅲ, Ⅴ, NT were 21%, 5.3%, 36.8%, 31.6%, 5.3%, respectively. Thus, the predominant serotypes of the infection groups were serotype lb and Ⅲ, and those of the carrier groups serotype Ⅲ and Ⅴ. All isolates were characterized by PFGE analysis. Isolates were assigned to three major phylogenetic Divisions (Ⅰ, Ⅱ, Ⅲ) and subsequently to 16 PFGE groups (A[A1, A2, A3], B[B1, B2, B3], C, D[D1. D2, D3, D4, D5, D6], E[E1, E2], F). Seventy five percentages of Division Ⅰ belonged to the infection group. Seventy eight percentages Division Ⅱ of belonged to the carrier group. One hundred percentages of Division Ⅲ belonged to the infection group. The study on the correlation between genotype and serotype among the isolates showed that serotype Ⅲ was predominant in A1 group (5 of 5 isolates), D1 group (4 of 5 isolates), and F group (2 of 2 isolates). In addition, serotype Ⅴ was the sole serotype in C and D5 group. Those results indicated that serotyping and PFGE group analysis were highly correlated.

      • 플라우의 경운저항 예측 관련 국내 논 토양의 물리적특성

        Two dimensional and three dimensional model for an inclined blade was evaluated to decide the soil physical properties which is necessary for draft prediction of moldboard plow. From the above model, soil cohesion, soil internal friction, soil-metal friction, soil-metal adhesion and bulk density were decided as soil parameters which are needed for computer simulation. Besides above soil parameters, soil moisture content and soil texture were measured for more information at the Iri, Namyang, Yeoju, Pyeongtaek, Asan, Suwon, Chungju, Milyang, Kimhae, Sangju, Kimpo and Kanghwa area from October to November and from March to May, from 1995 to 1996. The most popular soil texture was silty loam and loam The range of soil bulk density was 1500∼1700 kg/㎥. The range of soil cohesion was 10∼15 kPa and that of internal friction angle was 40∼60 degree. The range of soil-metal friction coefficient and adhesion was 0.3∼0.8 and 1.0∼3.0 kPa.

      • 트랙터 부착용 로더 기구부의 기구학적 시뮬레이션 프로그램 개발

        A loader mechanism mounted on tractor was modeled for kinematic analysis, and a computer simulation program was developed by using Cartesian coordinate approach, in order to remove the burden of routine derivation and computation from the engineer. The results of kinematic analysis was compared with those of a commercial CAD program. The results of this study was summarized as follows. 1. The results of the simulation showed good agreement with results of the commercial CAD program. This program could be used for predicting the kinematic properties of each links on the loader mechanism. 2. The velocity of bucket which affect the performance of loader increased very quickly just before the finishing point of boom operation, and acceleration also increased sharply at this point. Thus it is concluded that when bucket is rolling, maximum reaction force arises. 3. Velocity and acceleration of each links on the loader mechanism changed very quickly when the bucket cylinder started operation.

      • 農業機械의 走行性에 관한 硏究(Ⅲ) : 動力耕耘機의 軟弱地 作業性能을 中心으로 Performance of Power Tiller during Tillage Operations in Wetland Rice Field

        To evaluate the factors which influence the mobility of power tiller and to find means which can be used fer predicting the mobility of power tiller in soft paddy field during both plowing and rotary tilling operations, a series of field experiment was conducted with two models of power tilers ( 8ps and 6ps diesel). There is no significant difference in wheel sinkage between the tillage operations and the travelling without load. The upper limit of tillage operation of power tiller in soft paddy field is 15cm sinkage of rectangular plate(50 x 100mm, 30kg weight). Normal plowing operation can be obtained when the rectangular plate sin kage is less than 5cm (2.5kg/㎠ of cone index equivalent), and no more than 10cm (2kg/㎠ of cone index equivalent) for rotary tilling operation.

      • 軟弱地에서 動力耕耘機 沈下量 推定을 위한 平板의 適正形象 및 크기에 관한 硏究

        This study was conducted to determine the optimum shape and dimension of plate predicting the sink age of power tiller in soft paddy field. Various size of disc and rectangular plates were tried at five different soil moisture content, and the resules are summarized as follows. 1. Linear relationship is recognized between contact pressure and sinkage of plates using log-log paper at higher soil moisture content, but the plate sinkage is affected by hard pan in case of the lower moisture soil conditions. 2. Concerning the value of soil deformation coefficient K, the variation of measured values due to shape and size of plate can be practically negligible for the rectangular plates which have 2.5, 5 and 10cm width and with 2 or 3 aspect ratio, and the disc plate which has 20cm diameter. The variation of the exponent of sinkage n, can be practically negligible for the rectangular plates which have 5, 10 and 15cm width, and disc plates which have 20 and 30cm diameters. 3. Relationship between the sinkage of some type of plate and that of power tiller was linear. The closest relationship were found between the sinkage of power tiller and plates for 2.5 and 5cm width rectangular plates regardless of aspect ratio and for the disc plates having 10 and 20cm diameters. 4. Consequently, the optimum plate shape and size for predicting the sinkage of power tiller in soft paddy field are found as the rectangular plates which have 5 and 10cm width and 2 or 3 aspect ratio, and the disc plates having 10 and 20cm diameter.

      • Canine Distemper Virus 검출을 위한 정량 Real-time PCR

        Canine distemper virus(CDV) infection causes a fatal systemic disease in wide range of carnivore species, including domestic dogs. It has been a highly contagious disease of veterinary importance for centurise. Recently many researches show that qualitative RT-PCR for the detection of CDV was sensitive and specific method for the early and safe antemortem diagnosis of distemper by using whole blood regardless of clinical sign and pathological finding. Recently, results were correlated to clinical findings, anti-CDV neutralizing antibody titers, postmortem findings, and demonstration of CDV NP(nucleoprotein) antigen by immunohistochemistry. We have studied a quantitative real-time PCR assay for CDV in order to improve and broaden the clinical usefulness of RT-PCR assay. Total 37 samples including 9 clinically positive and 29 randomly selected ones have investigated. CDV RNA was extracted from whole blood. The isolated RNA was reverse-transcripted into cDNA followed by PCR amplification. The assay based on real-time PCR using SYBR Green I dye as quantitative method with a LightCycler. Calibration curves covered 30-3×10^(8) copies of CDV were created using cloned 240bp CDV cDNA of viral RNA. CDV was detected all in 8 positive samples whereas 8 in 29 randomly collected ones. The detection ranges of clinical CDV specimens were various from 3×10² to 3×10⁴ copies. Comparision of real-time PCR to conventional PCR showed equal sensitivity and specificity. But real-time PCR method has a merit of quantification as well as more rapid results than those of common RT-PCR. CDV can be accurately detected and quantified in the whole blood from the dog infected with CDV. This quantitative technique may improve the clinical utility of RT-PCR for the detection of CDV.
